    Kurzfassung: With the gradual deepening of mine excavation depth, the strong disturbance of deep strata becomes more and more obvious. Rock’s failure under blasting mainly depends on its dynamic tensile strength. The changes in rock’s dynamic properties are obviously affected by temperature and water. In order to study the dynamic tensile properties of annular sandstone specimens under the influence of temperature and water, deep sandstone was drilled, followed by water bath tests at eight temperatures (25~95 °C). It can be seen from the analysis of test results that the mass and volume growth rates of the annular and the intact sandstone specimens first increased and then decreased, while the density growth rate first decreased and then increased. The mass and volume growth rates of the annular sandstone specimens were smaller, but the density growth rate was larger. Because of the increase in water temperature, the dynamic compressive strength first increased and then decreased. The dynamic tensile strength of the annular sandstone specimen was lower. The average strain rate and peak strain also showed a quadratic function relationship of first decreasing and then increasing with the increase in water temperature. The average strain rate of the annular sandstone specimen was smaller, but the peak value changed greatly. The Brazilian disc validity condition is applicable to two failure conditions of sandstone specimens. Through XRD and SEM analysis, we found that the changes in the dynamic properties of sandstone specimens were not due to their own material composition, but to the damage to their structure caused by the temperature–water coupling effect.

    Kurzfassung: Different discharge processes of triggered lightning can radiate electromagnetic signals with different frequency bands. During the triggered-lightning experiment conducted at the Field Experiment Base on Lightning Sciences of China Meteorological Administration (CMA-FEBLS), three magnetic field (B-field) antennas with different frequency responses were deployed at about 80 m from the rocket-launching site. By using the synchronous observations, the quantitative relationship between the close-range B-field measurement and the channel-base current at different stages of triggered lightning were established in the investigation. The initial continuous current (ICC) waveform can be reconstructed by numerically integrating the B-field signals measured with the dB/dt antenna. However, the slow variations of ICC cannot be retrieved by the B-field signals measured with the LF-MF antenna because the antenna bandwidth cannot cover a frequency below 500 Hz. The B-field signals of the return stroke measured with the low-sensitivity antenna can be simulated by the MTLL return-stroke model, and the B-field signal shows a fairly good consistency with the return-stroke current. The analyses suggest that the current waveform of the natural return stroke that occurred within 1.5 km can be retrieved, or at least its peak value can be estimated by using the B-field measurements.

    Kurzfassung: (1) Background: Abnormal repair after alveolar epithelial injury drives the progression of idiopathic pulmonary fibrosis (IPF). The maintenance of epithelial integrity is based on the self-renewal and differentiation of alveolar type 2 (AT2) cells, which require sufficient energy. However, the role of glutamine metabolism in the maintenance of the alveolar epithelium remains unclear. In this study, we investigated the role of glutamine metabolism in AT2 cells of patients with IPF and in mice with bleomycin-induced fibrosis. (2) Methods: Single-cell RNA sequencing (scRNA-seq), transcriptome, and metabolomics analyses were conducted to investigate the changes in the glutamine metabolic pathway during pulmonary fibrosis. Metabolic inhibitors were used to stimulate AT2 cells to block glutamine metabolism. Regeneration of AT2 cells was detected using bleomycin-induced mouse lung fibrosis and organoid models. (3) Results: Single-cell analysis showed that the expression levels of catalytic enzymes responsible for glutamine catabolism were downregulated (p 〈 0.001) in AT2 cells of patients with IPF, suggesting the accumulation of unusable glutamine. Combined analysis of the transcriptome (p 〈 0.05) and metabolome (p 〈 0.001) revealed similar changes in glutamine metabolism in bleomycin-induced pulmonary fibrosis in mice. Mechanistically, inhibition of the key enzymes involved in glucose metabolism, glutaminase-1 (GLS1) and glutamic-pyruvate transaminase-2 (GPT2) leads to reduced proliferation (p 〈 0.01) and differentiation (p 〈 0.01) of AT2 cells. (4) Conclusions: Glutamine metabolism is required for alveolar epithelial regeneration during lung injury.

    Kurzfassung: The genus Convallaria (Asparagaceae) comprises three herbaceous perennial species that are widely distributed in the understory of temperate deciduous forests in the Northern Hemisphere. Although Convallaria species have high medicinal and horticultural values, studies related to the phylogenetic analysis of this genus are few. In the present study, we assembled and reported five complete chloroplast (cp) sequences of three Convallaria species (two of C. keiskei Miq., two of C. majalis L., and one of C. montana Raf.) using Illumina paired-end sequencing data. The cp genomes were highly similar in overall size (161,365–162,972 bp), and all consisted of a pair of inverted repeats (IR) regions (29,140–29,486 bp) separated by a large single-copy (LSC) (85,183–85,521 bp) and a small single-copy (SSC) region (17,877–18,502 bp). Each cp genome contained the same 113 unique genes, including 78 protein-coding genes, 30 transfer RNA genes, and 4 ribosomal RNA genes. Gene content, gene order, AT content and IR/SC boundary structure were nearly identical among all of the Convallaria cp genomes. However, their lengths varied due to contraction/expansion at the IR/LSC borders. Simple sequence repeat (SSR) analyses indicated that the richest SSRs are A/T mononucleotides. Three highly variable regions (petA-psbJ, psbI-trnS and ccsA-ndhD) were identified as valuable molecular markers. Phylogenetic analysis of the family Asparagaceae using 48 cp genome sequences supported the monophyly of Convallaria, which formed a sister clade to the genus Rohdea. Our study provides a robust phylogeny of the Asparagaceae family. The complete cp genome sequences will contribute to further studies in the molecular identification, genetic diversity, and phylogeny of Convallaria.

    Kurzfassung: Background: The nutrients-rich food (NRF) index provides a score of diet quality. Although high diet quality is associated with survival of ovarian cancer (OC), the associations between NRF index scores and OC survival remain unevaluated. Methods: The prospective cohort study enrolled 703 women with newly diagnosed epithelial OC to assess the correlations between NRF index scores and overall survival (OS) in OC patients. Dietary consumption was evaluated through a food frequency questionnaire and diet quality was calculated based on NRF index scores, including three limited nutrients and six (NRF6.3), nine (NRF9.3), or eleven (NRF11.3) benefit nutrients. All-cause deaths were ascertained through medical records combined with active follow-up. Immunohistochemistry (IHC) analyses were conducted to evaluate the expression of IHC indicators (including Estrogen Receptor, Progesterone Receptor, p53, Vimentin, and Wilms’ tumor 1), which were identified by two independent pathologists. The Cox proportional hazards regression models were applied for estimating the hazard ratios (HRs) and 95% confidence intervals (CIs). Moreover, we performed the penalized cubic splines model to assess the curvilinear associations of NRF index scores with OC survival. Results: During the median follow-up of 37.17 (interquartile: 24.73–50.17) months, 130 deaths were documented. Compared to the lowest tertiles, the highest tertile of index scores [NRF9.3 (HR = 0.63, 95% CI = 0.41–0.95), NRF6.3 (HR = 0.59, 95% CI = 0.39–0.89), and NRF11.3 (HR = 0.57, 95% CI = 0.38–0.87)] were correlated to better OS, showing an obvious linear trend (all p trend 〈 0.05). Interestingly, the curvilinear association between the NRF6.3 index score and OC survival was also observed (p non-linear 〈 0.05). Subgroup analyses, stratified by clinical, demographic, and IHC features, showed similar risk associations as the unstratified results. Furthermore, there were significant multiplicative interactions between NRF index scores and Progestogen Receptors as well as Wilms’ tumor 1 expressions (all p interaction 〈 0.05). Conclusions: Higher NRF index scores were associated with an improved OS in OC patients.

    Kurzfassung: Metastasis is the main cause of death for patients suffering gastric cancer. Epithelial-mesenchymal transition (EMT) and cancer stem cells (CSC) are critical attributes of metastasis, both of which are regulated tightly by DNA methylation and Wnt/β-catenin signaling. Here, we studied the functions of DNA dioxygenase TET1 in regulating Wnt signaling and in gastric cancer metastasis. Knocking-down and overexpressing TET1 in gastric cancer cells promoted and inhibited metastatic spreading to the liver in immune-deficient mice, respectively. TET1 showed inhibitory effects on metastasis-related features -EMT and CSC, which were reversed by interfering with Wnt/β-catenin signaling. RNA-sequencing identified FOXO4 as a direct transactivating target of TET1. FOXO4 directly interacted with β-catenin and recruited it in the cytoplasm, so as to inhibit β-catenin-mediated transcription of Wnt target genes, including CSC marker EpCAM. Moreover, modulation of FOXO4 could reverse the effects of TET1 manipulation on EMT and self-renewal of CSCs. The analysis with clinical samples confirmed the value of FOXO4 as an independent prognostic predictor of patients’ overall survival. Taken together, regulation of Wnt signaling by TET1/FOXO4 is essential for metastasis-associated cellular properties, and targeting TET1/FOXO4/β-catenin pathway may serve as promising therapeutics in the prevention and treatment of gastric cancer metastasis.

    Kurzfassung: The purpose of this study is to investigate the role that a deep learning approach could play in computational mechanics. In this paper, a convolutional neural network technique based on modified loss function is proposed as a surrogate of the finite element method (FEM). Several surrogate-based physics-informed neural networks (PINNs) are developed to solve representative boundary value problems based on elliptic partial differential equations (PDEs). According to the authors’ knowledge, the proposed method has been applied for the first time to solve boundary value problems with elliptic partial differential equations as the governing equations. The results of the proposed surrogate-based approach are in good agreement with those of the conventional FEM. It is found that modification of the loss function could improve the prediction accuracy of the neural network. It is demonstrated that to some extent, the deep learning approach could replace the conventional numerical method as a significant surrogate model.

    Kurzfassung: An urban agglomeration is the engine of regional and national economic growth, but also causes many ecological and environmental issues that emerge from massive land changes. In this study, the spatiotemporal evolution of an urban agglomeration was quantified and its impacts on the urban and regional landscape patterns were evaluated. It showed that the urbanized land area of the Pearl River Delta Urban Agglomeration (PRDUA) in China nearly quadrupled, having linearly increased from 1819.8 km2 to 7092.2 km2 between 1985 and 2015. The average annual growth rate presented a bimodal wave-like pattern through time, indicating that the PRDUA has witnessed two rounds of the urbanization process. The growth modes (e.g., leapfrog, edge-expansion, infilling) were detected and they exhibited co-existing but alternating dominating patterns during urbanization, demonstrating that the spatiotemporal evolution of the urban development of the PRDUA follows the “spiral diffusion-coalescence” hypothesis. The morphology of the PRDUA presented an alternating dispersal-compact pattern over time. The city-level and regional-level landscape patterns changed synchronously with the spatiotemporal evolution of the PRDUA over time. The urbanization of the PRDUA increased both the complexity and aggregation of the landscape, but also resulted in an increasing fragmentation and decreasing connectivity of the natural landscape in the Pearl River Delta region. These findings are helpful for better understanding how urban agglomerations evolve and in providing insights for regional urban planning and sustainable land management.

    Kurzfassung: Deoxynivalenol (DON) together with its acetylated derivatives cause detrimental effects on human health, and the purpose of this study was to assess the prevalence of DON and its acetylated derivatives from grains and grain products in Zhejiang province, China, and to assess the risk of DON and its acetylated derivatives due to multiple consumptions of grains and grain products among the Zhejiang population. Food samples numbering 713 were collected, and the LC-MS/MS method was used to determine the toxins. The levels of toxins from grains and grain products were relatively low: DON was the toxin at the highest levels. The food frequency questionnaire was used to collect food consumption data. The result of exposure assessments showed that the population was overall at low levels of toxin exposure. The probable mean group daily intake of toxins was 0.21 μg/kg bw/day, which was far from the group provisional maximum tolerable daily intake of 1 μg/kg bw/day, but 0.71% of participants were at high exposure levels. Rice and dried noodles (wheat-based food) were the main sources of toxin exposure, and reducing the consumption of rice and dried noodles while consuming more of other foods with lower levels of toxins is recommended.

    Kurzfassung: To develop feasible carbon fiber reinforced polymer (CFRP) retrofit schemes for the shear strengthening of real three-dimensional reinforced concrete (RC) beam-column joints, a series of parameters in relation to the contributions of the CFRP sheets externally bonded to joint panels was numerically investigated in this study. The parameters include CFRP reinforcement ratio, CFRP layout, transverse beam-to-joint panel width ratio, transverse beam-to-joint panel height ratio, location of transverse beam, and number of transverse beams. Strengthening efficiency, a new dimensionless index, was introduced to evaluate the residual effect of a CFRP-strengthening system weakened by the presence of transverse beams in comparison with the increase in joint shear capacity in relation to the one-way counterpart. The results obtained from 44 nonlinear finite element models, which were calibrated against experimental observations, confirmed the effectiveness of the CFRP strengthening technique with regard to the relatively wide ranges of the parameters. The significant differences among the roles of the parameters were revealed, and the reasons behind the differences were analyzed. Furthermore, the shear mechanism of the CFRP-retrofitted joint panels was discussed with the proposed strut-and-tie model.
