    Abstract: Measured impedance spectra of Li-ion battery cells are often reproduced with equivalent circuits or physical models to determine losses due to charge transfer processes at the electrodes. The identified model parameters can usually not readily or unambiguously be assigned to the anode and the cathode. A new measurement method is presented that enables the assignment of features of impedance spectra of full cells to single electrodes. To this end, temperature gradients are imprinted perpendicular to the electrode layers of a single-layered Li-ion battery cell while impedance spectra are measured. The method exploits different dependences of the charge transfer processes at the electrodes on temperature. An equivalent circuit model of RC-elements and the effect of temperature on the related electrode properties is discussed to demonstrate the feasibility of the method. A reliable assignment of the change of impedance spectra to the electrode processes is shown to be possible. The assignment can be used to identify if changes in an impedance spectrum originate from the anode or the cathode.

    Abstract: Angiotensin-converting enzyme 2 (ACE2) is a main receptor for SARS-CoV-2 entry to the host cell. Indeed, the first step in viral entry is the binding of the viral trimeric spike (S) protein to ACE2. Abundantly present in human epithelial cells of many organs, ACE2 is also expressed in the human brain. ACE2 is a type I membrane protein with an extracellular N-terminal peptidase domain and a C-terminal collectrin-like domain that ends with a single transmembrane helix and an intracellular 44-residue segment. This C-terminal segment contains a PDZ-binding motif (PBM) targeting protein-interacting domains called PSD-95/Dlg/ZO-1 (PDZ). Here, we identified the human PDZ specificity profile of the ACE2 PBM using the high-throughput holdup assay and measuring the binding intensities of the PBM of ACE2 against the full human PDZome. We discovered 14 human PDZ binders of ACE2 showing significant binding with dissociation constants’ values ranging from 3 to 81 μM. NHERF, SHANK, and SNX27 proteins found in this study are involved in protein trafficking. The PDZ/PBM interactions with ACE2 could play a role in ACE2 internalization and recycling that could be of benefit for the virus entry. Interestingly, most of the ACE2 partners we identified are expressed in neuronal cells, such as SHANK and MAST families, and modifications of the interactions between ACE2 and these neuronal proteins may be involved in the neurological symptoms of COVID-19.

    Abstract: Background: Squamous cell carcinoma of the anal canal (SCCA) is rare. Most cases are diagnosed in a localized setting. Metastatic SCCA is rare, and investigation has been limited in the past for these patients. We believe that hepatic-only metastatic disease could have a unique treatment landscape compared to diseases with diffuse metastatic involvement. Here, we describe cases at our institution. Methods: We reviewed eight SCCA cases with hepatic-only metastatic disease (diagnosed February 2018–January 2022). The objectives were to determine the overall survival and disease-free survival with this approach. Results: The median age was 62 years old (yo). Patients had an ECOG of 0–1. All patients received definitive chemoradiation to their primary anal tumor. A median of three months of neoadjuvant systemic therapy was provided. All patients had a response on their first scan after systemic therapy. Sixty-two percent received carboplatin + paclitaxel. A complete pathologic response was seen in 62% of patients. At their last follow-up, all patients were alive. Three patients had recurrent disease. The estimated 1-year disease-free survival probability was 56.2%. Conclusion: Our report shows the feasibility of a curative-intent approach for patients with hepatic-only metastatic SCCA following the neoadjuvant application of carboplatin + paclitaxel. This approach appears promising in these select patients and warrants further investigation.

    Abstract: Land cover maps contribute to a large diversity of geospatial applications, including but not limited to land management, hydrology, land use planning, climate modeling and biodiversity monitoring. In densely populated and highly fragmented landscapes as observed in the Walloon region (Belgium), very high spatial resolution is required to depict all the infrastructures, buildings and most of the structural elements of the semi-natural landscapes (like hedges and small water bodies). Because of the resolution, the vertical dimension needs explicit handling to avoid discontinuities incompatible with many applications. For example, how to map a river flowing under a bridge? The particularity of our data is to provide a two-digit land cover code to label all the overlapping items. The identification of all the overlaps resulted from the combination of remote sensing image analysis and decision rules involving ancillary data. The final product is therefore semantically precise and accurate in terms of land cover description thanks to the addition of 24 classes on top of the 11 pure land cover classes. The quality of the map has been assessed using a state-of-the-art validation scheme. Its overall accuracy is as high as 91.5%, with an average producer’s accuracy of 86% and an average user’s accuracy of 91%.

    Abstract: Sézary syndrome (SS) is an aggressive cutaneous T cell lymphoma with poor prognosis mainly characterized by the expansion of a tumor CD4+ T cell clone in both skin and blood. So far, the development of new therapeutic strategies has been hindered by a lack of reproducible in vivo models closely reflecting patients’ clinical features. We developed an SS murine model consisting of the intravenous injection of Sézary patients’ PBMC, together with a mixture of interleukins, in NOD-SCID-gamma mice. Thirty-four to fifty days after injection, mice showed skin disorders similar to that observed in patients, with the detection of epidermis thickening and dermal tumor T cell infiltrates. Although experimental variability was observed, Sézary cells could be tracked in the blood stream, confirming that our model could efficiently exhibit both skin and blood involvement. Using this model, we evaluated the therapeutic potential of RT39, a cell-penetrating peptide derived from the survival protein anti-apoptosis clone 11 (AAC-11), that we previously characterized as specifically inducing apoptosis of Sézary patients’ malignant clone ex vivo. Systemic administration of RT39 led to cutaneous tumor T cells depletion, demonstrating efficient malignant cells’ targeting and a favorable safety profile. These preclinical data confirmed that RT39 might be an innovative therapeutic tool for Sézary syndrome.
