    Abstract: At present, liquid starter fertilizer (LSF) application technologies experience problems with low fertilizer utilization efficiency. In this study, we adopted a method of precise application of LSF near the seeds on seed bed in point form during sowing. A precise application system that can detect seed information in real time and control the solenoid valve to open automatically was developed for this method. The LSF supply system and detection control system were studied in detail. Field experiments were conducted to evaluate the performance of the precise application system in terms of operation quality (qualified index of the length of the LSF, QIL; the amount of the LSF, FA; and qualified index of the distance between the seeds and the LSF, QID) at forward speeds of 4, 6, and 8 km/h and pressures of 0.10, 0.15, 0.20, 0.25, and 0.30 MPa. The results indicated that QIL was 96.4%, the range of FA was 1.34 to 13.86 mL, and QID was 82.6%, which signifies the developed system meets the demands of precise LSF application. This method achieves the target of improving fertilizer use efficiency and provides a reference for developing fertilization devices for precisely applying LSF.

    Abstract: Soybeans are an important cash crop and are widely used as a source of vegetable protein and edible oil. MicroRNAs (miRNA) are endogenous small RNA that play an important regulatory role in the evolutionarily conserved system of gene expression. In this study, we selected four lines with extreme phenotypes, as well as high or low protein and oil content, from the chromosome segment substitution line (CSSL) constructed from suinong (SN14) and ZYD00006, and planted and sampled at three stages of grain development for small RNA sequencing and expression analysis. The sequencing results revealed the expression pattern of miRNA in the materials, and predicted miRNA-targeted regulatory genes, including 1967 pairs of corresponding relationships between known-miRNA and their target genes, as well as 597 pairs of corresponding relationships between novel-miRNA and their target genes. After screening and annotating genes that were targeted for regulation, five specific genes were identified to be differentially expressed during seed development and subsequently analyzed for their regulatory relationship with miRNAs. The expression pattern of the targeted gene was verified by Real-time Quantitative PCR (RT-qPCR). Our research provides more information about the miRNA regulatory network in soybeans and further identifies useful genes that regulate storage during soy grain development, providing a theoretical basis for the regulation of soybean quality traits.

    Abstract: A P(V)-N activation method based on nucleoside phosphoropiperidate/DCI system has been developed for improved synthesis of diverse UDP-furanoses. The reaction conditions including temperature, amount of activator, and reaction time were optimized to alleviate the degradation of UDP-furanoses to cyclic phosphates. In addition, an efficient and facile phosphoramidite route was employed for the preparation of furanosyl-1-phosphates.

    Abstract: The Dicer protein is one of the most important components of RNAi machinery because it regulates the production of small RNAs (sRNAs) in eukaryotes. Here, Dicer1-like gene (Pit-DCL1) and Dicer2-like gene (Pit-DCL2) RNAi transformants were generated via pSilent-1 in Penicillium italicum (Pit), which is the causal agent of citrus blue mold. Neither transformant showed a change in mycelial growth or sporulation ability, but the pathogenicity of the Pit-DCL2 RNAi transformant to citrus fruits was severely impaired, compared to that of the Pit-DCL1 RNAi transformant and the wild type. We further developed a citrus wound-mediated RNAi approach with a double-stranded fragment of Pit-DCL2 generated in vitro, which achieved an efficiency in reducing Pi-Dcl2 expression and virulence that was similar to that of protoplast-mediated RNAi in P. italicum, suggesting that this approach is promising in the exogenous application of dsRNA to control pathogens on the surface of citrus fruits. In addition, sRNA sequencing revealed a total of 69.88 million potential sRNAs and 12 novel microRNA-like small RNAs (milRNAs), four of which have been predicated on target innate immunity or biotic stress-related genes in Valencia orange. These data suggest that both the Pit-DCL1 and Pit-DCL2 RNAi transformants severely disrupted the biogenesis of the potential milRNAs, which was further confirmed for some milRNAs by qRT-PCR or Northern blot analysis. These data suggest the sRNAs in P. italicum that may be involved in a molecular virulence mechanism termed cross-kingdom RNAi (ck-RNAi) by trafficking sRNA from P. italicum to citrus fruits.

    Abstract: To prevent citrus decay caused by Geotrichum citri-aurantii, 12 natural products were isolated from two endophytic fungi, in which cytosporone B was shown to have excellent bioactivity for control of G. citri-aurantii with median effect concentration (EC50) of 26.11 μg/mL and minimum inhibitory concentration (MIC) of 105 μg/mL, and also significantly reduced the decay of sugar orange during the in vivo trials. In addition, cytosporone B could alter the morphology of G. citri-aurantii by causing distortion of the mycelia and loss of membrane integrity. Differentially expressed genes (DEGs) between cytosporone B-treated and -untreated samples were revealed by Illumina sequencing, including 3540 unigenes. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses showed that most DEGs were related to metabolic production and cell membrane. These findings suggest cytosporone B is a promising biological preservative to control citrus decay and reveal the action mechanism of cytosporone B in relation to the destruction of the fungal cell membrane at both morphological and molecular levels.

    Abstract: The influenza vaccination coverage among children is low in China. We aimed to conduct a nationwide survey to quantify parental preferences and willingness to pay (WTP) for influenza vaccination for their children. Parents with children aged six months to 18 years from six provinces in China were investigated by a discrete choice experiment regarding six influenza vaccination attributes. Mixed logit models were used to estimate the relative importance of vaccine attributes and parents’ WTP. Interaction analysis and subgroup analysis were conducted to explore preference heterogeneity. A total of 1206 parents were included in the analysis. Parents reported vaccine effectiveness as the most important vaccine attribute. The mode of vaccine administration had no significant impact on parents’ preferences. Parents aged over 30 years with higher education or income levels were more likely to prefer no influenza vaccination for their children. The largest marginal WTP (CNY 802.57) for vaccination and the largest increase in vaccine uptake (41.85%) occurred with improved vaccine effectiveness from 30% to 80%. Parents from central regions or mid-latitude areas had a relatively lower WTP than those from other regions. No significant difference in the relative importance of vaccine attributes were observed among parents from various regions of China.

    Abstract: Interferometric synthetic aperture radar (InSAR) data from 6 Envisat ASAR descending tracks; spanning the 2003–2010 period; was used to measure interseismic strain accumulation across the Northeastern Tibetan Plateau. Mean line-of-sight (LOS) ratemaps are computed by stacking atmospheric-corrected and orbital-corrected interferograms. The ratemaps from one track with different atmospheric-corrected results or two parallel; partially overlapping tracks; show a consistent pattern of left-lateral motion across the fault; which demonstrates the MERIS and ECMWF atmospheric correction works satisfactorily for small stain measurement of this region; even with a limited number of interferograms. By combining the measurements of InSAR and GPS; a fine crustal deformation velocity and strain rate field was estimated on discrete points with irregular density depending on the fault location; which revealed that the present-day slip rate on the Haiyuan fault system varies little from west to east. A change (2–3 mm/year) in line-of-sight (LOS) deformation rate across the fault is observed from the Jinqianghe segment to its eastern end. Inversion from the cross-fault InSAR profiles gave a shallow locking depth of 3–6 km on the main rupture of the 1920 earthquake. We therefore infer that the middle-lower part of the seismogenic layer on the 1920 rupture is not yet fully locked since the 1920 large earthquake. Benefit from high spatial resolution InSAR data; a low strain accumulation zone with high strain rates on its two ends was detected; which corresponds to the creeping segment; i.e., the Laohushan fault segment. Contrary to the previous knowledge of squeezing structure; an abnormal tension zone is disclosed from the direction map of principal stress; which is consistent with the recent geological study. The distribution of principal stress also showed that the expanding frontier of the northeastern plateau has crossed the Liupan Shan fault zone; even arrived at the northeast area of the Xiaoguan Shan. This result agrees with the deep seismic reflection profile.
