Abstract: The Asia-Pacific Lightning Location Network (APLLN) is a lightning location system consisting of a series of very low-frequency signal detection sites. Since 2018, 16 detection sites have been deployed with an average baseline longer than 1000 km. The detection site used a trigger sampling method to record the lightning signal with a duration of 2 ms and calculates the lightning arrival time based on digital filtering and the Hilbert envelope method. APLLN used a time difference location algorithm and improved Levenberg–Marquardt non-linear least squares iterative algorithm to calculate and optimize the lightning location results. The analysis results of a strong thunderstorm process show that the average detection efficiency of APLLN was 55.34% for intracloud (IC) strokes, 63.55% for cloud-to-ground (CG) strokes and 61.83% for all strokes (IC + CG). The average location error of APLLN for this thunderstorm is 5–10 km.

Abstract: Lightning waveform plays an important role in lightning observation, location, and lightning disaster investigation. Based on a large amount of lightning waveform data provided by existing real-time very low frequency/low frequency (VLF/LF) lightning waveform acquisition equipment, an automatic and accurate lightning waveform classification method becomes extremely important. With the widespread application of deep learning in image and speech recognition, it becomes possible to use deep learning to classify lightning waveforms. In this study, 50,000 lightning waveform samples were collected. The data was divided into the following categories: positive cloud ground flash, negative cloud ground flash, cloud ground flash with ionosphere reflection signal, positive narrow bipolar event, negative narrow bipolar event, positive pre-breakdown process, negative pre-breakdown process, continuous multi-pulse cloud flash, bipolar pulse, skywave. A multi-layer one-dimensional convolutional neural network (1D-CNN) was designed to automatically extract VLF/LF lightning waveform features and distinguish lightning waveforms. The model achieved an overall accuracy of 99.11% in the lightning dataset and overall accuracy of 97.55% in a thunderstorm process. Considering its excellent performance, this model could be used in lightning sensors to assist in lightning monitoring and positioning.

Abstract: Differentiation from preadipocytes into mature adipocytes is a complex biological process in which miRNAs play an important role. Previous studies showed that miR-214-3p facilitates adipocyte differentiation of bone marrow-derived mesenchymal stem cells (BMSCs) in vitro. The detailed function and molecular mechanism of miR-214-3p in adipocyte development is unclear. In this study, the 3T3-L1 cell line was used to analyze the function of miR-214-3p in vitro. Using 5-Ethynyl-2′-deoxyuridine (EdU) staining and the CCK-8 assay, we observed that transfection with the miR-214-3p agomir visibly promoted proliferation of 3T3-L1 preadipocytes by up-regulating the expression of cell cycle-related genes. Interestingly, overexpression of miR-214-3p promoted 3T3-L1 preadipocyte differentiation and up-regulated the expression of key genes for lipogenesis: PPARγ, FABP4, and Adiponectin. Conversely, inhibition of miR-214-3p repressed 3T3-L1 preadipocyte proliferation and differentiation, and down-regulated the expression of cell cycle-related genes and adipogenic markers. Furthermore, we proved that miR-214-3p regulates 3T3-L1 preadipocyte differentiation by directly targeting the 3′-untranslated regions (3′UTR) of Ctnnb1, which is an important transcriptional regulatory factor of the Wnt/β-Catenin pathway. Taken together, the data indicate that miR-214-3p may positively regulate preadipocyte proliferation and enhance differentiation through the Wnt/β-Catenin signaling pathway.

Abstract: Background: We aimed to assess the extent to which drug persistence is better with non-vitamin K antagonist oral anticoagulants (NOACs) than vitamin K antagonists (VKAs) in atrial fibrillation (AF) patients and to estimate the difference in therapy persistence depending on NOAC dosing regimen (once daily (QD) vs. twice daily (BID)). Methods: Consecutive patients were followed for 1 year in phase III of the GLORIA-AF registry. Drug persistence was defined as the use of OAC without any discontinuation in 〉 30 days or switching to alternative therapy. Results: Among 21,109 eligible patients in phase III, 17,266 patients who were prescribed OAC at baseline and those who took ≥1 OAC dose were included. The 1-year proportion of patients receiving NOAC and VKA who persisted on treatment was 80% and 75%, respectively. The 1-year persistence with NOACs BID and NOACs QD was 81% and 80%, respectively. Female gender, hypertension, older age, alcohol use, permanent, asymptomatic, and minimally symptomatic AF were associated with better OAC persistence. Region, medication usage predisposing to bleeding, being a current smoker, treatment reimbursement, and proton pump inhibitors were associated with lower OAC persistence. Conclusions: Drug persistence was higher with NOACs (1-year persistence was 80%) than with VKAs (75%). There was little difference in 1-year persistence between NOAC dosing regimens.

Abstract: The preoperative differentiation of breast phyllodes tumors (PTs) from fibroadenomas (FAs) plays a critical role in identifying an appropriate surgical treatment. Although several imaging modalities are available, reliable differentiation between PT and FA remains a great challenge for radiologists in clinical work. Artificial intelligence (AI)-assisted diagnosis has shown promise in distinguishing PT from FA. However, a very small sample size was adopted in previous studies. In this work, we retrospectively enrolled 656 breast tumors (372 FAs and 284 PTs) with 1945 ultrasound images in total. Two experienced ultrasound physicians independently evaluated the ultrasound images. Meanwhile, three deep-learning models (i.e., ResNet, VGG, and GoogLeNet) were applied to classify FAs and PTs. The robustness of the models was evaluated by fivefold cross validation. The performance of each model was assessed by using the receiver operating characteristic (ROC) curve. The area under the curve (AUC), accuracy, sensitivity, specificity, positive predictive value (PPV), and negative predictive value (NPV) were also calculated. Among the three models, the ResNet model yielded the highest AUC value, of 0.91, with an accuracy value of 95.3%, a sensitivity value of 96.2%, and a specificity value of 94.7% in the testing data set. In contrast, the two physicians yielded an average AUC value of 0.69, an accuracy value of 70.7%, a sensitivity value of 54.4%, and a specificity value of 53.2%. Our findings indicate that the diagnostic performance of deep learning is better than that of physicians in the distinction of PTs from FAs. This further suggests that AI is a valuable tool for aiding clinical diagnosis, thereby advancing precision therapy.

Abstract: Mapping the spatial variation of forest aboveground biomass (AGB) at the national or regional scale is important for estimating carbon emissions and removals and contributing to global stocktake and balancing the carbon budget. Recently, several gridded forest AGB products have been produced for China by integrating remote sensing data and field measurements, yet significant discrepancies remain among these products in their estimated AGB carbon, varying from 5.04 to 9.81 Pg C. To reduce this uncertainty, here, we first compiled independent, high-quality field measurements of AGB using a systematic and consistent protocol across China from 2011 to 2015. We applied two different approaches, an optimal weighting technique (WT) and a random forest regression method (RF), to develop two observationally constrained hybrid forest AGB products in China by integrating five existing AGB products. The WT method uses a linear combination of the five existing AGB products with weightings that minimize biases with respect to the field measurements, and the RF method uses decision trees to predict a hybrid AGB map by minimizing the bias and variance with respect to the field measurements. The forest AGB stock in China was 7.73 Pg C for the WT estimates and 8.13 Pg C for the RF estimates. Evaluation with the field measurements showed that the two hybrid AGB products had a lower RMSE (29.6 and 24.3 Mg/ha) and bias (−4.6 and −3.8 Mg/ha) than all five participating AGB datasets. Our study demonstrated both the WT and RF methods can be used to harmonize existing AGB maps with field measurements to improve the spatial variability and reduce the uncertainty of carbon stocks. The new spatial AGB maps of China can be used to improve estimates of carbon emissions and removals at the national and subnational scales.

Abstract: Late Jurassic NE-trending A-type granitoids are widespread in the Shihang belt, South China, though their petrogenesis and geodynamic settings remain controversial. The Guiping complex is located on the southwest margin of the Shihang belt. In this study, the petrography, major and trace element geochemistry, whole-rock Sr-Nd isotopes, and zircon U-Pb geochronology of the Guiping complex were investigated. The Guiping complex is composed of the Fenghuangling and Xishan plutons; both plutons yielded zircon U-Pb ages of ca. 160 Ma. The Fenghuangling pluton has low SiO2 content of 54.26% to 60.31%, whereas the Xishan pluton exhibits high SiO2 content of 65.19% to 71.18%. Both of them are metaluminous and belong to the high-K calc-alkaline series and are enriched in large-ion lithophile elements (LILEs) such as Rb, Th, U, and Pb. The Fenghuangling and Xishan plutons showed enrichment in light rare earth elements (LREEs) and high-field strength elements (Nb, Ta, Zr, and Hf) and depletion in heavy rare earth elements (HREEs). Marked Nb and Ta negative anomalies were not observed. Due to the high contents of Zr + Ce + Nb + Y and high Ga/Al ratios, all the samples belonged to the group of A-type granites. The Fenghuangling and Xishan plutons had low ISr (mainly in the range of 0.7046–0.7058) and high εNd(t) (−0.60 to 1.94) values, though obviously different from those of the Precambrian basement in South China. Furthermore, they lie between the ocean island basalt (OIB) of the asthenosphere and the arc basaltic rocks of the enriched lithospheric mantle. Therefore, we proposed that the basaltic parental magma of the Guiping complex originated from partial melting of the enriched lithospheric mantle, which was metasomatized by asthenosphere-related OIB-type basaltic magma. Mafic microgranular enclaves in the Xishan pluton displayed positive Nb and Ta anomalies, which is consistent with OIB-type basalts. The enclaves also had similar Sr-Nd isotopic compositions to the Xishan pluton. That indicated that the enclaves were probably formed by mixing of the OIB-type basaltic magma and the Xishan pluton. In conclusion, the formation of the Late Jurassic NE-trending A-type granite belt was attributed to back-arc extension as a result of the rollback of the Paleo-Pacific Plate.

Abstract: Negative symptoms represent an unmet need for schizophrenia treatment. The effect of theta frequency transcranial alternating current stimulation (theta-tACS) applied during working memory (WM) tasks on negative symptoms has not been demonstrated as of yet. We conducted a randomized, double-blind, sham-controlled trial of 36 stabilized schizophrenia patients, randomized to receive either twice daily, 6 Hz 2 mA, 20 min sessions of in-phase frontoparietal tACS or sham for five consecutive weekdays. Participants were concurrently engaged in WM tasks during stimulation. The primary outcome measure was the change over time in the Positive and Negative Syndrome Scale (PANSS) negative subscale score measured from baseline through to the 1-month follow-up. Secondary outcome measures were other symptom clusters, neurocognitive performance, and relevant outcomes. The intention-to-treat analysis demonstrated greater reductions in PANSS negative subscale scores at the end of stimulation in the active (−13.84%) than the sham (−3.78%) condition, with a large effect size (Cohen’s d = 0.96, p = 0.006). The positive effect endured for at least one month. Theta-tACS also showed efficacies for cognitive symptoms, WM capacity, and psychosocial functions. Online theta-tACS offers a novel approach to modulate frontoparietal networks to treat negative symptoms of schizophrenia. The promising results require large-scale replication studies in patients with predominantly negative symptoms.

Abstract: Reduced left-lateralized electroencephalographic (EEG) frontal alpha asymmetry (FAA), a biomarker for the imbalance of interhemispheric frontal activity and motivational disturbances, represents a neuropathological attribute of negative symptoms of schizophrenia. Unidirectional high-frequency transcranial random noise stimulation (hf-tRNS) can increase the excitability of the cortex beneath the stimulating electrode. Yet, it is unclear if hf-tRNS can modulate electroencephalographic FAA in patients with schizophrenia. We performed a randomized, double-blind, sham-controlled clinical trial to contrast hf-tRNS and sham stimulation for treating negative symptoms in 35 schizophrenia patients. We used electroencephalography to investigate if 10 sessions of hf-tRNS delivered twice-a-day for five consecutive weekdays would modulate electroencephalographic FAA in schizophrenia. EEG data were collected and FAA was expressed as the differences between common-log-transformed absolute power values of frontal right and left hemisphere electrodes in the alpha frequency range (8–12.5 Hz). We found that hf-tRNS significantly increased FAA during the first session of stimulation (p = 0.009) and at the 1-week follow-up (p = 0.004) relative to sham stimulation. However, FAA failed to predict and surrogate the improvement in the severity of negative symptoms with hf-tRNS intervention. Together, our findings suggest that modulating electroencephalographic frontal alpha asymmetry by using unidirectional hf-tRNS may play a key role in reducing negative symptoms in patients with schizophrenia.
