    Abstract: Lysine 2-hydroxyisobutyrylation (Khib) is a novel type of histone acylation whose prevalence and function in plants remain unclear. Here, we identified 41 Khib sites on histones in Arabidopsis thaliana, which did not overlap with frequently modified N-tail lysines (e.g. H3K4, H3K9 and H4K8). Chromatin immunoprecipitation-sequencing (ChIP-seq) assays revealed histone Khib in 35% of protein-coding genes. Most Khib peaks were located in genic regions, and they were highly enriched at the transcription start sites. Histone Khib is highly correlated with acetylation (ac), particularly H3K23ac, which it largely resembles in its genomic and genic distribution. Notably, co-enrichment of histone Khib and H3K23ac correlates with high gene expression levels. Metabolic profiling, transcriptome analyses, and ChIP-qPCR revealed that histone Khib and H3K23ac are co-enriched on genes involved in starch and sucrose metabolism, pentose and glucuronate interconversions, and phenylpropanoid biosynthesis, and help fine-tune plant response to dark-induced starvation. These findings suggest that Khib and H3K23ac may act in concert to promote high levels of gene transcription and regulate cellular metabolism to facilitate plant adaption to stress. Finally, HDA6 and HDA9 are involved in removing histone Khib. Our findings reveal Khib as a conserved yet unique plant histone mark acting with lysine acetylation in transcription-associated epigenomic processes.

    Abstract: Induced pluripotent stem cells (iPSCs) generated from somatic cell sources are pluripotent and capable of indefinite expansion in vitro. They provide an unlimited source of cells that can be differentiated into lung progenitor cells for potential clinical use in pulmonary regenerative medicine. This review gives a comprehensive overview of recent progress toward the use of iPSCs to generate proximal and distal airway epithelial cells and mix lung organoids. Furthermore, their potential applications and future challenges for the field are discussed, with a focus on the technological hurdles that must be cleared before stem cell therapeutics can be used for clinical treatment.

    Abstract: This study aimed to investigate different methods of obtaining high-quality Computed Tomography pulmonary angiography (CTPA) images using low-dose scanning in patients with different body mass index (BMI) values. Sixty patients with suspected pulmonary embolism were grouped based on their BMI values (BMI & lt; 25, designated N, and BMI ≥ 25, designated O) and were assigned to receive either test bolus (TB) or bolus tracking (BT) at conventional (C) or low (L) dose. The effective dose (ED) in the N–TB–L group was lower than in the group N–TB–C (0.56 ± 0.05 vs. 3.78 ± 1.16, p  & lt; 0.001), with similar image quality (4.90 ± 0.31 vs. 4.70 ± 0.47, p = 0.120). The ED in the O–TB–L group was lower than in the O–TB–C group (0.54 ± 0.03 vs. 5.14 ± 1.34, p  & lt; 0.001), but the group O–TB–C’s image quality was higher (4.65 ± 0.59 vs. 3.95 ± 0.89, p = 0.006). Groups N–TB–L versus O–TB–L, groups N-TB-L versus N–BT-L and groups O–TB–C versus O–BT–C had similar EDs (all ps  & gt; 0.05), but the image quality was different (all ps  & lt; 0.05). In conclusion, the results showed that the image quality of low-dose CTPA scanning using TB was similar to that of the conventional-dose CTPA in patients with BMI & lt; 25 but was lower in patients with BMI ≥ 25. TB was better than BT for all patients, regardless of BMI, when receiving the same ED.

    Abstract: Obesity is a consideration in the pharmacologic intervention for estrogen receptor (ER) positive (ER+) breast cancer risk. Body mass index (BMI) and waist/hip ratio (WHR) have demonstrated different effects on breast cancer risk in relation to estrogen receptor (ER) status, but the results have been inconsistent. Furthermore, the situation in Chinese women remains unclear. Materials and Methods We conducted a case-control study including 1,439 breast cancer cases in Northern and Eastern China. Both ER and progesterone receptor (PR) statuses were available for 1,316 cases. Associations between body size-related factors and breast cancer risk defined by receptor status were assessed by multiple polytomous unconditional logistic regression analysis. Results Body mass index and WHR were positively associated with overall breast cancer risk. Body mass index was positively associated with both ER+/PR positive (PR+) and ER negative (ER−)/PR negative(PR−) subtype risks, although only significantly for ER+/PR+ subtype. Waist–hip ratio was only positively correlated with ER−/PR− subtype risk, although independent of BMI. Body mass index was positively associated with risk of ER+/PR+ and ER−/PR− subtypes in premenopausal women, whereas WHR was inversely correlated with ER+/PR− and positively with ER−/PR− subtype risks. Among postmenopausal women, WHR & gt;0.85 was associated with increased risk of ER−/PR− subtype. Conclusion Both general and central obesity contribute to breast cancer risk, with different effects on specific subtypes. General obesity, indicated by BMI, is more strongly associated with ER+/PR+ subtype, especially among premenopausal women, whereas central obesity, indicated by WHR, is more specific for ER−/PR− subtype, independent of menopausal status. These results suggest that different chemoprevention strategies may be appropriate in selected individuals.

    Abstract: Spatial genetic and phenotypic diversity within solid tumors has been well documented. Nevertheless, how this heterogeneity affects temporal dynamics of tumorigenesis has not been rigorously examined because solid tumors do not evolve as the standard population genetic model due to the spatial constraint. We therefore, propose a neutral spatial (NS) model whereby the mutation accumulation increases toward the periphery; the genealogical relationship is spatially determined and the selection efficacy is blunted (due to kin competition). In this model, neutral mutations are accrued and spatially distributed in manners different from those of advantageous mutations. Importantly, the distinctions could be blurred in the conventional model. To test the NS model, we performed a three-dimensional multiple microsampling of two hepatocellular carcinomas. Whole-genome sequencing (WGS) revealed a 2-fold increase in mutations going from the center to the periphery. The operation of natural selection can then be tested by examining the spatially determined clonal relationships and the clonal sizes. Due to limited migration, only the expansion of highly advantageous clones can sweep through a large part of the tumor to reveal the selective advantages. Hence, even multiregional sampling can only reveal a fraction of fitness differences in solid tumors. Our results suggest that the NS patterns are crucial for testing the influence of natural selection during tumorigenesis, especially for small solid tumors.

    Abstract: Pyroptosis has been found implicated in several diseases, however, whether it was involved in gouty arthritis remained unclear. Our study was performed to uncover the role of pyroptosis in gouty arthritis based on a mice model. Methods Mouse gouty arthritis model was established by injections of potassium oxonate (PO), monosodium urate (MSU) and pyroptosis suppressor disulfiram. The diameter of the ankle joints was measured, and ankle joints morphology was observed with hematoxylin–eosin (H & E) staining. Uric acid, creatinine and blood urea nitrogen (BUN) concentrations were measured, while cytokines level and xanthine oxidase (XOD) activity were quantified. Relative pyroptosis markers expressions were determined using quantitative real-time polymerase chain reaction (qRT-PCR) and Western blot as needed. Results In mouse model, PO and MSU injections cause damage to right ankle, increase the root thickness ratio and uric acid, creatinine and BUN levels in serum and decrease the uric acid and creatinine levels in urine. Also, under PO and MSU treatment, up-regulated XOD activity, inflammatory cytokines levels and pyroptosis markers expressions are observed. Negative regulation of mice injury by disulfiram treatment is also observed. Conclusion Pyroptosis inhibition might alleviate PO- and MSU-induced gouty arthritis, providing possible therapeutic strategies for gouty arthritis.

    Abstract: Plants have evolved complex mechanisms to reprogram growth in response to drought stress. In herbaceous perennial plant species, the rhizome, which is normally an organ for propagation and food storage, can also support plant growth in stressful environments, and allows the plant to perennate and survive stress damage. However, the mechanisms that regulate rhizome growth in perennial herbs during abiotic stresses are unknown. Here, we identified a chrysanthemum (Chrysanthemum morifolium) DEAD-box RNA helicase gene, CmRH56, that is specifically expressed in the rhizome shoot apex. Knock down of CmRH56 transcript levels decreased the number of rhizomes and enhanced drought stress tolerance. We determined that CmRH56 represses the expression of a putative gibberellin (GA) catabolic gene, GA2 oxidase6 (CmGA2ox6). Exogenous GA treatment and silencing of CmGA2ox6 resulted in more rhizomes. These results demonstrate that CmRH56 suppresses rhizome outgrowth under drought stress conditions by blocking GA biosynthesis.

    Abstract: The air pollutants in tobacco smoke cause serious harm to human health. To study the influence of plants and ventilation on the concentrations of air pollutants in tobacco smoke, three different experimental chambers were established to simulate different smoking environments: ventilated Form-1 with plants, closed Form-2 with plants and closed Form-3 without plants. The concentrations of four pollutants produced by a lit cigarette were measured. The results showed that the concentrations of pollutants in the chambers with plants were the lowest. The concentration of CO2 in Form-1 decreased the most quickly. The times required for the concentrations of formaldehyde and particulate matter to decrease to standard values was 2.3 and 8.3 hours shorter in Form-1 than Form-2, respectively. However, the concentration of total volatile organic compounds in the three chambers was consistently above the standard value at 12 hours. The removal efficiency was stable after 6 hours in Form-1, and it may take longer than 12 hours to remove pollutants in Form-2. The removal efficiency of pollutants in Form-1 was consistently higher than that in Form-2 and Form-3, indicating that the removal was more effective when the chambers were ventilated. Among CO2, HCHO, TVOCs and PM, the removal efficiency of PM was the highest. It is recommended to increase the leaf area and guide the airflow to bring the pollutants into contact with the plants to improve the removal effect.
