    Abstract: TianQin is a planned space-based gravitational wave (GW) observatory consisting of three Earth-orbiting satellites with an orbital radius of about $10^5 \, {\rm km}$. The satellites will form an equilateral triangle constellation the plane of which is nearly perpendicular to the ecliptic plane. TianQin aims to detect GWs between $10^{-4} \, {\rm Hz}$ and $1 \, {\rm Hz}$ that can be generated by a wide variety of important astrophysical and cosmological sources, including the inspiral of Galactic ultra-compact binaries, the inspiral of stellar-mass black hole binaries, extreme mass ratio inspirals, the merger of massive black hole binaries, and possibly the energetic processes in the very early universe and exotic sources such as cosmic strings. In order to start science operations around 2035, a roadmap called the 0123 plan is being used to bring the key technologies of TianQin to maturity, supported by the construction of a series of research facilities on the ground. Two major projects of the 0123 plan are being carried out. In this process, the team has created a new-generation $17 \, {\rm cm}$ single-body hollow corner-cube retro-reflector which was launched with the QueQiao satellite on 21 May 2018; a new laser-ranging station equipped with a $1.2 \, {\rm m}$ telescope has been constructed and the station has successfully ranged to all five retro-reflectors on the Moon; and the TianQin-1 experimental satellite was launched on 20 December 2019—the first-round result shows that the satellite has exceeded all of its mission requirements.

    Abstract: Dapoxetine on demand has been approved for premature ejaculation (PE) management in China; however, studies on the efficacy and safety of this treatment in the Chinese population are scarce. Aim The aim of this study was to evaluate the safety and effectiveness of dapoxetine 30 mg and 60 mg on demand in Chinese men with PE. Methods Phase IV real-world study on 1,252 patients with PE. If men reported no response to dapoxetine 30 mg after 4 weeks treatment, dapoxetine has been uptitrated at 60 mg for 4 weeks more. Main Outcome Measure Self-reported data were collected for demographics, general and sexual health characteristics, PE severity, and treatment safety and effectiveness, as measured by the PE profile questionnaire. Results Adverse events (AEs), such as nausea, thirst, headache, and dizziness, similarly to previous literature, were detected. The treatment-emergent AEs rate was higher in the patients treated with 30 and 60 mg (n = 192) compared with those treated with the dapoxetine 30 mg only (n = 1060) (34.4% vs 15.8%, respectively). No new safety concerns were observed. The overall effectiveness rates were 88.2% in subjects using 30 mg of dapoxetine, whereas a rescue from the previous failure was in 55.7% in the patients who received 60 mg after the initial 30 mg. Overall, 83.2% responded to dapoxetine at dosages equal to or lower than 60 mg. Conclusion The results in this study demonstrated in a large Chinese population that on-demand dapoxetine is a safe and effective symptomatic treatment in patients with PE.     Abstract: Tetrahydrobiopterin (BH4) is a critical determinant of the biological function of endothelial nitric oxide synthase. The present study was to investigate the role of valvular endothelial cell (VEC)-derived BH4 in aortic valve calcification. Methods and results Plasma and aortic valve BH4 concentrations and the BH4:BH2 ratio were significantly lower in calcific aortic valve disease patients than in controls. There was a significant decrease of the two key enzymes of BH4 biosynthesis, guanosine 5′-triphosphate cyclohydrolase I (GCH1) and dihydrofolate reductase (DHFR), in calcified aortic valves compared with the normal ones. Endothelial cell-specific deficiency of Gch1 in Apoe  −/− (Apoe  −/−  Gch1  fl/fl  Tie2  Cre) mice showed a marked increase in transvalvular peak jet velocity, calcium deposition, runt-related transcription factor 2 (Runx2), dihydroethidium (DHE), and 3-nitrotyrosine (3-NT) levels in aortic valve leaflets compared with Apoe  −/−  Gch1  fl/fl mice after a 24-week western diet (WD) challenge. Oxidized LDL (ox-LDL) induced osteoblastic differentiation of valvular interstitial cells (VICs) co-cultured with either si-GCH1- or si-DHFR-transfected VECs, while the effects could be abolished by BH4 supplementation. Deficiency of BH4 in VECs caused peroxynitrite formation increase and 3-NT protein increase under ox-LDL stimulation in VICs. SIN-1, the peroxynitrite generator, significantly up-regulated alkaline phosphatase (ALP) and Runx2 expression in VICs via tyrosine nitration of dynamin-related protein 1 (DRP1) at Y628. Finally, folic acid (FA) significantly attenuated aortic valve calcification in WD-fed Apoe  −/− mice through increasing DHFR and salvaging BH4 biosynthesis. Conclusion The reduction in endothelial-dependent BH4 levels promoted peroxynitrite formation, which subsequently resulted in DRP1 tyrosine nitration and osteoblastic differentiation of VICs, thereby leading to aortic valve calcification. Supplementation of FA in diet attenuated hypercholesterolaemia-induced aortic valve calcification by salvaging BH4 bioavailability.

    Abstract: To fully unlock the potential of pigs as both agricultural species for animal-based protein food and biomedical models for human biology and disease, a comprehensive understanding of molecular and cellular mechanisms underlying various complex phenotypes in pigs and how the findings can be translated to other species, especially humans, are urgently needed. Here, within the Farm animal Genotype-Tissue Expression (FarmGTEx) project, we build the PigBiobank (http:////pigbiobank.farmgtex.org) to systematically investigate the relationships among genomic variants, regulatory elements, genes, molecular networks, tissues and complex traits in pigs. This first version of the PigBiobank curates 71 885 pigs with both genotypes and phenotypes from over 100 pig breeds worldwide, covering 264 distinct complex traits. The PigBiobank has the following functions: (i) imputed sequence-based genotype-phenotype associations via a standardized and uniform pipeline, (ii) molecular and cellular mechanisms underlying trait-associations via integrating multi-omics data, (iii) cross-species gene mapping of complex traits via transcriptome-wide association studies, and (iv) high-quality results display and visualization. The PigBiobank will be updated timely with the development of the FarmGTEx-PigGTEx project, serving as an open-access and easy-to-use resource for genetically and biologically dissecting complex traits in pigs and translating the findings to other species.

    Abstract: In an optical tweezers system, the output signal of a photodiode quadrant detector and the temperature in a sample cell are two key factors for the quantitative measurements of mechanical properties of living biological objects such as cells, organelles and macro-molecules. In order to enhance the output of a quadrant detector and effectively control the temperature in a sample cell, the dependence of the temperature in the sample cell and the output of the quadrant detector for different illumination conditions are studied. The results show that appropriate illumination conditions can ensure both nearly constant temperatures in the cell and the desired output signal, which provides for the possibility of highprecision and damage-free analysis of living biological objects.

    Abstract: To investigate the broad-spectrum antifungal activity of Burkholderia sp. BV6, that is isolated from rice roots and its biocontrol potential against rice blast caused by Magnaporthe oryzae. Methods and Results We evaluated the ability to isolate BV6 in the biological control of rice blast disease and investigated its antifungal mechanisms. BV6 strongly inhibited the hyphal growth of M. oryzae Guy11 and other plant pathogenic fungi, and pot experiments showed that BV6 significantly decreases the disease index of rice blast from 47.5 to 24.6. The secreted small-molecule secondary metabolites were regarded as weapons during the antifungal process by inhibiting the germination of M. oryzae conidia and mycelial growth, and thereby prevent the following infection. Liquid chromatography–mass spectrometry analysis of the metabolites from the supernatant of isolate BV6 showed that the antifungal weapons of isolate BV6 are novel, small, molecular hydrophilic compounds that are different from reported antifungal compounds. Conclusions The isolate BV6 inhibits the M. oryzae infection by the production of small-molecule antifungal compounds. Significance and Impact of the Study The current study discovers the role of the Burkholderia sp. BV6 in the biocontrol of plant pathogenic fungi. Therefore, isolate BV6 is a potential candidate for developing a microbial formulation for the biocontrol of the most common disease of rice blast.

    Abstract: We present sensitive observations of NH2D $1_{11}^\mathrm{ a}\!-\!1_{01}^\mathrm{ s}$ at 110.153 599 GHz toward 50 Galactic massive star-forming regions with the Institut de Radioastronomie Millimétrique (IRAM) 30-m telescope. The NH2D $1_{11}^\mathrm{ a}\!-\!1_{01}^\mathrm{ s}$ transition is detected toward 36 objects, yielding a detection rate of 72 per cent. Column densities of NH2D, HC3N, and C18O for each source are derived by assuming local thermal equilibrium conditions with a fixed excitation temperature. The deuterium ratio of NH3, defined as the abundance ratio of NH2D to NH3, for 19 sources is also obtained with the NH3 information from the literature. The range of deuterium fractionation bends to be large in the late-stage star-forming regions in this work, with the value from 0.043 to 0.0006. The highest deuterium ratio of NH3 is 0.043 in G081.75+00.78 (DR21). We also find that the deuterium ratio of NH3 increases with Galactocentric distance and decreases with line width.

    Abstract: To understand the relation between NH2D and its physical environment, we mapped ortho-NH2D $1_{11}^s-1_{01}^a$ at 85.9 GHz toward 24 Galactic late-stage massive star-forming regions with Institut de Radioastronomie Millim${\rm \acute{e}}$trique (IRAM) 30-m telescope. Ortho-NH2D $1_{11}^s-1_{01}^a$ was detected in 18 of 24 sources. Comparing with the distribution of H13CN 1-0 as a dense gas tracer and radio recombination line H42α, ortho-NH2D $1_{11}^s-1_{01}^a$ present complex and diverse spatial distribution in these targets. 11 of the 18 targets, present a different distribution between ortho-NH2D $1_{11}^s-1_{01}^a$ and H13CN 1-0, while no significant difference between these two lines can be found in other 7 sources, mainly due to limited spatial resolution and sensitivity. Moreover, with H42α tracing massive young stellar objects, ortho-NH2D $1_{11}^s-1_{01}^a$ seems to show a relatively weak emission near the massive young stellar objects.
