    Kurzfassung: We assessed interactions between the astrocytic volume-regulated anion channel (VRAC) and aquaporin 4 (AQP4) in the supraoptic nucleus (SON). Acute SON slices and cultures of hypothalamic astrocytes prepared from rats received hyposmotic challenge (HOC) with/without VRAC or AQP4 blockers. In acute slices, HOC caused an early decrease with a late rebound in the neuronal firing rate of vasopressin neurons, which required activity of astrocytic AQP4 and VRAC. HOC also caused a persistent decrease in the excitatory postsynaptic current frequency, supported by VRAC and AQP4 activity in early HOC; late HOC required only VRAC activity. These events were associated with the dynamics of glial fibrillary acidic protein (GFAP) filaments, the late retraction of which was mediated by VRAC activity; this activity also mediated an HOC-evoked early increase in AQP4 expression and late subside in GFAP-AQP4 colocalization. AQP4 activity supported an early HOC-evoked increase in VRAC levels and its colocalization with GFAP. In cultured astrocytes, late HOC augmented VRAC currents, the activation of which depended on AQP4 pre-HOC/HOC activity. HOC caused an early increase in VRAC expression followed by a late rebound, requiring AQP4 and VRAC, or only AQP4 activity, respectively. Astrocytic swelling in early HOC depended on AQP4 activity, and so did the early extension of GFAP filaments. VRAC and AQP4 activity supported late regulatory volume decrease, the retraction of GFAP filaments, and subside in GFAP-VRAC colocalization. Taken together, astrocytic morphological plasticity relies on the coordinated activities of VRAC and AQP4, which are mutually regulated in the astrocytic mediation of HOC-evoked modulation of vasopressin neuronal activity.

    Kurzfassung: In this paper, the design of a self-developed EMP simulator with a 5 m height and an inverted conducting mono-cone antenna with a cone half-angle of 32° is introduced. The experimental region of the simulator is a circular area of 25 m in diameter around the cone vertex. Two feeding modes, feed-in over the ground and feed-in under the ground, are realized by two different high-voltage pulse sources. It can be concluded through radiation field testing that the radiation field waveform generated by the simulator has a rise time of 2–3 ns and a half-width of about 25 ns, meeting the specifications of the EMP experimental waveform in the IEC61000-2-9 standard. Meanwhile, the differences between the engineering implementation of the simulator and its ideal structure during the design process can lead to some distortion issues in the antenna radiation characteristics and the electromagnetic radiation field it generates. The radiation field waveform and the distribution of the EMP field generated by the simulator under different feeding methods, antenna wire quantities, antenna end processing methods, and different antenna resistive loading were studied, and the changes in the radiation field waveform and field distribution with different angles and distances of the monitoring point were analyzed. Based on the measurement results, the radiation characteristics of the antenna and the factors that affect the waveform of the field were studied and analyzed. Through the aforementioned work, a comprehensive understanding of the performance, radiation characteristics, and engineering factors affecting the electromagnetic environment generated by the simulator has been obtained. On this basis, the parameters of the antenna wire quantity, antenna end processing method, and test point position of the designed simulator were reconfirmed and optimized. In summary, this work has important reference significance for mastering the development technology of such simulators, understanding their antenna radiation characteristics, and conducting EMP-related assessments and effect experiments in the future.

    Kurzfassung: Introduction: The objective was to evaluate the poisoning severity score (PSS) as an early prognostic predictor in patients with wasp stings and identify associated clinical characteristics and risk factors for mortality. Methods: A total of 363 patients with wasp stings at Suining Central Hospital between January 2016 and December 2018 were enrolled. Within the first 24 h of admission, the poisoning severity score (PSS) and the Chinese expert consensus on standardized diagnosis and treatment of wasp stings (CECC) were utilized for severity classification, and their correlation was examined. Patients were then divided into survival and death groups based on discharge status. Logistic regression analysis was employed to analyze factors influencing patients’ outcomes. Results: The mortality of wasp sting patients was 3.9%. The PSS and CECC were found to correlate for severity classification. Additionally, female gender, age, number of stings, and PSS were identified as independent risk factors for mortality in wasp sting patients. Combining these four factors yielded an AUC of 0.962 for predicting death. Conclusions: PSS aids in early severity classification of wasp stings. Female gender, age, number of stings, and PSS were independent mortality risk factors in these patients.

    Kurzfassung: Traditional coupling of ligands for gold wet etching makes large-scale applications problematic. Deep eutectic solvents (DESs) are a new class of environment-friendly solvents, which could possibly overcome the shortcomings. In this work, the effect of water content on the Au anodic process in DES ethaline was investigated by combining linear sweep voltammetry (LSV) and electrochemical impedance spectroscopy (EIS). Meanwhile, we employed atomic force microscopy (AFM) to image the evolution of the surface morphology of the Au electrode during its dissolution and passivation process. The obtained AFM data help to explain the observations about the effect of water content on the Au anodic process from the microscopic perspective. High water contents make the occurrence of anodic dissolution of gold at higher potential, but enhances the rate of the electron transfer and gold dissolution. AFM results reveal the occurrence of massive exfoliation, which confirms that the gold dissolution reaction is more violent in ethaline with higher water contents. In addition, AFM results illustrate that the passive film and its average roughness could be tailored by changing the water content of ethaline.

    Kurzfassung: In order to further improve the insulation performance of fiber reinforce plastic (FRP) materials used in electromagnetic pulse (EMP) simulators, the flashover characteristics of FRP materials with different surface roughness and groove, i.e., those who are easily achieved and have a prominent effect, are investigated in 0.1 MPa SF6 under nanosecond pulse voltage with a rise time of 20–30 ns. The experimental results show that surfaces with different roughness have no significant influence on the flashover voltages of the FRP insulators, and both the convex grooves made of FRP and the convex grooves with nylon rings inlaid to form projections can improve the surface flashover voltage of epoxy FRP insulators under nanosecond pulse, in which the effect of the former surface is more obvious. For the insulators with convex grooves made of FRP, it is found that the root of the FRP protrusions breaks down after a number of shots with the occurrence of carbonization channels and spots, which is nonexistent for the nylon projections. Combined with the test results of surface characteristics, the surface roughness and the secondary electron emission yield (SEEY) are not key factors of flashover characteristics in SF6 under nanosecond pulse, arguably due to the fact that the energy needed for an incident electron to ionize an SF6 molecule is lower than that to excite two secondary electrons. Hence, the flashover performance cannot be improved by adjusting the surface roughness, and the flashover channel is principally governed by the macroscopic distribution of electrical field which can be changed by the convex groove. Breakdown phenomena of FRP protrusions indicate that the bulk insulation performance of resin FRP is weaker compared to pure resin because of its composite structure, as well as the impurities and voids introduced in the manufacturing process. The results are instructive for the design of FRP insulation structures in the compact EMP simulator.

    Kurzfassung: The dysbiosis of oral microbiota is linked to numerous diseases and is associated with personal lifestyles, such as alcohol drinking. However, there is inadequate data to study the effect of alcohol drinking on oral microbiota from the Chinese population. Here, we profiled the oral microbiota of 150 healthy subjects in the Chinese population by 16S rRNA gene sequencing. The results showed that drinkers had significantly higher alpha diversity than non-drinkers. A significant difference in overall microbiota composition was observed between non-drinkers and drinkers. Additionally, using DESeq analysis, we found genus Prevotella and Moryella, and species Prevotella melaninogenica and Prevotella tannerae were significantly enriched in drinkers; meanwhile, the genus Lautropia, Haemophilus and Porphyromonas, and species Haemophilus parainfluenzae were significantly depleted in drinkers. PICRUSt analysis showed that significantly different genera were mainly related to metabolism pathways. The oxygen-independent pathways, including galactose, fructose and mannose metabolism pathways, were enriched in drinkers and positively associated with genera enriched in drinkers; while the pyruvate metabolism pathway, an aerobic metabolism pathway, was decreased in drinkers and negatively associated with genera enriched in drinkers. Our results suggested that alcohol drinking may affect health by altering oral microbial composition and potentially affecting microbial functional pathways. These findings may have implications for better understanding the potential role those oral bacteria play in alcohol-related diseases.

    Kurzfassung: Increasing attention has been paid to removal of aqueous contaminations resulting from azo dyes in water by plasma technology. However, the influence factors and removal mechanism of plasma technology were still obscure, moreover, energy consumption and oxidized degradation efficiency of plasma reactor were also inferior. In the present study, a comparative analysis was performed using 100 mg/L of Methyl Orange (MO) in the simulated wastewater with a novel plasma reactor to achieve the ideal parameters involving voltage, discharge gap, and discharge needle numbers. Therefore, the optimal removal rate for MO could be up to 95.1% and the energy consumption was only 0.26 kWh/g after the plasma treatment for 60 min, when the voltage was set as 15 kV, the discharge gap was 20 mm, and the discharge needle numbers was 5. Based upon the response surface methodology (RSM), the removal rate of MO was predicted as 99.3% by massive optimization values in software, and the optimum conditions were confirmed with the plasma treatment period of 60 min, the voltage of 14.8 kV, the discharge gap of 20 mm, and the discharge needles of 5. Plasma associated with catalysts systems including plasma, plasma/Fe2+, plasma/PS, and plasma/PS/Fe2+ were further investigated, and the best removal rate for MO reached 99.2% at 60 min under the plasma/PS/Fe2+ system due to simultaneously synergistic reactions of HO• and SO4•−. Moreover, it was also revealed that –N=N– bond was attacked and broken by active species like HO•, and the oxidized by-products of benzenesulfonic acid and phenolsulfonic acid might be generated, via the analysis of the variation in the absorbances through UV-Vis spectrophotometry during the plasma treatment. As a result, the advanced plasma technique in this study presented excellent efficacy for MO removal from simulated wastewater with low energy consumption.

    Kurzfassung: Dark skin-type individuals have a greater tendency to have pigmentary disorders, among which melasma is especially refractory to treat and often recurs. Objective measurement of melanin amount helps evaluate the treatment response of pigmentary disorders. However, naked-eye evaluation is subjective to weariness and bias. We used a cellular resolution full-field optical coherence tomography (FF-OCT) to assess melanin features of melasma lesions and perilesional skin on the cheeks of eight Asian patients. A computer-aided detection (CADe) system is proposed to mark and quantify melanin. This system combines spatial compounding-based denoising convolutional neural networks (SC-DnCNN), and through image processing techniques, various types of melanin features, including area, distribution, intensity, and shape, can be extracted. Through evaluations of the image differences between the lesion and perilesional skin, a distribution-based feature of confetti melanin without layering, two distribution-based features of confetti melanin in stratum spinosum, and a distribution-based feature of grain melanin at the dermal–epidermal junction, statistically significant findings were achieved (p-values = 0.0402, 0.0032, 0.0312, and 0.0426, respectively). FF-OCT enables the real-time observation of melanin features, and the CADe system with SC-DnCNN was a precise and objective tool with which to interpret the area, distribution, intensity, and shape of melanin on FF-OCT images.

    Kurzfassung: Oral hygiene and the alteration of the oral microbiome have been linked to nasopharyngeal carcinoma (NPC). This study aimed to investigate whether the oral microbiome plays a mediating role in the relationship between oral hygiene and NPC, and identify differential microbial taxonomies that potentially mediated this association. We conducted a case–control study that involved 218 NPC patients and 192 healthy controls. The 16S rRNA gene sequencing of the V4 region was performed to evaluate the composition of the oral microbiome. Mediation analysis was applied to explore the relationship among oral hygiene, the oral microbiome and NPC. We found that dental fillings and poor oral hygiene score were associated with increased risks of NPC (OR = 2.51 (1.52–4.25) and OR = 1.54 (1.02–2.33)). Mediation analysis indicated that dental fillings increased the risk of NPC by altering the abundance of Erysipelotrichales, Erysipelotrichaceae, Solobacterium and Leptotrichia wadei. In addition, Leptotrichia wadei also mediated the association between oral hygiene score and the risk of NPC. Our study confirmed that poor oral hygiene increased the risk of NPC, which was partly mediated by the oral microbiome. These findings might help us to understand the potential mechanism of oral hygiene influencing the risk of NPC via the microbiome.

    Kurzfassung: Ambulatory blood pressure (BP) monitoring (ABPM) is vital for screening cardiovascular activity. The American College of Cardiology/American Heart Association guideline for the prevention, detection, evaluation, and management of BP in adults recommends measuring BP outside the office setting using daytime ABPM. The recommendation to use night–day BP measurements to confirm hypertension is consistent with the recommendation of several other guidelines. In recent studies, ABPM was used to measure BP at regular intervals, and it reduces the effect of the environment on BP. Out-of-office measurements are highly recommended by almost all hypertension organizations. However, traditional ABPM devices based on the oscillometric technique usually interrupt sleep. For all-day ABPM purposes, a photoplethysmography (PPG)-based wrist-type device has been developed as a convenient tool. This optical, noninvasive device estimates BP using morphological characteristics from PPG waveforms. As measurement can be affected by multiple variables, calibration is necessary to ensure that the calculated BP values are accurate. However, few studies focused on adaptive calibration. A novel adaptive calibration model, which is data-driven and embedded in a wearable device, was proposed. The features from a 15 s PPG waveform and personal information were input for estimation of BP values and our data-driven calibration model. The model had a feedback calibration process using the exponential Gaussian process regression method to calibrate BP values and avoid inter- and intra-subject variability, ensuring accuracy in long-term ABPM. The estimation error of BP (ΔBP = actual BP—estimated BP) of systolic BP was −0.1776 ± 4.7361 mmHg; ≤15 mmHg, 99.225%, and of diastolic BP was −0.3846 ± 6.3688 mmHg; ≤15 mmHg, 98.191%. The success rate was improved, and the results corresponded to the Association for the Advancement of Medical Instrumentation standard and British Hypertension Society Grading criteria for medical regulation. Using machine learning with a feedback calibration model could be used to assess ABPM for clinical purposes.
