    Abstract: The large displacements of functionally graded sandwich (FGSW) beams in thermal environment  are studied using a finite element formulation. The beams are composed of three layers, a homogeneous core and two functionally graded face sheets with volume fraction of constituents following a power gradation law. The material properties of the beams are considered to be temperature-dependent.  Based on Antman beam model and the total Lagrange formulation, a two-node nonlinear beam element taking the effect of temperature rise into account  is formulated and employed in the study. The element with explicit expressions for the internal force vector and tangent stiffness matrix is derived using linear interpolations and reduced integration technique to avoid the shear locking. Newton-Raphson based iterative algorithm is employed in combination with the arc-length control method to compute the large displacement response of a cantilever FGSW beam subjected to end forces.  The accuracy of the formulated element is confirmed through a comparison study. The effects of the material inhomogeneity, temperature rise and layer thickness ratio on the large deflection response of the beam are examined and highlighted.

    Abstract: Phototrophic dinoflagellates of the genus Symbiodinium (= zooxanthellae) are well known as symbionts of corals. They play an important role in maintaining the healthy as well as establishment of new reefs. In addition, products such as toxins or pigments from these symbiotic microalgae are potentially important for applications in the biomedical sciences, especially in new drug development. In this study, we isolated successfully a symbiotic microalga from soft coral Sinularia sp. which was collected in Cu Lao Cham coastal waters of Quang Nam province. Based on morphological characteristics and 18S rRNA gene sequences, isolated SHM8 strain was identified as Symbiodinium goreaui. This symbiont was cultured in different media such as ASP-A8, F2, ES, IMK, K, and L1. L1 culture medium supplemented with taurine concentration of 4.88 mM led to the cell density of SHM8 strain increased in 50% compared to control on day 2 in cultivation; amino acid mixture enhanced cell density by 31.7% on day 5 in cultivation. Supplementation of carbon source (bicarbonate salt of 16.8 g/L) and nitrogen source (nitrate salt of 0.055 g/L) enhanced the cell density by 40% and 27%, respectively on day 6 in culture. Obtained results in this study proved that this microalga can live independently without host.     Abstract: The overuse of antibiotics is losing its effectiveness due to increased antibiotic resistance in bacteria. Over the last two decades, the evaluation of antibiotic residues in the environment has greatly attracted the attention of researchers around the world. The more new groups of antibiotics were validated to use in medical treatment, the more antibiotic residues were discharged into the environment. In this work, a combined SPE-UPLC-MS/MS method was developed and validated for simultaneously analyzing 9 antibiotic compounds belonging 3 different groups which are tetracyclines (tetracycline – TET, chlortetracycline – CHL, oxytetracycline – OXY), fluoroquinolones (ciprofloxacin – CIP, norfloxacin – NOR, ofloxacin – OFL) and macrolides (azithromycin – AZI, erythromycin – ERY, clarithromycin – CLA) in surface water. All target analytes were separated on a reversed-phase column (Water BEH C18 column, 1.7 µm particle size, 100 x 2.1 mm) coupled with a mass in tandem employing positive electrospray ionization (+ESI). All target analytes were well separated with an overall run time of 16 minutes. The limit of detection was from 0.2 to 10 µg L-1. The extraction process has been successfully optimized using the HLB column (Oasis, 6mL, 200mg, Waters) with recoveries from 71 to 125%. The optimized method was finally applied to analyze ten surface water samples (lake and river). OFL, CLA, and ERY were most found in lake samples at concentrations from 35 – 570.3 ng.L-1 while only CIP was found in all river samples. The other compounds were also detected in both types of samples but the concentrations were lower than the quantification limit.

    Abstract: Five phenolic glycosides, clerodenoside A (1), seguinoside K (2), cistanoside D (3), verbascoside (4), and isoverbacoside (5) were isolated from the ethyl acetate soluble fraction of methanolic extract of the Clerodendrum inerme stems. Their structures were determined by spectroscopic analysis and comparison of spectral data with those reported in the literature. Compound 3 was isolated from this plant for the first time.

    Abstract: Bac thom 7 rice (BT7 rice) is one of the major elite rice cultivars in Vietnam with superior productivity and quality but very susceptible to bacterial leaf blight caused by Xanthomonas oryzae pv. oryzae. The gene OsSWEET14, belonging to the OsSWEET family which encodes sugar transport proteins, is considered to be a susceptible gene involved in the virulence of Xoo. At least three cis-elements (EBE - Effector-binding element), including Tal5, PthXo3 and AvrXa7 on BT7 OsSWEET14 promoter, bind specifically to well-known transcription activator–like effectors (TALEs) of many Asian Xoo bacterium strains. In this study, a T-DNA construct which expressed the protein-RNA complex CRISPR/Cas for editing three EBEs position on the OsSWEET14 promoter was designed. The recombinant binary vector was tested by PCR, restriction enzyme and finally sequencing and then successfully transferred to Bac thom 7 rice through Agrobacterium tumefaciens. 28 of 30 hygromycin-resistant regenerated rice lines that grew and developed normally under nethouse conditions were selected by PCR with specific primers. Among these, twelve transgenic rice lines were identified carrying one copy of the T-DNA construct. The presence of CRISPR/Cas9-induced mutations of the targeted promoter in the transgenic plants were confirmed by T7EI assay. These results provide the basis to determine the role of OsSWEET14 in the susceptibility of Bac thom 7 rice to Xanthomonas oryzae pv. oryzae -caused disease, towards the further goal of generating an improved Bac thom 7 rice variety with broad-spectrum bacterial leaf blight resistance using CRISPR/Cas9 technology.

    Abstract: Acute Hepatopancreatic Necrosis Disease (AHPND) is a widespread disease targeting cultured shrimps, which results in heavy losses in many regions around the world including Vietnam. The causative agent of the disease is Vibrio paraheamolyticus which is a Gram-negative bacterium present ubiquitously in marine environment. However, this normal floral organism once acquired a plasmid containing Pir- toxin encoding gene (AHPND plasmid) can transform into a highly toxic strain (AHPND strain) with the ability to kill a large number of cultured shrimps in a short period. To understand more on the virulence of V. parahaemolyticus, 17 different V. parahaemolyticus isolates collected from different locations in Southern Vietnam have been analyzed for the presence of AHPND plasmid using PCR method and their extracellular enzyme profile using agar- based method. Seven enzymes which are known to be important virulence factors of the bacterium were studied including caseinase, gelatinase, lecithinase, hemolysin, elastase, lipase and chitinase. Results showed that among 17 studied isolates, six were detected to have AHPND plasmid. Enzymatic activity was observed for caseinase, gelatinase and lecithinase while undetectable for others. The ability to produce these three enzymes varied among different V. parahaemolyticus isolates. Lecithinase appeared in all isolates while the presence of caseinase and gelatinase varied. This indicated that lecithinase seems to be core enzyme of V. paraheamolyticus and the extracellular enzymes do not correlate with the presence of AHPND plasmid. In conclusion, our data suggested that the virulence of Vibrio parahaemolyticus mostly depends on the presence of AHPND plasmid other than extracellular enzymes.

    Abstract: Currently, nearly 36.5 million people live with Alzheimer’s disease worldwide, and acetylcholinesterase inhibition is considered the main treatment strategy against it. Seaweeds (or macroalgae) are a natural source of high-value bioactive compounds and have great potential in the production of health foods/foods, pharmaceuticals, cosmetics, animal feeds, biofertilizers, and biofuels. The most studied and applied seaweed species include red seaweed (Rhodophyta), green seaweed (Chlorophyta) and brown seaweed (Phaeophyta). This study aimed to evaluate the antioxidant and neuroprotective activities of extracts/substances isolated with solvents including ethanol 75o and 96o, water, n-hexane, ethyl acetate under normal and ultrasonic conditions (power 80W, frequency 4.7 kHz and for 1 hour) from 5 economically important seaweed species belonging to genera Caulerpa, Sargassum, Gracilaria, Ulva and Kappaphycus collected in Ninh Thuan and Khanh Hoa provinces, Vietnam. The results have shown that 05/30 extracts and substances (including SaE96S, SaH, SaHW, SaEA and fucosterol) isolated from five seaweed species exhibited high antioxidant activity through the free radical screening method using 2,2-diphenyl-1-picrylhydrazyl (DPPH) (with EC50 values 〈 5 mg/mL for extracts and EC50 value 〈 2 mM for substance) in comparison with positive control ascorbic acid (EC50 = 0.015 mg/mL); acetylcholinesterase inhibitory activity using acetylcholinesterase inhibitor screening KIT (with IC50 values of 〈 200 µg/mL for both extracts and substances), compared with positive control galantamine (with IC50 value of 52,8 µg/mL). The extracts/substances were able to protect the cell against cytotoxity in the C6 alzheimer’s disease cell model induced by amyloid beta-protein fragment (Aβ25-35). The results achieved from this research have proven that the extracts/substances isolated from seaweed species were a potential source of medicinal agents for the prevention and treatment of alzheimer’s disease.

    Abstract: Recently, a contagious lung disease named coronavirus disease 2019 (COVID-19) caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), rapidly spread worldwide and has many serious consequences for human health. Human genetic polymorphisms may contribute to the variation of incidence, mortality as well as severity of COVID-19. To date, this factor in the Vietnamese population remains unknown. A cellular protease termed transmembrane protease serine 2 (TMPRSS2) was found to play a vital role in the entry of SARS-CoV-2 into host cells. In this study, we investigated polymorphisms in the TMPRSS2 gene from 270 whole exome sequencing data of Vietnamese peoples. We also employed bioinformatics tools including SIFT, Polyphen-2, and PROVEAN to predict the possible function of missense variants. A total of 34 TMPRSS2 variants were identified, of which, 29 were in non-coding regions and 14 were in coding regions. Variants found in exons included seven synonymous and seven non-synonymous point mutations, one of which was novel mutation (c.A1336C/p.R446R). Mutation c.G589A/p.V197M (rs12329760) possesses the highest frequency and was predicted to have the ability to damage protein by SIFT and Polyphen-2. In addition, the damaging possibility was also found in c.T244G/p.Y82D and c.C896T/p.A299V variants. This study contributes to the understanding of Vietnamese genetic variation databases relating to susceptibility to COVID-19.
