    Abstract: Depression is a frequent and serious comorbidity associated with diabetes which adversely affects prognosis and quality of life. Glucagon-like peptide-1 receptor (GLP-1R) agonists, widely used in the treatment of diabetes, are reported to exert neuroprotective effects in the central nervous system. Thus, we aim to evaluate whether GLP-1R agonist exendin-4 (EX-4) could alleviate depression-like behaviors in diabetic mice and to explore its underlying mechanism. The antidepressant effects of EX-4 were evaluated using behavioral tests in db/db mice. The effects of EX-4 on microglial pyroptosis and neuroinflammation were assessed in N9 microglial cells. EX-4 administration alleviated depression-like behaviors in diabetic db/db mice. GLP-1R activation by EX-4 significantly suppressed microglial pyroptosis and neuroinflammation by downregulation of gasdermin D (GSDMD) and interleukin (IL)-1β in diabetic mice and lipopolysaccharide (LPS)-primed N9 microglia. Mechanistically, GLP-1R activation improved mitochondrial function and promoted mitophagy by decreasing the accumulation of mitochondrial reactive oxygen species (mtROS) and intracellular ROS production. EX-4 exhibits antidepressant effects in depression associated with diabetes in diabetic mice, which may be mediated by inhibiting microglial pyroptisis via promoting mitophagy. It is supposed that GLP-1R agonists may be a promising therapy in depression associated with diabetes.

    Abstract: Si3N4 ceramics with a microscale rice leaf structure (MRLS) and titanium alloy were connected via brazing, and the influence of the surface microstructure on the ceramic connection was analyzed. MRLS fabrication is an efficient and high-degree-of-freedom method that can be used to change a material’s surface morphology and wettability. The MRLS was obtained at a laser power of 110 W, with line spacings of 100 and 50 μm. The laser-treated surface included nanoparticles and micro particles, exhibiting a coral-like structure after agglomeration. When the MRLS was used to braze the titanium alloy, no defects were observed at the brazing interface, and the formation was excellent. Throughout the brazed joint, the MRLS remained intact and formed a strong metallurgical bond with the brazing filler metal. A finite element analysis was performed to study the cross-sectional morphology after joint fracture; from the load-time curve, it was found that the MRLS on the surface not only helped improve the mechanical occlusion and brazing area at the interface, but also helped generate compressive stress on the Si3N4 side. Crack propagation was hindered, thereby increasing the joint strength.

    Abstract: Background: Luteolin is widely distributed among a number of vegetal species worldwide. The pharmacological effects of luteolin are diverse and amongst antioxidant, free radical scavenging, and anti-inflammatory activities. Preliminary study showed that luteolin can ameliorate hypertension. However, the precise mechanism needs further investigation. There is no evidence that luteolin affects the paraventricular nucleus of the hypothalamus (PVN), a brain nucleus associated with a critical neural regulator of blood pressure. Our main aim was to explore the effect of luteolin on the PI3K/Akt/NF-κB signaling pathway within the PVN of hypertensive rats. Methods: spontaneously hypertensive rats (SHRs) and corresponding normotensive control rats, the Wistar Kyoto (WKY) rats were divided into four groups and subsequently treated for 4 weeks with bilateral PVN injections of either luteolin (20 µg/0.11 µL, volume: 0.11 µL/h) or vehicle (artificial cerebrospinal fluid). Results: luteolin infusion to the PVN significantly decreased some hemodynamic parameters including the mean arterial pressure (MAP), heart rate (HR), circulating plasma norepinephrine (NE) and epinephrine (EPI). Additionally, there was a decrease in the expressions of the phosphatidylinositol 3-kinase (p-PI3K) and phosphorylated protein kinase-B (p-AKT), levels of reactive oxygen species (ROS), NAD(P)H oxidase subunit (NOX2, NOX4) in the PVN of SHRs. Meanwhile, the expression of inflammatory cytokines and the activity of nuclear factor κB (NF-κB) p65 in the PVN of SHRs were lowered. Furthermore, immunofluorescence results showed that injection of luteolin in the PVN reduced the expression of tyrosine hydroxylase (TH), and increased that of superoxide dismutase (SOD1) and the 67-kDa isoform of glutamate decarboxylase (GAD67) in the PVN of SHRs. Conclusion: Our novel findings revealed that luteolin lowered hypertension via inhibiting NF-κB-mediated inflammation and PI3K/Akt signaling pathway in the PVN.

    Abstract: If the layer of soil surrounding a pile is not taken into account during the engineering detection process, the velocity-time curve might show asymptotic diameter shrinkage or diameter expanding features, which would alter the interpretation of the test findings. In this study, we suggest combining multi-feature extraction and a convolutional neural network (CNN) to increase accuracy in pile defect recognition for layered soil conditions and traditional deep learning flaws. First, numerical simulations are run to create velocity–time curves for foundation piles under layered soil conditions. Then, the data are extracted from three dimensions: time domain, frequency domain, and time-frequency domain, respectively, and fused into a set of feature vectors. Finally, a foundation pile defect identification model combining multi-scale features and CNN is established. The findings demonstrate that the CNN model has 97.8% accuracy while the PNN has 28.6% accuracy, demonstrating that the approach is very reliable.

    Abstract: Thermal interface materials with high thermal conductivity and low hardness are crucial to the heat dissipation of high-power electronics. In this study, a high magnetic field was used to align the milled carbon fibers (CFs, 150 μm) in silicone rubber matrix to fabricate thermal interface materials with an ordered and discontinuous structure. The relationship among the magnetic field density, the alignment degree of CFs, and the properties of the resulting composites was explored by experimental study and theoretical analysis. The results showed higher alignment degree and enhanced thermal conductivity of composites under increased magnetic flux density within a certain curing time. When the magnetic flux density increased to 9 T, the CFs showed perfect alignment and the composite showed a high thermal conductivity of 11.76 W/(m·K) with only 20 vol% CF loading, owing to the ordered structure. Meanwhile, due to the low filler loading and discontinuous structure, a low hardness of 60~70 (shore 00) was also realized. Their thermal management performance was further confirmed in a test system, revealing promising applications for magnetic aligned CF–rubber composites in thermal interface materials.

    Abstract: This research aimed to investigate the compensation mechanism of iodine deficiency and excess in the mammary gland during lactation. Female rats were divided into the low iodine group (LI), the normal iodine group (NI), the 10-fold high iodine group (10HI) and the 50-fold high iodine group (50HI). We measured the iodine levels in the urine, blood, milk, and mammary gland. The protein expression of sodium/iodide symporter (NIS), DPAGT1, and valosin-containing protein (VCP) in the mammary gland was also studied. The 24-hour urinary iodine concentration, serum total iodine concentration, serum non-protein-bound iodine concentration, breast milk iodine concentration, and mammary gland iodine content in the 50HI group were significantly higher than those in the NI group (p 〈 0.05). Compared with the NI group, NIS expression in the 50HI group significantly decreased (p 〈 0.05). DAPGT1 expression was significantly higher in the LI group than in the NI group (p 〈 0.05). The expression level of VCP was significantly increased in the 10HI and 50HI groups. In conclusion, milk iodine concentration is positively correlated with iodine intake, and the lactating mammary gland regulates the glycosylation and degradation of NIS by regulating DPAGT1 and VCP, thus regulating milk iodine level. However, the mammary gland has a limited role in compensating for iodine deficiency and excess.

    Abstract: Macrophages play an important role in the progression of sporadic acute type A aortic dissection (ATAAD). The aim of this study was to characterize the cellular heterogeneity of macrophages in ATAAD tissues by scRNA-seq. Ascending aortic wall tissue from six ATAAD patients and three heart transplant donors was assessed by scRNA-seq and then analyzed and validated by various bioinformatic algorithms and histopathology experiments. The results revealed that the proportion of macrophages in ATAAD tissues (24.51%) was significantly higher than that in normal tissues (13.69%). Among the six macrophage subclusters, pro-inflammatory macrophages accounted for 14.96% of macrophages in the AD group and 0.18% in the normal group. Chemokine- and inflammation-related genes (CCL2, CCL20, S100A8, and S100A9) were expressed more intensively in macrophages in ATAAD tissue than in those in normal tissue. Additionally, intercellular communication analysis and transcription factor analysis indicated the activation of inflammation and degradation of the extracellular matrix in ATAAD tissue. Finally, immunohistochemistry, immunofluorescence, and Western blot experiments confirmed the overexpression of macrophage marker genes (CD68 and CD163) and matrix metalloproteinases (MMP9 and MMP2) in ATAAD tissue. Collectively, our study provides a preliminary evaluation of the role of macrophages in ATAAD, and the results could aid in the development of therapeutic options in the future.

    Abstract: Long noncoding RNAs (lncRNAs) are transcripts with lengths of more than 200 nt and limited protein-coding potential. They were found to play important roles in plant stress responses. In this study, the maize drought-tolerant inbred line AC7643 and drought-sensitive inbred line AC7729/TZSRW, as well as their recombinant inbred lines (RILs) were selected to identify drought-responsive lncRNAs in roots. Compared with non-responsive lncRNAs, drought-responsive lncRNAs had different sequence characteristics in length of genes and number of exons. The ratio of down-regulated lncRNAs induced by drought was significantly higher than that of coding genes; and lncRNAs were more widespread expressed in recombination sites in the RILs. Additionally, by integration of the modifications of DNA 5-methylcytidine (5mC), histones, and RNA N6-methyladenosine (m6A), it was found that the enrichment of histone modifications associated with transcriptional activation in the genes generated lncRNAs was lower that coding genes. The lncRNAs-mRNAs co-expression network, containing 15,340 coding genes and 953 lncRNAs, was constructed to investigate the molecular functions of lncRNAs. There are 13 modules found to be associated with survival rate under drought. We found nine SNPs located in lncRNAs among the modules associated with plant survival under drought. In conclusion, we revealed the characteristics of lncRNAs responding to drought in maize roots based on multiomics studies. These findings enrich our understanding of lncRNAs under drought and shed light on the complex regulatory networks that are orchestrated by the noncoding RNAs in response to drought stress.

    Abstract: The catalytic conversion of CO2 to C2 products through the CO2 reduction reaction (CO2RR) offers the possibility of preparing carbon-based fuels and valuable chemicals in a sustainable way. Herein, various Fen and Co5 clusters are designed to screen out the good catalysts with reasonable stability, as well as high activity and selectivity for either C2H4 or CH3CH2OH generation through density functional theory (DFT) calculations. The binding energy and cohesive energy calculations show that both Fe5 and Co5 clusters can adsorb stably on the N-doped carbon (NC) with one metal atom anchored at the center of the defected hole via a classical MN4 structure. The proposed reaction pathway demonstrates that the Fe5-NC cluster has better activity than Co5-NC, since the carbon–carbon coupling reaction is the potential determining step (PDS), and the free energy change is 0.22 eV lower in the Fe5-NC cluster than that in Co5-NC. However, Co5-NC shows a better selectivity towards C2H4 since the hydrogenation of CH2CHO to CH3CHO becomes the PDS, and the free energy change is 1.08 eV, which is 0.07 eV higher than that in the C-C coupling step. The larger discrepancy of d band center and density of states (DOS) between the topmost Fe and sub-layer Fe may account for the lower free energy change in the C-C coupling reaction. Our theoretical insights propose an explicit indication for designing new catalysts based on the transition metal (TM) clusters supported on N-doped carbon for multi-hydrocarbon synthesis through systematically analyzing the stability of the metal clusters, the electronic structure of the critical intermediates and the energy profiles during the CO2RR.

    Abstract: Background: Extensive knowledge of allergic multimorbidities is required to improve the management of allergic diseases with the industrialization of China. However, the demography and allergen distribution patterns of allergic multimorbidities in China remain unclear, despite the increasing prevalence of allergies. Methods: This was a real-world, cross-sectional study of 1273 outpatients diagnosed with one or more allergic diseases in Guangzhou, the most populated city of southern China, with leading industrial and commercial centers, between April 2021 and March 2022. Seven allergic diseases (allergic rhinitis (AR), asthma (AS)/cough variant asthma (CVA), atopic dermatitis (AD)/eczema, food allergy (FA), allergic conjunctivitis (AC), drug allergy (DA), and anaphylaxis) were assessed. Positive rates of sensitization to different allergens were measured using an allergen detection system of the UniCAP (Pharmacia Diagnostics, Sweden) instrument platform to compare the groups of allergic multimorbidities against a single entity. Results: There were 659 (51.8%) males and 614 (48.2%) females aged from 4 months to 74 years included in the analysis. The study participants who were diagnosed with allergic diseases had an average of 1.6 diagnoses. Overall, 46.5% (592 of 1273) of the patients had more than one allergic condition, and allergic rhinitis was the most common type of multimorbidity. Women were more likely to suffer from an allergic disease alone, whereas allergic multimorbidities were more likely to be diagnosed in men (p = 0.005). In addition, allergic multimorbidities were common in all age groups, with an incidence ranging from 37.1% to 57.4%, in which children and adolescents were more frequently diagnosed with allergic multimorbidities than adults (18–60 years old) (all p 〈 0.05). Allergic multimorbidity was observed throughout the year. A difference in the positive rate of allergens sensitization and total immunoglobulin E (tIgE) levels between different allergic multimorbidities was observed. Conclusions: Allergic multimorbidities were very commonly found in nearly half of all patients with allergies. The proportion of allergic multimorbidities varied with the type of disease, sex, age, and allergen distribution pattern. These findings may help clinicians to develop “One health” strategies for the clinical management of allergic diseases.
