    Abstract: A metabolic illness known as non-alcoholic fatty liver disease (NAFLD), affects more than one-quarter of the world’s population. Bile acids (BAs), as detergents involved in lipid digestion, show an abnormal metabolism in patients with NAFLD. However, BAs can affect other organs as well, such as the brain, where it has a neuroprotective effect. According to a series of studies, brain disorders may be extrahepatic manifestations of NAFLD, such as depression, changes to the cerebrovascular system, and worsening cognitive ability. Consequently, we propose that NAFLD affects the development of brain disease, through the bile acid signaling pathway. Through direct or indirect channels, BAs can send messages to the brain. Some BAs may operate directly on the central Farnesoid X receptor (FXR) and the G protein bile acid-activated receptor 1 (GPBAR1) by overcoming the blood–brain barrier (BBB). Furthermore, glucagon-like peptide-1 (GLP-1) and the fibroblast growth factor (FGF) 19 are released from the intestine FXR and GPBAR1 receptors, upon activation, both of which send signals to the brain. Inflammatory, systemic metabolic disorders in the liver and brain are regulated by the bile acid-activated receptors FXR and GPBAR1, which are potential therapeutic targets. From a bile acid viewpoint, we examine the bile acid signaling changes in NAFLD and brain disease. We also recommend the development of dual GPBAR1/FXR ligands to reduce side effects and manage NAFLD and brain disease efficiently.

    Abstract: Depression is a mental illness that has a serious negative impact on physical and mental health. The pathophysiology of depression is still unknown, and therapeutic medications have drawbacks, such as poor effectiveness, strong dependence, adverse drug withdrawal symptoms, and harmful side effects. Therefore, the primary purpose of contemporary research is to understand the exact pathophysiology of depression. The connection between astrocytes, neurons, and their interactions with depression has recently become the focus of great research interest. This review summarizes the pathological changes of neurons and astrocytes, and their interactions in depression, including the alterations of mid-spiny neurons and pyramidal neurons, the alterations of astrocyte-related biomarkers, and the alterations of gliotransmitters between astrocytes and neurons. In addition to providing the subjects of this research and suggestions for the pathogenesis and treatment techniques of depression, the intention of this article is to more clearly identify links between neuronal–astrocyte signaling processes and depressive symptoms.

    Abstract: Water conservancy personnel usually need to know the water level by water gauge images in real-time and with an expected accuracy. However, accurately recognizing the water level from water gauge images is still a complex problem. This article proposes a composite method applied in the Wuyuan City, Jiangxi Province, in China. This method can detect water gauge areas and number areas from complex and changeable scenes, accurately detect the water level line from various water gauges, and finally, obtain the accurate water level value. Firstly, FCOS is improved by fusing a contextual adjustment module to meet the requirements of edge computing and ensure considerable detection accuracy. Secondly, to deal with scenes with indistinct water level features, we also apply the contextual adjustment module for Deeplabv3+ to segment the water gauge area above the water surface. Then, the area can be used to obtain the position of the water level line. Finally, the results of the previous two steps are combined to calculate the water level value. Detailed experiments prove that this method solves the problem of water level recognition in complex hydrological scenes. Furthermore, the recognition error of the water level by this method is less than 1 cm, proving it is capable of being applied in real river scenes.

    Abstract: In pipe jacking, the continued growth of friction resistance around the pipe may cause problems, such as an insufficient jacking force, soil collapse, and even land subsidence, which seriously endangers the structural safety of the pipe and surrounding structures. Bentonite slurry is often used as a lubrication material, but the friction resistance increases due to the inappropriate slurry concentration, and this may cause construction safety problems. In addition, the slurry standing time increases the difficulty of re-jacking construction. To further study the above problems, the friction characteristics of a steel pipe-soil interface under different slurry concentrations and slurry standing times were studied using direct shear tests. The test results show that the peak shear stress and friction coefficient of the interface first decrease and then increase with the increase in the concentration, which is less than or equal to 20%. The peak shear stress and friction coefficient increase with the increase in the concentration, which is greater than 20%, and the position of shear failure changes from between the slurry soil mixture and pipe wall to between the slurry and pipe wall, and finally to the slurry interior. The influence of the slurry standing time on the friction characteristics of the interface is as follows: For the same slurry concentration, the peak shear stress and friction coefficient of the interface increase with the standing time, approximately increasing first and then stabilizing; for different slurry concentrations, the amplification of the peak shear stress and friction coefficient increase with the increase in the concentration.

    Abstract: (1) Background: Deubiquitinase (DUB) regulates various important cellular processes via reversing the protein ubiquitination. The N-terminal fragment of a giant tegument protein, UL36, encoded by the Marek’s disease (MD) virus (MDV), encompasses a putative DUB (UL36-DUB) and shares no homology with any known DUBs. The N-terminus 75 kDa fragment of UL36 exists in MD T lymphoma cells at a high level and participates in MDV pathogenicity. (2) Methods: To characterize deubiquitinating activity and substrate specificity of UL36-DUB, the UL36 N-terminal fragments, UL36(323), UL36(480), and mutants were prepared using the Bac-to-Bac system. The deubiquitinating activity and substrate specificity of these recombinant UL36-DUBs were analyzed using various ubiquitin (Ub) or ubiquitin-like (UbL) substrates and activity-based deubiquitinating enzyme probes. (3) Results: The results indicated that wild type UL36-DUBs show a different hydrolysis ability against varied types of ubiquitin chains. These wild type UL36-DUBs presented the highest activity to K11, K48, and K63 linkage Ub chains, weak activity to K6, K29, and K33 Ub chains, and no activity to K27 linkage Ub chain. UL36 has higher cleavage efficiency for K48 and K63 poly-ubiquitin than linear ubiquitin chain (M1-Ub4), but no activity on various ubiquitin-like modifiers. The mutation of C98 and H234 residues eliminated the deubiquitinating activity of UL36-DUB. D232A mutation impacted, but did not eliminated UL36(480) activity. The Ub-Br probe can bind to wild type UL36-DUB and mutants UL36(480)H234A and UL36(480)D232A, but not C98 mutants. These in vitro results suggested that the C98 and H234 are essential catalytic residues of UL36-DUB. UL36-DUB exhibited a strict substrate specificity. Inhibition assay revealed that UL36-DUB exhibits resistance to the Roche protease inhibitor cocktail and serine protease inhibitor, but not to the Solarbio protease inhibitor cocktail. (4) Conclusions: UL36-DUB exhibited a strict substrate preference, and the protocol developed in the current study for obtaining active UL36-DUB protein should promote the high-throughput screening of UL36 inhibitors and the study on the function of MDV-encoded UL36.

    Abstract: A Fe61Cr2Nb3Si12B22 amorphous alloy rod sample of 8.8 mm diameter has been successfully prepared through explosive consolidation. The structure and thermal stability of the as-synthesized sample have been analyzed through X-ray diffraction (XRD) and differential scanning calorimeter (DSC) analysis. The results demonstrate that the sample still retains an amorphous structure, and the glass transition temperature (Tg), the crystallization onset temperature (Tx), the supercooled liquid zone (ΔTx) (Tx − Tg) and the reduced glass transition temperatures (Trg) (Tg/Tm) are 784 K, 812 K, 28 K, and 0.556, respectively. Its microstructure has been investigated by optical microscopy (OM) and scanning electron microscopy (SEM). The average microhardness of the alumina compact is about 1069 HV.

    Abstract: The electro-hydraulic servo pump control system (EHSPCS) is a volume control system that uses a permanent magnet synchronous motor (PMSM) with a fixed displacement pump to directly drive and control the hydraulic cylinder. The energy transmission law of the system is very complicated due to the transformation of electrical, mechanical and hydraulic energy as well as other energy fields, and qualitative analysis of the energy transfer efficiency is difficult. Energy transfer analysis of the EHSPCS under different working conditions and loads is proposed in this paper. First, the energy flow transfer mechanism was analyzed, and the mathematical and energy transfer models of the key components of the system were established to explore the energy characteristic state transition rule. Second, a power bond diagram model was built, its state equation and state matrix were deduced, and a system simulation model was built. Finally, combined with the EHSPCS experimental platform, simulation experiments were carried out on the dynamic position following and steady-state position holding conditions of the system, and the variation rules of the power of each energy characteristic state and the system energy transfer efficiency under different loads were obtained. The research results provide a foundation for the study of power matching and energy-saving mechanism of the EHSPCS.

    Abstract: Currently, no consensus exists regarding Sotos syndrome in the Chinese population. Here, we present a case of neonatal Sotos syndrome, followed by a retrospective analysis of five cases of neonatal Sotos syndrome, reported in China. The study subject was a twin premature infant, heavier than gestational age, with characteristic facial features, limb shaking, and hypertonia. Transient hypoglycemia, abnormal cranial magnetic resonance imaging, multiple nodules in polycystic kidneys and liver, abnormal hearing, patent ductus arteriosus, and an atrial septal defect were also noted. The subject showed overgrowth and developmental retardation at 3 months of age. Sequencing revealed a novel missense mutation, c.5000C 〉 A, in the nuclear receptor binding the SET domain protein 1 gene, resulting in an alanine-to-glutamate substitution. The bioinformatics analysis suggested high pathogenicity at this site. This study provides insights into diagnosis of neonatal Sotos syndrome based on specific phenotypes. Subsequent treatment and follow-up should focus on developmental retardation, epilepsy, and scoliosis.
