    Abstract: Chicken products and chickens with colibacillosis are often reported to be a suspected source of extraintestinal pathogenic Escherichia coli (ExPEC) causing several diseases in humans. Such pathogens in healthy chickens can also contaminate chicken carcasses at the slaughter and then are transmitted to humans via food supply; however, reports about the ExPEC in healthy chickens are still rare. In this study, we determined the prevalence and characteristics of ExPEC isolates in healthy chickens in China. A total of 926 E. coli isolates from seven layer farms (371 isolates), one white-feather broiler farm (78 isolates) and 17 live poultry markets (477 isolates from yellow-feather broilers) in 10 cities in China, were isolated and analyzed for antibiotic resistance phenotypes and genotypes. The molecular detection of ExPEC among these healthy chicken E. coli isolates was performed by PCRs, and the serogroups and antibiotic resistance characteristics of ExPEC were also analyzed. Pulsed-field gel electrophoresis (PFGE) and Multilocus sequence typing (MLST) were used to analyze the genetic relatedness of these ExPEC isolates. We found that the resistance rate for each of the 15 antimicrobials tested among E. coli from white-feather broilers was significantly higher than that from brown-egg layers and that from yellow-feather broilers in live poultry markets (p 〈 0.05). A total of 22 of the 926 E. coli isolates (2.4%) from healthy chickens were qualified as ExPEC, and the detection rate (7.7%, 6/78) of ExPEC among white-feather broilers was significantly higher than that (1.6%, 6/371) from brown-egg layers and that (2.1%, 10/477) from yellow-feather broilers (p 〈 0.05). PFGE and MLST analysis indicated that clonal dissemination of these ExPEC isolates was unlikely. Serogroup O78 was the most predominant type among the six serogroups identified in this study, and all the six serogroups had been frequently reported in human ExPEC isolates in many countries. All the 22 ExPEC isolates were multidrug-resistant (MDR) and the resistance rates to ampicillin (100%) and sulfamethoxazole-trimethoprim (100%) were the highest, followed by tetracycline (95.5%) and doxycycline (90.9%). blaCTX-M was found in 15 of the 22 ExPEC isolates including 10 harboring additional fosfomycin resistance gene fosA3. Notably, plasmid-borne colistin resistance gene mcr-1 was identified in six ExPEC isolates in this study. Worryingly, two ExPEC isolates were found to carry both mcr-1 and blaNDM, compromising both the efficacies of carbapenems and colistin. The presence of ExPEC isolates in healthy chickens, especially those carrying mcr-1 and/or blaNDM, is alarming and will pose a threat to the health of consumers. To our knowledge, this is the first report of mcr-1-positive ExPEC isolates harboring blaNDM from healthy chickens.

    Abstract: In our chemical investigation into Penicillium sp. UJNMF0740 derived from mangrove sediment, fourteen indole diterpene analogs, including four new ones, are purified by multiple chromatographic separation methods, with their structures being elucidated by the analyses of NMR, HR-ESIMS, and ECD data. The antibacterial and neuroprotective effects of these isolates were examined, and only compounds 6 and 9 exhibited weak antibacterial activity, while compounds 5, 8, and 10 showed protective effects against the injury of PC12 cells induced by 6-hydroxydopamine (6-OHDA). Additionally, compound 5 could suppress the apoptosis and production of reactive oxygen species (ROS) in 6-OHDA-stimulated PC12 cells as well as trigger the phosphorylation of PI3K and Akt. Taken together, our work enriches the structural diversity of indole diterpenes and hints that compounds of this skeleton can repress the 6-OHDA-induced apoptosis of PC12 cells via regulating the PI3K/Akt signaling pathway, which provides evidence for the future utilization of this fascinating class of molecules as potential neuroprotective agents.

    Abstract: The present study assessed the willingness of the general population to receive COVID-19 vaccines and identified factors that influence vaccine hesitancy and resistance. A national online survey was conducted from 29 January 2021 to 26 April 2021 in China. Multinomial logistic regression analyses were conducted to identify factors that influence vaccine hesitancy and resistance. Of the 34,041 participants surveyed, 18,810 (55.3%) were willing to get vaccinated, 13,736 (40.3%) were hesitant, and 1495 (4.4%) were resistant. Rates of vaccine acceptance increased over time, with geographical discrepancies in vaccine hesitancy and resistance between provinces in China. Vaccine safety was the greatest concern expressed by most participants (24,461 [71.9%]), and the major reason for participants’ refusing vaccination (974 [65.2%] ). Government agencies (23,131 [68.0%]) and social media (20,967 [61.6%] ) were the main sources of COVID-19 vaccine information. Compared with vaccination acceptance, female, young and middle-aged, high income, and perceived low-risk of infection were associated with vaccine hesitancy. Histories of allergic reactions to other vaccines and depression symptoms were related to vaccine resistance. Common factors that influenced vaccine hesitancy and resistance were residing in cities and perceiving less protection with vaccines than with other protective measures. The results indicate that the rate of vaccine resistance is relatively low, but vaccine hesitancy is common. Individuals who are female, young and middle-aged, with a high income, and residing in cities are more likely to be hesitant for vaccination and should be the target populations for vaccination campaigns. Specific vaccine messaging from the government and social media could alleviate public concerns about vaccine safety and efficacy.

    Abstract: Recently, the most serious upsurge of the desert locust (Schistocerca gregaria) in the last 25 years is spreading across eastern Africa and southwestern Asia. Parts of the desert locust ‘invasion area’, namely the northern border areas of Pakistan and India, are very close to China, and whether locust swarms will invade China is of wide concern. To answer this question, we identified areas of potentially suitable habitat for the desert locust within China based on historical precipitation and temperature data, and found that parts of Xinjiang and Inner Mongolia provinces could provide ephemeral habitat in summer, but these places are remote from any other desert locust breeding areas. New generation adults of the desert locust in Pakistan and India present since April led to swarms spreading into the Indo-Pakistan border region in June, and so we examined historical wind data for this period. Our results showed that winds at the altitude of locust swarm flight blew eastward during April–June, but the wind speeds were quite slow and would not facilitate desert locust eastward migration over large distances. Simulated trajectories of desert locust swarms undertaking 10-day migrations mostly ended within India. The most easterly point of these trajectories just reached eastern India, and this is very close to the eastern border of the invasion area of desert locusts described in previous studies. Overall, the risk that the desert locust will invade China is very low.

    Abstract: Migration is a costly strategy in terms of reproduction output. Competition for limited internal resources leads to physiological management of migration-reproduction trade-offs in energy allocation. Migratory insects must choose to determine to allocate energy into reproduction or migration when confronted insufficient energy supply. Although nutrient shortage is known to stimulate insect migration to escape deteriorating habitat, little is known about when and how migratory insects make decisions when confronted by a nutritional shortage. Here Cnaphalocrocis medinalis (Lepidoptera: Pyralidae), a migratory rice pest in eastern Asia, was used to study the effects of starvation on reproductive traits, behavioral traits and energy allocation. The result showed that one or two days’ starvation before preoviposition did not significantly reduce the fertility (total egg per female laid) and flight capability (flight duration and distance) of both sexes C. medinalis. The preoviposition period was extended significantly only if moths were starved starting on the first day after emergence. Also, take-off percentage of moths starved since their first day increased significantly, and continued to increase even if supplemental nutrients were supplied as honey solution in later days. Moths starved on the first day appeared to allocate or transfer triglycerides into the thorax to maintain the migration process: the quantity of thoracic triglycerides did not differ with age, but abdominal triglycerides decreased with age if starvation continued. These results indicate that the first day post-emergence is a critical period for C. medinalis to decide to migrate or not in response to lack of food. This furthers our understanding of the population dynamics of migratory insects under natural conditions.

    Abstract: Seasonal weather systems that establish prevailing winds and seasonal rainfall on a large scale largely determine insect migration patterns, especially for micro-insects with completely windborne migration. Recent studies indicated that the summer migration of the brown planthopper (BPH, Nilaparvata lugens) in eastern China is related to the strength and position of the Western Pacific Subtropical High-Pressure (WPSH) system and its associated wind and rainfall patterns. Compared with the BPH, the white-backed planthopper (WBPH, Sogatella furcifera) has a similar diet, analogous body size, and strong long-distance migration ability. Thus, the migration pattern for the WBPH can be speculated to be similar to that of the BPH. However, the migration pattern of the WBPH and how this pattern relates to climatic conditions have scarcely been described. Based on almost three decades of data (1977–2003), it was suggested that the WBPH in southern China (south of approximately 27° N) migrates into the middle and lower reaches of the Yangtze River after the abrupt movements of the WPSH in mid-June, similar to the BPH. By contrast, the emigration of the WBPH in southern China begins in late May. Further analysis indicated that the migration of the WBPH in late May and early June was short or unsuccessful due to heavy precipitation during the preflood season in southern China from late May to middle June. The results herein demonstrate the differences in migration patterns between two rice planthoppers in the eastern Asia migration arena. We also provide new information that could assist with forecasting outbreaks and implementing control measures against these migratory pests.

    Abstract: Schistosomiasis is a helminth infection caused by the genus Schistosoma, which is still a threat in tropical and sub-tropical areas. In the China, schistosomiasis caused by Schistosoma japonicum is mainly endemic to the Yangtze River valley. The amphibious snail Oncomelania hupensis (O. hupensis) is the unique intermediate host of S. japonicum; hence, snail control is a crucial approach in the process of schistosomiasis transmission control and elimination. In 2016, a nationwide snail survey was conducted involving all snail habitats recorded since 1950 in all endemic counties of 12 provinces. A total of 53,254 existing snail habitats (ESHs) were identified, presenting three clusters in Sichuan Basin, Dongting Lake, and Poyang Lake. The overall habitat area was 5.24 billion m2, of which 3.58 billion m2 were inhabited by O. hupensis. The area inhabited by snails (AIS) in Dongting and Poyang Lakes accounted for 76.53% of the population in the country. Three typical landscape types (marshland and lakes, mountains and hills, and plain water networks) existed in endemic areas, and marshland and lakes had a predominant share (3.38 billion m2) of the AIS. Among the 12 endemic provinces, Hunan had a share of nearly 50% of AIS, whereas Guangdong had no ESH. Ditches, dryland, paddy fields, marshland, and ponds are common habitat types of the ESH. Although the AIS of the marshland type accounted for 87.22% of the population in the whole country, ditches were the most common type (35,025 or 65.77%) of habitat. Six categories of vegetation for ESHs were identified. A total of 39,139 habitats were covered with weeds, accounting for 55.26% of the coverage of the area. Multiple vegetation types of snail habitats appeared in the 11 provinces, but one or two of these were mainly dominant. Systematic sampling showed that the presence of living snails was 17.88% among the 13.5 million sampling frames. The occurrence varied significantly by landscape, environment, and vegetation type. The median density of living snails in habitats was 0.50 per frame (0.33 m × 0.33 m), and the highest density was 40.01 per frame. Furthermore, two main clusters with high snail densities and spatial correlations indicated by hotspot analysis were identified: one in Hunan and Hubei, the other in Sichuan. This national survey is the first full-scale census on the distribution of O. hupensis, which is significant, as transmission interruption and elimination are truly becoming the immediate goal of schistosomiasis control in China. The study discerns the detailed geographic distribution of O. hupensis with the hotspots of snail density in China. It is beneficial to understand the status of the snail population in order to finally formulate further national control planning.
