    Abstract: The primary aim of this study was to evaluate the efficacy of phage against mastitis induced by drug-resistant S. aureus in a mouse model. In this study, five S. aureus phages—4086-1, 4086-2, 4086-3, 4086-4, and 4086-6—were isolated from milk samples secreted by mastitis cows. Transmission electron microscopy showed that all the five phages had icosahedral heads and short non-contractile tails, which are typical characteristics of the family Podoviridae. All these phages were species-specific against S. aureus. The one-step growth curve showed a short latency period (10–20 min) and high burst size (up to 400 PFU/infected cell). To evaluate the effectiveness of the phage 4086-1 in the treatment against mastitis, a mouse model of mastitis was challenged with drug-resistant S. aureus. The results showed the proliferation of S. aureus in the mammary glands was significantly inhibited after treating by phage 4086-1. The concentrations of TNF-α and IL-6 decreased significantly, which demonstrated the phages could effectively alleviate the inflammatory responses. Furthermore, the histopathological analysis showed that inflammatory infiltration in the mammary glands was significantly reduced. These results demonstrate that phage may be a promising alternative therapy against mastitis caused by drug-resistant S. aureus.

    Abstract: Glycogen synthase kinase-3 (GSK-3) is a serine/threonine kinase that has been implicated in numerous oncogenic processes. GSK-3 inhibitor elraglusib (9-ING-41) has shown promising preclinical and clinical antitumor activity across multiple tumor types. Despite promising early-phase clinical trial results, there have been limited efforts to characterize the potential immunomodulatory properties of elraglusib. We report that elraglusib promotes immune cell-mediated tumor cell killing of microsatellite stable colorectal cancer (CRC) cells. Mechanistically, elraglusib sensitized CRC cells to immune-mediated cytotoxicity and enhanced immune cell effector function. Using western blots, we found that elraglusib decreased CRC cell expression of NF-κB p65 and several survival proteins. Using microarrays, we discovered that elraglusib upregulated the expression of proapoptotic and antiproliferative genes and downregulated the expression of cell proliferation, cell cycle progression, metastasis, TGFβ signaling, and anti-apoptotic genes in CRC cells. Elraglusib reduced CRC cell production of immunosuppressive molecules such as VEGF, GDF-15, and sPD-L1. Elraglusib increased immune cell IFN-γ secretion, which upregulated CRC cell gasdermin B expression to potentially enhance pyroptosis. Elraglusib enhanced immune effector function resulting in augmented granzyme B, IFN-γ, TNF-α, and TRAIL production. Using a syngeneic, immunocompetent murine model of microsatellite stable CRC, we evaluated elraglusib as a single agent or combined with immune checkpoint blockade (anti-PD-1/L1) and observed improved survival in the elraglusib and anti-PD-L1 group. Murine responders had increased tumor-infiltrating T cells, augmented granzyme B expression, and fewer regulatory T cells. Murine responders had reduced immunosuppressive (VEGF, VEGFR2) and elevated immunostimulatory (GM-CSF, IL-12p70) cytokine plasma concentrations. To determine the clinical significance, we then utilized elraglusib-treated patient plasma samples and found that reduced VEGF and BAFF and elevated IL-1 beta, CCL22, and CCL4 concentrations correlated with improved survival. Using paired tumor biopsies, we found that tumor-infiltrating immune cells had a reduced expression of inhibitory immune checkpoints (VISTA, PD-1, PD-L2) and an elevated expression of T-cell activation markers (CTLA-4, OX40L) after elraglusib treatment. These results address a significant gap in knowledge concerning the immunomodulatory mechanisms of GSK-3 inhibitor elraglusib, provide a rationale for the clinical evaluation of elraglusib in combination with immune checkpoint blockade, and are expected to have an impact on additional tumor types, besides CRC.

    Abstract: In the context of rapid urbanization and the “Healthy China” strategy, neighborhood environments play an important role in improving mental health among urban residents. While an increasing number of studies have explored the linear relationships between neighborhood environments and mental health, much remains to be revealed about the nonlinear health effects of neighborhood environments, the thresholds of various environmental factors, and the optimal environmental exposure levels for residents. To fill these gaps, this paper collected survey data from 1003 adult residents in Guangzhou, China, and measured the built and social environments within the neighborhoods. The random forest model was then employed to examine the nonlinear effects of neighborhood environments on mental health, evaluate the importance of each environmental variable, as well as identify the thresholds and optimal levels of various environmental factors. The results indicated that there are differences in the importance of diverse neighborhood environmental factors affecting mental health, and the more critical environmental factors included greenness, neighborhood communication, and fitness facility density. The nonlinear effects were shown to be universal and varied among neighborhood environmental factors, which could be classified into two categories: (i) higher exposure levels of some environmental factors (e.g., greenness, neighborhood communication, and neighborhood safety) were associated with better mental health; (ii) appropriate exposure levels of some environmental factors (e.g., medical, fitness, and entertainment facilities, and public transport stations) had positive effects on mental health, whereas a much higher or lower exposure level exerted a negative impact. Additionally, this study identified the exact thresholds and optimal exposure levels of neighborhood environmental factors, such as the threshold (22.00%) and optimal exposure level ( 〉 22.00%) of greenness and the threshold (3.80 number/km2) and optimal exposure level (3.80 number/km2) of fitness facility density.

    Abstract: Short chain fatty acids (SCFAs) are produced by the colonic microbiota through fermentation. Influences of maternal PM2.5 exposure on SCFAs of the offspring have not been well understood. Additionally, studies of dietary intervention have not been carried out yet. Here we performed a study that dams were received PM2.5 and quercetin intervention during gestation and lactation. SCFAs in colon of dams and their offspring (on postnatal day 21 and 35) were analyzed using gas chromatography. For male offspring, when compared with the control group levels of acetic acid, butyric acid, and valeric acid were lower in the PM2.5 group (p 〈 0.05), however, levels of isobutyric acid and isovaleric acid were higher in the PM2.5 group (p 〈 0.05). For female offspring, as compared with the control group, propanoic acid was lower in the PM2.5 group, however isovaleric acid was higher in the PM2.5 group (p 〈 0.05). 100 mg/kg and 200 mg/kg quercetin intervention could inhibit SCFAs production of male offspring, especially in isobutyric acid and isovaleric acid (p 〈 0.05). 100 mg/kg quercetin intervention could upgrade the level of propanoic acid of female offspring (p 〈 0.05). Taken together, these results suggest that PM2.5 tracheal exposure during gestation and lactation could influence SCFAs of offspring. Quercetin administration might have the potential to offset the effects of mater PM2.5 exposure on SCFAs in the offspring to some extent. The above effects were showed in a sex-dependent manner.

    Abstract: Human norovirus (HNV) is one of the emerging and rapidly spreading groups of pathogens and the main cause of epidemic viral gastroenteritis globally. Due to a lack of in vitro culture systems and suitable animal models for HNV infection, murine norovirus (MNV) has become a common model. A recent study showed that MNV activates NLRP3 inflammasome leading to pyroptosis. Jatrorrhizine (JAT) is a natural isoquinoline alkaloid isolated from Coptis Chinensis, which has been proven to have antibacterial, anti-inflammatory, and antitumor effects. However, whether JAT has an effect on norovirus gastroenteritis and the underlying molecular mechanism remain unclear. Here, we found that JAT could ameliorate NLRP3-N-GSDMD-dependent pyroptosis induced by MNV infection through inhibiting the MAPKs/NF-κB signaling pathways and decrease MNV replication in RAW264.7 macrophages, suggesting that JAT has the potential to be a therapeutic agent for treating norovirus gastroenteritis.

    Abstract: Sintering accounts for about 50% of the total NOx emissions of the iron and steel industry. NOx emissions from the sintering process can be simulated using the emissions from coke combustion. However, the generation and emission law for NOx burning in the sintering process of pulverized coal is still not clear. The formation characteristics of NOx during coal combustion catalyzed by iron ore fines and several iron-containing pure minerals were studied in this paper. The results showed that iron ore fines can improve the NOx emission rate and increase the total NOx emissions during coal combustion. The type and composition of the iron ore fines have an important impact on the generation and emission of NOx in the process of coal combustion. The peak concentration and emissions of NOx in coal combustion flue gas with limonite, hematite or specularite added increased significantly. The peak value for the NOx concentration in the coal combustion flue gas with magnetite or siderite added increased, but the emissions decreased. Therefore, the generation of NOx in the sintering process can to a certain extent be controlled by adjusting the type of iron-containing raw materials and the distribution of the iron-containing raw materials and coal.

    Abstract: The friction and wear behaviors of Fe-19Cr-15Mn-0.66N steel were investigated under applied loads of 5 N and 15 N at the wear-testing temperatures of 300 °C and 500 °C using a ball-on-disc tribometer. The wear tracks were evaluated by scanning electron microscopy (SEM) and laser scanning confocal microscopy (LSCM) to reveal the variation in morphologies. Energy-dispersive X-ray spectroscopy (EDS) and X-ray photoelectron spectroscopy (XPS) were used to determine the components of oxide layers formed on wear surfaces. The results demonstrated that the oxide layers are favorable for obtaining a low friction coefficient under all conditions. The average friction coefficient decreased with increasing load at 300 °C, while it increased with the increase in applied load at 500 °C. At 300 °C, severe abrasive wear characterized by grooves resulted in a high friction coefficient with 5 N applied, whereas the formation of a denser oxide layer consisting of Cr2O3, FeCr2O4, Fe2O3, etc., and the increased hardness caused by work hardening led to a decrease in friction characterized by mild adhesive wear. At 500 °C, the transformation of Fe2O3 to the relatively softer Fe3O4 and the high production of lubricating Mn2O3 resulted in a minimum average friction coefficient (0.34) when 5 N was applied. However, the softening caused by high temperature weakened the hardening effect, and thus the friction coefficient increased with 15 N applied at 500 °C.

    Abstract: Micro RNAs are regulatory factors in tissue development, organ formation, cell growth, apoptosis and other biological processes. In particular, several miRNAs are related to the development of hair follicles. Here, we investigated the effect of the targeting of PIK3R3 by miR-27a on the AKT/MTOR pathway and on the proliferation and apoptosis of hair follicle stem cells (HFSCs) in sheep. Knockdown of the expression of PIK3R3 was found to significantly inhibit the proliferation and promote the apoptosis of HFSCs. Similarly, a miR-27a mimic significantly inhibited the proliferation and promoted the apoptosis of HFSCs. The miR-27a mimic was also shown to significantly inhibit the expression of PIK3R3, AKT, and MTOR and the phosphorylation of AKT and MTOR, while a miR-27a inhibitor increased the expression of these genes. The presence of an miR-27a binding site in the 3′ UTR of PIK3R3 was identified by a bioinformatics analysis, and the interaction was verified with a dual-luciferase reporter assay. The expression of PIK3R3 mRNA and protein was negatively correlated with the presence of miR-27a, which suggests that this interaction may be involved in the biological impacts on proliferation and apoptosis. Thus, this study demonstrates that miR-27a plays a potential role in the proliferation and apoptosis of sheep hair follicle stem cells by targeting PIK3R3, which can be used to design new methods to improve sheep wool.

    Abstract: There are complex pore structures in shale reservoirs, and the nonlinear flow characteristics of shale reservoirs are complicated by stress sensitivity and boundary layer effects. In this research, a new flow mathematical model was established based on capillary model, stress sensitivity and boundary layer effect. The model was verified by the experimental results of Jilin shale oil reservoir, and the influencing factors of flow characteristics were analyzed. The results show that the new shale oil flow model has good fitting and high reliability with the experimental results. The boundary layer effect and stress sensitivity are the main factors affecting the seepage characteristics. With the increase in the pressure gradient, the influence of the boundary layer effect on the flow is gradually enhanced, and the influence of the stress sensitivity on the flow is gradually weakened. At the same time, this study developed a method for the evaluation of effective displacement distance of shale oil reservoir with CO2 injection through the limited injection-production well spacing method, and obtained the nonlinear flow curve of displacement distance with reservoir permeability under different injection and production pressure difference. This research deeply studies the flow characteristics of shale oil reservoir and provides a theoretical basis for the enhanced oil recovery of shale oil with CO2 injection.
