    Kurzfassung: The study of the origin of asymmetries in mirror β decay is extremely important to understand the fundamental nuclear force and the nuclear structure. The experiment was performed at the National Laboratory of Heavy Ion Research Facility in Lanzhou (HIRFL) to measure the β-delayed γ rays of 26P by silicon array and Clover-type high-purity Germanium (HPGe) detectors. Combining with results from the β decay of 26P and its mirror nucleus 26Na, the mirror asymmetry parameter δ ( ≡ft+/ft−− 1) was determined to be 46(13)% for the transition feeding the first excited state in the daughter nucleus. Our independent results support the conclusion that the large mirror asymmetry is close to the proton halo structure in 26P.

    Kurzfassung: In this study, both theoretical analysis and experimental validation are carried out for 3D surface measurement under different indoor/outdoor environmental conditions via combining the projected laser-beam-based sinusoidal optical signal, the optical filtering technique, and the single-shot approach based on Fourier transform profilometry. The designed optical signal generator used in this work is capable of ensuring that the projected fringe pattern is monochromatic, higher-contrast, time-invariant, and truly sinusoidal. The proposed and developed optical setup of 3D surface measurement is portable and is used for in-situ experiments of 3D surface measurements that have been carried out under different sunlight illuminations. The experimental results indicate that accurate reconstructions of measured objects with even or varying surface reflectivity can be obtained under windy conditions and strong environmental illuminations such as the background illuminance of 5600–35,000 Lux. The generated fringe-pattern signal is not sensitive to vibrations from environmental influences including the effects of the wind, which has overcome the outdoor-measurement restrictions of the traditional interferometric system and the profilometry approaches based on phase-shifting methods.

    Kurzfassung: Multi-hop machine reading comprehension is a challenging task in natural language processing as it requires more reasoning ability across multiple documents. Spectral models based on graph convolutional networks have shown good inferring abilities and lead to competitive results. However, the analysis and reasoning of some are inconsistent with those of humans. Inspired by the concept of grandmother cells in cognitive neuroscience, we propose a heterogeneous graph attention network model named ClueReader to imitate the grandmother cell concept. The model is designed to assemble the semantic features in multi-level representations and automatically concentrate or alleviate information for reasoning through the attention mechanism. The name ClueReader is a metaphor for the pattern of the model: it regards the subjects of queries as the starting points of clues, takes the reasoning entities as bridge points, considers the latent candidate entities as grandmother cells, and the clues end up in candidate entities. The proposed model enables the visualization of the reasoning graph, making it possible to analyze the importance of edges connecting entities and the selectivity in the mention and candidate nodes, which is easier to comprehend empirically. Evaluations on the open-domain multi-hop reading dataset WikiHop and drug–drug interaction dataset MedHop proved the validity of ClueReader and showed the feasibility of its application of the model in the molecular biology domain.

    Kurzfassung: Cellulosic ethanol is regarded as a perfect additive for petrol fuels for global carbon neutralization. As bioethanol conversion requires strong biomass pretreatment and overpriced enzymatic hydrolysis, it is increasingly considered in the exploration of biomass processes with fewer chemicals for cost-effective biofuels and value-added bioproducts. In this study, we performed optimal liquid-hot-water pretreatment (190 °C for 10 min) co-supplied with 4% FeCl3 to achieve the near-complete biomass enzymatic saccharification of desirable corn stalk for high bioethanol production, and all the enzyme-undigestible lignocellulose residues were then examined as active biosorbents for high Cd adsorption. Furthermore, by incubating Trichoderma reesei with the desired corn stalk co-supplied with 0.05% FeCl3 for the secretion of lignocellulose-degradation enzymes in vivo, we examined five secreted enzyme activities elevated by 1.3–3.0-fold in vitro, compared to the control without FeCl3 supplementation. After further supplying 1:2 (w/w) FeCl3 into the T. reesei-undigested lignocellulose residue for the thermal-carbonization process, we generated highly porous carbon with specific electroconductivity raised by 3–12-fold for the supercapacitor. Therefore, this work demonstrates that FeCl3 can act as a universal catalyst for the full-chain enhancement of biological, biochemical, and chemical conversions of lignocellulose substrates, providing a green-like strategy for low-cost biofuels and high-value bioproducts.

    Kurzfassung: Pyruvate dehydrogenase (PDH), a key enzyme on the mitochondrial outer membrane, has been found to decrease activity notably in early brain injury (EBI) after subarachnoid hemorrhage (SAH). It has been demonstrated that PDH is associated with the production of reactive oxygen species (ROS) and apoptosis. Hence, in this study, we aimed to determine the cause of the decreased PDH activity and explore the potential role of PDH in EBI. We investigated the expression changes of PDH and pyruvate dehydrogenase kinase (PDK) in vivo and in vitro. Then, we explored the possible effects of PDH and ROS after SAH. The results showed that early overexpression of PDK4 promoted the phosphorylation of PDH, inhibited PDH activity, and may play a protective role after SAH in vivo and in vitro. Finally, we investigated the levels of PDK4 and pyruvate, which accumulated due to decreased PDH activity, in the cerebrospinal fluid (CSF) of 34 patients with SAH. Statistical analysis revealed that PDK4 and pyruvate expression was elevated in the CSF of SAH patients compared with that of controls, and this high expression correlated with the degree of neurological impairment and long-term outcome. Taken together, the results show that PDK4 has the potential to serve as a new therapeutic target and biomarker for assisting in the diagnosis of SAH severity and prediction of recovery.

    Kurzfassung: Detecting litchis in a complex natural environment is important for yield estimation and provides reliable support to litchi-picking robots. This paper proposes an improved litchi detection model named YOLOv5-litchi for litchi detection in complex natural environments. First, we add a convolutional block attention module to each C3 module in the backbone of the network to enhance the ability of the network to extract important feature information. Second, we add a small-object detection layer to enable the model to locate smaller targets and enhance the detection performance of small targets. Third, the Mosaic-9 data augmentation in the network increases the diversity of datasets. Then, we accelerate the regression convergence process of the prediction box by replacing the target detection regression loss function with CIoU. Finally, we add weighted-boxes fusion to bring the prediction boxes closer to the target and reduce the missed detection. An experiment is carried out to verify the effectiveness of the improvement. The results of the study show that the mAP and recall of the YOLOv5-litchi model were improved by 12.9% and 15%, respectively, in comparison with those of the unimproved YOLOv5 network. The inference speed of the YOLOv5-litchi model to detect each picture is 25 ms, which is much better than that of Faster-RCNN and YOLOv4. Compared with the unimproved YOLOv5 network, the mAP of the YOLOv5-litchi model increased by 17.4% in the large visual scenes. The performance of the YOLOv5-litchi model for litchi detection is the best in five models. Therefore, YOLOv5-litchi achieved a good balance between speed, model size, and accuracy, which can meet the needs of litchi detection in agriculture and provides technical support for the yield estimation and litchi-picking robots.

    Kurzfassung: Flubendiamide, a ryanoid class insecticide, is widely used in agriculture. Several insecticides have been reported to promote adipogenesis. However, the potential influence of flubendiamide on adipogenesis is largely unknown. The current study was therefore to determine the effects of flubendiamide on adipogenesis utilizing the 3T3-L1 adipocytes model. Flubendiamide treatment not only enhanced triglyceride content in 3T3-L1 adipocytes, but also increased the expression of cytosine-cytosine-adenosine-adenosine-thymidine (CCAAT)/enhancer-binding protein α and peroxisome proliferator-activated receptor gamma-γ, two important regulators of adipocyte differentiation. Moreover, the expression of the most important regulator of lipogenesis, acetyl coenzyme A carboxylase, was also increased after flubendiamide treatment. Further study revealed that 5-aminoimidazole-4-carboxamide ribonucleotide (AICAR) or A769662, two Adenosine 5′-monophosphate (AMP)-activated protein kinase α activators, subverted effects of flubendiamide on enhanced adipogenesis. Together, these results suggest that flubendiamide promotes adipogenesis via an AMPKα-mediated pathway.

    Kurzfassung: (1) Background: DNA double strand breaks (DSBs) are the most serious form of DNA damage that affects oocyte maturation and the physiological state of follicles and ovaries. Non-coding RNAs (ncRNAs) play a crucial role in DNA damage and repair. This study aims to analyze and establish the network of ncRNAs when DSB occurs and provide new ideas for next research on the mechanism of cumulus DSB. (2) Methods: Bovine cumulus cells (CCs) were treated with bleomycin (BLM) to construct a DSB model. We detected the changes of the cell cycle, cell viability, and apoptosis to determine the effect of DSBs on cell biology, and further evaluated the relationship between the transcriptome and competitive endogenous RNA (ceRNA) network and DSBs. (3) Results: BLM increased γH2AX positivity in CCs, disrupted the G1/S phase, and decreased cell viability. Totals of 848 mRNAs, 75 long noncoding RNAs (lncRNAs), 68 circular RNAs (circRNAs), and 71 microRNAs (miRNAs) in 78 groups of lncRNA–miRNA–mRNA regulatory networks, 275 groups of circRNA–miRNA–mRNA regulatory networks, and five groups of lncRNA/circRNA–miRNA–mRNA co-expression regulatory networks were related to DSBs. Most differentially expressed ncRNAs were annotated to cell cycle, p53, PI3K-AKT, and WNT signaling pathways. (4) Conclusions: The ceRNA network helps to understand the effects of DNA DSBs activation and remission on the biological function of CCs.

    Kurzfassung: Background: Glioma is the most common primary tumor of the central nervous system with a high lethality rate. This study aims to mine fibroblast-related genes with prognostic value and construct a corresponding prognostic model. Methods: A glioma-related TCGA (The Cancer Genome Atlas) cohort and a CGGA (Chinese Glioma Genome Atlas) cohort were incorporated into this study. Variance expression profiling was executed via the “limma” R package. The “clusterProfiler” R package was applied to perform a GO (Gene Ontology) analysis. The Kaplan–Meier (K–M) curve, LASSO regression analysis, and Cox analyses were implemented to determine the prognostic genes. A fibroblast-related risk model was created and affirmed by independent cohorts. We derived enriched pathways between the fibroblast-related high- and low-risk subgroups using gene set variation analysis (GSEA). The immune infiltration cell and the stromal cell were calculated using the microenvironment cell populations-counter (MCP-counter) method, and the immunotherapy response was assessed with the SubMap algorithm. The chemotherapy sensitivity was estimated using the “pRRophetic” R package. Results: A total of 93 differentially expressed fibroblast-related genes (DEFRGs) were uncovered in glioma. Seven prognostic genes were filtered out to create a fibroblast-related gene signature in the TCGA-glioma cohort training set. We then affirmed the fibroblast-related risk model via TCGA-glioma cohort and CGGA-glioma cohort testing sets. The Cox regression analysis proved that the fibroblast-related risk score was an independent prognostic predictor in prediction of the overall survival of glioma patients. The fibroblast-related gene signature revealed by the GSEA was applicable to the immune-relevant pathways. The MCP-counter algorithm results pointed to significant distinctions in the tumor microenvironment between fibroblast-related high- and low-risk subgroups. The SubMap analysis proved that the fibroblast-related risk score could predict the clinical sensitivity of immunotherapy. The chemotherapy sensitivity analysis indicated that low-risk patients were more sensitive to multiple chemotherapeutic drugs. Conclusion: Our study identified prognostic fibroblast-related genes and generated a novel risk signature that could evaluate the prognosis of glioma and offer a theoretical basis for clinical glioma therapy.
