    Notes: The thermal and mechanical properties of phenolphthalein polyethersulfone/poly(phenylene sulfide) (PES-C/PPS) blends were studied using a differential scanning calorimeter, a dynamic mechanical analyzer, and mechanical characterization. The morphologies of fracture surfaces were observed by scanning electron microscopy. The blends are multiphase systems with strong interaction between the two phases. It is of interest that, although the strength and ductility of PPS are lower than those of PES-C, the addition of PPS can improve markedly the impact strength of PES-C without changing its higher strength. The PPS can also act as a flow aid for PES-C. © 1995 John Wiley & Sons, Inc.

    Notes: The effects of encapsulated metal salts on poly(DL-lactide-co-glycolide) (PLGA) water uptake and degradation properties were investigated in this work. Salts of varying aqueous solubility characteristics were incorporated into PLGA films either as particles or by codissolution in polymer solutions. Polymer films were characterized with respect to the kinetics of water uptake, morphology changes, degradation, and weight loss after hydration. It was found that these properties are strongly influenced by the presence and nature of encapsulated salts. Effects range from minor changes in water uptake profile with no significant difference in degradation kinetics to major alterations in water uptake kinetics together with a several-fold decrease in the polymer degradation rate. Possible mechanistic explanations for the observed effects are discussed. © 1997 John Wiley & Sons, Inc.

    Notes: Finite element partitioning (or substructuring) is employed to estimate the eigenproperties of large-scale structural systems. A homotopy equation is constructed and its solutions are characterized by a number of curves which connect the eigensolutions of the partitions with those of the complete system. A step-by-step tracing procedure is developed to follow these curves. At each step, prediction and correction are performed. The Rayleigh-Ritz procedure and the conjugate gradient method are used as predictor and corrector, respectively. Compared with the sole use of either the Rayleigh-Ritz or gradient methods, the proposed method is more reliable and more efficient for large-scale problems. Numerical implementation is well suited for supercomputers.

    Notes: New temporal and spatial discretization methods are developed for multiple scale structural dynamic problems. The concept of fast and slow time scales is introduced for the temporal discretization. The required time step is shown to be dependent only on the slow time scale, and therefore, large time steps can be used for high frequency problems. To satisfy the spatial counterpart of the requirement on time step constraint, finite-spectral elements and finite wave elements are developed. Finite-spectral element methods combine the usual finite elements with the fast convergent spectral functions to obtain a faster convergence rate; whereas, finite wave elements are developed in parallel to the temporal shifting technique. Therefore, the spatial resolution is increased substantially. These methods are especially applicable to structural acoustics and linear space structures. Numerical examples are presented to illustrate the effectiveness of these methods.

    Description: This paper addresses an autonomous navigation method for the autonomous underwater vehicle (AUV) C-Ranger applying information-filter-based simultaneous localization and mapping (SLAM), and its sea trial experiments in Tuandao Bay (Shangdong Province, P.R. China). Weak links in the information matrix in an extended information filter (EIF) can be pruned to achieve an efficient approach-sparse EIF algorithm (SEIF-SLAM). All the basic update formulae can be implemented in constant time irrespective of the size of the map; hence the computational complexity is significantly reduced. The mechanical scanning imaging sonar is chosen as the active sensing device for the underwater vehicle, and a compensation method based on feedback of the AUV pose is presented to overcome distortion of the acoustic images due to the vehicle motion. In order to verify the feasibility of the navigation methods proposed for the C-Ranger, a sea trial was conducted in Tuandao Bay. Experimental results and analysis show that the proposed navigation approach based on SEIF-SLAM improves the accuracy of the navigation compared with conventional method; moreover the algorithm has a low computational cost when compared with EKF-SLAM.

    Description: The recombinant ( S )-carbonyl reductase (SCR) in Escherichia coli catalyzed the reduction of 2-hydroxyacetophenone to ( S )-1-phenyl-1,2-ethanediol (PED) with low efficiency. In this work, its 6× histidine fusion gene his 6 - scr was cloned in Pichia pastoris under the control of the AOX1 methanol inducible promoter. The heterologous protein SCR was expressed through a Mut s phenotype. Under the optimal conditions: pH 7.0, initial OD 600 2.5, methanol daily addition concentration 1.0% and induction duration 4–5 days, the recombinant protein SCR was produced at the highest level. The enzyme activity in the cell-free exacts of P. pastoris was 0.38, which was over twofold than that of the recombinant E. coli -SCR. The enzyme was purified to homogeneity with a specific activity of 3.41 U mg −1 , and it catalyzed the biotransformation of ( S )-PED with a high optical purity of 96.9% in a high yield of 89.7% at optimum pH of 7.0. The developed effective system of P. pastoris -SCR will facilitate the preparation of pure chiral alcohol in industry.

    Description: Insulin is a secreted peptide hormone identified in human pancreas to promote glucose utilization. Insulin has been observed to induce cell proliferation and myogenesis in C2C12 cells. The precise mechanisms underlying the proliferation of C2C12 cells induced by insulin remain unclear. In this study, we observed for the first time that 10nM insulin treatment promotes C2C12 cell proliferation. Additionally, 50 nM and 100nM insulin treatment induces C2C12 cell apoptosis. By utilizing real-time PCR and Western blotting analysis, we found that the mRNA levels of cyclinD1 and BAD are induced upon 10nM and 50nM/100nM insulin treatment respectively. The similar results were observed in C2C12 cells expressing GATA-6 or PPARα. Our results identify for the first time the downstream targets of insulin, cyclin D1 and BAD, elucidate a new molecular mechanism of insulin in promoting cell proliferation and apoptosis. J. Cell.     Description: The next-generation anti-androgen MDV3100 prolongs overall survival of patients with metastatic castration-resistant prostate cancer (CRPC). However, patient responses are variable, and survival benefit remains relatively small. Developing effective modality to improve MDV3100 efficacy is urgently needed. Recent evidence suggests that constitutively-active androgen receptor splice variants (AR-Vs) drive resistance to MDV3100. In the present study, we show that methylselenol prodrug downregulates the expression and activity of both the full-length AR (AR-FL) and AR-Vs. The downregulation is independent of androgen and could be attributable to repressed transcription of the AR gene. Co-treatment with methylselenol prodrug and MDV3100 suppresses AR signaling more dramatically than either agent alone, and synergistically inhibits the growth CRPC cells in vitro . The combinatorial efficacy is observed in not only AR-V-expressing cells but also cells expressing predominantly AR-FL, likely owing to the ability of the two drugs to block the AR signaling cascade at distinct steps. Ectopic expression of AR-FL or AR-V7 attenuates the combinatorial efficacy, indicating that downregulating AR-FL and AR-V7 is importantly involved in mediating the combinatorial efficacy. Significantly, methylselenol prodrug also downregulates AR-FL and AR-Vs in vivo and substantially improves the antitumor efficacy of MDV3100. These findings support a potential combination therapy for improving MDV3100 efficacy, and provide a rationale for evaluating the clinical application of combining methylselenol prodrug with MDV3100 for the treatment of CRPC. © 2013 Wiley Periodicals, Inc.

    Description: BACKGROUND Castration-resistant progression of prostate cancer after androgen deprivation therapy remains a critical challenge in the clinical management of prostate cancer. Resurgent androgen receptor activity is an established driver of castration-resistant progression, and upregulation of androgen receptor expression has been implicated to contribute to the resurgent androgen receptor activity. We reported previously that methylselenocysteine can decrease the expression and activity of androgen receptor. Here we investigated the ability of methylselenocysteine to inhibit castration-resistant progression of prostate cancer. METHODS The regrowth of LNCaP prostate cancer xenografts after castration was monitored. The levels of prostate-specific antigen in mouse serum were measured by ELISA. Tumor cell proliferation and apoptosis were analyzed via Ki-67 immunohistochemistry and TUNEL assay, respectively. Intratumoral angiogenesis was assessed by immunohistochemistry staining of vascular endothelial growth factor and CD31. RESULTS We showed that methylselenocysteine delayed castration-resistant regrowth of LNCaP xenograft tumors after androgen deprivation. This was accompanied by decreased serum levels of prostate-specific antigen, inhibition of prostate cancer cell proliferation and tumor angiogenesis, as well as downregulation of androgen receptor and induction of apoptosis in the relapsed tumors. CONCLUSIONS The present study represents the first to show the preclinical efficacy of methylselenocysteine in delaying castration-resistant progression of prostate cancer. The findings provide a rationale for evaluating the clinical application of combining methylselenocysteine with androgen deprivation therapy for the treatment of advanced prostate cancer.
