    Abstract: This study investigated a method for improving the quality of images with a low number of excitations (NEXs) based on deep learning using T2-weighted magnetic resonance imaging (MRI) of the heads of normal Wistar rats to achieve higher image quality and a shorter acquisition time. A 7T MRI was used to acquire T2-weighted images of the whole brain with NEXs = 2, 4, 8, and 12. As a preprocessing step, non-rigid registration of the acquired low NEX images (NEXs = 2, 4, 8) and NEXs = 12 images was performed. A residual dense network (RDN) was used for training. A low NEX image was used as the input to the RDN, and the NEX12 image was used as the correct image. For quantitative evaluation, we measured the signal-to-noise ratio (SNR), peak SNR, and structural similarity index measure of the original image and the image obtained by RDN. The NEX2 results are presented as an example. The SNR of the cortex was 10.4 for NEX2, whereas the SNR of the image reconstructed with RDN for NEX2 was 32.1. (The SNR NEX12 was 19.6) In addition, the PSNR in NEX2 was significantly increased to 35.4 ± 2.0 compared to the input image and to 37.6 ± 2.9 compared to the reconstructed image (p = 0.05). The SSIM in NEX2 was 0.78 ± 0.05 compared to the input image and 0.91 ± 0.05 compared to the reconstructed image (p = 0.0003). Furthermore, NEX2 succeeded in reducing the shooting time by 83%. Therefore, in preclinical 7T MRI, supervised learning between the NEXs using RDNs can potentially improve the image quality of low NEX images and shorten the acquisition time.

    Abstract: This study aimed to evaluate cardiac function in a young mouse model of Duchenne muscular dystrophy (mdx) using cardiac magnetic resonance imaging (MRI) with feature tracking and self-gated magnetic resonance cine imaging. Cardiac function was evaluated in mdx and control mice (C57BL/6JJmsSlc mice) at 8 and 12 weeks of age. Preclinical 7-T MRI was used to capture short-axis, longitudinal two-chamber view and longitudinal four-chamber view cine images of mdx and control mice. Strain values were measured and evaluated from cine images acquired using the feature tracking method. The left ventricular ejection fraction was significantly less (p 〈 0.01 each) in the mdx group at both 8 (control, 56.6 ± 2.3% mdx, 47.2 ± 7.4%) and 12 weeks (control, 53.9 ± 3.3% mdx, 44.1 ± 2.7%). In the strain analysis, all strain value peaks were significantly less in mdx mice, except for the longitudinal strain of the four-chamber view at both 8 and 12 weeks of age. Strain analysis with feature tracking and self-gated magnetic resonance cine imaging is useful for assessing cardiac function in young mdx mice.

    Abstract: The aim of this paper was to assess the associations between prostate cancer aggressiveness and histogram-derived apparent diffusion coefficient (ADC) parameters and determine which ADC parameters may help distinguish among stromal hyperplasia (SH), glandular hyperplasia (GH), and low-grade, intermediate-grade, and high-grade prostate cancers. The mean, median, minimum, maximum, and 10th and 25th percentile ADC values were determined from the ADC histogram and compared among two benign prostate hyperplasia (BPH) groups and three Gleason score (GS) groups. Seventy lesions were identified in 58 patients who had undergone proctectomy. Thirty-nine lesions were prostate cancers (GS 6 = 7 lesions, GS 7 = 19 lesions, GS 8 = 11 lesions, GS 9 = 2 lesions), and thirty-one lesions were BPH (SH = 15 lesions, GH = 16 lesions). There were statistically significant differences in 10th percentile and 25th percentile ADC values when comparing GS 6 to GS 7 (p 〈 0.05). The 10th percentile ADC values yielded the highest area under the curve (AUC). Tenth and 25th percentile ADCs can be used to more accurately differentiate lesions with GS 6 from those with GS 7 than other ADC parameters. Our data indicate that the major challenge with ADC mapping is to differentiate between SH and GS 6, and SH and GS 7.

    Abstract: Chemical exchange saturation transfer (CEST) imaging is a non-invasive molecular imaging technique for indirectly measuring low-concentration endogenous metabolites. Conventional CEST has low specificity, owing to the effects of spillover, magnetization transfer (MT), and T1 relaxation, thus necessitating an inverse Z-spectrum analysis. We aimed to investigate the usefulness of inverse Z-spectrum analysis in creatine (Cr)-CEST in mice, by conducting preclinical 7T-magnetic resonance imaging (MRI) and comparing the conventional analysis metric magnetization transfer ratio (MTRconv) with the novel metric apparent exchange-dependent relaxation (AREX). We performed Cr-CEST imaging using 7T-MRI on mouse testes, using C57BL/6 mice as the control and a cisplatin-treated model. We prepared different doses of cisplatin to observe its dose dependence effect on testicular function. CEST imaging was obtained using an MT pulse with varying saturation frequencies, ranging from −4.8 ppm to +4.8 ppm. The application of control mouse testes improved the specificity of the CEST effect and image contrast between the testes and testicular epithelium. The cisplatin-treated model revealed impaired testicular function, and the Cr-CEST imaging displayed decreased Cr levels in the testes. There was a significant difference between the low- and high-dose models. The MTR values of Cr-CEST reflected the cisplatin dose dependence of testicular dysfunction.

    Abstract: Pelizaeus–Merzbacher disease (PMD) is an X-linked recessive disorder of the central nervous system. We performed 7 Tesla magnetic resonance imaging of the brain in Tama rats, a rodent PMD model, and control rats, as well as evaluated the diagnostic values. In the white matter of the Tama rats, the T2 values were prolonged, which is similar to that observed in patients with PMD (60.7 ± 1.8 ms vs. 51.6 ± 1.3 ms, p 〈 0.0001). The apparent diffusion coefficient values in the white matter of the Tama rats were higher than those of the control rats (0.68 ± 0.03 × 10−3 mm2/s vs. 0.64 ± 0.03 × 10−3 mm2/s, p 〈 0.05). In proton magnetic resonance spectroscopy, the N-acetylaspartate (6.97 ± 0.12 mM vs. 5.98 ± 0.25 mM, p 〈 0.01) and N-acetylaspartate + N-acetylaspartylglutamate values of the Tama rats were higher (8.22 ± 0.17 mM vs. 7.14 ± 0.35 mM, p 〈 0.01) than those of the control rats. The glycerophosphocholine + phosphocholine values of the Tama rats were lower than those of the control rats (1.04 ± 0.09 mM vs. 1.45 ± 0.04 mM, p 〈 0.001). By using Luxol fast blue staining, we confirmed dysmyelination in the Tama rats. These results are similar to those of patients with PMD and other PMD animal models.

    Abstract: Ribozymes are catalytic RNAs that are attractive platforms for the construction of nanoscale objects with biological functions. We designed a dimeric form of the Tetrahymena group I ribozyme as a unit structure in which two ribozymes were connected in a tail-to-tail manner with a linker element. We introduced a kink-turn motif as a bent linker element of the ribozyme dimer to design a closed trimer with a triangular shape. The oligomeric states of the resulting ribozyme dimers (kUrds) were analyzed biochemically and observed directly by atomic force microscopy (AFM). Formation of kUrd oligomers also triggered trans-splicing reactions, which could be monitored with a reporter system to yield a fluorescent RNA aptamer as the trans-splicing product.

    Abstract: This study validates the usefulness of myocardial strain analysis with cardiac cine magnetic resonance imaging (MRI) by evaluating the changes in the cardiac function and myocardial strain values longitudinally in a myocardial disease model. Six eight-week-old male Wistar rats were used as a model of myocardial infarction (MI). Cine images were taken in the short axis, two-chamber view longitudinal axis, and four-chamber view longitudinal axis directions in rats 3 and 9 days after MI and in control rats, with preclinical 7-T MRI. The control images and the images on days 3 and 9 were evaluated by measuring the ventricular ejection fraction (EF) and the strain values in the circumferential (CS), radial (RS), and longitudinal directions (LS). The CS decreased significantly 3 days after MI, but there was no difference between the images on days 3 and 9. The two-chamber view LS was −9.7 ± 2.1% at 3 days and −13.9 ± 1.4% at 9 days after MI. The four-chamber view LS was −9.9 ± 1.5% at 3 days and −11.9 ± 1.3% at 9 days after MI. Both the two- and four-chamber LS values were significantly decreased 3 days after MI. Myocardial strain analysis is, therefore, useful for assessing the pathophysiology of MI.

    Abstract: This study aimed to evaluate an intracerebral hemorrhage (ICH) model using quantitative susceptibility mapping (QSM) and chemical exchange saturation transfer (CEST) with preclinical 7T-magnetic resonance imaging (MRI) and determine the potential of amide proton transfer-CEST (APT-CEST) for use as a biomarker for the early detection of ICH. Six Wistar male rats underwent MRI, and another six underwent histopathological examinations on postoperative days 0, 3, and 7. The ICH model was created by injecting bacterial collagenase into the right hemisphere of the brain. QSM and APT-CEST MRI were performed using horizontal 7T-MRI. Histological studies were performed to observe ICH and detect iron deposition at the ICH site. T2-weighted images (T2WI) revealed signal changes associated with hemoglobin degeneration in red blood cells, indicating acute-phase hemorrhage on day 0, late-subacute-phase hemorrhage on day 3, and chronic-phase hemorrhage on day 7. The susceptibility alterations in each phase were detected using QSM. QSM and Berlin blue staining revealed hemosiderin deposition in the chronic phase. APT-CEST revealed high magnetization transfer ratios in the acute phase. Abundant mobile proteins and peptides were observed in early ICH, which were subsequently diluted. APT-CEST imaging may be a reliable noninvasive biomarker for the early diagnosis of ICH.

    Abstract: Quantitative susceptibility mapping (QSM) is used to obtain quantitative magnetic susceptibility maps of materials from magnitude and phase images acquired by three-dimensional gradient-echo using inverse problem-solving. Few preclinical studies have evaluated the intracerebral hemorrhage (ICH) model and asymmetric iron deposition. We created a rat model of ICH and compared QSM and conventional magnetic resonance imaging (MRI) during the longitudinal evaluation of ICH. Collagenase was injected in the right striatum of 12-week-old Wistar rats. QSM and conventional MRI were performed on days 0, 1, 7, and 28 after surgery using 7-Tesla MRI. Susceptibility, normalized signal value, and area of the hemorrhage site were statistically compared during image analysis. Susceptibility decreased monotonically up to day 7 but increased on day 28. Other imaging methods showed a significant increase in signal from day 0 to day 1 but a decreasing trend after day 1. During the area evaluation, conventional MRI methods showed an increase from day 0 to day 1; however, decreases were observed thereafter. QSM showed a significant increase from day 0 to day 1. The temporal evaluation of ICH by QSM suggested the possibility of detecting of asymmetric iron deposition for normal brain site.

    Abstract: The asymmetric development of the cerebellum has been reported in several mammalian species. The current study quantitatively characterized cerebellar asymmetry and sexual dimorphism in cynomolgus macaques using magnetic resonance (MR) imaging-based volumetry. Three-dimensional T1W MR images at 7-tesla were acquired ex vivo from fixed adult male (n = 5) and female (n = 5) monkey brains. Five transverse domains of the cerebellar cortex, known as cerebellar compartmentation defined by the zebrin II/aldolase expression pattern, were segmented on MR images, and the left and right sides of their volumes were calculated. Asymmetry quotient (AQ) analysis revealed significant left-lateralization at the population level in the central zone posterior to the cerebellar transverse domains, which included lobule VII of the vermis with the crura I and II of ansiform lobules, in males but not females. Next, the volume of the cerebellar hemispherical lobules was calculated. Population-level leftward asymmetry was revealed in the crus II regions in males using AQ analysis. The AQ values of the other hemispherical lobules showed no left/right side differences at the population level in either sex. The present findings suggest a sexually dimorphic asymmetric aspect of the cerebellum in cynomolgus macaques, characterized by a leftward lateralization of the crus II region in males, but no left/right bias in females.
