    Abstract: We aimed to investigate the microbial diversity, mine lignocellulose-degrading enzymes/proteins, and analyze the domain structures of the mined enzymes/proteins in humus samples collected from the Cuc Phuong National Park, Vietnam. Using a high-throughput Illumina sequencer, 52 Gbs of microbial DNA were assembled in 2,611,883 contigs, from which 4,104,872 open reading frames (ORFs) were identified. Among the total microbiome analyzed, bacteria occupied 99.69%; the five ubiquitous bacterial phyla included Proteobacteria, Bacteroidetes, Actinobacteria, Firmicutes, and Acidobacteria, which accounted for 92.59%. Proteobacteria (75.68%), the most dominant, was 5.77 folds higher than the second abundant phylum Bacteroidetes (13.11%). Considering the enzymes/proteins involved in lignocellulose degradation, 22,226 ORFs were obtained from the annotation analysis using a KEGG database. The estimated ratio of Proteobacteria/Bacteroidetes was approximately 1:1 for pretreatment and hemicellulases groups and 2.4:1 for cellulases. Furthermore, analysis of domain structures revealed their diversity in lignocellulose-degrading enzymes. CE and PL were two main families in pretreatment; GH1 and GH3-FN3 were the highest domains in the cellulase group, whereas GH2 and GH43 represented the hemicellulase group. These results validate that natural tropical forest soil could be considered as an important source to explore bacteria and novel enzymes/proteins for the degradation of lignocellulose.

    Abstract: Antiviral nucleoside analogues (ANA) are newly used therapeutics acting against the hepatitis C virus (HCV). This class of drug is well known to exhibit toxicity on mitochondrial DNA (mtDNA). People who inject drugs (PWID) are particularly affected by HCV infection and cumulated mitotoxic drug exposure from HIV treatments (antiretrovirals, ARV) and other illicit drugs. This study aims to explore the impact of direct-acting antiviral (DAA) treatments on mtDNA among PWID. A total of 470 actively injecting heroin users were included. We used quantitative PCR on whole blood to determine the mitochondrial copy number per cell (MCN) and the proportion of mitochondrial DNA deletion (MDD). These parameters were assessed before and after DAA treatment. MDD was significantly increased after HCV treatment, while MCN did not differ. MDD was even greater when subjects were cotreated with ARV. In multivariate analysis, we identified that poly-exposure to DAA and daily heroin injection or regular consumption of methamphetamines were positively associated with high MCN loss while DAA and ARV treatments or methadone use were identified as risk factors for having mtDNA deletion. These observations deserve attention since they were previously associated with premature cell ageing or cell transformation and therefore call for a long-term follow-up.

    Abstract: People who inject drugs (PWID) are a population exposed to many genotoxicants and with a high prevalence of HCV infection. Direct-acting antiviral (DAA) regimens are now widely used to treat chronic HCV infection. Although side effects to treatment are currently rare, the long-term effects such as suspicions of de novo hepatocellular carcinoma (HCC) occurrence or HCC recurrence and cardiac defects are still up for debate. Given the structure of DAAs, the molecules have a potential mitochondrial DNA (mtDNA) genotoxicity. We have previously reported acute mtDNA toxicity of three DAA regimens among PWID with a strong impact on the rate of mtDNA deletion, less on the quantity of mtDNA copy per cell at sustained viral response at 12 weeks (SVR12). Herein, we report the mtDNA parameters nine months after drug discontinuation. We observed that the percentage of the deleted mtDNA genome increased over time. No exposure to any other genotoxicants during this period was associated with a high deletion percentage, suggesting that the replicative advantage of the deleted molecules outweighed their elimination processes. Such observation calls for longer-term follow-up and may contribute to the molecular basis of subclinical side effects of DAA treatments.

    Abstract: Bacteria in rumen play pivotal roles in the digestion of nutrients to support energy for the host. In this study, metagenomic deep sequencing of bacterial metagenome extracted from the goats’ rumen generated 48.66 GB of data with 3,411,867 contigs and 5,367,270 genes. The genes were mainly functionally annotated by Kyoto Encyclopedia of Genes and Genomes (KEGG) Carbohydrate-Active enZYmes (CAZy), and HMMER database, and taxonomically classified by MEGAN. As a result, 65,554 genes encoding for 30 enzymes/proteins related to lignocellulose conversion were exploited, in which nine enzymes were seen for the first time in goat rumen. Prevotella was the most abundant genus, contributing 30% hemicellulases and 36% enzymes/proteins for lignocellulose pretreatment, and supporting 98.8% of feruloyl esterases and 71.7% acetylxylan esterases. In addition, 18 of the 22 most lignocellulose digesting- potential contigs belonged to Prevotella. Besides, Prevotella possessed many genes coding for amylolytic enzymes. One gene encoding for endoxylanase was successfully expressed in E. coli. The recombinant enzyme had high Vmax, was tolerant to some salts and detergents, worked better at pH 5.5–6.5, temperature 40–50 °C, and was capable to be used in practices. Based on these findings, we confirm that Prevotella plays a pivotal role for hemicellulose digestion and significantly participates in starch, cellulose, hemicellulose, and pectin digestion in the goat rumen.

    Abstract: Linezolid is an antibiotic of last resort for the treatment of infections caused by Gram-positive bacteria, including vancomycin-resistant enterococci. Enterococcus faecalis, a member of enterococci, is a significant pathogen in nosocomial infections. E. faecalis resistance to linezolid is frequently related to the presence of optrA, which is often co-carried with fex, phenicol exporter genes, and erm genes encoding macrolide resistance. Therefore, the common use of antibiotics in veterinary might promote the occurrence of optrA in livestock settings. This is a cross-sectional study aiming to investigate the prevalence of optrA positive E. faecalis (OPEfs) in 6 reservoirs in farms in Ha Nam province, Vietnam, and its associated factors and to explore genetic relationships of OPEfs isolates. Among 639 collected samples, the prevalence of OPEfs was highest in flies, 46.8% (51/109), followed by chickens 37.3% (72/193), dogs 33.3% (17/51), humans 18.7% (26/139), wastewater 16.4% (11/67) and pigs 11.3%, (14/80). The total feeding area and total livestock unit of the farm were associated with the presence of OPEfs in chickens, flies, and wastewater. Among 186 OPEfs strains, 86% were resistant to linezolid. The presence of optrA was also related to the resistant phenotype against linezolid and levofloxacin of E. faecalis isolates. Close genotypic relationships identified by Pulsed Field Gel Electrophoresis between OPEfs isolates recovered from flies and other reservoirs including chickens, pigs, dogs, and wastewater suggested the role of flies in the transmission of antibiotic-resistant pathogens. These results provided warnings of linezolid resistance although it is not used in livestock.

    Abstract: Understanding the contributions of participants from management practices and those from the local community to protected areas (PAs) as nature reserves, is crucial for forest protection and resource conservation. Thus, empirical studies have usually investigated whether patrolling has significantly improved forest protection action. Many previous studies have exhaustively examined the contributions to forest protection by rangers or local persons, but there has been little research on the contribution of both sides to patrol efforts with separate consideration given to each. This paper explores insights into the patrol efforts in the Nam Tien forest station. The total number of patrollers, walking distances, and working days across years were significantly different between the ranger and local person patrol groups. There was a considerable variation in the number of patrollers (Z = 2.02; p 〈 0.01), distance (Z = 2.45; p 〈 0.01) and working days (Z = 2.37; p 〈 0.01) between rangers and local persons related to monthly patrolling. For the traditional patrols in the various PAs, it is shown that, for long-term forest protection, local persons patrolling should be considered more, in order to achieve the sustainable conservation.

    Abstract: (1) Background: Hedera nepalensis (Araliaceae) is a recognized medicinal plant founded in Asia that has been reported to work in antioxidant, antifungal, antimicrobial, and antitumor capacities. (2) Methods: The subcritical fluid extraction of saponin from Hedera nepalensis leaves and the optimum of the extraction process based on yield of saponin contents (by calculating the hederacoside C contents in dried Hedera nepalensis leaves) are examined by response surface methodology (RSM). Furthermore, the antimicrobial activity of the extract is tested for potential drug applications in the future. (3) Results: Based upon RSM data, the following parameters are optimal: extraction time of 3 min, extraction temperature of 150 °C, and a sample/solvent ratio of 1:55 g/mL. Under such circumstances, the achieved yield of saponin is 1.879%. Moreover, the extracts inhibit the growth of some bacterial strains (Streptococcus pneumoniae, Streptococcus pyogenes, Haemophilus influenza) at a moderate to strong level with inhibition zone diameter values ranging from 12.63 to 19.50 mm. (4) Conclusions: The development of such a model provides a robust experimental process for optimizing the extraction factors of saponin contents from Hedera nepalensis extract using subcritical fluid extraction and RSM. Moreover, the current work reveals that saponin extracts of Hedera nepalensis leaves exhibit a potential antimicrobial activity, which can be used as scientific evidence for further study.

    Abstract: Rainwater is considered a promising alternative water source for coastal areas where freshwater resources are increasingly under pressure. This study evaluates rain regime characteristics that influence the ability to exploit rainwater in the coastal provinces of the Vietnamese Mekong Delta (VMD). In particular, it considers the impact of climate change and El Niño–Southern Oscillation (ENSO) processes. We analyzed rainfall data from 102 monitoring stations across the VMD from 1989 to 2017. Using statistical methods, we explored (1) characteristics of the rainy season, including the dates of onset and cessation and season length; (2) average rainfall volumes; and (3) the maximum number of consecutive nonrainy days during the rainy season and over the year. We also analyzed changes in these characteristics over time, in parallel with ENSO processes and climate change. Trend lines were determined using nonparametric methods, utilizing Sen’s slope estimation and the Mann–Kendall test. Results showed a tendency for the rainy season to start earlier and end later in the western coastal zone of the study area, with season length gradually decreasing towards the East Sea and inland. The shortest rainy season was found in the estuary zone (in the northeast of the VMD). Rainfall was abundant on the west coast, again diminishing gradually towards the East Sea and inland. Rain was also quite plentiful during four dry season observation months in the study area, but it lacked the predictability needed for effective exploitation. The number of consecutive days without rain averaged 96 annually, with a difference of 29 days between the largest and smallest observations. The difference between the provinces with the longest and shortest periods without rain averaged 41 days. Although the impact of climate change on the rain regime is complex, we can say that the rainy season now tends to start earlier, end later, and be lengthier, though without exhibiting clear trends. ENSO processes significantly impacted rainfall regime characteristics, especially the dates of onset and cessation, and season length.

    Abstract: Panax vietnamensis var. vietnamensis (PVV) and Panax vietnamensis var. fuscidiscus (PVF) both belong to Panax vietnamensis species and are chemically and morphologically similar, making it hard to distinguish for the consumer. Herein, 42 PVF and 12 PVV samples were collected in Quang Nam and Lai Chau Province, respectively, and subsequently characterized by ITSr-DNA sequence data to verify their origins. Next, untargeted metabolomics combined with multivariate statistical analysis was developed to differentiate PVV and PVF. The metabolic profiles of PVV and PVF were found to be distinct and classified well using Partial Least-Squares Discriminant Analysis (PLS-DA) in the training set. Among them, seven ginsenosides were of high abundance in PVV, while six were of high abundance in PVF. Next, the test set was used to validate 13 putative differential markers found in the training set, illustrating a complete match with the expression patterns of these ginsenosides in the training set. Finally, PLS-DA and linear Support Vector Machine models both indicated distinct ginsenoside profiles of PVV and PVF without misclassification in the test set. Conclusively, the developed untargeted metabolomics approach might serve as a powerful tool for the authentication of PVV and PVF at the metabolome level.

    Abstract: In this study, we present the fabrication of nitrile butadiene rubber/waste leather fiber (NBR/WLF) composites with different weight percentages of WLF and NBR (0/100, 20/80, 30/70, 40/60, 50/50, 60/40 wt/wt). WLF was prepared by cutting the scrap leathers from the waste product of the Vietnamese leather industry. Subsequently, in order to make the short fibers, it was mixed by a hammer mill. The characteristics of WLF/NBR composites such as mechanical properties (tensile strength, tear strength, hardness), dynamic mechanical properties, toluene absorption, and morphology were carefully evaluated. As a result, the tensile strength and tear strength become larger with increasing WLF content from 0 to 50 wt% and they decrease when further increasing WLF content. The highest tensile strength of 12.5 MPa and tear strength of 72.47 N/mm were achieved with the WLF/NBR ratio of 50/50 wt%. Both hardness and resistance of the developed materials with toluene increased with increasing WLF content. The SEM results showed a good adhesion of NBR matrix and the WLF. The increasing of storage modulus (E’) in comparison with raw NBR showed good compatibility between WLF and NBR matrix. This research showed that the recycled material from waste leather and NBR was successfully prepared and has great potential for manufacturing products such as floor covering courts and playgrounds, etc.
