    Abstract: Toxoplasma gondii (T. gondii) is the most common zoonotic protozoa and has infected about one-third of the population worldwide. Recombinant epitopes encapsulated in nanospheres have advantages over traditional T. gondii vaccines. For an efficient delivery system, poly (DL-lactide-co-glycolide) (PLGA) and chitosan are the most frequently used biodegradable polymeric nanospheres with strong safety profiles. In the present study, we first expressed and purified histone H2A1 of T. gondii using the prokaryotic expression system. The effects of recombinant TgH2A1 on the functions of murine macrophages were then studied. Purified recombinant TgH2A1 was then encapsulated in nanospheres with PLGA and chitosan. After subcutaneous vaccination in mice, the immune response was evaluated by double antibody sandwich ELISA kits. The results from this study showed that PLGA and chitosan loaded with rTgH2A1 could trigger a stronger Th1 oriented immune response and prolong the survival time of mice effectively. In conclusion, PLGA and chitosan nanospheres loaded with histone H2A1 are an effective method for the development of vaccines against T. gondii. Further studies should focus on evaluating the regulatory mechanism of TgH2A1, vaccine potency, and cellular response in chronic T. gondii infections.

    Abstract: Grain filling can directly influence rice yield. However, there is limited information on the growth relationship among grains at different positions on the entire panicle during grain filling. In this study, field experiments were conducted in 2014–2015 to compare the growth dynamics of grains at various positions for two rice cultivars (Nongle 1 and Guifeng 2). The results showed that a high similarity and a slow–fast–slow trend of dry-matter accumulation occurred in all primary branches. However, the maximum grain growth rates of the top primary branches were 86% and 44% higher than basal primary branches of Nongle 1 and Guifeng 2, respectively. Similarly, the maximum final grain weights were 32% and 18% greater in the top primary branches than in the basal primary branches of Nongle 1 and Guifeng 2, respectively. In contrast, the active grain filling duration was 1.5 and 1.3 times longer in the basal primary branches than the top primary branches of Nongle 1 and Guifeng 2, respectively. The time to reach the maximum rate of grain growth of the basal primary branches for Nongle 1 and Guifeng 2 was 2.2 and 2.5 times longer than those of the top primary branches, respectively. Based on cluster analysis of growth characteristics of all primary branches, Group I (superior primary-branches) was considered to be the fastest for grain filling and greatest for dry matter weight, followed by Group II (medium primary-branches). The poorest growth occurred in Group III (inferior primary-branches). Therefore, the yield of poor-filling grains at the basal panicle could be achieved primarily by improving the growth of Group III.

    Abstract: Sweet potatoes have become increasingly popular among consumers due to their health benefits, and, as a result, sweet potato production has been growing rapidly over the last decade in the United States. However, the industry is facing major challenges, including the risk of disease outbreaks and adverse weather events, which could potentially have a significant impact on the market. However, the economic literature on the sweet potato commodity is limited. This study models the U.S. sweet potato market price response to supply changes and derives elasticity estimates. This information is essential for understanding the sweet potato market and for simulating the impacts of potential supply shocks, given the challenges that the industry is facing. We found that prices are highly sensitive to supply. North Carolina, the largest sweet potato producer in the country, dominates the domestic market and exerts significantly larger influences on market prices than other producing states.

    Abstract: Porous magnesium oxysulfate (MOS) cement pastes were successfully fabricated by injecting presaturated bentonite into modified MOS cement paste. Their pore structure and hardened performance were investigated. The results indicated that the 20MgO-MgSO4·7H2O-18H2O system modified by citric acid (C6H8O7⋅H2O) and ethylene diamine tetraacetic acid was suitable to fabricate porous MOS cement paste. Bentonite slurry led to significant refinement of pores, generating nanosized pores in MOS cement pastes. When volume replacement of bentonite slurry in MOS cement paste rose between 0 and 60%, pore size corresponding to the peak in the pore size distribution curve of MOS cement-based materials decreased from 180.0 nm to 22.8 nm and then increased to 163.0 nm, and the porosity linearly increased from 21.1% to 58.1%. These small pores caused the successful preparation of porous MOS cement paste with dry bulk density of 760–1650 kg/m3, compressive strength of 7.8–69.8 MPa, and thermal conductivity of 0.25–0.85 W/(m·K).

    Abstract: As the demand for organic products quickly grows in the U.S., the domestic supply of organic commodities has stagnated and failed to meet the increasing needs. To expand the organic supply capacity in the country, it is crucial to identify the key factors that could effectively promote the scale of organic farming in the U.S. Using a multivariate analytic model and state level data from multiple sources and years, this study examines a wide range of determinants that could affect organic agriculture. The results show that research funding would significantly increase organic vegetable production and the number of organic farms. The development of organic farming could be greatly encouraged by the support of research institutions. In addition, the evidence shows that abundant farm workers are essential for the organic farming sector, which is labor-intensive. Finally, a large population base could create a more stable consumer group that would promote the development of organic agriculture. These results suggest the importance of exploring and expanding consumer groups for organic commodities. These findings provide insightful implications that research support, labor availability, and a solid consumer base are crucial to boost the organic sector in the U.S. and other countries.

    Abstract: The aim of this meta-analysis was to systematically review the relationships between lactation diet and the composition of mature breast milk through screening multiple databases and gray literatures, with priority given to quantitative articles published in Chinese and English. We identified 27 cross-sectional studies that included 4355 lactating women. According to these data, dietary patterns were categorized into four patterns based on the fat and protein energy supplying ratio, including rational-fat and rational-protein dietary (RR), high-fat and high-protein dietary (HH), rational-fat and high-protein dietary (RH), and high-fat and rational-protein dietary (HR). The results showed the fat content in mature milk was increased with increments of fat intake, while both the protein and lactose contents in mature milk did not increase with their intakes for lactating mothers. Among these four dietary patterns, the energy ratio of macronutrients in human milk at the RR was the closest to the estimated energy ratio for infants aged 0–6 M. In conclusion, our study represents that the rational dietary pattern should be advocated, and the irrational dietary patterns, especially high-fat and high-protein dietary patterns, should be avoided during lactation stage.

    Abstract: MicroRNA (miRNA) is a non-coding RNA that can regulate the expression of many target genes, and it is widely involved in various important physiological activities. MiR-124-3p was found to associate with the normal development of retinal vessels in our previous study, but the mechanism of its anti-angiogenic effect on pathological retinal neovascularization still needed to be explored. Therefore, this study aimed to investigate the effect and mechanism of miR-124-3p on retinal neovascularization in mice with oxygen-induced retinopathy (OIR). Here, we found that intravitreal injection of miR-124-3p agomir attenuated pathological retinal neovascularization in OIR mice. Moreover, miR-124-3p preserved the astrocytic template, inhibited reactive gliosis, and reduced the inflammatory response as well as necroptosis. Furthermore, miR-124-3p inhibited the signal transducer and activator of transcription 3 (STAT3) pathway and decreased the expression of hypoxia-inducible factor-1α and vascular endothelial growth factor. Taken together, our results revealed that miR-124-3p inhibited retinal neovascularization and neuroglial dysfunction by targeting STAT3 in OIR mice.

    Abstract: A material balance equation (MBE) is usually employed to estimate well-controlled dynamic reserves, but a more accurate calculation of these reserves is crucial. For abnormally pressured reservoirs with aquifers, the abnormally pressure and aquifer are difficult to determine, which leads to inaccurate calculations of well-controlled dynamic reserves. In this work, the MBE was established. Then, the correction function and abnormally pressure effect were deduced to determine well-controlled dynamic reserves calculations for weak aquifers and strong aquifers, respectively. For a weak aquifer, a pressure correction was determined to exist at a pressure coefficient of 1.2, and then the correction function was established to calculate the dynamic reserves. For a strong aquifer, the MBE derived by Walsh was employed to calculate the dynamic reserves and water influx. The relationship between the abnormally pressure effect and the dimensionless formation pressure was proposed. Classical, abnormally high-pressured gas reservoirs, Anderson L and Amu Darya, were selected to validate the accuracy and applicability of the newly developed methods. The results show that an abnormally high-pressure effect has a great influence on the dynamic reserves calculation in the early stage and the water influx has a stronger influence on the dynamic reserves calculation in the late stage.
