    Abstract: Respiratory disease continues to be the major cause of mortality in feedyard cattle, with bronchopneumonia (BP) and acute interstitial pneumonia (AIP) as the two most common syndromes. Recent studies described a combination of these pathological lesions with the presence of AIP in the caudodorsal lungs and BP in the cranioventral lungs of necropsied cattle. This pulmonary pathology has been described as bronchopneumonia with an interstitial pneumonia (BIP). The epidemiological characteristics of BIP in U.S. feedyard cattle are yet to be described. This study’s objectives were to describe the agreement between feedyard clinical and necropsy gross diagnosis and to characterize epidemiological factors associated with four gross pulmonary diagnoses (AIP, BIP, BP, and Normal pulmonary tissue) observed during feedyard cattle necropsies. Systemic necropsies were performed at six feedyards in U.S. high plains region, and gross pulmonary diagnoses were established. Historical data were added to the dataset, including sex, days on feed at death (DOFDEATH), arrival weight, treatment count, and feedyard diagnosis. Generalized linear models were used to evaluate epidemiological factors associated with the probability of each pulmonary pathology. Comparing feedyard clinical diagnosis with gross pathological diagnosis revealed relatively low agreement and the frequency of agreement varied by diagnosis. The likelihood of AIP at necropsy was higher for heifers than steers and in the 100–150 DOFDEATH category compared with the 0–50 DOFDEATH (p = 0.05). The likelihood of BIP increased after the first treatment, whereas the DOFDEATH 0–50 category had a lower likelihood compared with the 150–200 category (p = 0.05). These findings highlight the importance of necropsy for final diagnosis and can aid the development of future diagnosis and therapeutic protocols for pulmonary diseases.

    Abstract: The ErbB RTKs (EGFR, HER2, HER3, and HER4) have been well-studied in cancer. EGFR, HER2, and HER3 stimulate cancer proliferation, principally by activating the phosphatidylinositol-3-kinase and extracellular signal-regulated kinase (ERK) pathways, resulting in increased cancer cell survival and proliferation. Cancer cells have high densities of the EGFR, HER2, and HER3 causing phosphorylation of tyrosine amino acids on protein substrates and tyrosine amino acids near the C-terminal of the RTKs. After transforming growth factor (TGF) α binds to the EGFR, homodimers or EGFR heterodimers form. HER2 forms heterodimers with the EGFR, HER3, and HER4. The EGFR, HER2, and HER3 are overexpressed in lung cancer patient tumors, and monoclonal antibodies (mAbs), such as Herceptin against HER2, are used to treat breast cancer patients. Patients with EGFR mutations are treated with tyrosine kinase inhibitors, such as gefitinib or osimertinib. Peptide GPCRs, such as NTSR1, are present in many cancers, and neurotensin (NTS) stimulates the growth of cancer cells. Lung cancer proliferation is impaired by SR48692, an NTSR1 antagonist. SR48692 is synergistic with gefitinib at inhibiting lung cancer growth. Adding NTS to lung cancer cells increases the shedding of TGFα, which activates the EGFR, or neuregulin-1, which activates HER3. The transactivation process is impaired by SRC, matrix metalloprotease, and reactive oxygen species inhibitors. While the transactivation process is complicated, it is fast and occurs within minutes after adding NTS to cancer cells. This review emphasizes the use of tyrosine kinase inhibitors and SR48692 to impair transactivation and cancer growth.

    Abstract: There is a wide consensus within the polar science, meteorology, and oceanography communities that more in situ observations of the ocean, atmosphere, and sea ice are required to further improve operational forecasting model skills. Traditionally, the volume of such measurements has been limited by the high cost of commercially available instruments. An increasingly attractive solution to this cost issue is to use instruments produced in-house from open-source hardware, firmware, and postprocessing building blocks. In the present work, we release the next iteration of our open-source drifter and wave-monitoring instrument, which follows these solution aspects. The new design is significantly less expensive (typically by a factor of 5 compared with our previous, already cost-effective instrument), much easier to build and assemble for people without specific microelectronics and programming competence, more easily extendable and customizable, and two orders of magnitude more power-efficient (to the point where solar panels are no longer needed even for long-term deployments). Improving performance and reducing noise levels and costs compared with our previous generation of instruments is possible in large part thanks to progress from the electronics component industry. As a result, we believe that this will allow scientists in geosciences to increase by an order of magnitude the amount of in situ data they can collect under a constant instrumentation budget. In the following, we offer (1) a detailed overview of our hardware and software solution, (2) in situ validation and benchmarking of our instrument, (3) a fully open-source release of both hardware and software blueprints. We hope that this work, and the associated open-source release, will be a milestone that will allow our scientific fields to transition towards open-source, community-driven instrumentation. We believe that this could have a considerable impact on many fields by making in situ instrumentation at least an order of magnitude less expensive and more customizable than it has been for the last 50 years, marking the start of a new paradigm in oceanography and polar science, where instrumentation is an inexpensive commodity and in situ data are easier and less expensive to collect.

    Abstract: This study analyzes the effects of N fertilizer application rates on profitability of growing switchgrass and using the feedstock in a pyrolysis biorefinery facility to create a source of sustainable aviation fuel (SAF) supply in Tennessee. Switchgrass (Panicum virgatum L.) is a perennial bunchgrass native to North America with traits suitable for biofuel and co-product production. Previous chemical analysis has shown that ash content in switchgrass is related to the amount of nitrogen applied to the field, while at the biorefinery level, the percentage ash content reduces the biorefinery fuel output. To obtain optimal nitrogen (N) application rates for the switchgrass producers and the biorefinery, a two-part analysis is employed. First, a partial budgeting profitability analysis is conducted for this cropping enterprise at the farm-gate level without considering downstream implications of biomass quality, i.e., ash content. Second, the effects of higher ash content as a percentage of the feedstock on biorefinery output are analyzed. Results show farm-gate profit is maximized when N fertilizer is applied at 111 kg/ha, while as a result of increased production levels and decreased percentage ash content, biorefinery profit is maximized when N is applied at 157 kg/ha. Lower ash could lead to premium prices paid to switchgrass producers if higher quality feedstock were to be demanded as part of an integrated biofuel industry.

    Abstract: The deposition of suspended sediment is an important process that helps wetlands accrete surface material and maintain elevation in the face of sea level rise. Optical remote sensing is often employed to map total suspended solids (TSS), though algorithms typically have limited transferability in space and time due to variability in water constituent compositions, mixtures, and inherent optical properties. This study used in situ spectral reflectances and their first derivatives to compare empirical algorithms for estimating TSS using hyperspectral and multispectral data. These algorithms were applied to imagery collected by NASA’s Airborne Visible/Infrared Imaging Spectrometer-Next Generation (AVIRIS-NG) over coastal Louisiana, USA, and validated with a multiyear in situ dataset. The best performing models were then applied to independent spectroscopic data collected in the Peace–Athabasca Delta, Canada, and the San Francisco Bay–Delta Estuary, USA, to assess their robustness and transferability. A derivative-based partial least squares regression (PLSR) model applied to simulated AVIRIS-NG data showed the most accurate TSS retrievals (R2 = 0.83) in these contrasting deltaic environments. These results highlight the potential for a more broadly applicable generalized algorithm employing imaging spectroscopy for estimating suspended solids.

    Abstract: Aboveground biomass (AGB) plays a critical functional role in coastal wetland ecosystem stability, with high biomass vegetation contributing to organic matter production, sediment accretion potential, and the surface elevation’s ability to keep pace with relative sea level rise. Many remote sensing studies have employed either imaging spectrometer or synthetic aperture radar (SAR) for AGB estimation in various environments for assessing ecosystem health and carbon storage. This study leverages airborne data from NASA’s Airborne Visible/Infrared Imaging Spectrometer-Next Generation (AVIRIS-NG) and Uninhabited Aerial Vehicle Synthetic Aperture Radar (UAVSAR) to assess their unique capabilities in combination to estimate AGB in coastal deltaic wetlands. Here we develop AGB models for emergent herbaceous and forested wetland vegetation in coastal Louisiana. In addition to horizontally emitted, vertically received (HV) backscatter, SAR parameters are expressed by the Freeman–Durden polarimetric decomposition components representing volume and double-bounce scattering. The imaging spectrometer parameters include normalized difference vegetation index (NDVI), reflectance from 290 visible-shortwave infrared (VSWIR) bands, the first derivatives from those bands, or partial least squares (PLS) x-scores derived from those data. Model metrics and cross-validation indicate that the integrated models using the Freeman-Durden components and PLS x-scores improve AGB estimates for both wetland vegetation types. In our study domain over Louisiana’s Wax Lake Delta (WLD), we estimated a mean herbaceous wetland AGB of 3.58 Megagrams/hectare (Mg/ha) and a total of 3551.31 Mg over 9.92 km2, and a mean forested wetland AGB of 294.78 Mg/ha and a total of 27,499.14 Mg over 0.93 km2. While the addition of SAR-derived values to imaging spectrometer data provides a nominal error decrease for herbaceous wetland AGB, this combination significantly improves forested wetland AGB prediction. This integrative approach is particularly effective in forested wetlands as canopy-level biochemical characteristics are captured by the imaging spectrometer in addition to the variable structural information measured by the SAR.

    Abstract: Probiotic and omega-3 supplements have been shown to reduce inflammation, and dual supplementation may have synergistic health effects. We investigated if the novel combination of a multi-strain probiotic (containing B. lactis Bi-07, L. paracasei Lpc-37, L. acidophilus NCFM, and B. lactis Bl-04) alongside omega-3 supplements reduces low-grade inflammation as measured by high-sensitivity C-reactive protein (hs-CRP) in elderly participants in a proof-of-concept, randomized, placebo-controlled, parallel study (NCT04126330). In this case, 76 community-dwelling elderly participants (median: 71.0 years; IQR: 68.0–73.8) underwent an intervention with the dual supplement (n = 37) or placebo (n = 39) for eight weeks. In addition to hs-CRP, cytokine levels and intestinal permeability were also assessed at baseline and after the eight-week intervention. No significant difference was seen for hs-CRP between the dual supplement group and placebo. However, interestingly, supplementation did result in significant increases in the level of the anti-inflammatory marker IL-10. In addition, dual supplementation increased levels of valeric acid, further suggesting the potential of the supplements in reducing inflammation and conferring health benefits. Together, the results suggest that probiotic and omega-3 dual supplementation exerts modest effects on inflammation and may have potential use as a non-pharmacological treatment for low-grade inflammation in the elderly.

    Abstract: Analysis of the efficacy/pharmacology of long-term/lifetime medical treatment of acid hypersecretion in a large cohort of ZES patients in a prospective study. This study includes the results from all 303 patients with established ZES who were prospectively followed and received acid antisecretory treatment with either H2Rs or PPIs, with antisecretory doses individually titrated by the results of regular gastric acid testing. The study includes patients treated for short-term periods ( 〈 5 yrs), patients treated long-term ( 〉 5 yrs), and patients with lifetime treatment (30%) followed for up to 48 years (mean 14 yrs). Long-term/lifelong acid antisecretory treatment with H2Rs/PPIs can be successfully carried out in all patients with both uncomplicated and complicated ZES (i.e., with MEN1/ZES, previous Billroth 2, severe GERD). This is only possible if drug doses are individually set by assessing acid secretory control to establish proven criteria, with regular reassessments and readjustments. Frequent dose changes both upward and downward are needed, as well as regulation of the dosing frequency, and there is a primary reliance on the use of PPIs. Prognostic factors predicting patients with PPI dose changes are identified, which need to be studied prospectively to develop a useful predictive algorithm that could be clinically useful for tailored long-term/lifetime therapy in these patients.

    Abstract: Lettuce is often implicated in human norovirus (HuNoV) foodborne outbreaks. We identified H-like histo-blood group antigens (HBGAs) on lettuce leaves as specific binding moieties for virus-like particles (VLPs) of HuNoV GII.4/HS194/2009 strain. The objective of this study was to determine whether HuNoV-lettuce binding is mediated through the virus HBGA binding sites (HBS). Toward this objective, VLPs of historical HuNoV GII.4 strains (1987, 1997, 2002, 2004 and 2006) with known natural mutations in their HBS, two newly generated VLP mutants of GII.4/HS194/2009 (D374A and G443A) and a VLP mutant (W375A) of GI.1/Norwalk/1968 along with its wild type VLPs, which displays distinct HBS, were investigated for their binding to lettuce. ELISA revealed that historical GII.4 strains binding to lettuce was dependent on their HBGAs profiles. The VLP mutants D374A and G443A lost binding to HBGAs and displayed no to minimal binding to lettuce, respectively. The VLPs of GI.1/Norwalk/1968 strain bound to lettuce through an H-like HBGA and the binding was inhibited by fucosidase digestion. Mutant W375A which was previously shown not to bind to HBGAs, displayed significantly reduced binding to lettuce. We conclude that the binding of HuNoV GII.4 and GI.1 strains to lettuce is mediated through the virus HBS.
