    Abstract: Carbon doping in the buffer of AlGaN/GaN high-electron-mobility transistors (HEMTs) leads to the notorious current collapse phenomenon. In this paper, an HEMT structure with a source-connected p-GaN (SCPG) embedded in the carbon-doped semi-insulating buffer is proposed to suppress the buffer-induced current collapse effect. Two-dimensional transient simulation was carried out to show the successful suppression of buffer-induced current collapse in the SCPG-HEMTs compared with conventional HEMTs. The mechanism of suppressing dynamic on-resistance degradation by ejecting holes from the SCPG into the high resistive buffer layer after off-state stress is illustrated based on energy band diagrams. This paper contributes an innovative device structure to potentially solve the buffer-induced degradation of the dynamic on-resistance in GaN power devices.

    Abstract: The study was aimed to develop a predictive model to identify patients who may benefit from performing systematic random biopsies (SB) in addition to targeted biopsies (TB) in men suspected of having prostate cancer. A total of 198 patients with positive pre-biopsy MRI findings and who had undergone both TB and SB were prospectively recruited into this study. The primary outcome was detection rates of clinically significant prostate cancer (csPCa) in SB and TB approaches. The secondary outcome was net clinical benefits of SB in addition to TB. A logistic regression model and nomogram construction were used to perform a multivariate analysis. The detection rate of csPCa using SB was 51.0% (101/198) compared to a rate of 56.1% (111/198) for TB, using a patient-based biopsy approach. The detection rate of csPCa was higher using a combined biopsy (64.6%; 128/198) in comparison to TB (56.1%; 111/198) alone. This was statistically significant (p 〈 0.001). Age, PSA density and PIRADS score significantly predicted the detection of csPCa by SB in addition to TB. A nomogram based on the model showed good discriminative ability (C-index; 78%). The decision analysis curve confirmed a higher net clinical benefit at an acceptable threshold.

    Abstract: With rapid developments in society and economy, the concentrations of heavy metals in surface water in South China have increased significantly, which poses a serious threat to the regional water security. In this study, the Fuhe River watershed in South China was selected as the study area to analyze physicochemical characteristics and heavy metal (Cu, Zn, Pb, Cd and Cr) concentrations in river water in the dry and rainy seasons, in 2019, with the purpose of exploring their spatial–temporal variations and main influences and assessing the potential health risks of heavy metals. The pH value of river water varied from 5.82 to 7.97, and it fluctuated less overall in the dry season, but it oscillated greatly in the rainy season and was lower, especially in the lower reach of the Fuhe River. The electrical conductivity (EC) value changed between 33 μS/cm and 128 μS/cm and increased and fluctuated along the river flow in the two periods. The concentrations of Cd, Cr, Cu, Pb, and Zn in river water showed obvious differences between the two periods. The concentrations of Cd and Cu were lower with the ranges from 0.001 μg/L to 0.67 μg/L and from 1 μg/L to 12 μg/L, respectively, in the dry season than in the rainy season, while there were inverse cases for other heavy metals. Along the river flow, the Cr concentration was stable, whereas other heavy metals showed increasing trends. It was noted that the concentrations of heavy metals in the Fuhe River were, on average, lower than the Chinese drinking standard values, with the concentration of Pb in the dry season significantly higher than the drinking standard value of the World Health Organization (WHO). Principal component analysis and correlation analysis showed that rock weathering and anthropogenic inputs were the main controlling factors of Cu and Zn in the Fuhe River, and human activities were mainly responsible for Pb, Cr, and Cd concentrations. The health risk assessment results showed that the non-carcinogenic risk (HQingestion) value of Pb was greater than 1 in most sampling points in the middle and lower reaches in the dry season, suggesting a significant non-carcinogenic risk for adults and children by direct ingestion. The minimum carcinogenic risk (CRingestion) value of Cr was more than 10−4 in the rainy and dry seasons, and the CRingestion value of Cd in some sampling points was more than 10−4 in the rainy season, indicating significant cancer risks to adults and children. For areas with significant pollution and health risks in the Fuhe River watershed, it is urgent to strengthen the controls of industrial, agricultural, and urban wastewater discharge.

    Abstract: As the lightest metal structural material, magnesium and its alloys have the characteristics of low density, high specific strength and good biocompatibility, which gives magnesium alloys broad application prospects in fields of biomedicine, transportation, and aerospace. Laser selective melting technology has the advantages of manufacturing complex structural parts, high precision and high degree of freedom. However, due to some disadvantages of magnesium alloy, such as low boiling point and high vapor pressure, the application of it in laser selective melting was relatively undeveloped compared with other alloys. In this paper, the fabrication, microstructure, mechanical performance and corrosion resistance property of magnesium alloys were summarized, and the potential applications and the development direction of selective laser melting magnesium alloys in the future are prospected.

    Abstract: For the sound field reconstruction of large conical surfaces, current statistical optimal near-field acoustic holography (SONAH) methods have relatively poor applicability and low accuracy. To overcome this problem, conical SONAH based on cylindrical SONAH is proposed in this paper. Firstly, elementary cylindrical waves are transformed into those suitable for the radiated sound field of the conical surface through cylinder-cone coordinates transformation, which forms the matrix of characteristic elementary waves in the conical spatial domain. Secondly, the sound pressure is expressed as the superposition of those characteristic elementary waves, and the superposition coefficients are solved according to the principle of superposition of wave field. Finally, the reconstructed conical pressure is expressed as a linear superposition of the holographic conical pressure. Furthermore, to overcome ill-posed problems, a regularization method combining truncated singular value decomposition (TSVD) and Tikhonov regularization is proposed. Large singular values before the truncation point of TSVD are not processed and remaining small singular values representing high-frequency noise are modified by Tikhonov regularization. Numerical and experimental case studies are carried out to validate the effectiveness of the proposed conical SONAH and the combined regularization method, which can provide reliable evidence for noise monitoring and control of mechanical systems.

    Abstract: To develop new anti-inflammatory agents, a series of 7-O-amide hesperetin derivatives was designed, synthesized and evaluated for anti-inflammatory activity using RAW264.7 cells. All compounds showed inhibitory effect on LPS-induced NO production. Among them, 7-O-(2-(Propylamino)-2-oxoethyl)hesperetin (4d) and 7-O-(2-(Cyclopentylamino)-2-oxoethyl)hesperetin (4k) with hydrophobic side chains exhibited the most potent NO inhibitory activity (IC50 = 19.32 and 16.63 μM, respectively), showing stronger inhibitory effect on the production of pro- inflammatory cytokines tumor necrosis factor (TNF-α), interleukin-6 (IL-6) and interleukin-1β (IL-1β) than indomethacin and celecoxib at 10 μM. The structure-activity relationships (SARs) suggested that the 7-O-amide unit was buried in a medium-sized hydrophobic cavity of the bound receptor. Furthermore, compound 4d could also significantly suppress the expression of inducible nitric oxide synthase enzymes (iNOS) and cyclooxygenase-2 (COX-2), through the nuclear factor-kappa B (NF-κB) signaling pathway.

    Abstract: Background: Maize mesophyll (M) cells play important roles in various biological processes such as photosynthesis II and secondary metabolism. Functional differentiation occurs during M-cell development, but the underlying mechanisms for regulating M-cell development are largely unknown. Results: We conducted single-cell RNA sequencing (scRNA-seq) to profile transcripts in maize leaves. We then identified coregulated modules by analyzing the resulting pseudo-time-series data through gene regulatory network analyses. WRKY, ERF, NAC, MYB and Heat stress transcription factor (HSF) families were highly expressed in the early stage, whereas CONSTANS (CO)-like (COL) and ERF families were highly expressed in the late stage of M-cell development. Construction of regulatory networks revealed that these transcript factor (TF) families, especially HSF and COL, were the major players in the early and later stages of M-cell development, respectively. Integration of scRNA expression matrix with TF ChIP-seq and Hi-C further revealed regulatory interactions between these TFs and their targets. HSF1 and COL8 were primarily expressed in the leaf bases and tips, respectively, and their targets were validated with protoplast-based ChIP-qPCR, with the binding sites of HSF1 being experimentally confirmed. Conclusions: Our study provides evidence that several TF families, with the involvement of epigenetic regulation, play vital roles in the regulation of M-cell development in maize.

    Abstract: Intraguild predation (IGP) can have a significant impact on phytoplankton biomass, but its effects on their diversity and assemblage composition are not well understood. In this study, we constructed an IGP model based on the common three-trophic food chain of “fish (or shrimp)–large branchiopods (Daphnia)–phytoplankton”, and investigated the effects of IGP on phytoplankton assemblage composition and diversity in outdoor mesocosms using environmental DNA high-throughput sequencing. Our results indicated that the alpha diversities (number of amplicon sequence variants and Faith’s phylogenetic diversity) of phytoplankton and the relative abundance of Chlorophyceae increased with the addition of Pelteobagrus fulvidraco, while similar trends were found in alpha diversities but with a decrease in the relative abundance of Chlorophyceae in the Exopalaemon modestus treatment. When both predators were added to the community, the strength of collective cascading effects on phytoplankton alpha diversities and assemblage composition were weaker than the sum of the individual predator effects. Network analysis further showed that this IGP effect also decreased the strength of collective cascading effects in reducing the complexity and stability of the phytoplankton assemblages. These findings contribute to a better understanding of the mechanisms underlying the impacts of IGP on lake biodiversity, and provide further knowledge relevant to lake management and conservation.

    Abstract: DNA G-quadruplexes (G4s) are non-canonical four-stranded DNA structures involved in various biological processes in eukaryotes. Molecularly crowded solutions and monovalent cations have been reported to stabilize in vitro and in vivo G4 formation. However, how K+ and Na+ affect G4 formation genome-wide is still unclear in plants. Here, we conducted BG4-DNA-IP-seq, DNA immunoprecipitation with anti-BG4 antibody coupled with sequencing, under K+ and Na+ + PEG conditions in vitro. We found that K+-specific IP-G4s had a longer peak size, more GC and PQS content, and distinct AT and GC skews compared to Na+-specific IP-G4s. Moreover, K+- and Na+-specific IP-G4s exhibited differential subgenomic enrichment and distinct putative functional motifs for the binding of certain trans-factors. More importantly, we found that K+-specific IP-G4s were more associated with active marks, such as active histone marks, and low DNA methylation levels, as compared to Na+-specific IP-G4s; thus, K+-specific IP-G4s in combination with active chromatin features facilitate the expression of overlapping genes. In addition, K+- and Na+-specific IP-G4 overlapping genes exhibited differential GO (gene ontology) terms, suggesting they may have distinct biological relevance in rice. Thus, our study, for the first time, explores the effects of K+ and Na+ on global G4 formation in vitro, thereby providing valuable resources for functional G4 studies in rice. It will provide certain G4 loci for the biotechnological engineering of rice in the future.
