    Abstract: Human spermatozoa proteomics exposed to some physical, biological or chemical stressors is being explored. However, there is a lack of optimized sample preparation methods to achieve in-depth protein coverage for sperm cells. Meanwhile, it is not clear whether antibiotics can regulate proteins to affect sperm quality. Here, we systematically compared a total of six different protein extraction methods based the combination of three commonly used lysis buffers and physical lysis strategies. The urea buffer combined with ultrasonication (UA-ultrasonication) produced the highest protein extraction rate, leading to the deepest coverage of human sperm proteome (5685 protein groups) from healthy human sperm samples. Since the antibiotics, amoxicillin and clarithromycin, have been widely used against H. pylori infection, we conduct a longitudinal study of sperm proteome via data-independent acquisition tandem mass spectrometry (DIA-MS/MS) on an infected patient during on and off therapy with these two drugs. The semen examination and morphological analysis were performed combined with proteomics analysis. Our results indicated that antibiotics may cause an increase in the sperm concentration and the rate of malformed sperm and disrupt proteome expression in sperm. This work provides an optimized extraction method to characterize the in-depth human sperm proteome and to extend its clinical applications.

    Abstract: African swine fever (ASF) is a swine disease with a very high fatality rate caused by a complex double-stranded DNA virus. The fluorescence PCR detection method is widely used for virus nucleic acid detection. Surface plasmon resonance (SPR) is a label-free and real-time detection method, unlike the fluorescence PCR detection method. In this research, we detected the loop-mediated isothermal amplification (LAMP) products of the African swine fever virus by using the SPR and fluorescence methods separately and simultaneously. By comparing the positive and negative control results, we found that the SPR response unit is completely different before and after the LAMP process. In addition, the fluorescence results on a chip showed that with an increase in the concentration of the sample, the cycle threshold (CT) value decreased, which is consistent with commercial instruments. Both the decline rate of the SPR response unit and the CT value of the fluorescence realized were used to distinguish the positive control from the negative control and water, which indicates that the SPR method can be combined with fluorescence to detect LAMP products. This research provides a label-free and simple method for detecting LAMP products.

    Abstract: Objective: To monitor severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) RNA contamination in samples linked to imported cold-chain food and assess the situation from the implementation of a centralized supervision warehouse system in Guangzhou, Guangdong Province, China. Methods: Swabs of workers and frozen-food-related samples were collected between July 2020 and December 2023 in Guangzhou, Guangdong Province. SARS-CoV-2 RNA was extracted and analyzed by a real-time quantitative polymerase chain reaction using the commercially available SARS-CoV-2 nucleic acid test kit. The risk level and food source were monitored simultaneously. Results: A total of 283 positive cold-chain events were found in Guangzhou since the first cold-chain-related event of the coronavirus disease 2019 pandemic was identified in July 2020. Most positive samples were a low-to-medium risk, and the cycle threshold value was 〉 30. No live virus was detected, and no staff came into direct contact with a live virus. In total, 87.63% of positive events were identified through sampling and testing at the centralized food warehouse. Conclusion: Cold-chain food has a relatively low risk of transmitting SARS-CoV-2. Centralized food storage can be used as an effective method to control this risk, and this measure can also be used for other food-related, contact-transmitted diseases.

    Abstract: H2S is a poisonous gas that is widespread in nature and human activities. Its rapid and sensitive detection is essential to prevent it from damaging health. Herein, we report Pd- and Pt-bimetallic-nanoparticle-doped In2O3 hollow microspheres that are synthesized using solvothermal and in situ reduction methods for H2S detection. The structure of as-synthesized 1 at% Pd/Pt-In2O3 comprises porous hollow microspheres assembled from In2O3 nanosheets with Pd and Pt bimetallic nanoparticles loaded on its surface. The response of 1 at% Pd/Pt-In2O3 to 5 ppm H2S is 140 (70 times that of pure In2O3), and the response time is 3 s at a low temperature of 50 °C. In addition, it can detect trace H2S (as low as 50 ppb) and has superior selectivity and an excellent anti-interference ability. These outstanding gas-sensing performances of 1 at% Pd/Pt-In2O3 are attributed to the chemical sensitization of Pt, the electronic sensitization of Pd, and the synergistic effect between them. This work supplements the research of In2O3-based H2S sensors and proves that Pd- and Pt-bimetallic-doped In2O3 can be applied in the detection of H2S.

    Abstract: Although the two species of chamois (Rupicapra rupicapra and R. pyrenaica) are currently classified as least-concern by the IUCN (International Union for Conservation of Nature), inconsistencies on the subspecies classification reported in literature make it challenging to assess the conservation status of the single subspecies. Previous studies relying on mitochondrial genes, sometimes in combination with nuclear or Y-chromosome markers, reported the presence of clusters corresponding to the geographic distribution but highlighting ambiguities in the genus phylogeny. Here we report novel de novo assembled sequences of the mitochondrial genome from nine individuals, including previously unpublished R. r. balcanica and R. r. tatrica subspecies, and use them to untangle the genus phylogeny. Our results based on the full mitogenome inferred phylogeny confirm the previously reported genus subdivision in three clades and its monophyletic positioning within the Caprinae. Phylogeny and taxonomy of Rupicapra species thus remain controversial prompting for the inclusion of archeological remains to solve the controversy.

    Abstract: In this work, a high sensitivity micro-thermal conductivity detector (μTCD) with four thermal conductivity cells was proposed. Compared with conventional TCD sensors, the thermal conductivity cell in this work was designed as a streamlined structure; the thermistors were supported by a strong cantilever beam and suspended in the center of the thermal conductivity cell, which was able to greatly reduce the dead volume of the thermal conductivity cell and the heat loss of the substrate, improving the detection sensitivity. The experimental results demonstrated that the μTCD shows good stability and high sensitivity, which could rapidly detect light gases with a detection limit of 10 ppm and a quantitative repeatability of less than 1.1%.

    Abstract: Lung homeostasis and regeneration depend on lung epithelial progenitor cells. Lkb1 (Liver Kinase B1) has known roles in the differentiation of airway epithelial cells during embryonic development. However, the effects of Lkb1 in adult lung epithelial progenitor cell regeneration and its mechanisms of action have not been determined. In this study, we investigated the mechanism by which Lkb1 regulates lung epithelial progenitor cell regeneration. Organoid culture showed that loss of Lkb1 significantly reduced the proliferation of club cells and alveolar type 2 (AT2) cells in vitro. In the absence of Lkb1, there is a slower recovery rate of the damaged airway epithelium in naphthalene-induced airway epithelial injury and impaired expression of surfactant protein C during bleomycin-induced alveolar epithelial damage. Moreover, the expression of autophagy-related genes was reduced in club cells and increased in AT2 cells, but the expression of Claudin-18 was obviously reduced in AT2 cells after Lkb1 knockdown. On the whole, our findings indicated that Lkb1 may promote the proliferation of lung epithelial progenitor cells via a niche-dependent pathway and is required for the repair of the damaged lung epithelium.

    Abstract: Stress uniformity within the gauge zone of a cruciform specimen significantly affects materials’ in-plane biaxial mechanical properties in material testing. The stress uniformity depends on the load transmission of the cruciform specimen from the fixtures to the gauge zone. Previous studies failed to alter the nature of the load transmission of the geometric features using parametric optimisations. To improve stress uniformity in the gauge zone, we optimised the cross-arms to design a centre-reduced cruciform specimen with topology and shape optimisations. The simulations show that the optimised specimen obtains significantly less stress variation and range in the gauge zone than the optimised specimen under different observed areas, directions, and load ratios of von Mises, S11, S22, and S12. In the quantified gauge zone, a more uniform stress distribution could be generated by optimizing specimen geometry, whose value should be estimated indirectly each time through simulations. We found that topology and shape optimisations could markedly improve stress uniformity in the gauge zone, and stress concentration at the cross-arms intersection. We first optimised the cruciform specimen structure by combining topology and shape optimisations, which provided a cost-effective way to improve stress uniformity in the gauge zone and reduce stress concentration at the cross-arms intersection, helping obtain reliable data to perform large strains in the in-plane biaxial tensile test.

    Abstract: Given the lack of primary data on heat-resistant composites under high-temperature conditions, the focus of this paper is the development of an in-plane biaxial apparatus under high temperatures and complex loads. Besides loading complex loads up to 80 kN, the apparatus can load under high-temperatures up to 2500 °C. A C/C tensile/compression test at 1700 °C illustrates the successful use of high-temperature digital speckle pattern technology to evaluate the in-plane mechanical properties of heat-resistant composites at 1700 °C under biaxial stress. A high-temperature impact test of a graphite specimen at 2500 °C shows that this apparatus can load at a high temperature in a vacuum and inert gas atmosphere. The yield characteristics of the Q235 steel sheet under in-plane stress show that the apparatus can conduct various mechanical loads, including tension–tension, tension–compression, and compression–compression loads. The proposed equipment can measure the in-plane mechanical properties of composite materials, particularly heat-resistant composites. The obtained results can be applied to structural design, life prediction, and reliability evaluation, as well as for the development, research, and design of aerospace instruments and critical materials.
