    Abstract: Lint percentage is one of the most essential yield components and an important economic index for cotton planting. Improving lint percentage is an effective way to achieve high-yield in cotton breeding worldwide, especially upland cotton (Gossypium hirsutum L.). However, the genetic basis controlling lint percentage has not yet been systematically understood. Here, we performed a genome-wide association mapping for lint percentage using a natural population consisting of 189 G. hirsutum accessions (188 accessions of G. hirsutum races and one cultivar TM-1). The results showed that 274 single-nucleotide polymorphisms (SNPs) significantly associated with lint percentage were detected, and they were distributed on 24 chromosomes. Forty-five SNPs were detected at least by two models or at least in two environments, and their 5 Mb up- and downstream regions included 584 makers related to lint percentage identified in previous studies. In total, 11 out of 45 SNPs were detected at least in two environments, and their 550 Kb up- and downstream region contained 335 genes. Through RNA sequencing, gene annotation, qRT-PCR, protein–protein interaction analysis, the cis-elements of the promotor region, and related miRNA prediction, Gh_D12G0934 and Gh_A08G0526 were selected as key candidate genes for fiber initiation and elongation, respectively. These excavated SNPs and candidate genes could supplement marker and gene information for deciphering the genetic basis of lint percentage and facilitate high-yield breeding programs of G. hirsutum ultimately.

    Abstract: Understanding groundwater geochemistry is crucial for water supply in arid regions. The present research was conducted in the arid Mo river watershed on the Tibetan plateau to gain insights into the geochemical characteristics, governing processes and quality of groundwater in arid endorheic watersheds. A total of 28 groundwater samples were collected from the phreatic and confined aquifers for hydrochemical analysis. The results showed that the groundwater was slightly alkaline in all aquifers of the watershed. The phreatic groundwater samples (PGs) and confined groundwater samples (CGs) had the TDS value in the ranges of 609.19–56,715.34 mg/L and 811.86–2509.51 mg/L, respectively. PGs were salter than CGs, especially in the lower reaches. Both the PGs and CGs were dominated by the Cl-Na type, followed by the mixed Cl-Mg·Ca type. The toxic elements of NO2− (0.00–0.20 mg/L for PGs and 0.00–0.60 mg/L for CGs), NH4+ (0.00–0.02 mg/L for PGs and 0.00–0.02 mg/L for CGs) and F− (0.00–4.00 mg/L for PGs and 1.00–1.60 mg/L for CGs) exceeded the permissible limits of the Chinese guidelines at some sporadic sites. Water–rock interactions, including silicates weathering, mineral dissolution (halite and sulfates) and ion exchange, were the main contributions to the groundwater chemistry of all aquifers. The geochemistry of PGs in the lower reach was also greatly influenced by evaporation. Agricultural sulfate fertilizer input was responsible for the nitrogen pollutants and salinity of PGs. All CGs and 73.91% of PGs were within the Entropy-weighted water quality index (EWQI) of below 100 and were suitable for direct drinking purposes. Precisely 8.70 and 17.39% of PGs were within the EWQI value in the range of 100–150 (medium quality and suitable for domestic usage) and beyond 200 (extremely poor quality and not suitable for domestic usage), respectively. The electrical conductivity, sodium adsorption ratio, sodium percentage and permeability index indicated that groundwater in most parts of the watershed was suitable for irrigation, and only a small portion might cause salinity, sodium or permeability hazards. Groundwater with poor quality was mainly distributed in the lower reaches. CGs and PGs in the middle-upper reaches could be considered as the primary water resources for water supply. Agricultural pollution should be paid more attention to safeguard the quality of groundwater.

    Abstract: Verticillium dahliae (V. dahliae) infects roots and colonizes the vascular vessels of host plants, significantly reducing the economic yield of cotton and other crops. In this study, the protein VdTHI20, which is involved in the thiamine biosynthesis pathway, was characterized by knocking out the corresponding VdTHI20 gene in V. dahliae via Agrobacterium tumefaciens-mediated transformation (ATMT). The deletion of VdTHI20 resulted in several phenotypic defects in vegetative growth and conidiation and in impaired virulence in tobacco seedlings. We show that VdTHI20 increases the tolerance of V. dahliae to UV damage. The impaired vegetative growth of ΔVdTHI20 mutant strains was restored by complementation with a functional copy of the VdTHI20 gene or by supplementation with additional thiamine. Furthermore, the root infection and colonization of the ΔVdTHI20 mutant strains were suppressed, as indicated by green fluorescent protein (GFP)-labelling under microscope observation. When the RNAi constructs of VdTHI20 were used to transform Nicotiana benthamiana, the transgenic lines expressing dsVdTHI20 showed elevated resistance to V. dahliae. Together, these results suggest that VdTHI20 plays a significant role in the pathogenicity of V. dahliae. In addition, the pathogenesis-related gene VdTHI20 exhibits potential for controlling V. dahliae in important crops.

    Abstract: Urban traffic pollution, which is strongly influenced by the complex urban morphology, has posed a great threat to human health. In this study, we performed a high-resolution simulation of traffic pollution in a typical city block in Baoding, China, based on the Parallelized Large-eddy simulation Model (PALM), to examine the distribution patterns of traffic-related pollutants and explore their relationship with urban morphology. Based on the model results, we conducted a multi-linear regression (MLR) analysis and found that the distribution of air pollutants inside the city block was dominated by both traffic emissions and urban morphology, which explained about 70% of the total variance in spatial distribution of air pollutants. Excluding the contribution of emissions, over 50% of the total variance can still be explained by the urban morphology. Among these urban morphological factors, the key factors determining the spatial distribution of air pollution are “Distance from the road” (DR), “Building Coverage Ratio” (BCR) and “Aspect Ratio” (H/W) of the street canyon. Specifically, urban areas with lower Aspect Ratio, lower BCR and larger DR are less affected by traffic pollution. Compiling these individual factors, we developed a complex Urban Morphology Pollution Index (UMPI). Each unit increase in UMPI is associated with a one percent increase of nearby traffic pollution contribution. This index can help urban planners to semi-quantitatively evaluate building groups which tend to trap or ventilate traffic pollution and thus help to reduce human exposure to street canyon level pollution through either traffic emission control or urban morphology amelioration.

    Abstract: In the context of the global economic downturn caused by COVID-19, e-commerce has become the first choice for people to shop. Many merchants choose to launch promotion activities through some social media platforms. Price discounts can dramatically increase sales volume in social e-commerce due to the interaction of online consumers. It is urgent for e-commerce merchants to learn rules about discount information dissemination in social media, so as to formulate reasonable discount strategies and achieve sustainable business. This paper constructs an evolutionary game model for e-commerce platforms and merchants when they implement promotion strategies through social media, investigates discount information dissemination among consumers under multiple situations by introducing price discount parameters, and further discusses the influence mechanism of discount size and platform reputation on consumers’ purchase behavior. Results show that in low-reputation e-commerce platforms, the price discount is the main motivation to purchase. Consumers’ preference for the high discount is weakened by the increase in platforms’ reputation. Discounts should be set according to the different reputations. Businesses in a high-reputation environment are relatively more profitable. E-commerce businesses should work together, apply reasonable pricing, and improve their quality to create a green and healthy shopping environment, in order to get benefits and sustainable development.

    Abstract: This work aims to investigate the impact of antibiotics and extracellular antibiotic resistance genes (eARGs) on the dynamics of gastrointestinal antimicrobial resistance (AMR). The antibiotic resistance gene (ARG) levels of different segments of the gastrointestinal tract of mouse models were analyzed and compared after exposure to clinical concentrations of sulfadiazine and environmental levels of eARGs carried by the conjugative plasmid pR55. Exposure to sulfadiazine and eARGs led to significant changes in ARG levels by as many as four log-folds. Further analysis showed that the response of ARG levels appeared from 12–16 days after exposure and diminished 20 days after exposure. The responses in ARG levels were also restricted to different gastrointestinal segments for sulfadiazine and eARGs. Combined exposure of sulfadiazine and eARGs was unable to further increase ARG levels. From these findings, we concluded that the short-term consumption of environmental levels of eARGs and uptake of clinical levels of antibiotics lead to a spatially and temporally confined response in gastrointestinal AMR. These findings further clarify the detrimental impacts of antibiotic and eARG uptake, and the complexity of AMR development and dissemination dynamics in the gastrointestinal tract.

    Abstract: This study aimed to investigate the practical validity of the environmental DNA (eDNA) method for evaluating fish composition and diversity in different habitats. We evaluated the fish composition and diversity characteristics of seven different habitats in the Ma’an Archipelago Special Protected Area in April 2020. The results showed that a total of twenty-seven species of fishes belonging to six orders, eighteen families, and twenty-three genera of the Actinopterygii were detected in the marine waters of the Ma’an Archipelago Special Protected Area. The dominant species in each habitat were Larimichthys crocea, Paralichthys olivaceus, and Lateolabrax maculatus. The mussel culture area had the highest number of species, with 19 fish species, while the offshore bulk load shedding platform had the lowest number of species, with 12 fish species. The rest of the habitat was not significantly different. The results showed that the mussel culture area had the highest diversity index (average value of 2.352 ± 0.161), and the offshore bulk load shedding platform had the lowest diversity index (average value of 1.865 ± 0.127); the rest of the habitat diversity indices did not differ significantly. A comparison with historical surveys showed that the eDNA technique can detect species not collected by traditional methods such as gillnets and trawls. Our study demonstrates the role of eDNA technology in obtaining fish diversity in different habitats and provides a theoretical basis for the continuous monitoring and management of fish biodiversity in protected areas.

    Abstract: Verticillium wilt that is caused by Verticillium dahliae, does result in massive annual yield losses and fiber quality decline in cotton. Control by conventional mechanisms is not possible due to a wide host range and the longevity of dormant fungi in the soil in the case of absence of a suitable host. Plants have developed various mechanisms to boost their immunity against various diseases, and one is through the induction of various genes. In this research, we carried out RNA sequencing and then identified the members of the adenosine triphosphate (ATP)-binding cassette (ABC) proteins to be critical in enhancing resistance to V. dahliae infection. A total of 166 proteins that are encoded by the ABC genes were identified in Gossypium raimondii with varying physiochemical properties. A novel ABC gene, Gorai.007G244600 (ABCF5), was found to be highly upregulated, and its homolog in the tetraploid cotton Gh_D11G3432 (ABCF5), was then silenced through virus induced gene silencing (VIGS) in G. hirsutum, tetraploid upland cotton. The mutant cotton seedlings ability to tolerate V. dahliae infection was significantly reduced. Based on the evaluation of oxidant enzymes, hydrogen peroxide (H2O2) and malondialdehyde (MDA) showed significantly increased levels in the leaves of the mutant compared to the wild type. In addition, antioxidant enzymes, peroxidase (POD), catalase (CAT), and superoxide dismutase (SOD) concentrations were reduced in the mutant cotton leaves after treatment with V. dahliae fungi as compared to the wild type. Moreover, expression levels of the biotic stress genes, cotton polyamine oxidase (GhPAO), cotton ribosomal protein L18 (GhRPL18), and cotton polygalacturonase-inhibiting protein-1 (GhPGIP1), were all downregulated in the mutant but they were highly upregulated in the various tissues of the wild cotton seedlings. This research has shown that ABC genes could play an important role in enhancing the immunity of cotton to V. dahliae infection, and thus can be explored in developing more resilient cotton genotypes with improved resistance to V. dahliae infection in cotton.
