    Abstract: Shexiang Baoxin Pills (SBP) is widely used for the treatment of unstable angina pectoris (UAP) in China. However, the clinical evidence on the efficacy of SBP for the treatment of UAP is not well concluded. Methods: Seven electronic databases will be searched for eligible studies: MEDLINE, EMBASE, The Cochrane Library, Wanfang database, Chinese National Knowledge Infrastructure database, VIP database, and Chinese Biological and Medicine database. Data of included studies will be extracted, and quality will be evaluated. Data synthesis will be performed using RevMan software. Subgroup analysis and sensitivity analysis will also be carried out. Publication bias will be evaluated using funnel plot if included studies are sufficient. Results: This systemic review and meta-analysis will provide synthesized result of clinical efficacy of SBP for the treatment of UAP. Conclusions: This systemic review and meta-analysis will provide high-quality evidence on the clinical efficacy of SBP for the treatment of UAP. Registration: PEROSPERO CRD42019124668

    Abstract: Congenital heart disease (CHD) is the most serious congenital defect in newborns with higher mortality. Alternative splicing (AS) plays an essential role in numerous heart diseases. However, our understanding of the link between mRNA splicing and CHD in humans is limited. Here, we try to investigate the genome-wide AS events in CHD using bioinformatics methods. We collected available RNA-seq datasets of CHD-induced pluripotent stem cell-cardiomyocytes (iPSC-CMs) (including single ventricle disease [SVD] and tetralogy of Fallot [TOF] ) and non-CHD from the Gene Expression Omnibus database. Then, we unprecedentedly performed AS profiles in CHD-iPSC-CMs and non-CHD-iPSC-CMs. The rMAPS was used to generate RNA-maps for the analysis of RNA-binding proteins’ (RBPs) binding sites. We used StringTie to identify and quantify the transcripts from aligned RNA-Seq reads. A quantification matrix was generated with respect to different groups by extracting the transcripts per million values from StringTie outputs. Then, this matrix was used for correlation analysis between the expression level of RBP and AS level. Finally, we validated our AS results using RNA-seq data from CHD and non-CHD patient tissue samples. We identified CHD-related AS events using CHD-iPSC-CMs and CHD samples from patients. The results showed that functional enrichment of abnormal AS in SVD and TOF was transcription factor-related. Using rMAPS, RNA-binding proteins which regulated these AS were also determined, and RBP-AS regulatory network was constructed. Overall, we identified abnormal AS in CHD-iPSC-CMs and CHD samples from patients. We predicted AS regulators in SVD and TOF, respectively. At last, we concluded that AS played a key role in the pathogenesis of CHD.

    Abstract: The way to improve the long-term efficacy of fecal microbiota transplantation (FMT) in ulcerative colitis (UC) remains unclear. This study mainly dedicated to the UC patients’ satisfaction with FMT and the importance of patients’ adherence to repeated FMT for long-term clinical outcomes. Methods Patients with UC who underwent FMT at our center from November 2012 to September 2018 were included. We assessed patient satisfaction with efficacy, safety, and reliability of FMT, as well as adherence to the repeated FMT. Results One hundred and seventy-six patients were included in the analysis. The median follow-up duration of the study was 25.5 (interquartile range 13.0–46.5) months. The clinical response rate at 1 week, 1 month, 3 months, and 6 months after FMT was 48.9%, 69.3%, 49.4%, and 32.7%, respectively. 3.4% (6/176) of patients underwent colectomy after FMT during our long-term follow-up. Partial Mayo score at 1-month post-FMT ( P 〈 0.001) was an independent factor of patients’ satisfaction. The laboratory preparation process was related to the incidence of adverse events ( P 〈 0.05). 23.8% (29/122) of patients with a good adherence followed our recommendation to undergo the second course of FMT and achieved a longer clinical response compared with the patients with poor adherence ( P 〈 0.001). Conclusion Patients’ good adherence to repeated FMT is important to maintain long-term clinical benefits achieved from FMT in UC. Registration number: NCT01790061.

    Abstract: Adjuvant endocrine therapy is a vital portion of postoperative comprehensive treatment for breast cancer patients. In recent years, studies have shown that endocrine therapy has a certain impact on the serum lipids of breast cancer patients, and the changes of lipid profiles may bring a series of problems. However, very few studies focus on this issue to date. The results of these studies are inconsistent, and the influence of different adjuvant endocrine modalities on lipid profiles still remains controversial. In order to better explore this issue, we conduct this network meta-analysis. Method: The protocol followed preferred reporting items for systematic reviews and meta-analyses protocols. Three main databases (PubMed, Embase, and the Cochrane Library) will be searched systematically for eligible randomized controlled trials without language restriction. In addition, a manual search of the references of relevant published studies will also be considered. Two reviewers will conduct studies selection, data extraction, and risk of bias assessment independently. The primary outcome is the variation of biochemical parameters – the serum lipid profiles (cholesterol, triglyceride, high-density lipoprotein, low low-density lipoprotein). Results: The results will provide useful information about the side effects of different adjuvant endocrine drugs on lipid profiles in postoperative breast cancer patients (estrogen receptor-positive and/or progesterone receptor-positive). Conclusion: The findings of this study will be published in a peer-reviewed journal. Prospero Registration Number: CRD42019129850.

    Abstract: Idiopathic ventricular tachycardia ( VT ) is a type of cardiac arrhythmia occurring in structurally normal hearts. The heritability of idiopathic VT remains to be clarified, and numerous genetic factors responsible for development of idiopathic VT are as yet unclear. Variations in FGF 12 (fibroblast growth factor 12), which is expressed in the human ventricle and modulates the cardiac Na + channel Na V 1.5, may play an important role in the genetic pathogenesis of VT . Methods and Results We tested the hypothesis that genetic variations in FGF 12 are associated with VT in 2 independent Chinese cohorts and resequenced all the exons and exon–intron boundaries and the 5′ and 3′ untranslated regions of FGF 12 in 320 unrelated participants with idiopathic VT . For population‐based case–control association studies, we chose 3 single‐nucleotide polymorphisms—rs1460922, rs4687326, and rs2686464—which included all the exons of FGF 12 . The results showed that the single‐nucleotide polymorphism rs1460922 in FGF 12 was significantly associated with VT after adjusting for covariates of sex and age in 2 independent Chinese populations: adjusted P =0.015 (odds ratio: 1.54 [95% CI, 1.09–2.19]) in the discovery sample, adjusted P =0.018 (odds ratio: 1.64 [95% CI, 1.09–2.48]) in the replication sample, and adjusted P =2.52E‐04 (odds ratio: 1.59 [95% CI, 1.24–2.03]) in the combined sample. After resequencing all amino acid coding regions and untranslated regions of FGF 12 , 5 rare variations were identified. The result of western blotting revealed that a de novo functional variation, p.P211Q (1.84% of 163 patients with right ventricular outflow tract VT), could downregulate FGF 12 expression significantly. Conclusions In this study, we observed that rs1460922 of FGF 12 was significantly associated with VT and identified that a de novo variation of FGF 12 may be an important genetic risk factor for the pathogenesis of VT .

    Abstract: It remains unclear whether neuroimaging markers of cerebral small‐vessel disease (CSVD) affect the outcomes of patients with acute ischemic stroke receiving endovascular treatment (EVT). The aim of this systematic review and meta‐analysis was to evaluate the association between CSVD neuroimaging markers and outcomes in patients with acute ischemic stroke undergoing EVT. Methods We conducted a systematic search of PubMed and EMBASE databases up to July 2022 using keywords or Medical Subject Heading terms (“cerebral small‐vessel diseases,” “leukoaraiosis,” “microbleed,” “enlarged perivascular space,” “recent small subcortical infarct,” “atrophy,” “lacune,” and “thrombectomy”). The assessed clinical outcomes were a good functional outcome, 90‐day mortality, symptomatic intracranial hemorrhage, and early neurologic improvement after EVT. Results Overall, 30 studies on patients with acute ischemic stroke undergoing EVT were included. Patients with absent or mild white matter hyperintensities had higher good functional outcomes (odds ratio [OR], 2.94 [95% CI, 2.44–3.53] ; P 〈 0.001) and lower mortality rate (OR, 0.42 [95% CI, 0.11–1.59]; P 〈 0.001), whereas the presence of cerebral microbleeds increased only the risk of 90‐day mortality (OR, 0.60 [95% CI, 0.44–0.83]; P =0.002). Moreover, patients with moderate/severe CSVD burden had worse functional outcomes than those with none/mild CSVD burden (OR, 2.94 [95% CI, 2.44–3.53]; P 〈 0.001), but neither mortality nor symptomatic intracranial hemorrhage was significantly different between the 2 groups. Conclusions The existence of CSVD affected the outcomes of patients with acute ischemic stroke receiving EVT. Future multicenter prospective cohort studies with little heterogeneity should be prioritized to confirm our results.
