    Abstract: Maltooligosyltrehalose trehalohydrolase (MTHase) is an industrial enzyme for the production of trehalose. A DNA fragment of 1680 bp encoding for MTHase was cloned from Sulfobolus solfataricus DSM 1616 then fused with promoter acoA-amyE already amplified from pMSE3 vector by PCR to generate an expression cassette acoMTH. Afterward the cassette was inserted into pAC7 vector for expression of the gene in Bacillus subtilis WB800 – a conventional expression system. Gene MTH was inserted into the genome of B. subtilis WB800 by cross-exchange event of pAC7 vector with the host genome for expression of high quality and high quantity of extracellular recombinant protein. By crossing-exchange event at 3’amyE-5’amyE, the expressional cassette was integrated into B. subtilis WB800 genome. The expressional cassette was integrated into B. subtilis WB800 genome replacing 3’amyE-5’amyE, hindering the native amylase activity of the host. Expression of expected protein was confirmed by electrophoresis SDS-PAGE. From our results, it indicates that gene MTH was expressed successfully in B. subtilis WB800. After 0.5% acetoin induction for 48 h, the data showed that the protein with a molecular mass of ~64 kDa on SDS-PAGE was expressed. The level of recombinant protein in WBpAacoMTH was increased and reached 2.5%, 15.2% and 21.95%, respectively comparing with native B. subtilis WB800.

    Abstract: Triterpenoids are considered to be the major pharmaceutical active compounds found in the macrofungus Ganoderma lucidum (G. lucidum). Extraction conditions of triterpenoids from the Vietnamese red G. lucidum were optimized by response surface methodology using ultrasound –assisted extraction (UAE) method. A Box – Behnken experimental design was used to determine the simultaneous influences of three independent variables, namely solvent-to-material ratio, extraction time, and ultrasonic power on the yield of triterpenoids. The highest yield of triterpenoids obtained under optimum conditions including extraction time of 55 min, ultrasonic power of 480 W, solvent-to-material ratio of 27:1 mL/g was 9.5768 ± 0.228 mg/g according to ultraviolet-visible spectroscopy (UV-Vis) analysis. Besides, the hot water extraction method was carried out in order to compare with UAE in terms of the yield of triterpenoids. The results of anticancer evaluation of the extract obtained indicated that with the half-maximal inhibitory concentration value of 67.25 ± 0.82 µg/mL on the human Hep-G2 liver cancer cell lines, triterpenoids extracted from G. lucidum could be regarded as a potential agent for medicinal treatment.

    Abstract: Fermented wine is a complex process including many chain reactions to convert sugar into alcohol, CO2, the byproduct and energy. Fermentation processes and product quality depend on the fermentation conditions. The objective of this study is to find the a good condition for effective fermentation to produce good quality distillation and less toxins and impurities. Currently, the production technology of wine or beer in the world and in Vietnam is diverse, there are many different techniques to optimize production in order to create optimal products. Each technology depends on the scale of production, climatic characteristics, materials, strains used in fermentation,.... On this topic we focus on some basic factors affect on fermentation alcohol use barley malt, yeast Saccharomyces cerevisiae MS42 include: pH, temperature, concentration SO2 in the fluid of fermentation, the yeast cell count to the initial seed, sugar, kinetics of fermentation alcohol barley malt on the medium. The creteria for selection of optimal factors is based on the production of alcohol, toxic compounds, contaminants, cell biomass. The products produced sensory board for evaluation, combined analysis of chromatography on toxin components and impurities according to standard TCVN 7043: 2013. Carry out the fermentation experiments in 1000ml of fluid to determine optimal conditions, then perform experimental fermentation under the conditions of industrial production on a 500 liter scale. The optimal conditions for barley malt fermentaion by S. cerevisiae MS42 was: medium pH= 4.5-5.0; To= 25-30oC; K2S2O5: 0.2 - 0.3 (g/l); the starting seed: 7-10%; sugar content: 140 – 160 g/l; fermentation time for 132 to 138 hours.

    Abstract: Research on the growth of yeast is a critical requirement, occupying an important role in the technology of alcohol production. In reality, there are many factors affecting on the reproductive process and the growtht of yeast such as: temperature, pH, dissolved oxygen content, sugar content, nutrients, etc... The quantity and quality of yeast decide the quality on the wine products. When the amount of yeast is abundant, the fermentation speed is faster, which is an important factor in preventing infection from harmful microorganisms during fermentation. The study on influences affecting the growth of yeast is necessary to ensure the main aim of the propagation which is to create good quality breeding populations of yeast in a short time period. Furthermore, this has important implications for the fermentation process to create good products. Besides, increasing the speed and the number of yeast cells in practice, efficient, inexpensive materials used, as well as a common key element in the production process also bring high economic efficiency. In this study, basic factors affecting the growth of the species Saccharomyces cerevisiae MS42 we determined. Optimal conditions and compositions of the medium for breeding are shown in following: sugar content is 80 g/l: pH = 5.0; temperature from 28oC, the amount of dissolved oxygen from the initial is 7.0 mg/l, after 24 hours of incubation (since followed the same), the number of yeast cells has reached the maximum (219.5 ± 3.0 million cells/ml) and budding cell ratio (approximately 70 - 72%).

    Abstract: Lung cancer is the leading cause of death among Vietnamese people and can be divided into two major groups: small cell lung cancer (SCLC), and non-small cell lung cancer (NSCLC). It is estimated that 85% of patients was diagnosed as NSCLC. Therapy drugs that targeted lung cancer tumors with the epidermal growth factor receptor (EGFR) may initially provide benefit, but over time tumors can develop resistance. The recent discovery of EGFR-tyrosine kinase inhibitors (EGFR-TKIs) has provided a new target for the theurapeutic agents in non-small cell lung cancer treatment. In our study, homoisopogon A, a new compound isolated from the tubers of Ophiopogon japonicus (Mach mon, in Vietnamese) effectively showed the cytotoxic activity to the EGFR and TKI-resistant NSCLC cell lines including human lung carcinoma A549, human lung adenocarcinoma NCI-H1975 and human lung adenocarcinoma NCI-H1650. The IC50 values of homoisopogon A against A549, NCI-H1975 and NCI-H1650 were determined as 6.26, 19.51, and 24.66 µM, respectively. Preliminary study on the mechanism of action using flow cytometry analysis was performed. Homoisopogon A treatment of A549 cells at concentration of 25 μM generated apoptosis in 27.5% of cells (7% early apoptosis and 20.5% late apoptosis) after 24h of treatment. The effect increased significantly at the concentration of 50 μM, the homoisoflavanone generated apoptosis in 83.8% of cells (23.5% early apoptosis and 60.3% late apoptosis). Treatment with homoisopogon A for 48h also resulted in typical apoptotic morphological changes in A549 cells under microscopic observation. The results strongly suggested that homoisopogon A induces apoptosis in EGFR and TKI-resistant-A549 cells, thus resulting in the cytotoxicity.

    Abstract: A study on Mollusks’ species composition and distribution on dead coral from Quang Ngai to Cat Ba was conducted during the field survey between Vietnam and Russia on the AKADEMIC OPARIN RC vessel in 2021 at 39 reef stations in 14 study areas. By comparative morphology method, the authors have identified 135 taxa of Mollusca belonging to 48 families, 4 classes on coral reefs. The species composition of Mollusca was slightly different between study areas.

    Abstract:        Asian elephants are under high threats of extinction in Vietnam and worldwide. Population assessment of Asian elephants in Pumat National Park (NP) region, carried out in 2018−2019, showed that the wild Asian elephants population this region consisted of 3 groups with a total of 13−14 individuals. In Tuong Duong District, a group of 4-5 individuals lived in an area near Canh Ta stream (Tam Dinh commune, Vietnam) and Nam Xi stream (Muong Long commune, Laos), across the border between Vietnam and Laos. The group in Con Cuong district, with only one adult female, lived in forest compartments 780, 781,789 and 786 of Con Cuong Forestry Company, within the buffer zone of Pumat NP. The group in Anh Son district consisted of 8 individuals (5 adult females, 1 adult male, 1 sub-adult male and an juvenile born in 2016). They live in Cao Veu sector of Pumat NP core zone and buffer zone areas consisting of protected forests, rubber plantation forests and agricultural lands of households in Phuc Son Commune (Anh Son District) and Thanh Duc Commune (Thanh Chuong District).Wild Asian elephants in Pumat NP region are facing dangers of extinction due to their small number as well as direct and indirect threats from human. The elephant group in Anh Son district, with the largest number of individuals consisting of breeding males, breeding females and a calf, is the most important for the maintainance and future development of elephant population within Puman NP. However, they are facing a number of  serious threats such as: un-controlled wildlife hunting and trade; rapid decrease and degradation of natural forest outside of Pumat National Park; and increasingly serious human-elephant conflicts.

    Abstract: From the ethyl acetate extract of the leaves of Desmodium gangeticum collected in Me Linh, Ha Noi, we isolated 5 compounds including luteolin (1), luteolin tetramethyl ether (2),  N,N-dimethyl tetradecane-1-amin (3), D-pinitol (4)  and stigmasterol (5). In which, luteolin (1) was isolated from Desmodium gangeticum for the first time, luteolin tetramethyl ether (2) and  N,N-dimethyl tetradecane-1-amin (3) were first isolated from genus Desmodium. Their structures were determined by 1D and 2D NMR spectra.
