    Abstract: Commercial inactivated vaccines against H9N2 avian influenza (AI) have been developed in China since 1990s and show excellent immunogenicity with strong HI antibodies. However, currently approved vaccines cannot meet the clinical demand for a live-vectored vaccine. Newcastle disease virus (NDV) vectored vaccines have shown effective protection in chickens against H9N2 virus. However, preexisting NDV antibodies may affect protective efficacy of the vaccine in the field. Here, we explored avian paramyxovirus serotype 2 (APMV-2) as a vector for developing an H9N2 vaccine via intranasal delivery. APMV-2 belongs to the same genus as NDV, distantly related to NDV in the phylogenetic tree, based on the sequences of Fusion (F) and hemagglutinin-neuraminidase (HN) gene, and has low cross-reactivity with anti-NDV antisera. We incorporated hemagglutinin (HA) of H9N2 into the junction of P and M gene in the APMV-2 genome by being flanked with the gene start, gene end, and UTR of each gene of APMV-2-T4 to generate seven recombinant APMV-2 viruses rAPMV-2/HAs, rAPMV-2-NPUTR-HA, rAPMV-2-PUTR-HA, rAPMV-2-FUTR-HA, rAPMV-2-HNUTR-HA, rAPMV-2-LUTR-HA, and rAPMV-2-MUTR-HA, expressing HA. The rAPMV-2/HAs displayed similar pathogenicity compared with the parental APMV-2-T4 virus and expressed HA protein in infected CEF cells. The NP-UTR facilitated the expression and secretion of HA protein in cells infected with rAPMV-2-NPUTR-HA. Animal studies demonstrated that immunization with rAPMV-2-NPUTR-HA elicited effective H9N2-specific antibody (6.14 ± 1.2 log2) responses and conferred complete immune protection to prevent viral shedding in the oropharyngeal and cloacal swabs from chickens challenged with H9N2 virus. This study suggests that our recombinant APMV-2 virus is safe and immunogenic and can be a useful tool in the combat of H9N2 outbreaks in chicken.

    Abstract: The reversal of loss of the critical size of skeletal muscle is urgently required using biomaterial scaffolds to guide tissue regeneration. In this work, coaxial electrospun magnetic nanofibrous scaffolds were fabricated, with gelatin (Gel) as the shell of the fiber and polyurethane (PU) as the core. Iron oxide nanoparticles (Mag) of 10 nm diameter were added to the shell and core layer. Myoblast cells (C2C12) were cultured on the magnetic scaffolds and exposed to the applied magnetic fields. A mouse model of skeletal muscle injury was used to evaluate the repair guided by the scaffolds under the magnetic fields. It was shown that VEGF secretion and MyoG expression for the myoblast cells grown on the magnetic scaffolds under the magnetic fields were significantly increased, while, the gene expression of Myh4 was up-regulated. Results from an in vivo study indicated that the process of skeletal muscle regeneration in the mouse muscle injury model was accelerated by using the magnetic actuated strategy, which was verified by histochemical analysis, immunofluorescence staining of CD31, electrophysiological measurement and ultrasound imaging. In conclusion, the integration of a magnetic scaffold combined with the extra magnetic fields enhanced myoblast differentiation and VEGF secretion and accelerated the defect repair of skeletal muscle in situ.

    Abstract: This paper proposes a wide dynamic range (DR) and high-resolution discrete-time (DT) 2-order 4-bit sigma-delta modulator with a novel dynamic-modulated scaling-down (DM-SD) technology for non-invasive electroencephalogram (EEG) acquisition. The DM-SD technology can expand the input dynamic range and suppress large input offsets at the same time. The modulator was designed with 180nm CMOS technology with an area of 0.49 mm2. We achieve a 118.1 dB SNDR when the input signal is 437.5 Hz and the signal bandwidth is 1500 Hz. Due to the proposed DM-SD technology, the DR is expanded to 126 dB. The power consumption of the whole modulator is 1.6 mW and a 177.8 dB Schreier figure-of-merit (FoMs) is realized.

    Abstract: Vision-based sensors are widely used in lateral control of autonomous vehicles, but the large computational cost of the visual algorithms often induces uneven time delays. In this paper, a hierarchical vision-based lateral control scheme is proposed, where the upper controller is designed by robust H∞-based linear quadratic regulator (LQR) algorithm to compensate sensor-induced delays, and the lower controller is based on logic threshold method, in order to achieve strong convergence of the steering angle. Firstly, the vehicle lateral model is built, and the nonlinear uncertainties induced by time delays are linearized with Taylor expansion. Secondly, the state space of the system is augmented to describe such uncertainties with polytopic inclusions, which is controlled by an H∞-based LQR controller with a low cost of online computation. Then, a lower controller is designed for the control of the steering motor. According to the results of the vehicle experiment as well as the hardware-in-the-loop (HIL) experiment, the proposed control scheme shows good performance in vehicle’s lateral control task, and exhibits better robustness compared with a conventional LQR controller. The proposed control scheme provides a feasible solution for the lateral control of autonomous driving.

    Abstract: The Yellow River Basin is essential to China’s economic and social development and ecological security. In order to assess the temporal and spatial distribution of cloud water in the Yellow River Basin, we analyzed the distribution characteristics of water vapor content and cloud water content using ERA5 monthly averaged data from 1980 to 2019. The results showed that the high-value area of the annual average atmospheric water vapor content distribution was concentrated above the North China Plain in the eastern part of the basin, and the value was mostly between 21 and 24 mm. The low-value areas were mainly centered above the high mountain areas in the western part of the basin, and the value mostly fell between 3 and 6 mm. The seasonal distribution characteristics of the annual average water vapor content were relatively consistent with the annual average distribution characteristics. The high-value cloud ice water content area was in the northeastern part of the Qinghai-Tibet Plateau (4.5 g·m−2), while the low-value area appeared on the Loess Plateau (2 g·m−2). The high-value area of cloud liquid water content was on the north side of the West Qinling Mountains (12 g·m−2). The low-value area appeared on the Loess Plateau and the northern part of the Qinghai-Tibet Plateau (3 g·m−2). The cloud water content was higher in the eastern region than in the western region in the overall spatial distribution, and the content of cloud liquid water was higher than that of cloud ice water. The average annual atmospheric water vapor content was increasing, and the annual average content of cloud ice water and cloud liquid water was declining. The change in the total amount and spatial distribution of cloud water was not obvious in the Yellow River Basin.

    Abstract: Most of the coal mines in Southwest China are located in mountainous areas with high vegetation coverage, and most activities are carried out under the mountains. The deformation monitoring and mechanical behavior analysis of the mining area helps reveal the typical mountain deformation and failure mechanism caused by underground mining activities and reduce the risk of mountain collapse in the mining area. In this manuscript, a research method for mountain stability in mining areas is proposed, which combines InSAR deformation monitoring with numerical analysis. Based on the high-precision deformation information obtained by DS-InSAR and the landslide range, a three-dimensional explicit finite difference numerical analysis method was used to reconstruct the landslide model. According to the layout of the coal mining working face, the variation mechanism of overlying stratum stress and the mountain slip in the coal mining process is inverted, and the mechanism of mountain failure and instability in the mining area is analysed. Based on the sentinel data, the experiment performed time series monitoring and inversion analysis of the mountain collapse in Nayong, Guizhou, China. The results show that mining activities a certain distance from the mountain will affect mountain stability, and there are specific mechanisms. From 2015 to 2017, the stress redistribution of overlying strata above the goaf area resulted in dense longitudinal cracks in the landslide body due to coal mining. The mountain is in a continuous damage state, and the supporting force to prevent collapse continues to decrease, resulting in a gradual decrease in landslide stability. Both the time series DS-InSAR monitoring results and numerical simulation results verify the actual occurrence and development of the on-site subsidence.

    Abstract: The boundary layer structure is crucial to the formation and intensification of typhoons, but there is still a lack of high-precision turbulence observations in the typhoon boundary layer due to limitations of the observing instruments under typhoon conditions. Using joint observations from multiple ground-based Doppler wind lidars (DWL) collected by the Shanghai Typhoon Institute of China Meteorological Administration (CMA) during the transit of Typhoon Lekima (8–11 August 2019), the characteristics of the wind field and physical quantities (including turbulent kinetic energy (TKE) and typhoon boundary layer height (TBLH)) of the boundary layer of typhoon Lekima were analyzed. The magnitude of TKE was shown to be related not only to the horizontal wind speed but also to the presence of a strong downdraft, which leads to a rapid increase of TKE. The magnitudes of TKE in different quadrants of Typhoon Lekima were also found to differ. The TKE in the front right quadrant of the typhoon was 2.5–6.0 times that in the rear left quadrant and ~1.7 times that in the rear right quadrant. The TKE over the island was larger than that over the urban area. Before Typhoon Lekima made landfall, the TKE increased with decreasing distance to the typhoon center. After typhoon landfall, the TKE changes were different on the left and right sides of the typhoon center, with the TKE on the left decreasing rapidly, while that on the right changed little. The typhoon boundary layer height calculated by five methods was compared and was found to decrease gradually before typhoon landfall and increased gradually afterward. The trends of the TBLH calculated using helicity and TKE were consistent, and both determine the TBLH well, while the maximum tangential wind speed height (humax) was larger than the height calculated by other methods. This study confirms that DWL has a strong detecting capability for the finescale structure of the typhoon boundary layer and provides a powerful tool for the validation of numerical simulations of typhoon structure.

    Abstract: Three carbon-chain extension genes associated with fatty acid synthesis in upland cotton (Gossypium hirsutum), namely GhKAR, GhHAD, and GhENR, play important roles in oil accumulation in cotton seeds. In the present study, these three genes were cloned and characterized. The expression patterns of GhKAR, GhHAD, and GhENR in the high seed oil content cultivars 10H1014 and 10H1041 differed somewhat compared with those of 10H1007 and 2074B with low seed oil content at different stages of seed development. GhKAR showed all three cultivars showed higher transcript levels than that of 2074B at 10-, 40-, and 45-days post anthesis (DPA). The expression pattern of GhHAD showed a lower transcript level than that of 2074B at both 10 and 30 DPA but a higher transcript level than that of 2074B at 40 DPA. GhENR showed a lower transcript level than that of 2074B at both 15 and 30 DPA. The highest transcript levels of GhKAR and GhENR were detected at 15 DPA in 10H1007, 10H1014, and 10H1041 compared with 2074B. From 5 to 45 DPA cotton seed, the oil content accumulated continuously in the developing seed. Oil accumulation reached a peak between 40 DPA and 45 DPA and slightly decreased in mature seed. In addition, GhKAR and GhENR showed different expression patterns in fiber and ovule development processes, in which they showed high expression levels at 20 DPA during the fiber elongation stage, but their expression level peaked at 15 DPA during ovule development processes. These two genes showed the lowest expression levels at the late seed maturation stage, while GhHAD showed a peak of 10 DPA in fiber development. Compared to 2074B, the oil contents of GhKAR and GhENR overexpression lines increased 1.05~1.08 folds. These results indicated that GhHAD, GhENR, and GhKAR were involved in both seed oil synthesis and fiber elongation with dual biological functions in cotton.

    Abstract: The relatively stable lunar illumination may be used to realize radiometric calibration under low light. However, there is still an insufficient understanding of the accuracy of models and the influence of parameters when conducting research on low-light radiometric calibration. Therefore, this study explores the applicability of three atmospheric radiative transfer models under different nighttime conditions. The simulation accuracies of three nighttime atmospheric radiative transfer models (Night-SCIATRAN, Night-MODTRAN, and Night-6SV) were evaluated using the visible-infrared imaging radiometer suite day/night band (VIIRS/DNB) data. The results indicate that Night-MODTRAN has the highest simulation accuracy under DNB. The consistency between simulated top-of-atmosphere (TOA) radiance and DNB radiance is approximately 3.1%, and uncertainty is 2.5%. This study used Night-MODTRAN for parameter sensitivity analysis. The results indicate that for the lunar phase angle, aerosol optical depth, surface reflectance, lunar zenith angle, satellite zenith angle, and relative azimuth angle, the average change rates are 68%, 100%, 2561%, 75%, 20%, and 0%. This paper can help better understand the performance of models under different atmospheric and geographical conditions, as well as whether existing models can simulate the complex processes of atmospheric radiation.
