    Abstract: The benefits of traveling for older adults are extensively supported in the literature. Online mapping technology (OMT) is one of the most widely used applications by people during traveling. This study aimed to obtain insight into the acceptance of OMT among older adults. Additionally, an OMT acceptance model for older adults was developed in this study by integrating facilitating condition (FC), compatibility (COM), and self-satisfaction (SS) into the technology acceptance model (TAM). In this study, structural equation modeling was applied to the test of the OMT acceptance model. This study adopted a cross-sectional structured questionnaire survey for collecting quantitative data from older adults in China. Four hundred and sixteen Chinese older adults were involved in this survey. This study found that TAM was useful to explain the OMT acceptance among older adults. Additionally, FC was confirmed to be a positive factor in determining the perceived ease of use, while COM and SS were found to positively influence perceived usefulness. The results of this study are helpful for OMT developers to design OMT and adopt measures to enhance the use of OMT among older adults, thereby increasing their travel frequency.

    Abstract: Rett syndrome (RTT) is a severe neurodevelopmental disorder caused by MeCP2 mutations. Nonetheless, the pathophysiological roles of MeCP2 mutations in the etiology of intrinsic cardiac abnormality and sudden death remain unclear. In this study, we performed a detailed functional studies (calcium and electrophysiological analysis) and RNA-sequencing-based transcriptome analysis of a pair of isogenic RTT female patient-specific induced pluripotent stem-cell-derived cardiomyocytes (iPSC-CMs) that expressed either MeCP2wildtype or MeCP2mutant allele and iPSC-CMs from a non-affected female control. The observations were further confirmed by additional experiments, including Wnt signaling inhibitor treatment, siRNA-based gene silencing, and ion channel blockade. Compared with MeCP2wildtype and control iPSC-CMs, MeCP2mutant iPSC-CMs exhibited prolonged action potential and increased frequency of spontaneous early after polarization. RNA sequencing analysis revealed up-regulation of various Wnt family genes in MeCP2mutant iPSC-CMs. Treatment of MeCP2mutant iPSC-CMs with a Wnt inhibitor XAV939 significantly decreased the β-catenin protein level and CACN1AC expression and ameliorated their abnormal electrophysiological properties. In summary, our data provide novel insight into the contribution of activation of the Wnt/β-catenin signaling cascade to the cardiac abnormalities associated with MeCP2 mutations in RTT.

    Abstract: Chemo-resistance hinders treatment of patients with hepatocellular carcinoma. Although there are many models that can be found in the literature, the root mechanism to explain chemo-resistance is still not fully understood. To gain a better understanding of this phenomenon, a chemo-resistant line, R-HepG2, was developed from a chemo-sensitive HepG2 line through an exposure of doxorubicin (DOX). The R-HepG2 exhibited a cancer stem cell (CSC) phenotype with an over-expression of P-glycoprotein (P-gp), conferring it a significant enhancement in drug efflux and survival. With these observations, we hypothesize that metabolic alteration in this drug-resistant CSC is the root cause of chemo-resistance. Our results show that, unlike other metabolic-reprogrammed CSCs that exhibit glycolytic phenotype described by the “Warburg effect”, the R-HepG2 was metabolically quiescent with glucose independence, high metabolic plasticity, and relied on glutamine metabolism via the mitochondria for its chemo-resistance Intriguingly, drug efflux by P-gp in R-HepG2 depended on the mitochondrial ATP fueled by glutamine instead of glycolytic ATP. Armed with these observations, we blocked the glutamine metabolism in the R-HepG2 and a significant reduction of DOX efflux was obtained. We exploited this metabolic vulnerability using a combination of DOX and metformin in a glutamine-free condition to target the R-HepG2, resulting in a significant DOX sensitization. In conclusion, our findings highlight the metabolic modulation of chemo-resistance in CSCs. We delineate the altered metabolism that drives chemo-resistance and offer a new approach to target this CSC through metabolic interventions.

    Abstract: Cogeneration systems—also known as combined heat and power systems—form a promising technology for the simultaneous generation of power and thermal energy while consuming a single source of fuel at a site. A number of prior studies have examined the cogeneration systems used in residential, commercial, and industrial buildings. However, a systematic review of the economic and environmental evaluations of the system is not found in the literature. The present study aims to address this research gap by reviewing the most relevant studies on the cogeneration systems applied to buildings in different contexts (e.g., residential, commercial, and industrial) and provides systematic evaluation approaches from economic and environmental perspectives. Results show that the cogeneration system can significantly reduce energy consumption, operating costs, carbon dioxide equivalent emissions, and positive performance on other relevant parameters. The present study provides extensive evidence to show that the cogeneration system is simultaneously economically profitable and environmentally friendly in various application contexts. To achieve the maximum benefits from cogeneration systems, several practical suggestions are provided for their successful installation and implementation in real-life situations.

    Abstract: Truck-related accidents account for a substantial portion of traffic accidents. Risky driving behavior is a main cause of traffic accidents. Understanding the risky driving behavior of truck drivers is therefore important in reducing truck-related accidents. This study aimed to propose and validate a research model that integrated a theory of planned behavior, sensation seeking, and risk perception to explain the risky driving behavior of truck drivers. A total of 471 valid data were collected from Chinese truck drivers in this study. Structural equation modeling and mediation analysis were used to examine the influence of factors in the research model on the risky driving behavior of truck drivers. Results showed that sensation seeking and risk perception of truck drivers were influential in shaping their intention to drive riskily with the mediation of attitude toward risky driving. Risk perception and attitude toward risky driving also had a negative influence and positive influence on the intention, respectively. On the basis of the findings, practical recommendations for reducing the risky driving behavior of truck drivers were provided for concerned parties.

    Abstract: The use of cogeneration systems in the hotel industry leads to economic and environmental benefits, but its acceptance in the industry remains low. Hence, this study aimed to examine the factors that influence cogeneration system acceptance amongst hotel management. A cogeneration system acceptance model (CoSAM) was proposed by integrating the technology acceptance model with perceived cost, perceived benefit, risk perception, environmental awareness and facilitating conditions. The validity of the CoSAM was investigated using structural equation modelling based on 499 data points collected from hotel management personnel. Results showed that the intention to use the systems of hotel management personnel was positively determined by attitude towards using cogeneration systems, which was directly affected by perceived usefulness, risk perception and perceived benefit. Moreover, with perceived usefulness as a mediator, facilitating conditions and environmental awareness indirectly influenced attitude towards using cogeneration systems positively, while perceived cost indirectly influenced the attitude negatively. Based on the findings of this study, policy implications for promoting the adoption of cogeneration systems in the hotel industry were provided, thus saving energy and reducing the energy costs of hotels. This study is the first to remarkably contribute an in-depth understanding of the factors affecting cogeneration system acceptance to the literature.

    Abstract: High accident rates have been a complicated and persistent problem in the Hong Kong construction industry. This situation has stimulated this investigation into factors that influence the risk-taking propensity of construction workers. However, interviewing workers who had a bad experience is problematic because changes in attitude and perception may occur as a result of such an experience. Using quasi-expert interviews can reduce this problem. The objective of this study was to identify factors that influence the risk-taking propensity of construction workers. Semi-structured interviews were conducted with 16 safety professionals all with accident inspection experience and six super-safe workers with no incident record for the past five years. Seven factors that affect the risk-taking propensity of construction workers were successfully identified. Each factor is thoughtfully discussed, and this study shows that quasi-expert interview is a pragmatic approach for deepening the understanding of risk-taking propensity among construction workers. Findings of this study will hopefully help and encourage further quantitative research on the risk-taking propensity of construction workers with different perspectives.

    Abstract: Risk taking among construction workers is a critical topic in construction safety research. The aim of this study was to empirically investigate how optimism bias and safety climate influence construction worker risk-taking behavior. A survey with a designed questionnaire was conducted to collect data from construction workers. A total of 183 construction workers participated in this study and completed the designed questionnaire. The collected data were subjected to statistical analysis by using structural equation modeling. Results show that optimism bias related to work risks positively influences construction worker risk-taking behavior, whereas safety climate and optimism bias related to hazard perception skills negatively affect the risk-taking behavior. These findings can enrich the literature on construction worker risk-taking behavior from the perspective of optimism bias and safety climate. Practical implications are provided for discouraging construction workers from taking risks at work.
