    Notizen: The morphological relationships present in the areas of lymphoid infiltration found in the esophagus in Chrysemys scripta elegans were studied by the use of light and transmission electron microscopy. These infiltrations are wide-spread and can be observed in the lamina propria and the covering epithelium. Electron microscopy reveals the presence of epithelial cells with differing characteristics at the apex of the lymphoid infiltrations. The introduction of horseradish peroxidase (HRP) into the esophageal lumen revealed that these cells possess a micropinocytotic activity. On the basis of the morphological and experimental results obtained, a hypothesis is advanced of the existence, also in the turtle, of a local system of uptake of extraneous material, which induces local and systemic immunological processes.

    Notizen: Esterase activity studies on the areas of lymphoid infiltration in the bursa of Fabricius (diffusely infiltrated area) and the dorsal wall of the cloaca showed that the epithelial cells exhibit varying degree of diffuse esterase activity; it was possible to demonstrate the presence of spot-like esterase positivity in star-shaped cells in the epithelium itself. These cells can be found at various levels from the proximal to the distal region and have also been observed in the connective tissue of the tunica propria; as a result, the hypothesis has been advanced that they are connective tissue.

    Notizen: Polyurethane prepolymers were prepared in ethylacetate from TDI and poly(oxypropilene)glicols) (PE-PU) or poly(tetramethyleneglycoladipate)diols (PES-PU) of different molecular weights (M). Homogeneous membranes were prepared by spreading the pre-polymer solution added with a three, four or pentafunctional crosslinking agent, in the presence of stannous octoate as a catalyst. On increasing M, the PUs Tg decreases regularly in the range +30 to -47°C. PU's are amorphous with the exception of PES-PU obtained from highest M polyester diols. Permeability P, diffusivity D, solubility coefficient S of N2, O2, CO2, CH4, CO were measured at T=35°C (〉Tg).P, D, S do not depend on the crosslinker functionality, but mainly on M. P and D in PE-PU are linear function of T-Tg while in PES-PU the function is not linear. Gas solubility fulfills in general the regular solution theory. P, D, S of water vapor is also given. Permeability decreases in the order H2O〉CO2 〉 O2 〉 CH4 〉 CO 〉 N2. Some practical application based on particular selectivities is suggested.

    Notizen: Polyurethanes (PU) were prepared using polymeric diols, containing polar groups, as —O—C(O)—O—(carbonate) groups, carbonate and ether (—O—) groups, or carbonate and ester groups [—C(O)—O—]. PUs were prepared by the prepolymer two-step technique using ethyl acetate (EA) as the solvent; the diol was reacted at about +80°C with TDI (ratio 1:2) to give the prepolymer terminated with NCO, which was then cross-linked with triisopropanolamine (TIPA). The membranes were prepared using a Gardner knife and were characterized by differential thermal analysis (DSC). Most of the polymers prepared from low and medium molecular weight diols were amorphous and elastomeric at the temperature of gas transport measurement (35°C). The permeabilities (P) and the diffusion coefficients (D) of different gases (O2, N2, CO2, CH4, CO) were measured by a modified Lyssy apparatus, the solubility coefficient (S) was also calculated. The water-vapor transport properties D and S were measured according to ASTM. Diffusivity data follow the Fujita model and the solubility coefficients the regular solution theory, as developed by Prausnitz and Shair. © 1993 John Wiley & Sons, Inc.

    Beschreibung: The Himalaya and the Tibetan Plateau are uplifted by the ongoing northward underthrusting of the Indian continental lithosphere below Tibet resulting in lithospheric stacking. The layered structure of the Tibetan upper mantle is imaged by seismic methods, most detailed with the receiver function method. Tibet is considered as a place where the development of a future craton is currently under way. Here we study the upper mantle from Germany to northern Sweden with seismic S receiver functions and compare the structure below Scandinavia with that below Tibet. Below Proterozoic Scandinavia, we found two low velocity zones on top of each other, separated by a high velocity zone. The top of the upper low velocity zone at about 100km depth extends from Germany to Archaean northern Sweden. It agrees with the lithosphere-asthenosphere boundary (LAB) below Germany and Denmark. Below Sweden it is known as the 8°discontinuity, or as a mid-lithospheric discontinuity (MLD), similar to observations in North America. Seismic tomography places the LAB near 200km in Scandinavia, which is close to the top of our deeper low velocity zone. We also observed the bottom of the asthenosphere (the Lehmann discontinuity) deepening from 180km in Germany to 260km below Sweden. Remnants of old subduction in the upper about 100km below Scandinavia and Finland are known from controlled source seismic experiments and local earthquake studies. Recent tomographic studies indicate delamination of the lithosphere below southern Scandinavia and northern Germany. We are suggesting that the large scale layered structure in the Scandinavian upper mantle may be caused by processes similar to the ongoing lithospheric stacking in Tibet.

    Beschreibung: Globoid cell leukodystrophy (GLD) is a lysosomal storage disease caused by deficient activity of β-galactocerebrosidase (GALC). The infantile forms manifest with rapid and progressive central and peripheral demyelination, which represent a major hurdle for any treatment approach. We demonstrate here that neonatal lentiviral vector-mediated intracerebral gene therapy (IC GT) or transplantation of GALC-overexpressing neural stem cells (NSC) synergize with bone marrow transplant (BMT) providing dramatic extension of lifespan and global clinical–pathological rescue in a relevant GLD murine model. We show that timely and long-lasting delivery of functional GALC in affected tissues ensured by the exclusive complementary mode of action of the treatments underlies the outstanding benefit. In particular, the contribution of neural stem cell transplantation and IC GT during the early asymptomatic stage of the disease is instrumental to enhance long-term advantage upon BMT. We clarify the input of central nervous system, peripheral nervous system and periphery to the disease, and the relative contribution of treatments to the final therapeutic outcome, with important implications for treatment strategies to be tried in human patients. This study gives proof-of-concept of efficacy, tolerability and clinical relevance of the combined gene/cell therapies proposed here, which may constitute a feasible and effective therapeutic opportunity for children affected by GLD.

    Beschreibung: Arsenic-induced health effects may be associated with critically shortened telomeres. However, few data are available on the effects of arsenic exposure on telomere length. The aim of this study was to investigate the effects of chronic arsenic exposure on leukocyte telomere length (LTL) as well as the contribution of common polymorphisms in genes implicated in arsenic metabolism (GSTT1 and GSTM1) and DNA repair (hOGG1 and XRCC1). A group of 241 healthy subjects was enrolled from four areas of Italy known to be affected by natural or anthropogenic arsenic pollution. Urine samples were tested for inorganic As (iAs), monomethylarsinic (MMA) and dimethylarsinic acid (DMA). LTL was evaluated by quantitative reverse transcriptase polymerase chain reaction (qRT-PCR). Genotyping was carried out by PCR-RFLP on leukocyte DNA. In multiple linear regression analysis, LTL was significantly and inversely correlated with age (β = –0.231, P = 0.006) and showed a certain trend toward significance with iAs urinary concentration (log 10 iAs, β = –0.106, P = 0.08). The genotype distribution showed significant associations between GSTT1 and the As concentration (log 10 iAs, P = 0.01) and metabolite patterns (log 10 DMA, P = 0.05) in the urine. However, GST genes did not interact with arsenic exposure in the modulation of LTL. Conversely, the combined presence of a higher level of iAs + MMA + DMA ≥ 19.3 μg/l ( F = 6.0, P interaction = 0.01), Asi ≥ 3.86 ( F = 3.9, P interaction = 0.04) μg/l, iAs + MMA + DMA ≥ 15 μg/l ( F = 4.2, P interaction = 0.04) and hOGG1 Cys allele was associated with a significantly lower LTL. An interaction between XRCC1 Arg399Gln and arsenic exposure was also observed (all P interaction = 0.04). These findings suggest that telomere shortening may represent a mechanism that contributes to arsenic-related disease. The interaction of hOGG1 and XRCC1 DNA repair polymorphisms and exposure enhances telomeric DNA damage. Future studies are warranted to understand better the epidemiologic impact of arsenic on telomere function as well as to identify the subgroups of exposed subjects who need better health surveillance.

    Beschreibung: Mutations of FIG4 are responsible for Yunis-Varón syndrome, familial epilepsy with polymicrogyria, and Charcot-Marie-Tooth type 4J neuropathy (CMT4J). Although loss of the FIG4 phospholipid phosphatase consistently causes decreased PtdIns(3,5) P 2 levels, cell-specific sensitivity to partial loss of FIG4 function may differentiate FIG4-associated disorders. CMT4J is an autosomal recessive neuropathy characterized by severe demyelination and axonal loss in human, with both motor and sensory involvement. However, it is unclear whether FIG4 has cell autonomous roles in both motor neurons and Schwann cells, and how loss of FIG4/PtdIns(3,5) P 2 -mediated functions contribute to the pathogenesis of CMT4J. Here, we report that mice with conditional inactivation of Fig4 in motor neurons display neuronal and axonal degeneration. In contrast, conditional inactivation of Fig4 in Schwann cells causes demyelination and defects in autophagy-mediated degradation. Moreover, Fig4-regulated endolysosomal trafficking in Schwann cells is essential for myelin biogenesis during development and for proper regeneration/remyelination after injury. Our data suggest that impaired endolysosomal trafficking in both motor neurons and Schwann cells contributes to CMT4J neuropathy.

    Beschreibung: Oscillatory patterns permeate the universe and are essential requisite in living cells. Circadian clocks exist at the subcellular and single-cell level. Chromatin is assembled into rhythmic oscillatory domains driving stem cell growth/differentiation up to higher hierarchical embryo development. The cytoskeleton is organized as rhythmically oscillating networks, producing radioelectric fields that may turn local events into non-local, long-ranging paths. For decades, scientists have used chemical tools to affect cell behaviour. However, this view is now deeply challenged. Consonant with a major role of physical forces in living processes is the use of physical energies to modulate cell dynamics. To explore cell biology in the light of physics, we have recently established the Stem Wave Institute for Tissue Healing, within the context of GVM Care & Research—E.S. Health Science Foundation, Lugo, Italy. Here, we will discuss our recent findings that proper delivery of radioelectric fields is able to (i) finely tune stem cell multipotency, (ii) directly reprogramme human skin fibroblasts into cardiac-, neuronal-, and skeletal muscle-like cells, and (iii) revert stem cell senescence. We are dissecting vibrational modes as inherent properties of living cells, entailing nanomechanical signatures that can be used to direct stem cell fate. Mild mechanical forces are deployed to obtain human fluid tissues harbouring stem cells within their stromal–vascular niche. Synthetic molecules are designed to afford stem cell pluripotency. These discoveries prompt a deeper understanding of the interconnections between the physical universe and the living world in the attempt to further approach the information of Life.
