    Abstract: Nervous system diseases present significant challenges to the neuroscience community due to ethical and practical constraints that limit access to appropriate research materials. Somatic cell reprogramming has been proposed as a novel way to obtain neurons. Various emerging techniques have been used to reprogram mature and differentiated cells into neurons. This review provides an overview of somatic cell reprogramming for neurological research and therapy, focusing on neural reprogramming and generating different neural cell types. We examine the mechanisms involved in reprogramming and the challenges that arise. We herein summarize cell reprogramming strategies to generate neurons, including transcription factors, small molecules, and microRNAs, with a focus on different types of cells.. While reprogramming somatic cells into neurons holds the potential for understanding neurological diseases and developing therapeutic applications, its limitations and risks must be carefully considered. Here, we highlight the potential benefits of somatic cell reprogramming for neurological disease research and therapy. This review contributes to the field by providing a comprehensive overview of the various techniques used to generate neurons by cellular reprogramming and discussing their potential applications.

    Abstract: Alternative splicing (AS) is an important mechanism by which eukaryotes regulate transcription and protein diversity. The dynamic changes in AS that occur on a genome-wide scale during interactions between plant roots and pathogens remain unknown. Here, we used the interaction between Arabidopsis and Ralstonia solanacearum as a model to explore the AS changes that take place during the response of roots to infection by means of high-throughput RNA-sequencing. We showed that dynamic changes in AS occur much earlier than changes at the level of transcription during R.solanacearum infection. Comparing genes that are regulated at the transcriptional and AS levels indicated that there are few common genes between differentially spliced genes (DSGs) and differentially expressed genes (DEGs). The functional gene ontology (GO) analysis identified that the enriched GO terms for the DSGs were different from those of the DEGs. The DSGs were over-represented in GO terms associated with post-transcriptional and translational regulations, suggesting that AS may act on RNA stability and during post-translation, thus affecting the output of plant defense molecules. Meanwhile, changes in DSGs were infection stage-specific. Furthermore, the nucleotide binding domain and leucine-rich repeat proteins and receptor-like kinases, key regulators in plant immunity, were shown to undergo dynamic changes in AS in response to R. solanacearum. Taken together, AS, along with transcription, modulates plant root defense to R. solanacearum through transcriptome reprogramming.

    Abstract: Rice false smut caused by Ustilaginoidea virens is a serious disease of rice (Oryza sativa), severely reducing plant mass and yields worldwide. We performed genome-wide analysis of the CCHC-type zinc-finger transcription factor family in this pathogen. We identified and functionally characterized seven UvCCHC genes in U. virens. The deletion of various UvCCHC genes affected the stress responses, vegetative growth, conidiation, and virulence of U. virens. ∆UvCCHC5 mutants infected rice spikelets normally but could not form smut balls. Sugar utilization experiments showed that the ∆UvCCHC5 mutants were defective in the utilization of glucose, sucrose, lactose, stachyose, and trehalose. Deletion of UvCCHC5 did not affect the expression of rice genes associated with grain filling, as revealed by RT-qPCR. We propose that the ∆UvCCHC5 mutants are impaired in transmembrane transport, and the resulting nutrient deficiencies prevent them from using nutrients from rice to form smut balls. RNA-seq data analysis indicated that UvCCHC4 affects the expression of genes involved in mitochondrial biogenesis, ribosomes, transporters, and ribosome biogenesis. These findings improve our understanding of the molecular mechanism underlying smut ball formation in rice by U. virens.

    Abstract: In the computer vision field, underwater object detection has been a challenging task. Due to the attenuation of light in a medium and the scattering of light by suspended particles in water, underwater optical images often face the problems of color distortion and target feature blurring, which greatly affect the detection accuracy of underwater object detection. Although deep learning-based algorithms have achieved state-of-the-art results in the field of object detection, most of them cannot be applied to practice because of the limited computing capacity of a low-power processor embedded in unmanned underwater vehicles. This paper proposes a lightweight underwater object detection network based on the YOLOX model called LUO-YOLOX. A novel weighted ghost-CSPDarknet and simplified PANet were used in LUO-YOLOX to reduce the parameters of the whole model. Moreover, aiming to solve the problems of color distortion and unclear features of targets in underwater images, this paper proposes an efficient self-supervised pre-training joint framework based on underwater auto-encoder transformation (UAET). After the end-to-end pre-training process with the self-supervised pre-training joint framework, the backbone of the object detection network can extract more essential and robust features from degradation images when retrained on underwater datasets. Numerous experiments on the URPC2021 and detecting underwater objects (DUO) datasets verify the performance of our proposed method. Our work can assist unmanned underwater vehicles to perform underwater object detection tasks more accurately.

    Abstract: Balsaminaceae are world-famous ornamental flowers because of their high species diversity, rich variation, peculiar flower patterns, and long ornamental cycles. To study the species diversity, distribution patterns, and distribution hotspots of Balsaminaceae in China, we updated the list of Balsaminaceae by systematically searching the related literature. The distribution pattern and hotspots of Impatiens spp. were analyzed using the ArcGIS 10.8.2 software. Combining 19 meteorological factors and one elevation factor, the Maxent model was applied to analyze the dominant environmental factors that govern the distribution of Impatiens spp. As of February 2023, Balsaminaceae in China included 360 taxa in two genera, including one taxon in the genus of Hydrocera, 359 taxa in the genus Impatiens, 271 national endemic species, and 157 provincial endemic species. Impatiens spp. showed a diffusion pattern from the tropical and subtropical regions to the high-latitude and high-elevation regions concentrated in Southwest China, especially in the Hengduan Mountains in the broad sense, Southern Tibet, the Yunnan–Guizhou–Guangxi karst region, the Qinling–Daba Mountains, and the southeastern hills. The highest species richness was found in the 1200~1500 m elevation range, with 164 species of Impatiens spp. This high species richness was maintained at between 900 and 2700 m, the elevation range where Impatiens spp. are concentrated. When 100% of the species were screened out, 110 hotspots were found, including Southeast Yunnan, Northwest Yunnan, Southern Tibet, and Western Sichuan, where most of the hotspots were concentrated and overlapped with global biodiversity centers, but other hotspots were more scattered. Annual precipitation, the minimal temperature of the coldest month, the altitude and temperature annual range, and four environmental variables with a cumulative contribution of 93.7% were the dominant environmental factors affecting the distribution of Impatiens spp. in China. This study lays the foundation for subsequent studies of Balsaminaceae diversity and is conducive to the development and use of Impatiens spp. resources.

    Abstract: Immunogenicity is a major issue associated with the PK, efficacy, and safety evaluation of therapeutic protein products during pre-clinical and clinical studies. A multi-tiered approach consisting of screening, confirmatory, and titration assays has been widely adopted for anti-drug antibody testing. GQ1001, a recombinant humanized anti-human epidermal growth factor receptor 2 monoclonal antibody covalently linked to a cytotoxin of DM1, possesses a novel format of antibody–drug conjugates. In this study, we reported the development, validation, and application of an acid-dissociation bridging enzyme-linked immunosorbent assay for the detection of antibodies against GQ1001 in cynomolgus monkey serum. The sensitivity of the screening assay was 126.141 ng/mL in undiluted serum. The screening assay and confirmatory assay were neither affected by the naïve monkey serum nor by 2% and 5% (v/v) erythrocyte hemolysates. Moreover, the assay was not subject to interference by 2500 ng/mL of human IgG1 in the samples. Drug interference at low positive control (150 ng/mL) and high positive control (8000 ng/mL) of anti-GQ1001 antibodies was not observed when GQ1001 concentrations were below 3.125 μg/mL and 100 μg/mL, respectively. Furthermore, no hook effect was observed for the positive antibodies in the concentration range of 8 to 64 μg/mL. The validated assay was, thereafter, successfully applied to a single-dose toxicity study of GQ1001. Anti-drug antibody positive rates among dosing animals and testing samples were reported, and no significant impact was found on toxicokinetic outcomes.

    Abstract: Heat stress has a strong and detrimental effect on plant growth and yield. Goji berry or wolfberry (Lycium barbarum L.) is a dual-purpose medicinal and food plant but an increase in high temperatures has caused a serious decline in wolfberry yield and quality. In this study, we first explored the heat stress responses of Goji berry, and found that heat stress adaptation mechanisms fluctuated over 48 h. Moreover, L.barbarum 1402 was more heat resistant while L.barbarum Ningqi No. 7 (N7) was sensitive to high temperatures, in which amino acids and alkaloids played key roles; expression and accumulation timing was also crucial. That is, 1402 responded to heat stress rapidly starting at 1 h under high temperature, activated related genes, and accumulated metabolites earlier in the amino acid metabolic pathway compared to N7, which responded to heat stress starting at 3 h under high temperature. Thus, 1402 resisted high temperatures much earlier and better compared to N7. Furthermore, joint transcriptome and metabolome analysis results showed that L-phenylalanine, L-tyrosine, N-benzylformamide, N-benzylmethylene isomethylamine, lysoPC 19:1, and N-acetyl-D-glucosamine-1-phosthate, as well as their related genes, were higher in content, or earlier in expression, in 1402 compared to N7 under heat treatment. This study initially elucidates that Goji berry 1402 has a better tolerance to heat stress than N7 for earlier and higher expression or accumulation of amino acids and alkaloids when related to high temperatures.
