    Abstract: According to the movement characteristics of the surrounding rock of the gob-side entry retaining by roof cutting (GERRC), the structural evolution mechanism and stability control countermeasures are studied in this paper. Taking the roof cutting and bulking as the core point, the movement process of the surrounding rock of GERRC is divided into five stages: roof cutting, dynamic pressure support, gangue bulking, double arch roadway support, and roadway formation. Combined with mechanical analysis, the roof pressure of the mechanical model of the short-arm beam formed by roof cutting is calculated, and the roof breaking criterion is obtained, which provides a basis for roof control design and reasonable support strength calculation. A structural model of double-arch roadway protection under the action of hydrostatic roof cutting and gangue bulking was established, and the mechanism of roadway formation stability was studied. The gob-side bulking gangue and short-arm beam are formed by roof cutting, and the solid coal seam forms the immediate balance arch, the bulking gangue in goaf, articulated transfer rock beam, and solid coal seam forms the main balance arch. The two together are called double balance arch. With the support of the double-balanced arch on the overlying rock strata, the roadway below is located in the low-stress zone, which ensures the stability of the roadway surrounding rock. Through mechanical analysis and field observation, the law of bulking and deformation and mechanical characteristics of gob-side gangue in the above structural model are analyzed, and the reasonableness of the structural model of the double-arch guard lane is verified through numerical simulation of the cut top into lanes. and the reasonableness of the structural model of double-arch protection in GERRC is verified by using numerical simulation. In this paper, a mechanical double-arch model has been developed that allows us to understand the mechanism of stress transfer of roof cutting roadway from the perspective of a dynamic balance of roadway surrounding rock and overlying rock strata, helping us to specify efficient support countermeasures.

    Abstract: Fourier ptychographic microscopy (FPM) is a novel technique for computing microimaging that allows imaging of samples such as pathology sections. However, due to the influence of systematic errors and noise, the quality of reconstructed images using FPM is often poor, and the reconstruction efficiency is low. In this paper, a hybrid attention network that combines spatial attention mechanisms with channel attention mechanisms into FPM reconstruction is introduced. Spatial attention can extract fine spatial features and reduce redundant features while, combined with residual channel attention, it adaptively readjusts the hierarchical features to achieve the conversion of low-resolution complex amplitude images to high-resolution ones. The high-resolution images generated by this method can be applied to medical cell recognition, segmentation, classification, and other related studies, providing a better foundation for relevant research.

    Abstract: Background: Brugada syndrome (BrS) is an inheritable arrhythmia syndrome that can lead to sudden cardiac death in patients while the heart structure is normal. However, the genetic background of more than 65% of BrS probands remains unclear. Objectives: The purpose of this study is to report the variant spectrum in a Chinese cohort with suspected BrS and to analyze their distinct clinical and electrocardiographic features. Methods: Patients with suspected BrS from Tongji Hospital between 2008 and 2021 were analyzed retrospectively. Results: A total of 79 probands were included in this study. Patients with type 1 BrS electrocardiogram (ECG) had a prolonged QRS duration compared to patients with type 2/3 BrS ECG. Of them, 59 probands underwent genetic testing. Twenty-five patients (42.37%) showed abnormal genetic testing results, and eight of them (13.56%) carried pathogenic/likely pathogenic (P/LP) mutations. Mutation carriers presented much more prominent depolarization and repolarization abnormalities than non-carriers, including a prolonged P-wave duration, QRS duration, QTc interval, decreased QRS amplitude, and deviation of the electrocardiographic axes (T-wave axis and R-wave axis). Furthermore, our study identified four novel P/LP mutations: Q3508X in TTN, A990G in KCNH2, G1220E, and D372H (in a representative pedigree) in SCN5A. Conclusions: Our study showed the variant spectrum of a suspected Chinese BrS cohort, and we identified four novel P/LP mutations in TTN, KCNH2, and SCN5A.

    Abstract: Little information is available regarding the effect of melatonin on the quality and fertilization capability of sex-sorted bull sperm, and even less about the associated mechanism. Sex-sorted sperm from three individual bulls were washed twice in wash medium and incubated in a fertilization medium for 1.5 h, and each was supplemented with melatonin (0, 10−3 M, 10−5 M, 10−7 M, and 10−9 M). The reactive oxygen species (ROS) and endogenous antioxidant activity (glutathione peroxidase (GPx); superoxide dismutase (SOD); catalase (CAT)), apoptosis (phosphatidylserine [PS] externalization; mitochondrial membrane potential (Δψm)), acrosomal integrity events (malondialdehyde (MDA) level; acrosomal integrity), capacitation (calcium ion [Ca2+] i level; cyclic adenosine monophosphate (cAMP); capacitation level), and fertilization ability of the sperm were assessed. Melatonin receptor 1 (MT1) and 2 (MT2) expression were examined to investigate the involvement of melatonin receptors on sex-sorted bull sperm capacitation. Our results show that treatment with 10−5 M melatonin significantly decreased the ROS level and increased the GPx, SOD, and CAT activities of sex-sorted bull sperm, which inhibited PS externalization and MDA levels, and improved Δψm, acrosomal integrity, and fertilization ability. Further experiments showed that melatonin regulates sperm capacitation via MT1. These findings contribute to improving the fertilization capacity of sex-sorted bull sperm and exploring the associated mechanism.

    Abstract: Digital holography (DH) is a novel, real-time, non-destructive, and quantitative phase-contrast imaging method that is particularly suitable for label-free live biological cell imaging and real-time dynamic monitoring. It is currently a research hotspot in the interdisciplinary field of optics and biomedical sciences, both domestically and internationally. This article proposes an improved angle spectrum algorithm based on holographic technology, which reconstructs a cellular hologram based on phase information. Optical images and chromosome cell images, reconstructed using holographic technology at different diffraction distances under the improved angle spectrum algorithm, were analyzed and compared. The optimal diffraction distance for reconstructing chromosome cell images was selected, and chromosome cell images reproduced using traditional angle spectrum algorithms, angle spectrum algorithms combined with GS, and improved angle spectrum algorithms were compared. Comparative experiments with the different models show that the proposed algorithm is superior to traditional angle spectrum algorithms in reconstructing cell images based on phase information. Furthermore, experiments have shown that images reconstructed using the improved algorithm can resolve high signal-to-noise ratio information. This algorithmic improvement provides new applications for cellular detection in clinical diagnostics and is more suitable for cell phase reconstruction in practical applications.

    Abstract: Chronic obstructive pulmonary disease (COPD) is preventable and treatable. However, many patients remain undiagnosed and untreated due to the underutilization or unavailability of spirometers. Accordingly, we used Spirobank Smart, an app-based spirometer, for facilitating the early detection of COPD in outpatient clinics. This prospective study recruited individuals who were at risk of COPD (i.e., with age of ≥40 years, ≥10 pack-years of smoking, and at least one respiratory symptoms) but had no previous COPD diagnosis. Eligible participants were examined with Spirobank Smart and then underwent confirmatory spirometry (performed using a diagnostic spirometer), regardless of their Spirobank Smart test results. COPD was defined and confirmed using the postbronchodilator forced expiratory volume in 1 s/forced vital capacity values of 〈 0.70 as measured by confirmatory spirometry. A total of 767 participants were enrolled and examined using Spirobank Smart; 370 participants (94.3% men, mean age of 60.9 years and mean 42.6 pack-years of smoking) underwent confirmatory spirometry. Confirmatory spirometry identified COPD in 103 participants (27.8%). At the optimal cutoff point of 0.74 that was determined using Spirobank Smart for COPD diagnosis, the area under the receiver operating characteristic was 0.903 (95% confidence interval (CI) = 0.860–0.947). Multivariate logistic regression revealed that participants who have an FEV1/FVC ratio of 〈 74% that was determined using Spirobank Smart (odds ratio (OR) = 58.58, 95% CI = 27.29–125.75) and old age (OR = 3.23, 95% CI = 1.04–10.07 for 60 ≤ age 〈 65; OR = 5.82, 95% CI = 2.22–15.27 for age ≥ 65) had a higher risk of COPD. The Spirobank Smart is a simple and adequate tool for early COPD detection in outpatient clinics. Early diagnosis and appropriate therapy based on GOLD guidelines can positively influence respiratory symptoms and quality of life.

    Abstract: Lithium-sulfur (Li-S) batteries are deemed to be one of the most optimal solutions for the next generation of high-energy-density and low-cost energy storage systems. However, the low volumetric energy density and short cycle life are a bottleneck for their commercial application. To achieve high energy density for lithium-sulfur batteries, the concept of synergistic adsorptive–catalytic sites is proposed. Base on this concept, the TiN@C/S/Ta2O5 sulfur electrode with about 90 wt% sulfur content is prepared. TiN contributes its high intrinsic electron conductivity to improve the redox reaction of polysulfides, while Ta2O5 provides strong adsorption capability toward lithium polysulfides (LiPSs). Moreover, the multidimensional carbon structure facilitates the infiltration of electrolytes and the motion of ions and electrons throughout the framework. As a result, the coin Li-S cells with TiN@C/S/Ta2O5 cathode exhibit superior cycle stability with a decent capacity retention of 56.1% over 300 cycles and low capacity fading rate of 0.192% per cycle at 0.5 C. Furthermore, the pouch cells at sulfur loading of 5.3 mg cm−2 deliver a high areal capacity of 5.8 mAh cm−2 at low electrolyte/sulfur ratio (E/S, 3.3 μL mg−1), implying a high sulfur utilization even under high sulfur loading and lean electrolyte operation.

    Abstract: The subgenus Cerasus and its relatives include many crucial economic drupe fruits and ornamental plants. Repetitive elements make up a large part of complex genomes, and some of them play an important role in gene regulation that can affect phenotypic variation. However, the variation in their genomes remains poorly understood. This work conducted a comprehensive repetitive sequence identification across the draft genomes of eight taxa of the genus Prunus, including four of the Prunus subgenus Cerasus (Prunus pseudocerasus, P. avium, P. yedoensis and P. × yedoensis) as well as congeneric species (Prunus salicina, P. armeniaca, P. dulcis and P. persica). Annotation results showed high proportions of transposable elements in their genomes, ranging from 52.28% (P. armeniaca) to 61.86% (P. pseudocerasus). The most notable differences in the contents of long terminal repeat retrotransposons (LTR-RTs) and tandem repeats (TRs) were confirmed with de novo identification based on the structure of each genome, which significantly contributed to their genome size variation, especially in P. avium and P.salicina. Sequence comparisons showed many similar LTR-RTs closely related to their phylogenetic relationships, and a highly similar monomer unit of the TR sequence was conserved among species. Additionally, the predicted centromere-associated sequence was located in centromeric regions with FISH in the 12 taxa of Prunus. It presented significantly different signal intensities, even within the diverse interindividual phenotypes for Prunus tomentosa. This study provides insight into the LTR-RT and TR variation within Prunus and increases our knowledge about its role in genome evolution.
