    Abstract: Herein, gold–nitrogen-doped graphene quantum dot (Au/NGQD) composite modified electrodes were fabricated and applied as “on–off” nanosensors for the photo-electrochemical (PEC) detection of caffeic acid under visible-light irradiation. An effective and simple strategy was established for the preparation of Au/NGQD composites by hydrothermal and calcination methods. Owing to the quantum confinement effect of NGQDs, the local surface plasmon resonance (LSPR) effect of Au nanoparticles (NPs), and the synergistic effect between Au and NGQDs NPs, the Au/NGQDs showed excellent PEC performance, with wide linear concentration ranges (0.11 to 30.25 μM and 30.25 to 280.25 μM), a low detection limit (0.03 μM), excellent sensitivity, and high stability. The present study may provide an advanced strategy for the simple design of Au/NGQD composites to allow their effective application for selective and sensitive sensing of small biological molecules.

    Abstract: In recent years, with the development of geological prospecting from shallow ore to deep and hidden ore, the difficulty of prospecting is increasing day by day, so the application of computer technology and new methods of geological and mineral exploration is paid more and more attention. The mining and prediction of geological prospecting information based on deep learning have become the frontier field of earth science. However, as a deep artificial intelligence algorithm, deep learning still has many problems to be solved in the big data mining and prediction of geological prospecting, such as the small number of training samples of geological and mineral images, the difficulty of building deep learning network models, and the universal applicability of deep learning models. In this paper, the training samples and convolutional neural network models suitable for geochemical element data mining are constructed to solve the above problems, and the model is successfully applied to the prediction research of gold, silver, lead and zinc polymetallic metallogenic areas in South China. Taking the Pangxidong research area in the west of Guangdong Province as an example, this paper carries out prospecting target prediction research based on a 1:50000 stream sediment survey original data. Firstly, the support vector machine (SVM) model and statistical method were used to determine the ore-related geochemical element assemblage. Secondly, the experimental data of geochemical elements were augmented and a dataset was established. Finally, ResNet-50 neural network model is used for data training and prediction research. The experimental results show that the areas numbered 9, 29, 38, 40, 95, 111, 114, 124, 144 have great metallogenic potential, and this method would be a promising tool for metallogenic prediction. By applying the ResNet-50 neural network in metallogenic prediction, it can provide a new idea for the future exploration of mineral resources. In order to verify the generality of the research method in this paper, we conducted experimental tests on the geochemical dataset of B area, another deposit research area in South China. The results show that 100% of the prediction area obtained by using the proposed method covers the known ore deposit area. This model also provides method support for further delineating the prospecting target area in study area B.

    Abstract: Yaks are often subject to long-term starvation and a high prevalence of respiratory diseases and mortality in the withered season, yet the mechanisms that cause this remain unclear. Research has demonstrated that β-hydroxybutyrate (BHB) plays a significant role in regulating the immune system. Hence, we hypothesize that the low glucose and high BHB condition induced by severe starvation might have an effect on the pro-inflammatory response of the alveolar macrophages (AMs) in yaks. To validate our hypothesis, we isolated and identified primary AMs from freshly slaughtered yaks and cultured them in a medium with 5.5 mM of glucose or 2.8 mM of glucose plus 1–4 mM of BHB. Utilizing a real-time quantitative polymerase chain reaction (RT-qPCR), immunoblot assay, and enzyme-linked immunosorbent assay (ELISA), we evaluated the gene and protein expression levels of GPR109A (G-protein-coupled receptor 109A), NF-κB p65, p38, and PPARγ and the concentrations of pro-inflammatory cytokines interleukin (IL)-1β and IL-6 and tumor necrosis factor (TNF)-α in the supernatant. The results demonstrated that AMs exposed to low glucose plus BHB had significantly higher levels of IL-1β, IL-6, and TNF-α (p 〈 0.05) and higher activity of the GPR109A/NF-κB signaling pathway. A pretreatment of either pertussis toxin (PTX, inhibitor of GPR109A) or pyrrolidinedithiocarbamic (PDTC, inhibitor of NF-κB p65) was effective in preventing the elevated secretion of pro-inflammatory cytokines induced by low glucose plus BHB (p 〈 0.05). These results indicated that the low glucose plus BHB condition would induce an enhanced pro-inflammatory response through the activation of the GPR109A/NF-κB signaling pathway in primary yak AMs, which is probably the reason why yaks experience a higher rate of respiratory diseases and mortality. This study will offer new insight into the prevention and treatment of bovine respiratory diseases.

    Abstract: Superoxide dismutases (SODs) play critical roles in plants, especially in the maintenance of redox homeostasis. The response of SODs in Citrus (Citrus sinensis (L.) Osbeck) to citrus bacterial canker (CBC) infection were investigated. The CsSODs were identified, and their gene structures, phylogeny, conserved domains and motifs, predicted interactions, and chromosomal distribution were analyzed. CsSOD expression in response to stress-related plant hormones (salicylic acid, SA; methyl jasmonate, MeJA; and abscisic acid, ABA) and Xanthomonas citri subsp. citri (Xcc) infection were also investigated. Thirteen CsSODs were identified in C. sinensis, including four Fe/MnSODs and nine Cu/ZnSODs with typical functional domains. The CsSODs were distributed on chromosomes 3, 5, 7, and 8. Specific hormone-response motifs were identified in the gene promoter regions. Ten genes were induced by MeJA treatment, as shown by qRT-PCR, and were upregulated in the CBC-susceptible Wanjincheng citrus variety, while CsSOD06 and CsSOD08 were upregulated by ABA in both the Wanjincheng and the CBC-resistant Kumquat varieties. Xcc infection significantly altered the levels of most CsSODs. The overexpression of CsSOD06 and CsSOD08 resulted in increased hydrogen peroxide levels and SOD activity. Our findings highlight the significance of SOD enzymes in the plant response to pathogen infection and have a potential application for breeding CBC-tolerant citrus varieties.
