    Abstract: Seed aging detection and viable seed prediction are of great significance in alfalfa seed production, but traditional methods are disposable and destructive. Therefore, the establishment of a rapid and non-destructive seed screening method is necessary in seed industry and research. In this study, we used multispectral imaging technology to collect morphological features and spectral traits of aging alfalfa seeds with different storage years. Then, we employed five multivariate analysis methods, i.e., principal component analysis (PCA), linear discrimination analysis (LDA), support vector machines (SVM), random forest (RF) and normalized canonical discriminant analysis (nCDA) to predict aged and viable seeds. The results revealed that the mean light reflectance was significantly different at 450~690 nm between non-aged and aged seeds. LDA model held high accuracy (99.8~100.0%) in distinguishing aged seeds from non-aged seeds, higher than those of SVM (87.4~99.3%) and RF (84.6~99.3%). Furthermore, dead seeds could be distinguished from the aged seeds, with accuracies of 69.7%, 72.0% and 97.6% in RF, SVM and LDA, respectively. The accuracy of nCDA in predicting the germination of aged seeds ranged from 75.0% to 100.0%. In summary, we described a nondestructive, rapid and high-throughput approach to screen aged seeds with various viabilities in alfalfa.

    Abstract: Akkermansia muciniphila, a potential probiotic, has been proven to lessen the effects of several diseases. As established, the relative abundance of Akkermansia is positively correlated with tryptophan metabolism. However, the reciprocal interaction between tryptophan and Akkemansia is still unclear. Herein, for the first time, the possible effects of tryptophan and its derived metabolites on A. muciniphila were preliminarily investigated, including growth, physiological function, and metabolism. Obtained results suggested that 0.4 g/L of tryptophan treatment could significantly promote the growth of A. muciniphila. Notably, when grown in BHI with 0.8 g/L of tryptophan, the hydrophobicity and adhesion of A. muciniphila were significantly improved, potentially due to the increase in the rate of cell division. Furthermore, A. muciniphila metabolized tryptophan to indole, indole-3-acetic acid, indole-3-carboxaldehyde, and indole-3-lactic acid. Indoles produced by gut microbiota could significantly promote the growth of A. muciniphila. These results could provide a valuable reference for future research on the relationship between tryptophan metabolism and A. muciniphila.

    Abstract: Alfalfa (Medicago sativa) is a high-quality legume forage crop worldwide, and alfalfa production is often threatened by abiotic environmental stresses. GRAS proteins are important transcription factors that play a vital role in plant development, as well as in response to environmental stress. In this study, the availability of alfalfa genome “Zhongmu No.1” allowed us to identify 51 GRAS family members, i.e., MsGRAS. MsGRAS proteins could be classified into nine subgroups with distinct conserved domains, and tandem and segmental duplications were observed as an expansion strategy of this gene family. In RNA-Seq analysis, 14 MsGRAS genes were not expressed in the leaf or root, 6 GRAS genes in 3 differentially expressed gene clusters were involved in the salinity stress response in the leaf. Moreover, qRT-PCR results confirmed that MsGRAS51 expression was induced under drought stress and hormone treatments (ABA, GA and IAA) but down-regulated in salinity stress. Collectively, our genome-wide characterization, evolutionary, and expression analysis suggested that the MsGRAS proteins might play crucial roles in response to abiotic stresses and hormonal cues in alfalfa. For the breeding of alfalfa, it provided important information on stress resistance and functional studies on MsGRAS and hormone signaling.

    Abstract: Pomegranate (Punica granatum L.) is a kind of fruit with significant economic, ecological and health values. AP2/ERF transcription factors belong to a large group of factors mainly found in plants and play key roles in plant growth and development. However, AP2/ERF genes in pomegranate and their implication in development and postharvest preservation have been little described. In this study, 116 PgAP2/ERF genes in pomegranate were identified and renamed based on their chromosomal distributions. Phylogenetic relationship with genes from other species, structures, duplications, annotations, cis-elements in promoter sequences, and protein-protein interaction networks among PgAP2/ERF proteins were comprehensively explored. Expression analysis revealed several PgAP2/ERFs associated with the phenotypes of pomegranate seed hardness, including PgAP2/ERF5, PgAP2/ERF36, PgAP2/ERF58, and PgAP2/ERF86. Subsequent analysis indicated that many differentially expressed PgAP2/ERF genes are potentially important regulators of pomegranate fruit development. Furthermore, expression of more than one-half of PgAP2/ERFs was repressed in ‘Tunisian soft seed’ pomegranate fruit under low-temperature cold storage. The results showed that 1-MCP implicated in promoting postharvest preservation of ‘Tunisian soft seed’ pomegranate upregulated the PgAP2/ERF4, PgAP2/ERF15, PgAP2/ERF26, PgAP2/ERF30, PgAP2/ERF35 and PgAP2/ERF45 genes compared to those under low-temperature cold storage. This indicates that these genes are important candidate genes involved in pomegranate postharvest preservation. In summary, the findings of the present study provide an important basis for characterizing the PgAP2/ERF family genes and provide information on the candidate genes involved in pomegranate fruit development and postharvest preservation.

    Abstract: Overweight or obesity is related to a decrease in cognitive control, especially conflict control. However, research on conflict control in overweight/obese individuals are still controversial. This study was conducted to explore general and food-related conflict control in overweight Chinese females (OWs) with a color–word Stroop task and a food-related conflict task. Event-related potentials (ERPs) were recorded during the food-related conflict task. Behavioral results showed that, OWs had a longer reaction time (RT) than normal-weight Chinese females (NWs), in both tasks. ERP results in the food-related conflict task showed that there was a reduction of N2 and N450 response strength in OWs, and the P3 and late positive component (LPC) response strength was enhanced. Results indicated that OWs might be less efficient in monitoring and resolving conflict, and OWs tended to have a higher motivational or emotional involvement in processing food-related stimuli, which was likely to contribute to their difficulty in losing weight.
