An important step in this radical-chain reaction is the cis-selective migration of an ester group.     Notes: Two-dimensional gels offer a powerful method for separating complex protein mixtures, but subsequent methods for analysing individual components, such as protein sequencing and Western immunoblotting, are laborious and slow. The identification of proteins can be accelerated by using a combination of protease digestion and matrix assisted laser desorption-mass spectrometry (MALDI-MS). The peptide mass spectrum of a protein represents a unique fingerprint determined by the amino acid sequence and the cleavage properties of the protease. Software has been developed so that peptide masses can be used to search a mass-based peptide database generated from established protein sequence databases. A list of the closest matching proteins is produced to allow identification of the sample. The strategy was applied to 52 protein spots from human myocardial tissue separated by two-dimensional electrophoresis (2-DE) gels and analysed blind. Conditions for optimal trypsin digestion of proteins electroblotted onto polyvinylidene difluoride (PVDF) membranes are described. Mass data were generated from both Coomassie Brilliant Blue and sulforhodamine B-stained proteins, though the former required destaining prior to digestion. Alkylation of cysteine and oxidation of methionine were significant modifications that influenced the successful identification of a protein spot. Examples are presented to illustrate the advantages and disadvantages of this approach.

    Description: The potential impacts of climate change are an increasing focus of research and ever-larger climate projection ensembles are available, making standard impact assessments more onerous. An alternative way of estimating impacts involves response surfaces, which present the change in a given indicator for a large number of plausible climatic changes defined on a regular sensitivity domain. Sets of climate change projections can then be overlaid on the response surface and impacts estimated from the nearest corresponding points of the sensitivity domain, providing a powerful method for fast impact estimation for multiple projections and locations. However, the effect of assumptions necessary for initial response surface development must be assessed. This paper assesses the uncertainty introduced by use of a sensitivity framework for estimating changes in 20-year return period flood peaks in Britain. This sensitivity domain involves mean annual and seasonal precipitation changes, and a number of simplifications were necessary for consistency and to reduce dimensionality. The effect of these is investigated for nine catchments across Britain, representing nine typical response surfaces (response types), using three sets of climate projections. The results show that catchments can have different causes of uncertainty, and some catchments have an overall higher level of uncertainty than others. These differences are compatible with the underlying climatological and hydrological differences between the response types, giving confidence in generalisation of the results. This enables the development of uncertainty allowances by response type, to be used alongside the response surfaces to provide more robust impact estimates. This article is protected by copyright.     Description: In this paper we present the first decadal reanalysis simulation of the biogeochemistry of the North West European shelf, along with a full evaluation of its skill and value. An error-characterized satellite product for chlorophyll was assimilated into a physical-biogeochemical model of the North East Atlantic, applying a localized Ensemble Kalman filter. The results showed that the reanalysis improved the model predictions of assimilated chlorophyll in 60% of the study region. Model validation metrics showed that the reanalysis had skill in matching a large dataset of in situ observations for ten ecosystem variables. Spearman rank correlations were significant and higher than 0.7 for physical-chemical variables (temperature, salinity, oxygen), ∼0.6 for chlorophyll and nutrients (phosphate, nitrate, silicate), and significant, though lower in value, for partial pressure of dissolved carbon dioxide ( ∼ 0.4). The reanalysis captured the magnitude of pH and ammonia observations, but not their variability. The value of the reanalysis for assessing environmental status and variability has been exemplified in two case studies. The first shows that between 340,000-380,000 km 2 of shelf bottom waters were oxygen deficient potentially threatening bottom fishes and benthos. The second application confirmed that the shelf is a net sink of atmospheric carbon dioxide, but the total amount of uptake varies between 36-46 Tg C yr −1 at a 90% confidence level. These results indicate that the reanalysis output dataset can inform the management of the North West European shelf ecosystem, in relation to eutrophication, fishery, and variability of the carbon cycle.     Description: ABSTRACT Climate change is almost certain to affect snow and ice processes. Even at lower latitudes, changes in snow cover at high altitudes can significantly affect catchment hydrology. This article uses data from a transient Regional Climate Model projection (HadRM3Q0) for 1950–2099 (A1B emissions) to drive hydrological models for three nested catchments on the river Dee in north-east Scotland, to assess potential changes in flood frequency and timing using annual maxima and moving-window analyses. Some results are also shown for an upland catchment in northern England. Modelling is performed both with and without a snow module, to demonstrate the effects of snowfall/melt and how these change through time and vary between catchments. Modelled changes in flood magnitude and timing are nonlinear, with most changes for daily mean flows not significant. For longer duration (30-day) flows with snow there are significant decreases in peak magnitude, particularly for the smaller higher altitude Dee catchments, with peaks occurring months earlier in future (changes without snow are generally not significant). There is a general convergence in results with and without snow later in the period, as snow processes become less important, but convergence occurs at different times for different catchments and occurs differently for daily and 30-day peak flows due to the differential effects of snow at different durations. This not only highlights the importance of including snow processes for such catchments, particularly for longer duration flows, but also highlights the complexity of interactions: Physical catchment properties, the balance between precipitation occurrence and temperature, and how this balance alters as the climate changes will each be critical in determining the impact on the magnitude and timing of peak flows, making it hard to generalize results.

    Description: Background In chronic spontaneous urticaria (CSU) mast cell activation together with inflammatory changes in the skin are well documented and may play an important role in mechanisms of tissue oedema Objective To confirm and extend these observations by measuring microvascular markers, leukocytes and mast cells numbers in lesional and uninvolved skin and to compare findings with a control group. Methods Paired biopsies (one from 4- 8 hour spontaneous weals and one from uninvolved skin) were taken from eight patients with CSU and nine control subjects and studied by immunohistochemistry and confocal microscopy using the lectin UEA-1. Results Lesional skin in CSU contained significantly more CD31+ endothelial cells and CD31+ blood vessels, neutrophils, eosinophils, basophils, macrophages and CD3+ T cells, than non-lesional skin. Increased vascularity was confirmed by confocal imaging using the lectin UEA-1. Uninvolved skin from CSU contained significantly more CD31+ endothelial cells, CD31+ blood vessels and eosinophils compared to the control subjects. There was a three-fold increase in mast cell numbers when CSU was compared to controls but no difference was observed between lesional and uninvolved skin. Conclusions Increased vascular markers together with eosinophil and neutrophil infiltration are features of lesional skin in CSU and might contribute to tissue oedema. Eosinophils and microvascular changes persist in uninvolved skin which, together with increased mast cells, suggests that non-lesional skin is primed for further wealing.
