    Abstract: The cellulose of the plant cell wall indirectly affects the cell shape and straw stiffness of the plant. Here, the novel brittleness mutant brittle stalk-5 (bk-5) of the maize inbred line RP125 was characterized. We found that the mutant displayed brittleness of the stalk and even the whole plant, and that the brittleness phenotype existed during the whole growth period from germination to senescence. The compressive strength was reduced, the cell wall was thinner, and the cellulose content was decreased compared to that of the wild type. Genetic analysis and map-based cloning indicated that bk-5 was controlled by a single recessive nuclear gene and that it was located in a 90.2-Kb region on chromosome 3 that covers three open reading frames (ORFs). Sequence analysis revealed a single non-synonymous missense mutation, T-to-A, in the last exon of Zm00001d043477 (B73: version 4, named BK-5) that caused the 951th amino acid to go from leucine to histidine. BK-5 encodes a cellulose synthase catalytic subunit (CesA), which is involved with cellulose synthesis. We found that BK-5 was constitutively expressed in all tissues of the germinating stage and silking stage, and highly expressed in the leaf, auricula, and root of the silking stage and the 2-cm root and bud of the germinating stage. We found that BK-5 mainly localized to the Golgi apparatus, suggesting that the protein might move to the plasma membrane with the aid of Golgi in maize. According to RNA-seq data, bk-5 had more downregulated genes than upregulated genes, and many of the downregulated genes were enzymes and transcription factors related to cellulose, hemicellulose, and lignin biosynthesis of the secondary cell wall. The other differentially expressed genes were related to metabolic and cellular processes, and were significantly enriched in hormone signal transduction, starch and sucrose metabolism, and the plant–pathogen interaction pathway. Taken together, we propose that the mutation of gene BK-5 causes the brittle stalk phenotype and provides important insights into the regulatory mechanism of cellulose biosynthesis and cell wall development in maize.

    Abstract: Gibberellin is an important plant growth regulator that has been widely used in agricultural production with great market prospects. However, the low yield from Gibberella fujikuroi restricts its application. To improve the production of gibberellin A4 (GA4), the response surface methodology was used in this study to explore the effect of different types and concentrations of vegetable oil and precursors on the production of GA4. Based on a single factor experiment, the Behnken box and central composite designs were used to establish the fermentation condition model, and the response surface method was used for analysis. The results indicated that the optimum formula was 0.55% palm oil, 0.60% cottonseed oil, 0.64% sesame oil, 0.19 g/L pyruvic acid, 0.21 g/L oxaloacetic acid, and 0.21 g/L citric acid for 48 h, which produced a yield 4.32 times higher than that without optimization. This suggests that the mathematical model is valid for predicting GA4 production in Gibberella fujikuroi QJGA4-1.

    Abstract: The beneficial role of combining fiber reinforcement with lime stabilization in altering soil behavior has been established in the literature. However, the coupling effect of their combination still remains unclear in terms of its magnitude and microscopic mechanism, especially for natural fibers with special microstructures. The objective of this study was to investigate the coupling effect of wheat straw fiber reinforcement and lime stabilization on the mechanical behavior of Hefei clayey soil. To achieve this, an experimental program including unconsolidated–undrained (UU) triaxial tests and SEM analysis was implemented. Static compaction test samples were prepared on untreated soil, fiber-reinforced soil, lime-stabilized soil, and lime-stabilized/fiber-reinforced soil at optimum moisture content with determining of the maximum dry density of the untreated soil. The lime was added in three different contents of 2%, 4%, and 6%, and 13 mm long wheat straw fiber slices with a cross section one-quarter that of the intact ones were mixed in at 0.2%, 0.4%, and 0.6% by dry weight of soil. Analysis of the derived results indicated that the addition of a small amount of wheat straw fibers into lime-stabilized soil improved the intensity of the strain-softening behavior associated with mere lime stabilization. The observed evidence that the shear strength increase brought by a combination of 0.4% fiber reinforcement and 4% lime stabilization was smaller than the summation of the shear strength increases brought by their presence alone in a sample demonstrated a coupling effect between fiber reinforcement and lime stabilization. This coupling effect was also detected in the comparisons of the secant modulus and failure pattern between the combined treatment and the individual treatments. These manifestations of the coupling effect were explained by a microscopic mechanism wherein the fiber reinforcing effect was made more effective by the ways in which lime chemically stabilized the soil and lime stabilization development was quickened by the water channels passing through the surfaces and honeycomb pores of the wheat straw fibers.

    Abstract: Exponential fertilization has been regarded as an important technique for improving seedling quality at the initial plant-growth stage. In our study, containerized one-year-old Chinese fir (Cunninghamia lanceolata (Lamb.) Hook) rooted cuttings were reared at four nitrogen (N) fertilizer levels (0, 0.5, 1.0, 2.0 g cutting−1 season−1) under two topdressing methods (conventional and exponential) for a 210-day greenhouse rotation to evaluate growth and nutrient loading capacity of seedlings. N fertilizer was applied 20 times at an interval of 10 days during the study period. The results indicated that the schedule and rate of fertilization significantly affected the height, ground diameter, and biomass of Chinese fir rooted cuttings. The nitrogen concentration of different plant organs followed the order of leaves 〉 root 〉 stem. Compared to the CK, the N concentrations in root, stem, and leaves increased by 39.6%, 16.6%, and 41.1% in the conventional fertilizer treatment, and by 22.6% to 81.4%, 27.3% to 152.6%, and 73.6% to 135.5% in exponential fertilization treatments, respectively. The N concentrations of root, stem, and leaves of Chinese fir rooted cuttings under EF2 (1.0 N g cutting−1) were significantly higher than that of conventional fertilization (p 〈 0.05). However, there was no significant difference of phosphorus and potassium concentrations among different plant organs. Steady-state nutrition and superior growth performance were achieved by rooted cuttings fertilized exponentially at the rate of 1.0 g cutting−1 yielding (EF2).

    Abstract: A facile and scalable method is reported to fabricate Si-graphene nanocomposite as anode material for Li-ion batteries (LIBs) with high capacity and capacity retention performance. The Si-graphene electrode showed an initial discharge capacity of 1307 mAh g−1 at a current rate of 0.1C. At the 25th cycle, the electrode retained a discharge capacity of 1270 mAh g−1, with an excellent capacity retention of 97%. At the 50th cycle, the electrode still retained high capacity retention of 89%. The improved capacity retention of Si-graphene anode compared with Si anode is attributed to the mechanical flexibility of graphene that compromises the volume expansion of Si during the lithiation/delithiation process. The electrochemical impedance measurement further confirms the enhanced electrical conductivity and the denser solid-electrolyte-interface of the Si-graphene electrode. This fabrication approach is cost-effective and easy to scale up compared to known techniques, making it a promising candidate for commercializing Si-based anode for LIBs.

    Abstract: The DExD/H-box protein family encompasses a large number of RNA helicases that are involved in RNA metabolism and a variety of physiological functions in different species. However, there is limited knowledge of whether DExD/H-box proteins play a role in the pathogenicity of plant fungal pathogens. In the present work, the DExD/H-box protein MoDHX35, which belongs to the DEAH subfamily, was shown to be crucial in appressoria formation and full virulence of the rice blast fungus, Magnaporthe oryzae. The predicted protein sequence of MoDHX35 had typical DEAH-box domains, showed 47% identity to DHX35 in Homo species, but had no orthologs in Saccharomyces cerevisiae. Deletion of the MoDHX35 gene resulted in reduced tolerance of the mutants to doxorubicin, a nucleic acid synthesis disturbing agent, suggesting the involvement of MoDHX35 in RNA metabolism. MoDHX35-deleted mutants exhibited normal vegetative growth, conidia generation and conidial germination, but showed a reduced appressorium formation rate and attenuated virulence. Our work demonstrates the involvement of DEAH-box protein functions in the pathogenicity of plant fungal pathogens.

    Abstract: This retrospective study investigated the effect of iron status on peritonitis by analyzing longitudinal iron parameters in peritoneal dialysis (PD) patients. Patients who received PD at our center from 1 January 2006 to 31 December 2015 were included and followed up until 31 December 2017. According to the joint quartiles of baseline transferrin saturation and ferritin, iron status was categorized as reference iron status (RIS), absolute iron deficiency (AID), functional iron deficiency (FID), and high iron status (HIS). Generalized estimating equations and Cox regression models with time-dependent covariates were used. A total of 1258 PD patients were included; 752 (59.8%) were male, with a mean (±standard deviation) age of 47.4 (±14.9) years. During a median follow-up period of 35.5 (interquartile range, 18.4–60.0) months, 450 (34.3%) patients had 650 episodes of peritonitis. By analyzing longitudinal data, patients with AID were independently positively associated with the occurrence (adjusted odds ratio (AOR) = 1.45) and treatment failure of peritonitis (adjusted hazard ratio (AHR) = 1.85). Patients with HIS were positively associated with the treatment failure of peritonitis (AHR = 2.70). Longitudinal AID and HIS were associated with the episodes and poor prognosis of peritonitis. Active clinical monitoring and correction of iron imbalance in patients with PD are needed.
