    Kurzfassung: Reactive oxygen species (ROS) act as a signaling intermediate to promote cellular adaptation to maintain homeostasis by regulating autophagy during pathophysiological stress. However, the mechanism by which ROS promotes autophagy is still largely unknown. Here, we show that the ATM/CHK2/ULK1 axis initiates autophagy to maintain cellular homeostasis by sensing ROS signaling under metabolic stress. We report that ULK1 is a physiological substrate of CHK2, and that the binding of CHK2 to ULK1 depends on the ROS signal and the phosphorylation of threonine 68 of CHK2 under metabolic stress. Further, CHK2 phosphorylates ULK1 on serine 556, and this phosphorylation is dependent on the ATM/CHK2 signaling pathway. CHK2-mediated phosphorylation of ULK1 promotes autophagic flux and inhibits apoptosis induced by metabolic stress. Taken together, these results demonstrate that the ATM/CHK2/ULK1 axis initiates an autophagic adaptive response by sensing ROS, and it protects cells from metabolic stress-induced cellular damage.

    Kurzfassung: Chigger mites (Acari) are common ectoparasites on rodents, and they are the exclusive vector of scrub typhus. South China field mouse (Apodemus draco) and Lantsang field mouse (A. ilex) are two sibling rodent species. Based on field investigations in southwest China (2001–2015), this paper compared the infestation of these two mouse species with chiggers. Of 42 chigger species identified from two mouse species, 36 were found on A. draco, 11 on A. ilex and 5 common species on both mice. Jaccard similarity index (J = 0.12, J 〈 0.25) showed a very different species composition of chiggers on two mouse species, and some parameters of the chigger community were also different. The overall mean intensity of chiggers on A. draco (MI = 4.26) was higher than that on A. ilex (MI = 3.91, p 〈 0.05). The dominant chigger species on A. draco were Trombiculindus yunnanus, Leptotrombidium scutellare (a major vector species in China) and L. sinicum with a total constituent ratio Cr = 42.9% (106/247). Leptorombidium sinicum and L. scutellare independently occurred on A. draco with an association coefficient V = 0.09 (V ≈ 0). The dominant chigger species on A. ilex were L. rusticum, L. densipunctatum and L. gongshanense, with a total Cr = 58.14% (25/43). Leptorombidium rusticum and L. densipunctatum on A. ilex had a slight positive association (V = 0.49, 0.5 〈 V 〈 1). All dominant chigger species were unevenly distributed among different individuals of two mouse species. Chigger infestation showed sex bias on different sexes of two mouse species. The species abundance of the chigger community on A. draco was revealed as a log-normal distribution pattern.

    Kurzfassung: One significant source of decoherence in superconducting circuits is known as two-level systems (TLSs), found in amorphous oxide layers. These circuits can, however, also be utilized as spectral and temporal TLS probes. Comprehensive investigations on the physics of TLSs are now possible thanks to recent advancements in superconducting qubits. Here, we simultaneously measure the tunable Xmon qubit decoherence time as well as the resonance frequency for more than 3 days to investigate stochastic fluctuations. Time-domain Allan deviation and frequency-domain power spectral density analysis indicate that two TLSs in near resonance with the qubit are responsible for the fluctuations. From the extracted oscillation in T1 decay, we locate the two TLSs near the junctions.

    Kurzfassung: Background and Objective: Overweight or obesity, as an independent risk factor for chronic diseases, has been on the rise globally. Adopting a healthy lifestyle is positive to weight control. Mobile-based lifestyle interventions have shown potential benefits in weight loss, but most studies were carried out among non-elderly population, so it is necessary to perform well-designed randomized controlled trials among the elderly with overweight or obesity. The purpose of this study is to assess the effect of mobile-based lifestyle intervention on weight loss among the overweight and obese elderly population in China. Methods: This is a prospective, open-labeled, three-month, multicenter, randomized controlled trial involving 750 participants from five cities who were randomly assigned to dietary and physical activity interventions group (DPG; mobile phone with the App and bracelet), physical activity interventions group (PG; mobile phone with the App and bracelet) and control group (CG; no interventions and kept their lifestyle as before). The outcomes evaluated were changes in weight, body mass index (BMI), waist circumference (WC), and hip circumference (HC). Results: In total, 642 (85.6%) participants completed the study, 237 (94.8%), 203 (81.2%), and 202 (80.8%) for DPG, PG, and CG respectively. Comparing with PG and CG, the DPG showed a significant decrease in all outcomes after three months, including body weight (−4.1 kg vs. −1.0 kg; −4.1 kg vs. −0.8 kg; p 〈 0.05), BMI (−1.6 kg/m2 vs. −0.4 kg/m2; −1.6 kg/m2 vs. −0.3 kg/m2; p 〈 0.05), WC (−2.8 cm vs. −0.1 cm; −2.8 cm vs. −0.5 cm; p 〈 0.05), and HC (−3.8 cm vs. −1.3 cm; −3.8 cm vs. −1.3 cm; p 〈 0.05). Similar effects were seen across sex and BMI subgroups. Conclusions: Mobile-based lifestyle intervention obtained beneficial effect in weight loss among the elderly with overweight or obesity. Nevertheless, further studies are needed to confirm the effectiveness and its sustainability.

    Kurzfassung: The Guangshigou deposit is the largest pegmatite-type uranium deposit in the Shangdan domain of the North Qinling Orogenic Belt, which is characterized by the enrichment of uraninite hosted in biotite granitic pegmatites. At Guangshigou, uraninite commonly occurs as mineral inclusions in quartz, K-feldspar and biotite or in interstices of these rock-forming minerals with magmatic characteristics (e.g., U/Th 〈 100, high ThO2, Y2O3 and REE2O3 contents and low concentrations of CaO, FeO and SiO2). It crystallized at 407.6 ± 2.9 Ma from fractionated calc-alkaline high-K pegmatitic melts under conditions of 470–700 °C and 2.4–3.4 kbar as deduced by the compositions of coexisting peritectic biotite. The primary uranium mineralization took place during the Late Caledonian post-collisional extension in the North Qinling Orogen. After this magmatic event, uraninite has experienced multiple episodes of fluid-assisted metasomatism, which generated an alteration halo of mineral assemblages. The alteration halo (or radiohalo) was the result of the combined effects of metamictization and metasomatism characterized by an assemblage of goethite, coffinite and an unidentified aluminosilicate (probably clay minerals) around altered uraninite. This fluid-assisted alteration was concomitant with the albitization of K-feldspar subsequently followed by the coffinitization of uraninite during the major period of 84.9–143.6 Ma, as determined by U-Th-Pb chemical ages. Further investigations revealed that the metasomatic overprinting on uraninite initially and preferentially took place along microcracks or cavities induced by metamictization and promoted their amorphization, followed by the release of U and Pb from structure and the incorporation of K, Ca and Si from the fluids, finally resulting in various degrees of uraninite coffinitization. The released U and Pb were transported by alkali-rich, relatively oxidizing fluids and then re-precipitated locally as coffinite and an amorphous U-Pb-rich silicate under low to moderate temperature conditions (85–174 °C). The compositional changes in primary uraninite, its structure amorphization together with the paragenetic sequence of secondary phases, therefore, corroborate a combined result of intense metamictization of uraninite and an influx of alkali–metasomatic fluids during the Late Mesozoic Yanshanian magmatic event in the region. Hence, the remobilization and circulation of uranium in the North Qinling Orogen was most likely driven by post-Caledonian magmatism and hydrothermal activities related to large-scale tectonic events. In this regards, Paleozoic pegmatite-type uranium mineralization may represent a significant uranium source for Mesozoic hydrothermal mineralization identified in the Qinling Orogenic Belt.

    Kurzfassung: Fusarium graminearum causes Fusarium head blight (FHB), a devastating disease of wheat. Salicylic acid (SA) is involved in the resistance of wheat to F. graminearum. Cell wall mannoprotein (CWM) is known to trigger defense responses in plants, but its role in the pathogenicity of F. graminearum remains unclear. Here, we characterized FgCWM1 (FG05_11315), encoding a CWM in F. graminearum. FgCWM1 was highly expressed in wheat spikes by 24 h after initial inoculation and was upregulated by SA. Disruption of FgCWM1 (ΔFgCWM1) reduced mannose and protein accumulation in the fungal cell wall, especially under SA treatment, and resulted in defective fungal cell walls, leading to increased fungal sensitivity to SA. The positive role of FgCWM1 in mannose and protein accumulation was confirmed by its expression in Saccharomyces cerevisiae. Compared with wild type (WT), ΔFgCWM1 exhibited reduced pathogenicity toward wheat, but it produced the same amount of deoxynivalenol both in culture and in spikes. Complementation of ΔFgCWM1 with FgCWM1 restored the WT phenotype. Localization analyses revealed that FgCWM1 was distributed on the cell wall, consistent with its structural role. Thus, FgCWM1 encodes a CWM protein that plays an important role in the cell wall integrity and pathogenicity of F. graminearum.

    Kurzfassung: Chigger mites are the exclusive vector of scrub typhus. Based on field investigations of 91 survey sites in 5 provincial regions of Southwest China, this paper reported variations of chigger infestation on the oriental house rat (Rattus tanezumi) along various environmental gradients. A total of 149 chigger species were identified from 2919 R. tanezumi in the 5 provincial regions, and Leptotrombidium deliense (a major vector of scrub typhus in China) was the first dominant chigger species, followed by Ascoschoengastia indica and Walchia ewingi. Rattus tanezumi had a stable overall prevalence (PM = 21.10%), mean abundance (MA = 7.01), and mean intensity (MI = 33.20) of chiggers with the same dominant mites in the whole Southwest China in comparison with a previous report in Yunnan Province, but chigger infestations on R. tanezumi varied along different environmental gradients. Rattus tanezumi in mountainous landscape had a higher infestation load of chiggers with higher species diversity than in flatland landscape. The infestation was higher at lower altitudes and latitudes. A high intensity of vegetation coverage was associated with high infestations. The results reflect the environmental heterogeneity of chiggers on the same host species. Warm climate and high relative humidity are beneficial to chigger infestation on R. tanezumi.
