    Abstract: This work studied the regulation of hole concentration and mobility in p-InGaN layers grown by metalorganic chemical vapor deposition (MOCVD) under an N-rich environment. By adjusting the growth temperature, the hole concentration can be controlled between 6 × 1017/cm3 and 3 × 1019/cm3 with adjustable hole mobility from 3 to 16 cm2/V.s. These p-InGaN layers can meet different requirements of devices for hole concentration and mobility. First-principles defect calculations indicate that the p-type doping of InGaN at the N-rich limiting condition mainly originated from Mg substituting In (MgIn). In contrast with the compensation of nitrogen vacancy in p-type InGaN grown in a Ga-rich environment, the holes in p-type InGaN grown in an N-rich environment were mainly compensated by interstitial Mg (Mgi), which has very low formation energy.

    Abstract: A water-soluble acacetin prodrug has been synthesized and reported by our group previously. Acetaminophen (APAP) overdose is a leading cause of acute liver injury. We found that subcutaneous injection of acacetin prodrug (5, 10, 20 mg/kg) decreased serum ALT, AST, and ALP, corrected the abnormal MDA and GSH in liver, and improved intrahepatic hemorrhage and destruction of liver structures in APAP (300 mg/kg)-treated mice. Molecular mechanism analysis revealed that the expressions of endoplasmic reticulum (ER) stress markers ATF6, CHOP, and p-PERK, apoptosis-related protein BAX, and cleaved caspase 3 were decreased by acacetin in a dose-dependent manner in vivo and in vitro. Moreover, via the acacetin-upregulated peroxisome-proliferator-activated receptor gamma (PPARγ) of HepG2 cells and liver, the suppressive effect of acacetin on ER stress and apoptosis was abolished by PPARγ inhibitor (GW9662) or PPARγ-siRNA. Molecular docking revealed that acacetin can bind to three active pockets of PPARγ, mainly by hydrogen bond. Our results provide novel evidence that acacetin prodrug exhibits significant protective effect against APAP-induced liver injury by targeting PPARγ, thereby suppressing ER stress and hepatocyte apoptosis. Acacetin prodrug is likely a promising new drug candidate for treating patients with acute liver injury induced by APAP.

    Abstract: Melittin is a membrane-active peptide with strong anticancer activity against various cancers. Despite decades of research, the role of the singular Trp in the anticancer activity and selectivity of melittin remains poorly understood. Here, we propose a theranostic solution based on the substitution of Trp19 with a noncanonical fluorescent amino acid (DapAMCA). The introduction of DapAMCA residue in melittin stabilized the helical structure of the peptide, as evaluated by circular dichroism spectra and molecular dynamics simulations. In vitro hemolytic and anticancer activity assays revealed that introducing DapAMCA residue in melittin changed its mode of action with the cell membrane, resulting in reduced hemolytic toxicity and an improved the selectivity index (SI), with up to a five-fold increase compared to melittin. In vitro fluorescence imaging of DapAMCA-labeled melittin (MELFL) in cancer cells demonstrated high membrane-penetrating activity, with strong nuclear and nucleolar localization ability. These findings provide implications for novel anticancer therapies based on Trp-substituted designs and nuclear/nucleolar targeted therapy.

    Abstract: In the present study, a pyridoxal-5′-phosphate (PLP)-dependent L-aspartate-α-decarboxylase from Tribolium castaneum (TcPanD) was selected for protein engineering to efficiently produce β-alanine. A mutant TcPanD-R98H/K305S with a 2.45-fold higher activity than the wide type was selected through error-prone PCR, site-saturation mutagenesis, and 96-well plate screening technologies. The characterization of purified enzyme TcPanD-R98H/K305S showed that the optimal cofactor PLP concentration, temperature, and pH were 0.04% (m/v), 50 °C, and 7.0, respectively. The 1mM of Na+, Ni2+, Co2+, K+, and Ca2+ stimulated the activity of TcPanD-R98H/K305S, while only 5 mM of Ni2+ and Na+ could increase its activity. The kinetic analysis indicated that TcPanD-R98H/K305S had a higher substrate affinity and enzymatic reaction rate than the wild enzyme. A total of 267 g/L substrate l-aspartic acid was consumed and 170.5 g/L of β-alanine with a molar conversion of 95.5% was obtained under the optimal condition and 5-L reactor fermentation.

    Abstract: Hyperthyroidism is common and can induce cardiomyopathy, but there is no effective therapeutic strategy. The purpose of this study was to investigate the molecular mechanism of hyperthyroidism-induced cardiomyopathy (HTC) and the effect of N-acetylcysteine (NAC), an ROS inhibitor, on the pathophysiology of HTC in vivo and in vitro. Compared with those in the control groups in vivo and in vitro, TT3 and TT4 were significantly increased, the structure of myocardial cells was enlarged and disordered, and interstitial fibrosis and the apoptosis of myocardial cells were markedly increased in the L-Thy group. The ROS and inflammatory response were increased in the hyperthyroidism group. In the NAC group, the contents of TT3 and TT4 were decreased, the myocardial cell structure was slightly disturbed, fibrosis and apoptosis were significantly reduced, and the ROS level and inflammatory response were significantly reduced. Interestingly, L-Thy decreased the viability of fibroblasts and H9c2 cells, suggesting that L-Thy-induced fibrosis was not caused by the proliferation of fibroblasts. The molecular mechanism of HTC could be explained by the fact that L-Thy could cause cardiac hypertrophy, inflammation, and fibrosis by regulating the Ca2+/calpain/Rcan1-dependent signalling pathway, the Ca2+/Rcan1/NF-κB/p65-dependent signalling pathway, and the Ca2+/ROS/Bcl-2/caspase-3-dependent signalling pathway. In conclusion, NAC can alleviate the pathophysiology of hyperthyroidism-induced cardiomyopathy, probably by regulating the ROS/Ca2+-dependent pathway.

    Abstract: This paper presents a comprehensive review of energy management control strategies utilized in hybrid electric vehicles (HEVs). These can be categorized as rule-based strategies and optimization-based strategies. Rule-based strategies, as the most basic strategy, are widely used due to their simplicity and practical application. The focus of rule-based strategies is to determine and optimize the optimal threshold for mode switching; however, they fall into a local optimal solutions. To have better performance in energy management, optimization-based strategies were developed. The categories of the existing optimization-based strategies are identified from the latest literature, and a brief study of each strategy is discussed, which consists of the main research ideas, the research focus, advantages, disadvantages and improvements to ameliorate optimality and real-time performance. Deterministic dynamic programming strategy is regarded as a benchmark. Based on neural network and the large data processing technology, data-driven strategies are put forward due to their approximate optimality and high computational efficiency. Finally, the comprehensive performance of each control strategy is analyzed with respect to five aspects. This paper not only provides a comprehensive analysis of energy management control strategies for HEVs, but also presents the emphasis in the future.

    Abstract: Over decades, fiber-optic temperature sensors based on conventional single-mode fibers (SMF) have been demonstrated with either high linearity and stability in a limited temperature region or poor linearity and thermal hysteresis in a high-temperature measurement range. For high-temperature measurements, isothermal annealing is typically necessary for the fiber-optic sensors, aiming at releasing the residual stress, eliminating the thermal hysteresis and, thus, improving the high-temperature measurement linearity and stability. In this article, an annealing-free fiber-optic high-temperature (1100 °C) sensor based on a diaphragm-free hollow-core fiber (HCF) Fabry-Perot interferometer (FPI) is proposed and experimentally demonstrated. The proposed sensor exhibits an excellent thermal stability and linearity (R2 〉 0.99 in a 100–1100 °C range) without the need for high-temperature annealing. The proposed sensor is extremely simple in preparation, and the annealing-free property can reduce the cost of sensor production significantly, which is promising in mass production and industry applications.

    Abstract: The Bowman‒Birk inhibitor (BBI), a protease inhibitor derived from soybeans, has been extensively studied in anti-tumor and anti-inflammation research. We recently reported that BBI has an anti-HIV-1 property in primary human macrophages. Because HSV-2 infection plays a role in facilitating HIV-1 sexual transmission, we thus examined whether BBI has the ability to inhibit HSV-2 infection. We demonstrated that BBI could potently inhibit HSV-2 replication in human cervical epithelial cells (End1/E6E7). This BBI-mediated HSV-2 inhibition was partially through blocking HSV-2-mediated activation of NF-κB and p38 MAPK pathways. In addition, BBI could activate the JAK/STAT pathway and enhance the expression of several antiviral interferon-stimulated genes (ISGs). Furthermore, BBI treatment of End1/E6E7 cells upregulated the expression of tight junction proteins and reduced HSV-2-mediated cellular ubiquitinated proteins’ degradation through suppressing the ubiquitin‒proteasome system. These observations indicate that BBI may have therapeutic potential for the prevention and treatment of HSV-2 infections.

    Abstract: Antimicrobial-resistant bacteria causing intractable and even fatal infections are a major health concern. Resistant bacteria residing in the intestinal tract of healthy individuals present a silent threat because of frequent transmission via conjugation and transposition. Plasmids harboring quinolone resistance genes are increasingly detected in clinical isolates worldwide. Here, we investigated the molecular epidemiology of plasmid-mediated quinolone resistance (PMQR) in Gram-negative bacteria from healthy service trade workers. From 157 rectal swab samples, 125 ciprofloxacin-resistant strains, including 112 Escherichia coli, 10 Klebsiella pneumoniae, two Proteus mirabilis, and one Citrobacter braakii, were isolated. Multiplex PCR screening identified 39 strains harboring the PMQR genes (including 17 qnr,19 aac(6′)-Ib-cr, and 22 oqxA/oqxB). The genome and plasmid sequences of 39 and 31 strains, respectively, were obtained by short- and long-read sequencing. PMQR genes mainly resided in the IncFIB, IncFII, and IncR plasmids, and coexisted with 3–11 other resistance genes. The high PMQR gene carriage rate among Gram-negative bacteria isolated from healthy individuals suggests the high-frequency transmission of these genes via plasmids, along with other resistance genes. Thus, healthy individuals may spread antibiotic-resistant bacterial, highlighting the need for improved monitoring and control of the spread of antibiotic-resistant bacteria and genes in healthy individuals.

    Abstract: Bentonite is frequently utilized as a landfill lining material due to its high impermeability. Due to the fact that heavy metal ions in leachate can alter the permeability of bentonite liner, the impermeability and metal adsorption effect of bentonite liner is typically enhanced by the use of external admixtures. In this investigation, zeolite was combined with stone chips and bentonite. Using a flexible wall permeation test, zeta potential test, and X-ray diffraction test, the effect of zeolite on the permeability and adsorption properties of the mixture was investigated. The results indicate that the addition of zeolite can enhance the impermeability of the mixed soil. The permeability coefficient of the mixed soil in DIW is 3.74 × 10−7 cm/s when bentonite is incorporated at 11% and decreases to 6.55 × 10−8, 4.65 × 10−8, and 5.10 × 10−8 cm/s when 12.50%, 25%, and 50% of zeolite are incorporated; the permeability coefficient of the mixed soil in DIW was 3.74 × 10−7 cm/s when the permeate concentration was 0.01 mol/L of ZnCl2 solution, the permeation coefficients were 5.73 × 10−7, 5.98 × 10−8, 5.8 × 10−8, and 5.7 × 10−8 cm/s when the zeolite doping was 0, 12.50, 25, or 50%, respectively, and the Zn2+ concentration of the leachate decreased compared to the no-zeolite case by 92.48, 97.29, and 98.65%, respectively; the competitive adsorption of metal ions by zeolites in ionic solutions of different concentrations reduced the ionic concentration in the solution and decreased the inhibition of bentonite swelling, while the adsorption characteristics of stone chip-bentonite-zeolite mixture on Zn2+ were measured by the Langmuir and Freundlich et al. model.
