    Abstract: We evaluated the efficacy and safety of bevacizumab beyond progression (BBP) in Japanese patients with newly diagnosed glioblastoma and explored predictors of response to bevacizumab. This phase II study evaluated a protocol-defined primary therapy by radiotherapy with concurrent and adjuvant temozolomide plus bevacizumab, followed by bevacizumab monotherapy, and secondary therapy (BBP: bevacizumab upon progression). Ninety patients received the protocol-defined primary therapy (BBP group, n = 25). Median overall survival (mOS) and median progression-free survival (mPFS) were 25.0 and 14.9 months, respectively. In the BBP group, in which O6-methylguanine-DNA methyltransferase (MGMT)-unmethylated tumors predominated, mOS and mPFS were 5.8 and 1.9 months from BBP initiation and 16.8 and 11.4 months from the initial diagnosis, respectively. The primary endpoint, the 2-year survival rate of the BBP group, was 27.0% and was unmet. No unexpected adverse events occurred. Expression profiling using RNA sequencing identified that Cluster 2, which was enriched with the genes involved in macrophage or microglia activation, was associated with longer OS and PFS independent of the MGMT methylation status. Cluster 2 was identified as a significantly favorable independent predictor for PFS, along with younger age and methylated MGMT. The novel expression classifier may predict the prognosis of glioblastoma patients treated with bevacizumab.

    Abstract: HMG-CoA (3-hydroxy-3-methylglutaryl-coenzyme A) reductase inhibitors (statins) have been suggested to attenuate abdominal aortic aneurysm (AAA) growth. However, the effects of statins in human AAA tissues are not fully elucidated. The aim of this study was to investigate the direct effects of statins on proinflammatory molecules in human AAA walls in ex vivo culture. Simvastatin strongly inhibited the activation of nuclear factor (NF)-κB induced by tumor necrosis factor (TNF)-α in human AAA walls, but showed little effect on c-jun N-terminal kinase (JNK) activation. Simvastatin, as well as pitavastatin significantly reduced the secretion of matrix metalloproteinase (MMP)-9, monocyte chemoattractant protein (MCP)-2 and epithelial neutrophil-activating peptide (CXCL5) under both basal and TNF-α-stimulated conditions. Similar to statins, the Rac1 inhibitor NSC23766 significantly inhibited the activation of NF-κB, accompanied by a decreased secretion of MMP-9, MCP-2 and CXCL5. Moreover, the effect of simvastatin and the JNK inhibitor SP600125 was additive in inhibiting the secretion of MMP-9, MCP-2 and CXCL5. These findings indicate that statins preferentially inhibit the Rac1/NF-κB pathway to suppress MMP-9 and chemokine secretion in human AAA, suggesting a mechanism for the potential effect of statins in attenuating AAA progression.

    Abstract: Acute lung injury is characterized by the infiltration of neutrophils into lungs and the subsequent impairment of lung function. Here we explored the role of TDAG8 in lung injury induced by lipopolysaccharide (LPS) administrated intratracheally. In this model, cytokines and chemokines released from resident macrophages are shown to cause neutrophilic inflammation in the lungs. We found that LPS treatment increased TDAG8 expression in the lungs and confirmed its expression in resident macrophages in bronchoalveolar lavage (BAL) fluids. LPS administration remarkably increased neutrophil accumulation without appreciable change in the resident macrophages, which was associated with increased penetration of blood proteins into BAL fluids, interstitial accumulation of inflammatory cells, and damage of the alveolar architecture. The LPS-induced neutrophil accumulation and the associated lung damage were enhanced in TDAG8-deficient mice as compared with those in wild-type mice. LPS also increased several mRNA and protein expressions of inflammatory cytokines and chemokines in the lungs or BAL fluids. Among these inflammatory mediators, mRNA and protein expression of KC (also known as CXCL1), a chemokine of neutrophils, were significantly enhanced by TDAG8 deficiency. We conclude that TDAG8 is a negative regulator for lung neutrophilic inflammation and injury, in part, through the inhibition of chemokine production.

    Abstract: The amount of anodically dissolved charge of silver by linear sweep stripping voltammetry has been observed to be smaller than that of the potentiostatically deposited charge. The imbalance in the charge is opposite to the participation in the double-layer capacitance. This can be explained in terms of the negative capacitive current, which is caused by dipoles of generated redox charge (Ag+) with counterions (NO3−). Lower concentrations of counterions may suppress the capacitance to retain the equality of the charge. This prediction is examined in this work by the oxidation of silver film at various concentrations of NO3− by anodic stripping voltammetry. The capacitance decreased with a decrease in the salt concentrations less than 0.05 mol dm−3. Low concentrations of salts prevent loss of the anodic charge in electroanalysis. This dependence was related with the lifespan of generated silver nitrate dipoles and is described theoretically.

    Abstract: Previous studies have reported conflicting results on the clinical impact of alcohol consumption on the glomerular filtration rate (GFR). This retrospective cohort study aimed to assess the dose-dependent association between alcohol consumption and the slope of the estimated GFR (eGFR) in 304,929 participants aged 40–74 years who underwent annual health checkups in Japan between April 2008 and March 2011. The association between the baseline alcohol consumption and eGFR slope during the median observational period of 1.9 years was assessed using linear mixed-effects models with the random intercept and random slope of time adjusting for clinically relevant factors. In men, rare drinkers and daily drinkers with alcohol consumptions of ≥60 g/day had a significantly larger decline in eGFR than occasional drinkers (difference in multivariable-adjusted eGFR slope with 95% confidence interval (mL/min/1.73 m2/year) of rare, occasional, and daily drinkers with ≤19, 20–39, 40–59, and ≥60 g/day: −0.33 [−0.57, −0.09] , 0.00 [reference], −0.06 [−0.39, 0.26] , −0.16 [−0.43, 0.12], −0.08 [−0.47, 0.30] , and −0.79 [−1.40, −0.17], respectively). In women, only rare drinkers were associated with lower eGFR slopes than occasional drinkers. In conclusion, alcohol consumption was associated with the eGFR slope in an inverse U-shaped fashion in men but not in women.

    Abstract: Face attribute estimation can be used for improving the accuracy of face recognition, customer analysis in marketing, image retrieval, video surveillance, and criminal investigation. The major methods for face attribute estimation are based on Convolutional Neural Networks (CNNs) that solve face attribute estimation as a multiple two-class classification problem. Although one feature extractor should be used for each attribute to explore the accuracy of attribute estimation, in most cases, one feature extractor is shared to estimate all face attributes for the parameter efficiency. This paper proposes a face attribute estimation method using Merged Multi-CNN (MM-CNN) to automatically optimize CNN structures for solving multiple binary classification problems to improve parameter efficiency and accuracy in face attribute estimation. We also propose a parameter reduction method called Convolutionalization for Parameter Reduction (CPR), which removes all fully connected layers from MM-CNNs. Through a set of experiments using the CelebA and LFW-a datasets, we demonstrate that MM-CNN with CPR exhibits higher efficiency of face attribute estimation in terms of estimation accuracy and the number of weight parameters than conventional methods.

    Abstract: In this paper, we propose a method to refine the depth maps obtained by Multi-View Stereo (MVS) through iterative optimization of the Neural Radiance Field (NeRF). MVS accurately estimates the depths on object surfaces, and NeRF accurately estimates the depths at object boundaries. The key ideas of the proposed method are to combine MVS and NeRF to utilize the advantages of both in depth map estimation and to use NeRF for depth map refinement. We also introduce a Huber loss into the NeRF optimization to improve the accuracy of the depth map refinement, where the Huber loss reduces the estimation error in the radiance fields by placing constraints on errors larger than a threshold. Through a set of experiments using the Redwood-3dscan dataset and the DTU dataset, which are public datasets consisting of multi-view images, we demonstrate the effectiveness of the proposed method compared to conventional methods: COLMAP, NeRF, and DS-NeRF.

    Abstract: Chronoamperometric curves for the oxidation of a ferrocenyl derivative via a potential step, calculated using the Cottrell equation, showed less diffusion-controlled currents on a platinum wire electrode. This lower deviation cannot be explained via Butler–Volmer heterogeneous kinetics, but was ascribed to the negatively capacitive current associated with a redox reaction. The deviation in fully oxidized electrical potential corresponds to the non-zero concentration at the electrode surface, which cannot be predicted using the Nernst equation. This equation expresses the relationship between the electrical potential and activity at the electrode surface rather than the concentration. The diffusion equation determines the relationship between the current and surface concentration rather than activity. Negative capacitance or a non-zero concentration may arise from structure formation on the electrode owing to dipole–dipole interactions, which are similar to the generation of double-layer capacitance, including frequency dispersion. Following this concept, we derive expressions for a lowered diffusion-controlled current and time-dependent surface concentration. The negatively capacitive current shows the time dependence of t−0.9, which is similar to the decay of double-layer capacitive currents. The surface concentration decays with t−0.4-dependence.

    Abstract: Variations of the double layer capacitances (DLCs) at a platinum electrode with concentrations and kinds of salts in aqueous solutions were examined in the context of facilitating orientation of solvent dipoles. With an increase in ionic concentrations, the DLCs increased by ca. a half and then kept constant at concentrations over 1 mol dm−3. This increase was classically explained in terms of the Gouy–Chapman (GC) equation combined with the Stern model. Unfortunately, measured DLCs were neither satisfied with the Stern model nor the GC theory. Our model suggests that salts destroy hydrogen bonds at the electrode–solution interface to orient water dipoles toward the external electric field. A degree of the orientation depends on the interaction energy between the salt ion and a water dipole. The statistical mechanic calculation allowed us to derive an equation for the DLC as a function of salt concentration and the interaction energy. The equation took the Langmuir-type in the relation with the concentration. The interaction energy was obtained for eight kinds of salts. The energy showed a linear relation with the interaction energy of ion–solvent for viscosity, called the B-coefficient.

    Abstract: Ongoing aortic wall degeneration and subsequent aneurysm exclusion failure are major concerns after an endovascular aneurysm repair with a stent-graft. An ideal solution would be a drug therapy that targets the aortic wall and inhibits wall degeneration. Here, we described a novel drug delivery system, which allowed repetitively charging a graft with therapeutic drugs and releasing them to the aortic wall in vivo. The system was composed of a targeted graft, which was labeled with a small target molecule, and the target-recognizing nanocarrier, which contained suitable drugs. We developed the targeted graft by decorating a biotinylated polyester graft with neutravidin. We created the target-recognizing nanocarrier by conjugating drug-containing liposomes with biotinylated bio-nanocapsules. We successfully demonstrated that the target-recognizing nanocarriers could bind to the targeted graft, both in vitro and in blood vessels of live mice. Moreover, the drug released from our drug delivery system reduced the expression of matrix metalloproteinase-9 in mouse aortas. Thus, this hybrid system represents a first step toward an adjuvant therapy that might improve the long-term outcome of endovascular aneurysm repair.
