    Abstract: Protein post-translational modification (PTM) is an efficient biological mechanism to regulate protein structure and function, but its role in plant responses to heavy metal stress is poorly understood. The present study performed quantitative succinyl-proteome profiling using liquid chromatography–mass spectrometry analysis to explore the potential roles of lysine succinylation modification in turnip seedlings in response to cadmium (Cd) stress (20 μM) under hydroponic conditions over a short time period (0–8 h). A total of 547 succinylated sites on 256 proteins were identified in the shoots of turnip seedlings. These succinylated proteins participated in various biological processes (e.g., photosynthesis, tricarboxylic acid cycle, amino acid metabolism, and response to stimulation) that occurred in diverse cellular compartments according to the functional classification, subcellular localization, and protein interaction network analysis. Quantitative analysis showed that the intensities of nine succinylation sites on eight proteins were significantly altered (p 〈 0.05) in turnip shoots after 8 h of Cd stress. These differentially succinylated sites were highly conserved in Brassicaceae species and mostly located in the conserved domains of the proteins. Among them, a downregulated succinylation site (K150) in the glycolate oxidase protein (Gene0282600.1), an upregulated succinylation site (K396) in the catalase 3 protein (Gene0163880.1), and a downregulated succinylation site (K197) in the glutathione S-transferase protein (Gene0315380.1) may have contributed to the altered activity of the corresponding enzymes, which suggests that lysine succinylation affects the Cd detoxification process in turnip by regulating the H2O2 accumulation and glutathione metabolism. These results provide novel insights into understanding Cd response mechanisms in plants and important protein modification information for the molecular-assisted breeding of Brassica varieties with distinct Cd tolerance and accumulation capacities.

    Abstract: The triterpenes in bitter gourd (Momordica charantia) show a variety of medicinal activities. Oxidosqualene cyclase (OSC) plays an indispensable role in the formation of triterpene skeletons during triterpene biosynthesis. In this study, we identified nine genes encoding OSCs from bitter gourd (McOSC1–9). Analyses of their expression patterns in different tissues suggested that characteristic triterpenoids may be biosynthesized in different tissues and then transported. We constructed a hairy root system in which McOSC7 overexpression led to an increased accumulation of camaldulenic acid, enoxolone, and quinovic acid. Thus, the overexpression of McOSC7 increased the active components content in bitter gourd. Our data provide an important foundation for understanding the roles of McOSCs in triterpenoid synthesis.

    Abstract: Calcium-dependent protein kinases (CDPKs) are crucial calcium ions (Ca2+) sensors in plants with important roles in signal transduction, plant growth, development, and stress responses. Here, we identified 24 genes encoding CDPKs in Dendrobium officinale using genome-wide analysis. The phylogenetic analysis revealed that these genes formed four groups, with similar structures in the same group. The gene expression patterns following hormone treatments and yeast two-hybrid of homologous CDPK gene pairs with Rbohs showed differences, indicating functional divergence between homologous genes. In addition, the rapid accumulation of hydrogen peroxide (H2O2) and stomatal closure was observed in response to salicylic acid (SA)/jasmonic acid (JA) stress. Our data showed that CDPK9-2 and CDPK20-4 interacted with Rboh D and Rboh H, respectively, and were implicated in the generation of H2O2 and regulation of the stomatal aperture in response to salicylic acid/jasmonic acid treatment. We believe these results can provide a foundation for the functional divergence of homologous genes in D. officinale.

    Abstract: Land use decision-making is a vital livelihood strategy associated with the rational collocation of livelihood asset endowments by rural households. Based on the perspective of livelihood heterogeneity, this paper collected the data from 540 farm households in 27 villages in three Sichuan Province counties to identify the land use decision-making characteristics of the household groups. A land use decision-making framework (LUDF) based on the sustainable livelihood framework (SLF) was established and dynamic and dual indicators were developed to divide the sample into six household groups. The household livelihood capital, livelihood strategies, and livelihood diversity were then analyzed at the regional and household group level, and the land use decisions of these household groups were explored, from which the following was found. (1) Overall livelihood capital in the study area was low, and except for human assets, there were few other assets, with households in the survey areas being more inclined to engage in non-farming livelihood activities; however, households in Nanjiang and Qionglai had greater livelihood activities choices than Luxian. (2) The LL-type household was the chief household group; the household group distribution in Qionglai was well-proportioned but uneven in Nanjiang and Luxian; and the HL-type, ML-type, and LL-type household livelihood strategy choices were polarized. (3) Most households were involved in land self-cultivation, followed by land transfer-in, land transfer-out, and land abandonment households. Specifically, there were more households that cultivated fragmented landholdings than specialized households with large-scale landholdings, the land transfer rate was relatively low, the transfer-in land area was far greater than the transfer-out land areas, and a small number of households that had abandoned their land were still involved in agricultural production. (4) There were obvious discordant human–land relationships and inefficient land uses in the study area. Based on these findings, relevant policy recommendations are given to improve farm household livelihood capital, optimize livelihood strategies, and assist in land use decision-making.
