    Abstract: The intramuscular fat (IMF), or so-called marbling, is known as potential determinant of the high quality beef in China, Korea, and Japan. Of the methods that affect IMF content in cattle, castration is markedly regarded as an effective and economical way to improve the deposition of IMF but with little attention to its multi-omics in early-castrated cattle. The aim of this study was to investigate the liver transcriptome and metabolome of early-castrated Holstein cattle and conduct a comprehensive analysis of two omics associated with the IMF deposition using transcriptomics and untargeted metabolomics under different treatments: non−castrated and slaughtered at 16 months of age (GL16), castrated at birth and slaughtered at 16 months of age (YL16), and castrated at birth and slaughtered at 26 months of age (YL26). The untargeted metabolome was analyzed using ultrahigh-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry. The transcriptome of the hepatic genes was analyzed to identify marbling-related genes. Using untargeted metabolomics, the main altered metabolic pathways in the liver of cattle, including those for lipid and amino acid metabolism, were detected in the YL16 group relative to the GL16 and YL26 groups. Significant increases in the presence of betaine, alanine, and glycerol 3-phosphate were observed in the YL16 group (p 〈 0.05), which might have contributed to the improved beef-marbling production. Compared to the GL16 and YL26 groups, significant increases in the presence of glutathione, acetylcarnitine, and riboflavin but decreases in diethanolamine and 2-hydroxyglutarate were identified in YL16 group (p 〈 0.05), which might have been beneficial to the beef’s enhanced functional quality. The gene expressions of GLI1 and NUF2 were downregulated and that of CYP3A4 was upregulated in the YL16 group; these results were strongly correlated with the alanine, betaine, and leucine, respectively, in the liver of the cattle. In conclusion, implementation of early castration modified the hepatic metabolites and the related biological pathways by regulating the relevant gene expressions, which could represent a better rearing method for production of high marbled and healthier beef products.

    Abstract: Forecasting the prices of hogs has always been a popular field of research. Such information has played an essential role in decision-making for farmers, consumers, corporations, and governments. It is hard to predict hog prices because too many factors can influence them. Some of the factors are easy to quantify, but some are not. Capturing the characteristics behind the price data is also tricky considering their non-linear and non-stationary nature. To address these difficulties, we propose Heterogeneous Graph-enhanced LSTM (HGLTSM), which is a method that predicts weekly hog price. In this paper, we first extract the historical prices of necessary agricultural products in recent years. Then, we utilize discussions from the online professional community to build heterogeneous graphs. These graphs have rich information of both discussions and the engaged users. Finally, we construct HGLSTM to make the prediction. The experimental results demonstrate that forum discussions are beneficial to hog price prediction. Moreover, our method exhibits a better performance than existing methods.

    Abstract: (1) Background: Huperzine A, a natural cholinesterase (AChE) inhibitor isolated from the Chinese herb Huperzia Serrata, has been used as a dietary supplement in the United States and a drug in China for therapeutic intervention on Alzheimer’s disease (AD). This review aims to determine whether Huperzine A exerts disease-modifying activity through systematic analysis of preclinical studies on rodent AD models. (2) Methods: Sixteen preclinical studies were included based on specific criteria, and the methodological qualities were analyzed by SYRCLE’s risk of bias tool. Some outcomes were meta-analyzed: latencies and time spent in quadrant of Morris water maze, soluble amyloid-β (Aβ) level measured by ELISA in the cortex and hippocampus, Aβ plaque numbers measured by immunohistochemistry in hippocampus, choline acetyltransferase (ChAT) activity, and AChE activity. Finally, the mechanisms of Huperzine A on AD models were summarized. (3) Conclusions: The outcomes showed that Huperzine A displayed AChE inhibition, ChAT activity enhancement, memory improvement, and Aβ decreasing activity, indicating the disease-modifying effect of Huperzine A. However, due to the uneven methodological quality, the results need to be rationally viewed, and extensively repeated.

    Abstract: The cuticle is the outermost layer of plant aerial tissue that interacts with the environment and protects plants against water loss and various biotic and abiotic stresses. ADP ribosylation factor guanine nucleotide exchange factor proteins (ARF-GEFs) are key components of the vesicle trafficking system. Our study discovers that AtMIN7, an Arabidopsis ARF-GEF, is critical for cuticle formation and related leaf surface defense against the bacterial pathogen Pseudomonas syringae pathovar tomato (Pto). Our transmission electron microscopy and scanning electron microscopy studies indicate that the atmin7 mutant leaves have a thinner cuticular layer, defective stomata structure, and impaired cuticle ledge of stomata compared to the leaves of wild type plants. GC–MS analysis further revealed that the amount of cutin monomers was significantly reduced in atmin7 mutant plants. Furthermore, the exogenous application of either of three plant hormones—salicylic acid, jasmonic acid, or abscisic acid—enhanced the cuticle formation in atmin7 mutant leaves and the related defense responses to the bacterial Pto infection. Thus, transport of cutin-related components by AtMIN7 may contribute to its impact on cuticle formation and related defense function.

    Abstract: Plants evolve a prompt and robust immune system to defend themselves against pathogen infections. Pathogen-associated molecular pattern (PAMP)-triggered immunity (PTI) is the first battle layer activated upon the PAMP’s perception, which leads to multiple defense responses. The plasma membrane (PM) H+-ATPases are the primary ion pumps to create and maintain the cellular membrane potential that is critical for various essential biological processes, including plant growth, development, and defense. This study discovered that the PM H+-ATPase AHA5 is negatively involved in Arabidopsis PTI against the virulent pathogen Pseudomonas syringae pvr. tomato (Pto) DC3000 infection. The aha5 mutant plants caused the reduced stomata opening upon the Pto infection, which was associated with the salicylic acid (SA) pathway. In addition, the aha5 mutant plants caused the increased levels of callose deposition, defense-related gene expression, and SA accumulation. Our results also indicate that the PM H+-ATPase activity of AHA5 probably mediates the coupling of H2O2 generation and the apoplast alkalization in PTI responses. Moreover, AHA5 was found to interact with a vital defense regulator, RPM1-interacting protein 4 (RIN4), in vitro and in vivo, which might also be critical for its function in PTI. In summary, our studies show that AHA5 functions as a novel and critical component that is negatively involved in PTI by coordinating different defense responses during the Arabidopsis–Pto DC3000 interaction.

    Abstract: Microbial-induced calcium carbonate precipitation (MICP) has the potential to be an environmentally friendly technique alternative to traditional methods for sustainable coastal stabilization. This study used a non-pathogenic strain that exists in nature to experimentally investigate the application of the MICP technique on mitigating sandy beach erosion. First, the unconfined compressive strength (UCS) test was adopted to explore the consolidation performance of beach sand after the MICP treatment, and then model tests in a wave flume were conducted to investigate the MICP ability to mitigate beach erosion by plunger waves. This study also employed field emission scanning electron microscopy (FE-SEM) and energy-dispersive X-ray spectroscopy (EDS) to observe the crystal forms of MICP-treated sand after wave action. The results reveal that the natural beach sand could be consolidated by the MICP treatment, and the compressive strength increased with the increase in the cementation media concentration. In this study, the maximum compressive strength could be achieved was 517.3 kPa. The one-phase and two-phase MICP treatment strategies were compared of sandy beach erosion tests with various spray and injection methods on the beach surface. The research results indicate that the proper MICP treatment could mitigate beach erosion under various wave conditions; the use of MICP reduced beach erosion up to 33.9% of the maximum scour depth.

    Abstract: Severe fever with thrombocytopenia syndrome virus (SFTSV) is an emerging tick-borne bunyavirus that causes severe disease in humans with case-fatality rates of up to 30%. There are currently very limited treatment options for SFTSV infection. We conducted a drug repurposing program by establishing a two-tier test system to rapidly screen a Food and Drug Administration- (FDA)-approved drug library for drug compounds with anti-SFTSV activity in vitro. We identified five drug compounds that inhibited SFTSV replication at low micromolar concentrations, including hexachlorophene, triclosan, regorafenib, eltrombopag, and broxyquinoline. Among them, hexachlorophene was the most potent with an IC50 of 1.3 ± 0.3 µM and a selectivity index of 18.7. Mechanistic studies suggested that hexachlorophene was a virus entry inhibitor, which impaired SFTSV entry into host cells by interfering with cell membrane fusion. Molecular docking analysis predicted that the binding of hexachlorophene with the hydrophobic pocket between domain I and domain III of the SFTSV Gc glycoprotein was highly stable. The novel antiviral activity and mechanism of hexachlorophene in this study would facilitate the use of hexachlorophene as a lead compound to develop more entry inhibitors with higher anti-SFTSV potency and lower toxicity.

    Abstract: Phenotypic differentiation of Abies alba was identified among selected provenances originating from the northern edge of this species’ natural geographic range. We posited that although the seed sources of the study populations were not geographically distant, progenies would differ with respect to growth, needle pigment concentration, and values of photochemical parameters in response to contrasting light conditions. Potted seedlings of six provenances were grown for two seasons under low light (LL, 40% of full irradiance) or high light (HL, 100% of full irradiance). The results showed that the provenances differed in diameter at root collar (DRC) and in relative growth rates of height (RGR). Seedlings grown in full light had a greater mean value of DRC than those in LL. LL increased total needle chlorophyll (Chltot) and carotenoid concentrations compared with HL, and Chltot was modified by the provenance of the seedlings. The provenance “Syców”, localized at the northern limit of the species range, showed the lowest value of maximal electron transfer rate (ETRmax) and lower values of growth parameters compared with the other study provenances. Relative growth rate, Chltot, and non-photochemical quenching of fluorescence (NPQ) were correlated with some climatic characteristics of seed stands and formed clusters indicating the local adaptation of the study populations. These results provide evidence of low intraspecific A. alba differentiation in terms of growth dynamics, photochemical capacity, and tolerance to different light regimes. The intraspecific diversity found in our study among the northern lead populations could be, however, important for the natural expansion of this species further north and for the selection of provenances to be used in the programme of the A. alba restoration in the Sudety Mountains, southern Poland, where decline of this species has been observed.

    Abstract: After the General Data Protection Regulation (GDPR) was introduced, some organizations and big data companies shared data without conducting any privacy protection and compliance authentication, which endangered user data security, and were punished financially for this reason. This study proposes a blockchain-based GDPR compliance data sharing scheme, aiming to promote compliance with regulations and provide a tool for interaction between users and service providers to achieve data security sharing. The zero-knowledge Succinct Non-Interactive Arguments of Knowledge (zk-SNARK) algorithm is adopted for protecting data and ensure that the user’s private data can satisfy the individual requirements of the service provider without exposing user data. The proposed scheme ensures mutual authentication through the Proof of Authority consensus based on the Committee Endorsement Mechanism (CEM-PoA), and prevents nodes from doing evil using the reputation incentive mechanism. Theoretical analysis and performance comparison indicate that the scheme meets the confidentiality, availability, and other indicators. It has superiority in efficiency and privacy protection compared with other schemes.

    Abstract: UM-164 is a dual inhibitor of c-Src and p38 MAPK, and has been a lead compound for targeting triple-negative breast cancer. UM-164 shows stronger binding to the active sites of Src compared with the conventional Src inhibitor Dasatinib. While Dasatinib has displayed some inhibitory effects on glioma growth in clinical trials, whether UM-164 can suppress glioma growth has not been reported. Here we show that UM-164 suppressed the proliferation, migration and spheroid formation of glioma cells, and induced cell cycle arrest in the G1 phase. Moreover, UM-164 triggered YAP translocation to the cytoplasm and reduced the activity of YAP, as evidenced by a luciferase assay. Accordingly, UM-164 markedly decreased the expression levels of YAP target genes CYR61 and AXL. Importantly, ectopic expression of wild-type YAP or YAP-5SA (YAP constitutively active mutant) could rescue the anti-proliferative effect induced by UM-164. Intriguingly, p38 MAPK appears to play a greater role than Src in UM-164-mediated inhibition of YAP activity. Furthermore, the in vitro anti-glioma effect mediated by UM-164 was confirmed in a xenograft glioma model. Together, these findings reveal a mechanism by which UM-164 suppresses the malignant phenotypes of glioma cells and might provide a rationale for UM-164-based anti-glioma clinical trials.
