    Abstract: This study focused on elucidating the possibilities of improving current trapping methods for Ips typographus (Linnaeus, 1758). Three field experiments were conducted simultaneously in one study area in the German federal state of Saxony. A comparison of six different commercial attractants revealed a significant superiority of Typosan®, especially for adult beetles after hibernation in the phase of their first swarming. It also attracted fewer individuals of Thanasimus spp. than the other highly attractive products Pheroprax® and IT Ecolure Extra®. Increasing the Pheroprax® application rate by using four instead of one dispenser in a single trap increased the total catch of I. typographus only by 15.5%. In contrast, Thanasimus spp. catch increased by 195.5% when four dispensers were used. A test of different trap types showed a species-specific catching capability, with the 12-funnel WitaTrap® being the most effective in catching I. typographus. The quantity of Thanasimus spp. bycatch in multiple-funnel traps demonstrated the necessity of a selective mechanism to minimize impacts on predator populations. Although we were not able to identify new milestones towards mass trapping, this study contributes to necessary improvements of current trapping methods. Especially in future stands with a smaller share of Norway spruce (Picea abies Karsten, 1881) the weakened beetle population in spring could be effectively reduced by properly conducted mass trapping.

    Abstract: Grapes provide a rich source of polyphenols and fibers. This study aimed to evaluate the effect of the daily consumption of 46 g of whole grape powder, providing the equivalent of two servings of California table grapes, on the gut microbiome and cholesterol/bile acid metabolism in healthy adults. This study included a 4-week standardization to a low-polyphenol diet, followed by 4 weeks of 46 g of grape powder consumption while continuing the low-polyphenol diet. Compared to the baseline, 4 weeks of grape powder consumption significantly increased the alpha diversity index of the gut microbiome. There was a trend of increasing Verrucomicrobia (p = 0.052) at the phylum level, and a significant increase in Akkermansia was noted. In addition, there was an increase in Flavonifractor and Lachnospiraceae_UCG-010, but a decrease in Bifidobacterium and Dialister at the genus level. Grape powder consumption significantly decreased the total cholesterol by 6.1% and HDL cholesterol by 7.6%. There was also a trend of decreasing LDL cholesterol by 5.9%, and decreasing total bile acid by 40.9%. Blood triglyceride levels and body composition were not changed by grape powder consumption. In conclusion, grape powder consumption significantly modified the gut microbiome and cholesterol/bile acid metabolism.

    Abstract: TRPA1, a versatile ion channel of the Transient Receptor Potential (TRP) channel family, detects a large variety of chemicals and can contribute to signal processing of other stimuli, e.g., due to its sensitivity to cytosolic calcium elevation or phosphoinositolphosphate modulation. At first, TRPA1 was found on sensory neurons, where it can act as a sensor for potential or actual tissue damage that ultimately may elicit pain or itch as warning symptoms. This review provides an update regarding the analgesic and antipruritic potential of TRPA1 modulation and the respective clinical trials. Furthermore, TRPA1 has been found in an increasing amount of other cell types. Therefore, the main focus of the review is to discuss the non-analgesic and particularly the disease-modifying potential of TRPA1. This includes diseases of the respiratory system, cancer, ischemia, allergy, diabetes, and the gastrointestinal system. The involvement of TRPA1 in the respective pathophysiological cascades is so far mainly based on pre-clinical data.

    Abstract: The mechanism of acetaminophen (APAP) analgesia is at least partially unknown. Previously, we showed that the APAP metabolite N-acetyl-p-benzoquinone imine (NAPQI) activated Kv7 channels in neurons in vitro, and this activation of Kv7 channels dampened neuronal firing. Here, the effect of the Kv7 channel blocker XE991 on APAP-induced analgesia was investigated in vivo. APAP had no effect on naive animals. Induction of inflammation with λ-carrageenan lowered mechanical and thermal thresholds. Systemic treatment with APAP reduced mechanical hyperalgesia, and co-application of XE991 reduced APAP’s analgesic effect on mechanical pain. In a second experiment, the analgesic effect of systemic APAP was not antagonized by intrathecal XE991 application. Analysis of liver samples revealed APAP and glutathione-coupled APAP indicative of metabolization. However, there were no relevant levels of these metabolites in cerebrospinal fluid, suggesting no relevant APAP metabolite formation in the CNS. In summary, the results support an analgesic action of APAP by activating Kv7 channels at a peripheral site through formation of the metabolite NAPQI.

    Abstract: Transient receptor potential cation channel subfamily A member 1 (TRPA1), an ion channel primarily expressed on sensory neurons, can be activated by substances occurring during myocardial infarction. Aims were to investigate whether activation, inhibition, or absence of TRPA1 affects infarcts and to explore underlying mechanisms. In the context of myocardial infarction, rats received a TRPA1 agonist, an antagonist, or vehicle at different time points, and infarct size was assessed. Wild type and TRPA1 knockout mice were also compared in this regard. In vitro, sensory neurons were co-cultured with cardiomyocytes and subjected to a model of ischemia-reperfusion. Although there was a difference between TRPA1 activation or inhibition in vivo, no experimental group was different to control animals in infarct size, which also applies to animals lacking TRPA1. In vitro, survival probability of cardiomyocytes challenged by ischemia-reperfusion increased from 32.8% in absence to 45.1% in presence of sensory neurons, which depends, at least partly, on TRPA1. This study raises doubts about whether TRPA1 is a promising target to reduce myocardial damage within a 24 h period. The results are incompatible with relevant enlargements of infarcts by TRPA1 activation or inhibition, which argues against adverse effects when TRPA1 is targeted for other indications.

    Abstract: Background: Preclinical studies suggest that methylglyoxal (MG) increases within the myocardium upon acute myocardial infarction (AMI) and thereafter contributes to adverse postinfarct remodeling. The aims of this study were to test whether MG increases in plasma of humans after AMI and whether this increase is related to the left ventricular ejection fraction (LVEF). Methods: The plasma samples of 37 patients with ST elevation AMI undergoing primary percutaneous coronary intervention (pPCI) acquired in a previously conducted randomized controlled trial testing remote ischemic conditioning (RIC) were analyzed by means of high-performance liquid chromatography. Time courses of the variables were analyzed by means of mixed linear models. Multiple regression analyses served to explore the relationship between MG levels and the LVEF. Results: Compared to the MG levels upon admission due to AMI, the levels were increased 2.4-fold (95% CI, 1.6–3.6) 0.5 h after reperfusion facilitated by pPCI, 2.6-fold (1.7–4.0) after 24 h and largely returned to the baseline after 30 d (1.1-fold, 0.8–1.5). The magnitude of the MG increase was largely independent of that of cardiac necrosis markers. Overall, the highest MG values within 24 h after AMI were associated with the lowest LVEF after 4 d. While markers of myocardial necrosis and stretch quantified within the first 24 h explained 52% of the variance of the LVEF, MG explained additional 23% of the variance (p 〈 0.001). Conclusions: Considering these observational data, it is plausible that the preclinical finding of MG generation after AMI negatively affecting the LVEF also applies to humans. Inhibition of MG generation or MG scavenging might provide a novel therapeutic strategy to target post-AMI myocardial remodeling and dysfunction.

    Abstract: The development of X-ray-absorbing scintillating nanoparticles is of high interest for solving the short penetration depth problem of visible and infrared light in photodynamic therapy (PDT). Thus, these nanoparticles are considered a promising treatment for several types of cancer. Herein, gadolinium oxide nanoparticles doped with europium ions (Gd2O3:Eu3+) were obtained by using polyvinyl alcohol as a capping agent. Hybrid silica nanoparticles decorated with europium-doped gadolinium oxide (SiO2-Gd2O3:Eu3+) were also prepared through the impregnation method. The synthesized nanoparticles were structurally characterized and tested to analyze their biocompatibility. X-ray diffraction, scanning electron microscopy, and transmission electron microscopy confirmed the high crystallinity and purity of the Gd2O3:Eu3+ particles and the homogeneous distribution of nanostructured rare earth oxides throughout the fumed silica matrix for SiO2-Gd2O3:Eu3+. Both nanoparticles displayed stable negative ζ-potentials. The photoluminescence properties of the materials were obtained using a Xe lamp as an excitation source, and they exhibited characteristic Eu3+ bands, including at 610 nm, which is the most intense transition band of this ion. Cytotoxicity studies on mouse glioblastoma GL261 cells indicated that these materials appear to be nontoxic from 10 to 500 μg·mL−1 and show a small reduction in viability in non-tumor cell lines. All these findings demonstrate their possible use as alternative materials in PDT.

    Abstract: The ever-increasing complexity of cyber-physical systems is driving the need for assurance of critical infrastructure and embedded systems. However, traditional methods to secure cyber-physical systems—e.g., using cyber best practices, adapting mechanisms from information technology systems, and penetration testing followed by patching—are becoming ineffective. This paper describes, in detail, Verification Evidence and Resilient Design In anticipation of Cybersecurity Threats (VERDICT), a language and framework to address cyber resiliency. When we use the term resiliency, we mean hardening a system such that it anticipates and withstands attacks. VERDICT analyzes a system in the face of cyber threats and recommends design improvements that can be applied early in the system engineering process. This is done in two steps: (1) Analyzing at the system architectural level, with respect to cyber and safety requirements and (2) by analyzing at the component behavioral level, with respect to a set of cyber-resiliency properties. The framework consists of three parts: (1) Model-Based Architectural Analysis and Synthesis (MBAAS); (2) Assurance Case Fragments Generation (ACFG); and (3) Cyber Resiliency Verifier (CRV). The VERDICT language is an Architecture Analysis and Design Language (AADL) annex for modeling the safety and security aspects of a system’s architecture. MBAAS performs probabilistic analyses, suggests defenses to mitigate attacks, and generates attack-defense trees and fault trees as evidence of resiliency and safety. It can also synthesize optimal defense solutions—with respect to implementation costs. In addition, ACFG assembles MBAAS evidence into goal structuring notation for certification purposes. CRV analyzes behavioral aspects of the system (i.e., the design model)—modeled using the Assume-Guarantee Reasoning Environment (AGREE) annex and checked against cyber resiliency properties using the Kind 2 model checker. When a property is proved or disproved, a minimal set of vital system components responsible for the proof/disproof are identified. CRV also provides rich and localized diagnostics so the user can quickly identify problems and fix the design model. This paper describes the VERDICT language and each part of the framework in detail and includes a case study to demonstrate the effectiveness of VERDICT—in this case, a delivery drone.
