    Abstract: Dwarfism is a desirable trait in eggplant breeding, as it confers higher lodging resistance and allows simplified management and harvest. However, a few dwarf mutants have been reported, and the molecular mechanism underlying dwarfism in eggplant is completely unknown. Here, we report a dwarf mutant (dwf) isolated from an ethyl methyl sulfonate (EMS)-induced mutant library. The hypocotyl length, plant height, and length of internode cells of dwf were significantly decreased compared to those of the wild-type parent ‘14-345’ (WT). Differential gene expression analysis revealed that GA-related genes, including GA2ox and DELLA, were up-regulated whereas the gibberellin (GA3) content decreased in dwf. Moreover, exogenous GA3 treatment significantly increased the relative growth rate of dwf compared to WT, further indicating the important roles of GA in regulating the dwarf phenotype of dwf. Collectively, our findings shed light on GA-mediated dwarfism in dwf plants and offer a good germplasm that could be used for eggplant dwarfism breeding in the future.

    Abstract: The dynamic drug release mechanisms from Carbomer 940 (CP) hydrogels have not been systematically explored elsewhere. This study aimed to investigate the quantitative structure−activity relationship of licorice flavonoids (LFs) compounds on their drug release from CP hydrogels based on LFs-CP interactions and drug solubility in the release medium. Ten LFs-CP hydrogels were formulated, and their in vitro release study was conducted. The intermolecular forces of LFs-CP systems were characterized by FTIR, molecular docking and molecular dynamic simulation. Ten LFs compounds were classified into I (high-release capability) LFs and II (low-release capability) LFs according to the different negative correlations between drug release percent at 48 h and intermolecular forces of drugs-CP, respectively. Moreover, high-release LFs possessed significantly lower log P and higher drug solubility in the release medium than low-release LFs. All I LFs release behaviors best followed the first-order equation, while II LFs release characteristics best fitted the zero-order equation except for isoliquiritigenin. Log P mainly affect the hydrogel relaxation process for I drugs release and the drug diffusion process for II drugs release. Higher log P values for LFs resulted in higher intermolecular strength for I drugs-CP systems and lower drug solubility in the release medium for II drugs, which hindered drug release. Hydrophobic association forces in drug-CP hydrogel played a more and more dominant role in hindering I LFs release with increasing release time. On the other hand, lower drug solubility in the release medium restricted II LFs release, and the dominant role of drug solubility in the release medium increased in 24 h followed by a significant decline after 36 h. Collectively, log P of LFs served as a bridge to determine LFs compound release behaviors and classification from CP hydrogels, which provided guidelines for reasonable design of LFs hydrogels in pharmaceutical topical formulations.

    Abstract: Mangrove ecosystems can be both significant sources and sinks of greenhouse gases. The restoration of mangrove forests is increasingly used as a natural climate solution tool to mitigate climate change. However, the estimates of carbon exchanges remain unclear, especially from restored mangroves. In this study, we observed the temporal variations in carbon dioxide (CO2) and methane (CH4) fluxes and their biophysical controls for 4 years, based on a closed-path eddy covariance (EC) system. The measurements were conducted in a mangrove wetland park with 14-year-old restored mangroves surrounded by open waters in Guangdong Province, China. The EC measurements showed that the mangrove ecosystem acted as a CO2 source with a net CO2 ecosystem exchange (NEE) of 305 g C m−2 from January 2019 to May 2020 by the 5-m tower measurement. After the tower was adjusted to 10 m, the mangrove showed a CO2 sink with an NEE of −345 g C m−2 from June 2020 to December 2022. The change in tower height influenced the interpretation of interannual trends on NEE. There were no significant interannual trends in the gross primary productivity (GPP) and the ecosystem respiration (Re) values. The change from CO2 source to sink may be attributed to the decrease in land surface proportion by the tower replacement, which reduces the proportion of the mangrove canopy respiration and, therefore, captures lower CO2 fluxes from open waters. The restored mangroves indicated strong CH4 sources of 23.2–26.3 g C m−2 a−1. According to the random forest analysis, the land surface proportion, radiation, and relative humidity were the three most important predictors of NEE, while the CH4 flux was most sensitive to air temperature. Compared to the natural and long-term restored mangroves, this 14-year-old restored mangrove had not yet achieved a maximum carbon sequestration capability. Our study highlights the need for the careful design of long-term observations from restored mangroves and proposes future needs in the context of carbon neutrality.

    Abstract: Deep learning has achieved great successes in performing many visual recognition tasks, including object detection. Nevertheless, existing deep networks are computationally expensive and memory intensive, hindering their deployment in resource-constrained environments, such as mobile or embedded devices that are widely used by city travellers. Recently, estimating city-level travel patterns using street imagery has been shown to be a potentially valid way according to a case study with Google Street View (GSV), addressing a critical challenge in transport object detection. This paper presents a compressed deep network using tensor decomposition to detect transport objects in GSV images, which is sustainable and eco-friendly. In particular, a new dataset named Transport Mode Share-Tokyo (TMS-Tokyo) is created to serve the public for transport object detection. This is based on the selection and filtering of 32,555 acquired images that involve 50,827 visible transport objects (including cars, pedestrians, buses, trucks, motors, vans, cyclists and parked bicycles) from the GSV imagery of Tokyo. Then a compressed convolutional neural network (termed SVDet) is proposed for street view object detection via tensor train decomposition on a given baseline detector. The method proposed herein yields a mean average precision (mAP) of 77.6% on the newly introduced dataset, TMS-Tokyo, necessitating just 17.29 M parameters and a computational capacity of 16.52 G FLOPs. As such, it markedly surpasses the performance of existing state-of-the-art methods documented in the literature.

    Abstract: Signal Transducers and Activators of Transcription (STATs) are a family of transcription factors that regulate cell proliferation, differentiation, apoptosis, immune and inflammatory responses, and angiogenesis. Cumulative evidence has established that STAT3 has a critical role in the development of multiple cancer types. Because it is constitutively activated during disease progression and metastasis in a variety of cancers, STAT3 has promise as a drug target for cancer therapeutics. Recently, STAT3 was found to have an important role in maintaining cancer stem cells in vitro and in mouse tumor models, suggesting STAT3 is integrally involved in tumor initiation, progression and maintenance. STAT3 has been traditionally considered as nontargetable or undruggable, and the lag in developing effective STAT3 inhibitors contributes to the current lack of FDA-approved STAT3 inhibitors. Recent advances in cancer biology and drug discovery efforts have shed light on targeting STAT3 globally and/or specifically for cancer therapy. In this review, we summarize current literature and discuss the potential importance of STAT3 as a novel target for cancer prevention and of STAT3 inhibitors as effective chemopreventive agents.

    Abstract: This paper first presents an adaptive expectation-maximization (AEM) control algorithm based on a measurement-data-driven model to reduce the variance of microelectromechanical system (MEMS) accelerometer sensor under multi disturbances. Significantly different characteristics of the disturbances, consisting of drastic-magnitude, short-duration vibration in the external environment, and slowly-varying, long-duration fluctuation inside the sensor are first constructed together with the measurement model of the accelerometer. Next, through establishing a data-driven model based on a historical small measurement sample, the window length of filter of the presented algorithm is adaptively chosen to estimate the sensor state and identify these disturbances simultaneously. Simulation results of the proposed AEM algorithm based on experimental test are compared with the Kalman filter (KF), least mean square (LMS), and regular EM (REM) methods. Variances of the estimated equivalent input under static condition are 0.212 mV, 0.149 mV, 0.015 mV, and 0.004 mV by the KF, LMS, REM, and AEM, respectively. Under dynamic conditions, the corresponding variances are 35.5 mV, 2.07 mV, 2.0 mV, and 1.45 mV, respectively. The variances under static condition based on the proposed method are reduced to 1.9%, 2.8%, and 27.3%, compared with the KF, LMS, and REM methods, respectively. The corresponding variances under dynamic condition are reduced to 4.1%, 70.1%, and 72.5%, respectively. The effectiveness of the proposed method is verified to reduce the variance of the MEMS resonant accelerometer sensor.

    Abstract: Indigenous pig breeds are important biological resources and their diversity has been severely damaged. The Liangshan pig is a typical mountain-type local pig breed in southwest China. Here, the meat quality, amino acid, and fatty acid composition of Liangshan pigs were compared at seven stages within the weight range of 50–90 kg. A score for comprehensive factors of meat quality was maintained after rising and kept in a plateau within 74.9–91.5 kg of body weight. The total amount of amino acids in the longissimus dorsi muscle remained stable, and the total fatty acids showed an upward trend. Amino acid composition analysis revealed that as the body weight of Liangshan pigs increased, umami, basic, and acidic amino acid contents decreased, while the essential amino acids (EAA) content and the ratio of basic amino acids to acidic amino acids increased. Fatty acid composition analysis revealed that as body weight increased, the content of polyunsaturated fatty acids (PUFA) exhibited a downward trend, while the content of saturated fatty acids (SFA) exhibited an upward trend. This study is a primary step towards the development and utilization of Liangshan pigs and provides useful information for local pork processing and genetic improvement.

    Abstract: The KERNEL NUMBER PER ROW6 (KNR6)-mediated phosphorylation of an adenosine diphosphate ribosylation factor (Arf) GTPase-activating protein (AGAP) forms a key regulatory module for the numbers of spikelets and kernels in the ear inflorescences of maize (Zea mays L.). However, the action mechanism of the KNR6–AGAP module remains poorly understood. Here, we characterized the AGAP-recruited complex and its roles in maize cellular physiology and agronomically important traits. AGAP and its two interacting Arf GTPase1 (ARF1) members preferentially localized to the Golgi apparatus. The loss-of-function AGAP mutant produced by CRISPR/Cas9 resulted in defective Golgi apparatus with thin and compact cisternae, together with delayed internalization and repressed vesicle agglomeration, leading to defective inflorescences and roots, and dwarfed plants with small leaves. The weak agap mutant was phenotypically similar to knr6, showing short ears with fewer kernels. AGAP interacted with KNR6, and a double mutant produced shorter inflorescence meristems and mature ears than the single agap and knr6 mutants. We hypothesized that the coordinated KNR6–AGAP–ARF1 complex modulates vegetative and reproductive traits by participating in vesicle trafficking in maize. Our findings provide a novel mechanistic insight into the regulation of inflorescence development, and ear length and kernel number, in maize.
