Abstract: Pacific white shrimp (Penaeus vannamei or Litopenaeus vannamei) is native to South America, high economic value, and widely cultivated in the world and Vietnam. Over the last two decades, viral diseases have seriously threatened the shrimp aquaculture industry. Among the viral diseases, white spot syndrome virus (WSSV) is the most important viral pathogens of shrimp farming. WSSV causes a cumulative mortality can reach 100% within 3–10 days. Genome sequencing and assembly has been an important step for deciphering molecular mechanisms and accelerating genetic improvements of traits of interest in economically important species. This study aims at constructing and annotating the genome of white spot syndrome virus - infected Pacific white shrimp in Vietnam. The whole genome sequencing data was de novo assembled using SOAP denovo2 to obtained draft genome of WSSV- infected L. vannamei shrimp. The draft genome contained 3,180,049 scaffolds (genome size ~1.67 Gb) with the length arranging from 200 bp to 137,569 bp and with N50 as 616 bp. Applying gene prediction method, we have been able to identify 187,948 putative genes. The results have shown that 33,611 genes were annotate in NT database and 133,548 genes were annotated in UniProtKB/Swissprot database. These results are only the initial information about white spot syndrome virus - infected Pacific white shrimp but they are really important for future studies relating to white spot syndrome virus – resistance L. vannamei shrimp in Vietnam.

Abstract: White spot syndrome virus (WSSV) is a dangerous virus causing great damage to shrimp farming. Many genes related to disease resistance mechanisms have been identified and studied. In this study, we amplified and sequenced nine fragments of DNA harboring SNPs in the genes involved in WSSV resistance of white-legged shrimp, Litopenaeous vannamei. Allele frequencies at the SNP loci were recorded and calculated by SPSS statistical software (version 22) in the study groups: the disease shrimp (were infected with WSSV and died) and the resistance shrimp (were infected with WSSV but survived). Six SNPs (in AIF, ALF1, HAE, P53, Rab5B, and TRAF6 genes) were in accordance with Hardy–Weinberg Equilibrium (HWE) (p 〉 0.05) while three SNPs (in ALF2, BGB, and CAL) were not (p 〈 0.05). For AIF and ALF1 genes, the frequencies of GG and CC genotypes were significantly different between two groups and were associated with an decreased resistance with WSSV compared to the AA and TT genotypes (p = 0.021 and p = 0.017, respectively). The G and C alleles were associated with a decreased resistance with WSSV (p = 0.000 and p = 0.001, respectively). For HAE gene, the frequency of TT genotype was significantly different between two groups and was associated with a increased resistance with WSSV compared to the TC+CC genotype (OR = 68.750; 95% CI: 11.462–412.381; p = 0.000). For Rab5B gene, the frequency of CC genotype was significantly different between two groups and was associated with an increased resistance with WSSV compared to the TT genotype in all three tested models (p 〈 0.05). The C allele was associated with an increased resistance with WSSV (OR = 3.974; 95% CI: 1.754–9.003; p = 0.001). The above evaluation results suggested that the potential SNPs of these AIF, ALF1, HAE, and Rab5B genes can be used as the molecular markers for breeding selection the resistance to white spot disease in white-legged shrimp L. vannamei.

Abstract: Excessive salt consumption has been found to cause many negative effects on human health. To date, many researches have been conducted to reduce the sodium content of the diet. This study focused on the comparison of saltiness intensity and other parameters in time-intensity curves of four commercial grain salts in four regions of Viet Nam (Bac Lieu, Thanh Hoa, Sa Huynh, Vung Tau), one flower salt in Sa Huynh and a control sample with two particle sizes of 1-2 mm and 2-3 mm to find out their taste perception characteristics. A modified sensory evaluation technique focusing on time-intensity was used to train a panel to evaluate the temporal profile of saltiness of salt crystals. The results showed a significant difference in the perception ability and time intensity of saltiness in salt products by region and particle size. Sensory measures exhibited the differences in the specificity of salt samples by region and a strong correlation with their physical properties. Sa Huynh flower salt and sea salt showed different characteristics compared to other salt samples with higher intensity of salty taste, faster time to reach the maximum intensity, and shorter duration of salt taste intensity. These results demonstrated that flower salt could reduce the level of sodium intake while maintaining the saltiness perception in products, which is a potential ingredient in food, spices, etc.

Abstract: Effect of zirconia nanoparticles (zirconia NPs/ZrO2 NPs) modified by 3 wt.% (glycidyloxypropyl) triethoxysilane – GPTES (m-ZrO2 NPs) on some properties of epoxy coating such as mechanical properties, thermal stability and anti-corrosion performance was investigated. The obtained results indicated that the addition of zirconia nanoparticles to epoxy coating could enhance the properties of this coating. The addition of 2 wt.% of pure ZrO2 NPs (u-ZrO2 NPs) to the epoxy matrix could increase the mechanical properties (hardness and adhesion to the steel substrate) by approximately 10 %, the onset temperature of thermal degradation of the epoxy/u-ZrO2 NPs coating was 4.4 oC higher, and the anti-corrosion performance of epoxy coating was improved in comparison with the neat epoxy coating. Organically modified ZrO2 NPs had higher improvement for epoxy coating’s properties than pure ZrO2 NPs. The epoxy coating filled with m-ZrO2 NPs had 19.7 % higher in relative hardness, 88.73 % more in adhesion to steel substrate, 25.6 oC higher in the onset temperature of thermal degradation, and higher anti-corrosion performance in comparison with the epoxy coating filled with 2 wt.% of pure zirconia nanoparticles. The cross-section FESEM images of the epoxy/m-ZrO2 NPs coating showed that m-ZrO2 NPs could regularly disperse in epoxy polymer matrix while unmodified zirconia nanoparticles (u-ZrO2 NPs) were agglomerated to big cluster in the epoxy coating. This was the reason for the high performance of the epoxy coating filled with zirconia NPs modified by GPTES.

Abstract: The studies on the impact of the ENSO phenomenon on climate and environment change have shown that the Bien Dong (Southeast Asia Sea) is strongly influenced by this phenomenon. This paper focuses on analyzing sea surface temperature (SST) monthly data for 16 years (7/2002–12/2017) in the coastal waters of Vietnam from MODIS satellite images of National Aeronautics and Space Administration (US NASA). The analysis results showed a clear pattern of ENSO impacts on SST in the coastal waters of Vietnam. However, the intensity of the ENSO phenomenon affecting the three studied areas is very different, and in terms of trend, the intensity gradually increases to the north. The period of impact of ENSO on SST in the coastal waters of Vietnam is often about one month later than the ONI index and the influenced duration lasts about 6 to 9 months. In addition, in the La Niña years, the values of the SST anomaly reflect quite well in terms of the intensity of the La Niña phenomenon. However, in the El Niño years, the SST anomaly index is not generally proportional to the ONI index. Specifically, in the years when El Niño phenomenon was considered to be weak, the SST anomaly index of the coastal waters of Vietnam is much higher than in the years when El Niño phenomenon was considered to be moderate or very strong. The above results are contributing to improving the understanding of the impact of the vagaries of the climate on the coastal waters of Vietnam, supporting scientists and managers in making measures to prevent and efficiently avoid, limit and mitigate the damage caused by ENSO, and to have reasonable alternatives in the protection of biodiversity and environment in the coastal waters of Vietnam. It could be considered as a “small piece” of the overall picture of the impacts of the ENSO phenomenon on global climate change.

Abstract: Panax bipinnatifidus Seem. belonging to the family Araliaceae has been used as a folk medicine in Viet Nam. Our ongoing study on bioactive compounds from Panax bipinnatifidus Seem. collected in Sapa, Lao Cai resulted in the isolation of two saponins together with the well-known saponin Stipuleanoside R2. Their structures were identified as 3-O-β-D-glucopyranosyl-(1→3)-β-D-glucuronopyranosyloleanolic acid (1) and Stipuleanoside R1 (2) based on NMR spectroscopic data as well as comparison with the literature data. For the first time, 3-O-β-D-glucopyranosyl-(1→3)-β-D-glucuronopyranosyloleanolic acid (1) was found in genus Panax. On biological screening, the crude extract and the principal saponin, stipuleanoside R2 showed weak inhibitory effect on markedly nitric oxide production in lipopolysaccharide-treated RAW 264.7 cells.

Abstract: Y-chromosome microsatellites or short tandem repeats (STRs) have been proved to be ideal markers to delineate the differences between individuals in human population. Nowadays, Y-STR testing using the PowerPlex® Y23 amplification kit is considered as an extremely sensitive analysis method and has the potential to be used to perform forensic caseworks, and to explore the complexity in population substructures. However, little is known about the forensic Y-chromosome databases in the Vietnam population. In this study, 23 Y-STR loci (DYS576, DYS389I, DYS389 II, DYS448, DYS19, DYS391, DYS481, DYS549, DYS533, DYS438, DYS437, DYS570, DYS635, DYS390, DYS439, DYS392, DYS393, DYS458 DYS456, DYS643, YGATAH4, and DYS385a/b) were investigated in 120 non-related males of the Kinh population in Northern Vietnam using PowerPlex® Y23 system kit (Promega). Our results showed that allele frequencies of 23 loci in the sample population, with the calculated average gene diversity (GD) for each locus, ranged from 0.24 (DYS438) to 0.92 (DYS385a/b). In addition, a total of 120 different haplotypes were found, all of them were unique. Therefore, we found that the haplotype diversity was 1 with a discrimination capacity of 100%, which serves as an essential prerequisite for using Y-chromosomal STR with PowerPlex® Y23 System kit in forensic application in Vietnam. We also compared genetic distances between Kinh population and 10 other neighboring populations from Y-chromosome haplotype reference database (YHRD). The Kinh population is significantly different from other populations. In conclusion, it was indicated that the 23 Y-STR loci were highly genetically polymorphic in the Kinh population in Vietnam and might be of great value in forensic application.
