    Abstract: Chinese Shang Ring adult male circumcision (SC) is a safe and effective procedure which is easy to learn and to perform. By a specially designed small device, male circumcision (MC) can be performed in 5 min. Compared with conventional adult MC (CC), SC has shorter operation time, less blood loss, less pain score, higher appearance satisfaction rate, and lower complication rate. SC was first developed in China. As recent studies have demonstrated that MC reduced the risk of acquiring human immunodeficiency virus (HIV) infection via vaginal intercourse in African countries, SC was introduced into Africa to fight HIV. Other sexually transmitted diseases such as human papillomavirus infection may also be prevented by SC. In conclusion, Chinese Shang Ring, a small device, provides an easy, quick, safe, and effective method to perform adult MC.

    Abstract: 〈 b 〉 〈 i 〉 Background/Aims: 〈 /i 〉 〈 /b 〉 The topological organization of brain functional networks is impaired in Parkinson's disease (PD). However, the altered patterns of functional network hubs in different subtypes of PD are not completely understood. 〈 b 〉 〈 i 〉 Methods: 〈 /i 〉 〈 /b 〉 3T resting-state functional MRI and voxel-based graph-theory analysis were employed to systematically investigate the intrinsic functional connectivity patterns of whole-brain networks. We enrolled 31 patients with PD (12 tremor dominant [TD] and 19 with postural instability/gait difficulty [PIGD] ) and 22 matched healthy controls. Whole-brain voxel-wise functional networks were constructed by measuring the temporal correlations of each pair of brain voxels. Functional connectivity strength was calculated to explore the brain network hubs. 〈 b 〉 〈 i 〉 Results: 〈 /i 〉 〈 /b 〉 We found that both the TD and PIGD subtypes had comprehensive disrupted regions. These mainly involved the basal ganglia, cerebellum, superior temporal gyrus, pre- and postcentral gyri, inferior frontal gyrus, middle temporal gyrus, lingual gyrus, insula, and parahippocampal gyrus. Furthermore, the PIGD subgroup had more disrupted hubs in the cerebellum than the TD subgroup. These disruptions of hub connectivity were not correlated with the HY stage or disease duration. 〈 b 〉 〈 i 〉 Conclusion: 〈 /i 〉 〈 /b 〉 Our results emphasize the subtype-specific PD-related degeneration of brain hubs, providing novel insights into the pathophysiological mechanisms of connectivity dysfunction in different PD subgroups.

    Abstract: 〈 b 〉 〈 i 〉 Aims: 〈 /i 〉 〈 /b 〉 To determine FOXO1 (forkhead box O1) expression in the human endometrium during the menstrual cycle and decidua of early pregnancy, as well as FOXO1 regulation in endometrial stromal cells (ESC). 〈 b 〉 〈 i 〉 Methods: 〈 /i 〉 〈 /b 〉 Immunohistochemistry, RT-qPCR, and Western blot analyses evaluated cellular localization and altered FOXO1 expression in the endometrium during the menstrual cycle and decidua of early pregnancy (proliferative phase, n = 12; early-secretory phase, n = 7; mid-secretory phase, n = 10; late-secretory phase, n = 10; early pregnancy, n = 12). Using RT-qPCR and Western blot, we studied the regulation of FOXO1 by 8-bromo-cAMP, 4-pregnene-3,20-dione, 17β-estradiol, and human chorionic gonadotrophin in ESC (n = 5). 〈 b 〉 〈 i 〉 Results: 〈 /i 〉 〈 /b 〉 The expression level of FOXO1 in human endometrial tissue fluctuated with the menstrual cycle. If pregnancy occurred, the expression of FOXO1 was further increased (p 〈 0.05) and cAMP regulated FOXO1 expression in ESC. In addition, 4-pregnene-3,20-dione cooperatively stimulated FOXO1 expression with cAMP. We also observed FOXO1 expression during in vitro cAMP-induced decidualization. 〈 b 〉 〈 i 〉 Conclusions: 〈 /i 〉 〈 /b 〉 The pattern of FOXO1 expression suggests a potential role for FOXO1 in implantation and decidualization.

    Abstract: The novel coronavirus (COVID-19 or 2019-nCoV) is a respiratory virus that can exist in the mouth and saliva of patients and spreads through aerosol dispersion. Therefore, stomatological hospitals and departments have become high-infection-risk environments. Accordingly, oral disinfectants that can effectively inactivate the virus have become a highly active area of research. Hexadecyl pyridinium chloride, povidone-iodine, and other common oral disinfectants are the natural primary choices for stomatological hospitals. Therefore, this study investigated the inhibitory effect of hexadecyl pyridinium chloride on severe acute respiratory syndrome coronavirus (SARS-CoV-2) in vitro. Vero cells infected with SARS-CoV-2 were used to determine the disinfection effect; the CCK-8 method was used to determine cytotoxicity, and viral load was determined by real-time PCR. The results showed that hexadecyl pyridinium chloride has no obvious cytotoxic effect on Vero cells in the concentration range of 0.0125–0.05 mg/mL. The in vitro experiments showed that hexadecyl pyridinium chloride significantly inhibits the virus at concentrations of 0.1 mg/mL or above at 2 min of action. Thus, the results provide experimental support for the use of hexadecyl pyridinium chloride in stomatological hospitals.

    Abstract: 〈 b 〉 〈 i 〉 Background: 〈 /i 〉 〈 /b 〉 It has been reported that coronary slow flow (CSF) is associated with acute myocardial infarction, ventricular tachycardia, ventricular fibrillation, and even sudden cardiac death. Although studies concerning the etiopathogenesis of CSF are scarce, diffuse atherosclerosis and endothelial dysfunction are thought to play important roles. It has been suggested that a high plasma thrombomodulin (TM) level seems to play an important role in the pathogenesis of atherosclerosis and endothelial dysfunction. 〈 b 〉 〈 i 〉 Objectives: 〈 /i 〉 〈 /b 〉 We hypothesized that a high plasma TM level might be associated with CSF and aimed to research the relationship between plasma TM level and CSF. 〈 b 〉 〈 i 〉 Methods: 〈 /i 〉 〈 /b 〉 Fifty-two CSF patients with angiographically proven CSF and 44 cases with normal coronary flow were included in this study. Coronary flow velocity was determined by the thrombolysis in myocardial infarction (TIMI) frame count method. Plasma TM levels were measured in all the study subjects. 〈 b 〉 〈 i 〉 Results: 〈 /i 〉 〈 /b 〉 Plasma TM levels were significantly higher in the CSF group compared to the control group (3.9 ± 0.5 vs. 3.6 ± 0.3 ng/mL, 〈 i 〉 p 〈 /i 〉 = 0.01). There was a positive relationship ( 〈 i 〉 r 〈 /i 〉 = 0.31, 〈 i 〉 p 〈 /i 〉 = 0.002) between plasma TM level and mean TIMI frame count (TFC). Factors associated with mean TFC were plasma TM level (β = 0.206, 〈 i 〉 p 〈 /i 〉 = 0.038) and red cell distribution width (β = 0.088, 〈 i 〉 p 〈 /i 〉 = 0.009) in multiple linear regression analysis. 〈 b 〉 〈 i 〉 Conclusions: 〈 /i 〉 〈 /b 〉 Patients with CSF have a higher plasma TM level, and this may play an important role in the pathogenesis of CSF. An elevated plasma TM level may be a predictor of CSF. Future studies are needed to confirm these results.

    Abstract: 〈 i 〉 Objective: 〈 /i 〉 Previous studies have demonstrated that an imbalance of matrix metalloproteinases (MMPs)/tissue inhibitors of metalloproteinase-1 (TIMP-1) and a loss of fibronectin are associated with postmyocardial infarction remodeling in rats. The present study was designed to examine this issue in patients with congestive heart failure (CHF). 〈 i 〉 Methods: 〈 /i 〉 We measured plasma levels and the cardiac protein expression of MMPs/TIMP-1 and fibronectin in 39 patients with CHF and 38 controls. 〈 i 〉 Results: 〈 /i 〉 Plasma levels of MMP-2, MMP-3, and MMP-9 tended to be higher in patients with CHF (NYHA II: 276 ± 18, 613 ± 118, and 245 ± 43 µg/l, respectively; NYHA III: 302 ± 20, 850 ± 132, and 310 ± 39 µg/l, respectively; NYHA IV: 367 ± 15, 998 ± 99, and 392 ± 27 µg/l, respectively) than in controls (213 ± 23, 485 ± 102, and 158 ± 31 µg/l, respectively), while the plasma TIMP-1 level tended to be lower in patients with CHF (NYHA II: 126 ± 12 µg/l, NYHA III: 83 ± 11 µg/l, and NYHA IV: 61 ± 12 µg/l) than in controls (208 ± 15 µg/l). Interestingly, the changes in protein expression of MMPs/TIMP-1 were consistent with their plasma concentration. Furthermore, the fibronectin level in the patients with CHF was significantly lower than in the controls. 〈 i 〉 Conclusions: 〈 /i 〉 These data suggest that human CHF is associated with an imbalance of MMPs/TIMP-1 and a concurrent loss of fibronectin.

    Abstract: 〈 i 〉 Background: 〈 /i 〉 Antibodies induced from glycoprotein B (gB) by antigenic domain (AD)-1 demonstrate broad neutralizing activity across different human cytomegalovirus (HCMV) types. This study aimed to prepare a novel HCMV vaccine using the modified adenoviral vector Ad5F35 to direct the expression of the conserved HCMV epitope AD-1 and to determine its transfer and expression in peripheral blood mononuclear cells (PBMCs). 〈 i 〉 Methods: 〈 /i 〉 AD-1 genes were amplified from AD169 HCMV strain and cloned into the Ad5F35 vector. Ad5F35-AD-1 virus vaccine was prepared by packaging Ad5F35-AD-1 into HEK293 cells. RT-PCR and fluorescence detection were used to detect the expression of AD-1 in HEK293 cells. PBMCs were stimulated in vitro with Ad5F35-AD-1 virus vaccine. The AD-1 expression in PBMCs was determined with immunocytochemistry and cell viability was measured to observe the possible adverse effects of AD-1 on PBMCs. 〈 i 〉 Results: 〈 /i 〉 We constructed an Ad5F35-AD-1 vector and transferred it into HEK293 cells to prepare the Ad5F35-AD-1 virus vaccine successfully. The AD-1 gene was proved to be expressed in HEK293 cells. In vitro stimulation of PBMCs with Ad5F35-AD-1 showed the highly efficient expression of AD-1 and low cytopathic activity in PBMCs. 〈 i 〉 Conclusion: 〈 /i 〉 The novel vaccine Ad5F35-AD-1 is a promising candidate for clinical trials and may be of utility in prime-boost strategies for HCMV prevention and control.

    Abstract: 〈 b 〉 〈 i 〉 Objective: 〈 /i 〉 〈 /b 〉 This study aimed to detect the genotype distributions and allele frequencies of methylenetetrahydrofolate reductase (MTHFR) C677T, A1298C and methionine synthase reductase (MTRR) A66G polymorphisms of pregnant women in Jiaodong region in China, and to investigate whether folic acid supplementation affect the pregnancy complications. 〈 b 〉 〈 i 〉 Setting: 〈 /i 〉 〈 /b 〉 A total of 7,812 pregnant women from the Jiaodong region in Shandong province in China. 〈 b 〉 〈 i 〉 Methods: 〈 /i 〉 〈 /b 〉 By using Taqman-MGB, 2,928 pregnant women (case group) were tested for the genotype distributions and allele frequencies of MTHFR C677T, A1298C and MTRR A66G polymorphisms. Folic acid metabolism ability was ranked at four levels and then pregnant women in different rank group were supplemented with different doses of folic acid. Their pregnancy complications were followed up and compared with 4,884 pregnant women without folic acid supplementation (control group) in the same hospital. 〈 b 〉 〈 i 〉 Results: 〈 /i 〉 〈 /b 〉 The allele frequencies of MTHFR C677T were 49.1 and 50.9%; those of MTHFR A1298C were 80.2 and 19.8%, and those of MTRR A66G were 74.1 and 25.9%. After supplemented with folic acid, the complication rates in different age groups were significantly reduced, especially for gestational diabetes mellitus and hypertension. 〈 b 〉 〈 i 〉 Conclusion: 〈 /i 〉 〈 /b 〉 Periconceptional folic acid supplementation and healthcare following gene polymorphism testing may be a powerful measure to decrease congenital malformations.

    Abstract: 〈 b 〉 〈 i 〉 Background: 〈 /i 〉 〈 /b 〉 This study was to explore the expression profile of endometrial carcinoma (EC) and identify the potential molecular mechanism and therapeutic targets. 〈 b 〉 〈 i 〉 Methods: 〈 /i 〉 〈 /b 〉 Differentially expressed genes (DEGs) and differentially expressed miRNAs (DEMs) were identified in EC using mRNA and miRNA sequencing data released by the Cancer Genome Atlas database; then, gene function and pathway of DEGs were analyzed based on the Gene Ontology and Kyoto Encyclopedia of Genes and Genomes pathway databases; finally, the transcription factors (TFs) latently regulating the DEGs and DEMs were predicted and a TF-miRNA-Gene network was then established to summarize the regulatory links between TFs, DEMs and DEGs. 〈 b 〉 〈 i 〉 Results: 〈 /i 〉 〈 /b 〉 One thousand five hundred and forty two upregulated and 1,885 downregulated DEGs, 34 upregulated and 12 downregulated DEMs were identified. The principal DEGs-enriched functions were cell differentiation, cell migration, and cell surface receptor signaling pathway. The DEGs-enriched cell signaling pathways including the MAPK, Wnt signaling pathway, and the p53 signaling pathway. As shown in the TF-miRNA-Gene network, TFs such as CPBP and GKLF, miRNAs such as miR-141-3p and miR-130b-3p, regulated most of DEGs and DEMs. 〈 b 〉 〈 i 〉 Conclusion: 〈 /i 〉 〈 /b 〉 These results may contribute to the study of the molecular mechanism and therapeutic targets in EC, and facilitate the discovery of new biomarkers for screening, diagnosis and monitoring.

    Abstract: 〈 b 〉 〈 i 〉 Aim: 〈 /i 〉 〈 /b 〉 To study whether infertile patients with endometriosis have a higher prevalence of endometrial polyps, and to clarify the characteristics of the pathology of combined polyps. 〈 b 〉 〈 i 〉 Methods: 〈 /i 〉 〈 /b 〉 Infertile patients who had undergone both hysteroscopy and laparoscopy in Reproductive Hospital Affiliated with Shandong University from January 2014 to May 2017 were enrolled. Patients with and without endometriosis, diagnosed by laparoscopy, were staged and included in the study group and control group, respectively, and the prevalence of polyps was compared. The pathological types of endometrial polyps were analyzed. 〈 b 〉 〈 i 〉 Results: 〈 /i 〉 〈 /b 〉 A total of 414 cases were enrolled in the study group and 3,048 cases in the control group; polyps were diagnosed, with endoscopy, in 1,107 patients. Endometrial polyps were detected by hysteroscopy in 47.83% of the endometriosis group and 29.82% of the control group. The prevalence of endometrial polyps was significantly higher in the endometriosis group than in the control group ( 〈 i 〉 p 〈 /i 〉 & #x3c; 0.001) but not significantly different between stages of endometriosis ( 〈 i 〉 p 〈 /i 〉 = 0.580). The pathological diagnosis included 899 endometrial polyps and 208 polypoid hyperplasia; 66.5% of endometrial polyps were combined with simple hyperplasia. 〈 b 〉 〈 i 〉 Conclusions: 〈 /i 〉 〈 /b 〉 The infertile patients with endometriosis had a higher prevalence of endometrial polyps, and those polyps are often combined with simple hyperplasia.
