    Abstract: Passive systems that utilize a natural power such as a pond, plant, or microorganisms, is expected to be a cost-effective method for acid mine drainage (AMD) treatment. The Ningyo-toge mine, a non-operational uranium mine located in Okayama Prefecture, Japan, generates AMD containing arsenic and iron. To quantitatively study arsenic and iron ion removal in an artificial wetland and pond, chemical reactions were modeled and incorporated into the GETFLOWS (general-purpose terrestrial fluid-flow simulator) software. The chemical reaction models consisted of arsenite and ferrous oxidation equations and arsenic adsorption on ferrihydrite. The X-ray diffraction analysis of sediment samples showed ferrihydrite patterns. These results were consistent with the model for arsenite/ferrous oxidation and arsenic adsorption on ferrihydrite. Geofluid simulation was conducted to simulate mass transfer with the utilized topographic model, inlet flow rate, precipitation, and evaporation. The measured arsenic and iron ions concentrations in solution samples from the wetland and pond, fitted well with the model. This indicated that the main removal mechanism was the oxidation of arsenite/ferrous ions and that arsenic was removed by adsorption rather than dilution.

    Abstract: During sexual reproduction/conjugation of the ciliate Tetrahymena thermophila, the germinal micronucleus undergoes meiosis resulting in four haploid micronuclei (hMICs). All hMICs undergo post-meiotic DNA double-strand break (PM-DSB) formation, cleaving their genome. DNA lesions are subsequently repaired in only one ‘selected’ hMIC, which eventually produces gametic pronuclei. DNA repair in the selected hMIC involves chromatin remodeling by switching from the heterochromatic to the euchromatic state of its genome. Here, we demonstrate that, among the 15 Tetrahymena Snf2 family proteins, a core of the ATP-dependent chromatin remodeling complex in Tetrahymena, the germline nucleus specific Iswi in Tetrahymena IswiGTt and Rad5Tt is crucial for the generation of gametic pronuclei. In either gene knockout, the selected hMIC which shows euchromatin markers such as lysine-acetylated histone H3 does not appear, but all hMICs in which markers for DNA lesions persist are degraded, indicating that both IswiGTt and Rad5Tt have important roles in repairing PM-DSB DNA lesions and remodeling chromatin for the euchromatic state in the selected hMIC.

    Abstract: In Japan, population health with life expectancy (LE) and healthy life expectancy (HALE) as indicators varies across the 47 prefectures (administrative regions). This study investigates how health examination results, including attitude toward improving life habits, are associated with population health. The association between health checkup variables and summary population health outcomes (i.e., life expectancy and healthy life expectancy) was investigated using a cross-sectional ecological design with prefectures as the unit of analysis. The medical records, aggregated by prefecture, gender, and age in the National Database of Health Insurance Claims and Specific Health Checkups of Japan (NDB) Open Data Japan, were used as health checkup variables. Body weight, blood pressure, liver enzymes, drinking habits, smoking habits, diabetes, serum lipids, and answers to questions regarding attitude toward improving health habits were significantly correlated to population health outcomes. Multiple regression analysis also revealed significant influence of these variables on population health. This study highlights that health examination results, including attitude toward improving health habits, are positively associated with population health. Consequently, implementing measures to improve health habits in response to the examination results could help the population maintain a healthy life.

    Abstract: Near infrared photoimmunotherapy (NIR-PIT) is a newly developed and highly selective cancer treatment that employs a monoclonal antibody (mAb) conjugated to a photo-absorber dye, IRDye700DX, which is activated by 690 nm light. Cancer cell-targeted NIR-PIT induces rapid necrotic/immunogenic cell death (ICD) that induces antitumor host immunity including re-priming and proliferation of T cells. Interleukin-15 (IL-15) is a cytokine that activates natural killer (NK)-, B- and T-cells while having minimal effect on regulatory T cells (Tregs) that lack the IL-15 receptor. Here, we hypothesized that IL-15 administration with cancer cell-targeted NIR-PIT could further inhibit tumor growth by increasing antitumor host immunity. Three syngeneic mouse tumor models, MC38-luc, LL/2, and MOC1, underwent combined CD44-targeted NIR-PIT and short-term IL-15 administration with appropriate controls. Comparing with the single-agent therapy, the combination therapy of IL-15 after NIR-PIT inhibited tumor growth, prolonged survival, and increased tumor infiltrating CD8+ T cells more efficiently in tumor-bearing mice. IL-15 appears to enhance the therapeutic effect of cancer-targeted NIR-PIT.

    Abstract: This study aimed to clarify whether infection by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is prevalent among the staff of a hospital providing treatment to patients with severe coronavirus disease 2019 (COVID-19) using radioligand assay (RLA). One thousand samples from the staff of a general hospital providing treatment to patients with severe COVID-19 were assayed for SARS-CoV-2 nucleocapsid protein (N) IgG using RLA. Nine patients with COVID-19 who had been treated in inpatient settings and had already recovered were used as control subjects, and 186 blood donor samples obtained more than 10 years ago were used as negative controls. Four of the 1000 samples showed apparently positive results, and approximately 10 or more samples showed slightly high counts. Interestingly, a few among the blood donor samples also showed slightly high values. To validate the results, antibody examinations using ELISA and neutralizing antibody tests were performed on 21 samples, and chemiluminescence immunoassay (CLIA) was performed on 201 samples, both resulting in a very high correlation. One blood donor sample showed slightly positive results in both RLA and CLIA, suggesting a cross-reaction. This study showed that five months after the pandemic began in Japan, the staff of a general hospital with a tertiary emergency medical facility had an extremely low seroprevalence of the antibodies against SARS-CoV-2. Further investigation will be needed to determine whether the slightly high results were due to cross-reactions or a low titer of anti-SARS-CoV-2 antibodies. The quantitative RLA was considered sensitive enough to detect low titers of antibodies.

    Abstract: The origin of deep-sea sediments in the western North Pacific Ocean, which are significantly enriched in rare-earth elements and yttrium (REY), and its paleoceanographic implications have been poorly constrained. Here, we investigated stratigraphic variations in the chemical compositions and textures of ferromanganese (Fe–Mn) micronodules separated from western North Pacific sediments. The characteristics of the micronodules of an extremely REY-rich mud layer vary from almost purely diagenetic to relatively hydrogenetic. This indicates the abundant supply of organic matter to the sediment together with fish debris that accumulates REY at the onset of the REY-enrichment of the mud, followed by the exposure of the seafloor to oxic water masses during the latter half of the formation of the REY-rich mud. These results support a previously proposed formation mechanism based on which enhanced bottom water currents caused pelagic fish proliferation via the upwelling of nutrients and fish debris was physically sorted and selectively accumulated on the seafloor. After the main REY-enrichment, the micronodules exhibit varying diagenetic signatures, suggesting changes in the bottom current intensities after the main REY-enrichment. However, the bulk REY contents do not increase. This implies that a sufficient increase in the fish productivity is an essential factor affecting the formation of REY-rich mud.

    Abstract: Current approaches to the quality control of water are unsatisfying due to either a high cost or the inability to capture all of the relevant information. In this study, near-infrared spectroscopy (NIRS) with aquaphotomics as a novel approach was assessed for the discrimination of natural, processed and aged water samples. Temperature perturbation of water samples was employed to probe the aqueous systems and reveal the hidden information. A radar chart named an aquagram was used to visualize and compare the absorbance spectral patterns of waters at different temperatures. For the spectra acquired at a constant temperature of 30 °C, the discrimination analysis of different water samples failed to produce satisfying results. However, under perturbation by increasing the temperature from 35 to 60 °C, the absorbance spectral pattern of different waters displayed in aquagrams revealed different, water-specific dynamics. Moreover, it was found that aged processed water changed with the temperature, whereas the same processed water, when freshly prepared, had hydrogen bonded structures unperturbed by temperature. In summary, the aquaphotomics approach to the NIRS analysis showed that the water absorbance spectral pattern can be used to describe the character and monitor dynamics of each water sample as a complex molecular system, whose behavior under temperature perturbation can reveal even subtle changes, such as aging and the loss of certain qualities during storage.

    Abstract: In a plating process, the steel plate is conveyed 20–50 m in the vertical direction for drying, during which it is negligibly supported by rollers and other mechanisms. This produces plating without uniformity owing to the generation of vibration and other factors, which prevent the increase in productivity. We have developed a noncontact guide system for a high-speed traveling elastic steel plate in which electromagnetic forces are applied by actuators at the edges of the plate to control the plate’s position. In this study, we investigated the vibration phenomenon when changing the steady current value of the electromagnet used for controlling the position. In addition, we conducted mode analysis of the steel plate to enable stable control even at low steady current values and verified whether stable guide can be provided by using it together with a permanent magnet. As a result, by arranging the permanent magnets, stable guidance was possible even at a low steady current value. In addition, it became clear that vibration damping performance is also improved.

    Abstract: Deep-sea sediments with total rare-earth elements and yttrium (ΣREY) concentrations exceeding 400 ppm, which are termed REY-rich mud, are widely distributed in the world oceans. Specifically, deep-sea sediments within the Japanese exclusive economic zone (EEZ) surrounding Minamitorishima Island in the western North Pacific have attracted significant attention as a new REY resource, because they contain REY-enriched layers exceeding 2000 ppm of ΣREY. However, neither the sediments deeper than 15 m below the seafloor (mbsf) nor those outside the Minamitorishima EEZ have ever been studied. Recently, a number of distinct geochemical features which are aligned in stratigraphic order were recognized in these sediments, based on multi-elemental composition data. Chemostratigraphy enables us to laterally correlate three REY peaks among apparently featureless pelagic clays. Here, we apply chemostratigraphic correlation to 19 new cores collected from the northern Pigafetta Basin and several small basins within the Marcus-Wake Seamounts. This study revealed that the REY-enriched layers occur at greater depths than the piston core length in a wider area than previously investigated (e.g., ~20.5 mbsf at Ocean Drilling Program Site 801A). This finding suggests that the depositional environments in these areas were basically similar, although local geographic conditions could have affected the continuity of REY peaks.
