    Abstract: Karst collapse columns (KCCs) are naturally formed geological structures that are widely observed in North China. Given their influence on normal mining operations and the progress of mining work, collapse columns pose a hidden danger in coal mining under the influence of manual mining. By communicating often with the aquifer, the water inrush from KCCs poses a serious threat to construction projects. This paper adopts three flow field models, namely, Darcy aquifer laminar flow, Forchheimer flow, and Navier–Stokes turbulent flow, based on the changes in the water inrush flow pattern in the aquifer and laneway, and uses COMSOL Multiphysics software to produce the numerical solutions of these models. As the water inrush flow velocity increases, the Forchheimer flow shows the effect of additional force (inertial resistance) on flow in KCCs, in addition to the effect of viscous resistance. After the joint action of viscous resistance and inertial resistance, the inertial resistance ultimately dominates and gradually changes the water inrush from the KCCs to fluid seepage. Forchheimer flow can comprehensively reflect the nonlinear flow process in the broken rock mass of KCCs, demonstrate the dynamic process from the Darcy aquifer to the final tunnel turbulence layer, and quantitatively show the changes in the flow patterns of the water inrush from KCCs.

    Abstract: A combination of chemotherapeutic drugs and siRNA is emerging as a new modality for cancer therapy. A safe and effective carrier platform is needed for combination drug delivery. Here, a functionalized mixed micelle-based delivery system was developed for targeted co-delivery of methotrexate (MTX) and survivin siRNA. Linolenic acid (LA) was separately conjugated to branched polyethlenimine (b-PEI) and methoxy-polyethyleneglycol (mPEG). MTX was then conjugated to LA-modified b-PEI (MTX-bPEI-LA) to form a functionalized polymer-drug conjugate. Functionalized mixed micelles (M-MTX) were obtained by the self-assembly of MTX-bPEI-LA and LA-modified mPEG (mPEG-LA). M-MTX had a narrow particle size distribution and could successfully condense siRNA at an N/P ratio of 16/1. M-MTX/siRNA was selectively taken up by HeLa cells overexpressing the folate receptor (FR) and facilitated the release of the siRNA into the cytoplasm. In vitro, M-MTX/siRNA produced a synergy between MTX and survivin siRNA and markedly suppressed survivin protein expression. In tumor-bearing mice, M-MTX/Cy5-siRNA showed an elevated tumor uptake. In addition, M-MTX/siRNA inhibited tumor growth. Immunohistochemistry and a western blot analysis showed a significant target gene downregulation. In conclusion, M-MTX/siRNA was highly effective as a delivery system and may serve as a model for the targeted co-delivery of therapeutic agents.

    Abstract: Lycium barbarum L. (LB) fruits have high nutritive values and therapeutic effects. The aim of this study was to comprehensively evaluate the differences in phenolic composition of LB fruits from different geographical regions. Different methods of characterization and statistical analysis of data showed that different geographic sources of China could be significantly separated from each other. The highest total phenolic compound (TPC) content was observed in LB fruits from Ningxia (LBN), followed by those from Gansu (LBG) and Qinghai (LBQ). The Fourier transform infrared (FTIR) spectra of LB fruits revealed that LBQ had a peak at 2972 cm−1 whereas there was no similar peak in LBG and LBQ. A new HPLC method was established for the simultaneous determination of 8 phenolic compounds by quantitative analysis of multiple components by a single marker (QAMS), including 4 phenolic acids (chlorogenic acid, caffeic acid, 4-hydroxycinnamic acid, and ferulic acid), 1 coumarin (scopoletin), and 3 flavonoids (kaempferol-3-O-rutinoside, rutin, and narcissoside). It was showed that rutin was the most dominant phenolic compound in LBQ, although the average content of 4 phenolic acids was also high in LBQ, and scopoletin was the richest in LBG. UHPLC-Q-TOF-MS was used to qualitatively analyze the phenolics, which showed LBN was abundant in phenolic acids, LBQ was rich in flavonoids, and coumarins were the most plentiful in LBG. In conclusion, this study can provide references for the quality control and evaluation of phenolics in LB fruits and their by-products.

    Abstract: This study aimed to compare the effect of hot roller (HR) drying and hot air (HA) drying on the sensory evaluation, chemical quality, antioxidant activity, and metabolic profile of Yihong Congou black tea processed from E’cha NO1. The Yihong Congou black tea dried with HA obtained higher sensory scores and better chemical qualities such as the hue of tea brew color (a and b), content of theaflavins, thearubigins, water extract, free amino acids, tea polyphenol, and the ratio of polyphenol to amino acids as well as higher antioxidant capacities compared to that dried with HR. The HA drying tea increased the contents of volatile compounds that had positive correlation with sweet and flowery flavor, while the HR drying tea increased the contents of volatile compounds related to fruity flavor. Moreover, non-targeted metabolomics data indicated that the levels of most free amino acids significantly increased, while the levels of most soluble sugars reduced in the HA drying method compared to the HR drying method. The metabolic analysis was also consistent with the above results and revealed that D-ribose and gallic acid were the main characteristic metabolites of HA drying. Our results could provide a technical reference and theoretical guide to processing a high quality of Yihong Congou black tea.

    Abstract: This study investigated the effects of microwave modification, alkali polyphenol (ferulic acid) covalently combined modification, and microwave-alkali polyphenol covalently combined modification on the gel properties of soy protein emulsions. The results showed that the properties of soy protein emulsions were improved significantly by the three modification methods. After three kinds of modification, the viscoelasticity of soy protein emulsion gel increased, and a gel system with stronger elasticity was formed. The texture, water-holding, and hydration properties of the emulsion gel increased significantly. The SEM and ClSM results showed that the modified soy protein emulsion gel had a more compact and uniform porous structure, and the oil droplets could be better embedded in the network structure of the gel. Among the three modification methods, the microwave-alkali method polyphenol covalently combining the compound modification effect was best, and the microwave modification effect was least effective compared to the other two methods. Our obtained results suggested that for gel property modification of soy protein emulsion gels, microwave pretreatment combined with the covalent binding of polyphenols by an alkaline method is an effective method.

    Abstract: Subirrigation (SI), where water is provided to container seedlings from below and rises through the growing media via capillary action, is regarded as an environmentally-responsible method of delivering water and fertilizer to nursery-grown plants, resulting in more uniform crops and improved production efficiency. While a concern around adopting this method is that a potential higher salt concentration in the upper layers of growing media under SI may inhibit root growth and result in decreased plant quality, few studies have focused on how root morphology is altered by SI. Therefore, a balanced two-factor factorial design with three rates of fertilization (50, 100, and 150 mg N seedling−1) and two irrigation methods (SI or overhead irrigation (OI)) was used to examine the growth response of Prince Rupprecht’s larch (Larix principis-rupprechtii Mayr) seedlings for one nursery season. Associated changes between rhizosphere electrical conductivity (EC) and root morphology of different root size classes were analyzed. Results show that (1) height, root-collar diameter, and root volume were similar between seedlings grown under SI and OI. However, (2) compared to seedlings receiving OI, SI-seedlings had less root mass, length, and surface area but greater average root diameter (ARD). (3) Morphological differences were evident primarily in root diameter size classes I–III (D ≤ 1.0 mm). (4) Fertilizer rate influenced root length and surface area up to 130 days after sowing but affected ARD throughout the growing season such that seedlings treated with 50 mg N had smaller ARD than seedlings treated with 100 mg N. (5) As the growing season progressed, SI-media had significantly higher EC compared to OI-media and EC increased with increasing fertilizer rate under SI but not under OI. These results indicate that SI can produce larch seedlings of similar height and root collar diameter (RCD) compared to OI, but root systems are smaller overall with fewer small-diameter roots, which may be related to high EC levels in SI-media, which is exacerbated by the use of high rates of fertilizer. Therefore, the EC in the media should be monitored and adjusted by reducing fertilizer rates under SI.

    Abstract: The shape of the leaf is the primary phenotype which determines the commercial value of leaf mustard (Brassica juncea). However, there arefew reports on the lobed-leaf gene of B. juncea, and the molecular regulatory mechanisms underlying leaf margin formation are unknown. In this study, an F2 population derived from ‘MN001’ and ‘MU056’ was constructed. Genetic analysis revealed that the lobed-leaf trait is controlled by a major gene, and lobed leavesare dominant compared to round leaves. The GradedPool-Seq analysis combined with the re-sequencing results of parents identified a major interval on chromosome 10 of B. juncea’s genome A. The BjLMI1 gene (BjuA040054) was confirmed to be a candidate gene by gene ontology (GO) analysis, and it is homologous with LMI1 and encodes HD-Zip protein ATHB-51. A base substitution was observed in the conserved domain, and a 63 bp fragment deletion was found in the exon region between the two parents in the CDs region. The expression of BjLMI1 was significantly higher in the lobed-leaf parent than in the round-leaf parent. These findings provide insights into the molecular mechanism underlying leaf margin formation and will be valuable in the development of an ideal leaf shape in B. juncea.
