    Abstract: A new off-axis optical design alternative to that of the GRACE Follow-on mission for future NGGM missions is considered. In place of the triple-mirror assembly of the GRACE Follow-on mission, a laser retro-reflector is instead generated by means of lens systems. The receiving (RX) beam and transmitting (TX) beam are enforced to be anti-parallel by a control loop with differential wavefront sensing (DWS) signals as readout, and a fast-steering mirror is employed to actuate the pointing of the local beam. The tilt-to-length (TTL) coupling noise of the new off-axis optical bench layout is carefully studied in the present work. Local TTL originated from piston noise as well as assembly and alignment errors of optical components are studied. Effort is also made to have an in depth understanding of global TTL due to relative attitude jitter between spacecraft. The margin of TTL noise in the position noise budget for laser ranging is examined. With an open loop control of the offset between the reference point of the optical bench and the centre of mass of a satellite, the TTL noise of the new off-axis optical bench design may be suppressed efficiently.

    Abstract: Heavy oil reservoirs are often characterized by high viscosity and poor mobility, which is more complex with the presence of bottom water. The conventional vertical well development method has low oil recovery efficiency and limited controlled reserves of a single well. In addition, water cut can increase dramatically when the edge-bottom water breaks through. Horizontal well and CO2 huff-n-puff is an effective alternative development model for heavy oil reservoirs. This development method makes efficient use of CO2 and accords with the “Carbon Capture, Utilization, and Storage (CCUS)”. The horizontal well can increase the drainage area. The dissolution of CO2 improves the mobility of crude oil and increases formation energy. In this paper, we established numerical simulation models based on the Liuguanzhuang oilfield in Dagang. The characteristics and producing rules of the horizontal well and CO2 huff-n-puff development in the heavy oil reservoir were studied. The results show that the production characteristics of horizontal well and CO2 huff-n-puff were similar to Steam-Assisted Gravity Drainage (SAGD). CO2 forms a viscosity reduction area above the horizontal well and the heavy oil flows into the wellbore due to gravity after viscosity reduction. The CO2 huff-n-puff can effectively enhance the production area of horizontal wells compared with the depletion development. However, the improvement in the production area gradually decreased as CO2 huff-n-puff cycles continued. There was a boundary of production area against the horizontal well, with the main production of heavy oil occurring at the upper and either end of the horizontal well. The CO2 huff-n-puff has a restraining effect on the edge-bottom water, which is confirmed via the proposed theoretical model.

    Abstract: Taraxasterol (TAX), one of the active components in Dandelion, demonstrated strong antitumor properties in several cancers. However, the effect and underlying mechanism of TAX in non-small cell lung cancer (NSCLC) is unclear. In this study, we showed that TAX inhibited the proliferation of cells by inducing S-phase cell cycle arrest and prevented cell migration by interfering epithelial-mesenchymal transition (EMT) in Lewis lung cancer (LLC) cells and lung carcinoma SPC-A1 cells. The pharmacological network analysis predicted that induction of apoptosis might be the potential mechanism of TAX-mediated cell deaths. Further in vitro experiments showed that TAX could significantly induce cancer cell apoptosis as verified by increased pro-apoptotic molecules including Bax, caspase-9, and PARP1 downregulated anti-apoptotic protein Bcl-2; and decreased mitochondrial potential. The LLC subcutaneous tumor model demonstrated that TAX inhibited tumor growth by induction of apoptosis and inhibition of proliferation in vivo, which is consistent with the in vitro data. Importantly, TAX administration downregulated the proportion of Treg cells and upregulated CD107a+ NK cells in the tumor microenvironment in the tumor model. Together, these data reveal that TAX performs its antitumor effect by inducing apoptosis and modulating the tumor microenvironment, providing evidence that TAX could serve as a potential natural drug for lung cancer therapy.

    Abstract: Human papillomavirus (HPV) is a significant etiologic driver of penile squamous cell carcinoma (PSCC). The integration pattern of HPV and its carcinogenic mechanism in PSCC remain largely unclear. We retrospectively reviewed 108 PSCC cases who received surgery between 2008 and 2017. Using high-throughput viral integration detection, we identified 35 HPV-integrated PSCCs. Unlike cervical cancer, the HPV E2 oncogene was not prone to involvement in integration. Eleven of the 35 (31.4%) HPV-integrated PSCCs harbored intact HPV E2; these tumors had lower HPV E6 and E7 expression and higher expression of p53 and pRb proteins than those with disrupted E2 did (p 〈 0.001 and p = 0.024). Integration breakpoints are preferentially distributed in or near host genes, including previously reported hotspots (KLF5, etc.) and newly identified hotspots (CADM2, etc.), which are mainly involved in oncogenic signaling pathways (MAPK, JAK/STAT, etc.). Regarding the phosphorylation levels of JNK, p38 was higher in HPV-positive tumors with MAPK-associated integration than those in HPV-positive tumors with other integration and those in HPV-negative tumors. In vitro, KLF5 knockdown inhibited proliferation and invasion of PSCC cells, while silencing CADM2 promoted migration and invasion. In conclusion, this study enhances our understanding of HPV-induced carcinogenesis in PSCC, which may not only rely on the E6/E7 oncogenes, but mat also affect the expression of critical genes and thus activate oncogenic pathways.

    Abstract: MicroRNAs (miRNAs) are a class of small, endogenous, noncoding RNAs. Recent research has proven that miRNAs play an essential role in the occurrence and development of ischemic stroke. Our previous studies confirmed that 20(R)-ginsenosideRg3 [20(R)-Rg3] exerts beneficial effects on cerebral ischemia–reperfusion injury (CIRI), but its molecular mechanism has not been elucidated. In this study, we used high-throughput sequencing to investigate the differentially expressed miRNA and mRNA expression profiles of 20(R)-Rg3 preconditioning to ameliorate CIRI injury in rats and to reveal its potential neuroprotective molecular mechanism. The results show that 20(R)-Rg3 alleviated neurobehavioral dysfunction in MCAO/R-treated rats. Among these mRNAs, 953 mRNAs were significantly upregulated and 2602 mRNAs were downregulated in the model group versus the sham group, whereas 437 mRNAs were significantly upregulated and 35 mRNAs were downregulated in the 20(R)-Rg3 group in contrast with those in the model group. Meanwhile, the expression profile of the miRNAs showed that a total of 283 differentially expressed miRNAs were identified, of which 142 miRNAs were significantly upregulated and 141 miRNAs were downregulated in the model group compared with the sham group, whereas 34 miRNAs were differentially expressed in the 20(R)-Rg3 treatment group compared with the model group, with 28 miRNAs being significantly upregulated and six miRNAs being significantly downregulated. Furthermore, 415 (391 upregulated and 24 downregulated) differentially expressed mRNAs and 22 (17 upregulated and 5 downregulated) differentially expressed miRNAs were identified to be related to 20(R)-Rg3′s neuroprotective effect on stroke recovery. The Kyoto Encyclopedia of Genes and Genomes (KEGG) results showed that 20(R)-Rg3 could modulate multiple signaling pathways related to these differential miRNAs, such as the cGMP-PKG, cAMP and MAPK signaling pathways. This study provides new insights into the protective mechanism of 20(R)-Rg3 against CIRI, and the mechanism may be partly associated with the regulation of brain miRNA expression and its target signaling pathways.

    Abstract: Contemporary Chinese artist villages (CCAVs) have been developing rapidly and have become an important locus for developing cultural and creative industries, providing a strong pathway for China to achieve high-quality development and promote new urbanization. However, in practice, there are still many CCAVs that struggle with sustainable development. At present, the academic research on CCAVs mainly focuses on demonstrating the role of artist villages in promoting development in economic, social, artistic, and other fields without forming a systematic, sustainable development model or targeted policy recommendations. Therefore, the CCAV must be taken as a research object, and the influencing factors and mechanisms of its development must be analyzed through systematic quantitative research to provide a theoretical basis for the sustainable development (SD) of CCAVs. This paper identifies the factors influencing the sustainable development of CCAVs through grounded theory and expert interviews and creates a list of 21 influencing factors. Based on internal and external considerations, these influencing factors can be divided into the following four categories: artists, government, company/capital, and outside environment. This paper constructs a mechanism model for the SD of CCAVs with the help of a structural equation model, which is a quantitative analysis tool based on empirical research. The key mechanisms influencing the SD of CCAVs are obtained through research. Using quantitative research, the paper determines four categories of influencing factors and the mechanisms influencing the SD of CCAVs. This study provides an important resource for governments at all levels to promote the sustainable development of artist villages in the region.

    Abstract: As a development form of the creative industry, the Artist Village is a high concern of the Chinese government, and it has achieved rapid developments in China in recent years. However, many Artist Villages come to an untimely end a short period after their birth and fail to realize sustainable development. In this study, an empirical analysis on 80 Artist Villages in China was carried out. A field investigation and research on eight Artist Villages was implemented. Attention was paid to thoroughly analyze three typical Artist Villages of Guangzhou Xiaozhou Artist Village, Chongqing Gujianshan Artist Village and Beijing Songzhuang Artist Village. It has been found from studies that the development of Artist Villages in China has experienced initiation, development and maturity stages. The development of Artist Villages in China generally has many problems, such as an insufficient endogenous impetus, the excessive intervention of government, market capital “squeezing out” the original artists, and so on. This paper proposed a sustainable development model for Artist Villages during urbanization in China based on the findings and conclusions of this study. This study not only enriches research contents in this field, but can also provide meaningful references for the sustainable development of Artist Villages in China.

    Abstract: Self-incompatibility (SI) is a type of reproductive barrier within plant species and is one of the mechanisms for the formation and maintenance of the high diversity and adaptation of angiosperm species. Approximately 40% of flowering plants are SI species, while only 10% of orchid species are self-incompatible. Intriguingly, as one of the largest genera in Orchidaceae, 72% of Dendrobium species are self-incompatible, accounting for nearly half of the reported SI species in orchids, suggesting that SI contributes to the high diversity of orchid species. However, few studies investigating SI in Dendrobium have been published. This study aimed to address the following questions: (1) How many SI phenotypes are in Dendrobium, and what are they? (2) What is their distribution pattern in the Dendrobium phylogenetic tree? We investigated the flowering time, the capsule set rate, and the pollen tube growth from the representative species of Dendrobium after artificial pollination and analysed their distribution in the Asian Dendrobium clade phylogenetic tree. The number of SI phenotypes exceeded our expectations. The SI type of Dendrobium chrysanthum was the primary type in the Dendrobium SI species. We speculate that there are many different SI determinants in Dendrobium that have evolved recently and might be specific to Dendrobium or Orchidaceae. Overall, this work provides new insights and a comprehensive understanding of Dendrobium SI.
