    Abstract: Migraine onset is associated with the abnormal release of vasoactive neurotransmitters from perivascular nerves, and these neurotransmitters are involved in the pathophysiology of migraine. Hyperalgesia is a key feature of migraine, and accumulating evidence indicates that electroacupuncture (EA) at the single acupuncture point (Fengchi [GB20 ] ) is effective in ameliorating hyperalgesia. In clinical practice, multiple acupuncture points are widely used, especially GB20 and Yanglingquan (GB34). However, the role played by vasoactive neurotransmitters in acupuncture antihyperalgesic effect at the single or multiple acupuncture points remains unknown. We aimed to determine whether EA would exert its antihyperalgesic effects by modulating vasoactive neurotransmitter release from the perivascular nerves. Furthermore, we examined whether targeting multiple acupuncture points would be more effective than targeting a single point in reducing hyperalgesia. The mechanical and thermal hyperalgesia were evaluated by measuring the facial and hind-paw mechanical withdrawal thresholds, tail-flick and hot-plate latencies. Plasma concentrations of vasoactive neurotransmitters were determined using rat-specific ELISA kits from jugular vein, including calcitonin gene-related peptide (CGRP), substance P (SP), vasoactive intestinal peptide (VIP), neuropeptide Y (NPY), pituitary adenylate cyclase-activating polypeptide (PACAP), nitric oxide (NO), and endothelin-1 (ET-1). The result suggested that EA significantly ameliorated the mechanical and thermal hyperalgesia, reduced c-Fos levels in the trigeminal ganglion, and attenuated plasma and dural levels of vasoactive neurotransmitters, especially in the multiple acupuncture points group (GB20+GB34). In conclusion, EA exerts antihyperalgesic effect in a rat model of conscious recurrent migraine, possibly via modulation of the vasoactive neurotransmitters. Furthermore, targeting multiple acupuncture points is more effective than targeting a single point in reducing hyperalgesia.

    Abstract: Chronic rhinosinusitis (CRS) is a complex condition brought on for many reasons, and its prevalence is rising gradually around the world. Xanthii Fructus (XF) has been used in the treatment of CRS for decades and is effective. The chemical and pharmacological profiles of XF, on the other hand, are still unknown and need to be clarified. The potential mechanisms of XF in CRS treatment were investigated using a network pharmacology approach in this study. OB and DL were in charge of screening the bioactive components in XF and drug-likeness. TCMSP and PubChem databases were used to identify prospective XF proteins, whereas GeneCards and the DisGeNET database were used to identify potential CRS genes. An interactive network of XF and CRS is built using the STRING database based on common goals identified by the online tool Venny. Cytoscape was used to visualize the topological characteristics of nodes, while the biological function pathways were identified by GO Knowledge Base, KEGG. There were 26 bioactive components and 115 potential targets in XF that bind to CRS or are considered therapeutically relevant. Five significant signaling pathways have been found for CRS by the pathway analysis including the HIF-1 signaling pathway, TNF signaling pathway, Toll-like receptor signaling pathway, NOD-like receptor signaling pathway, and PI3K-Akt signaling pathway. We simultaneously confirmed that the PI3K-Akt pathway promotes the development of CRS. Finally, this study took a holistic approach to the pharmacological actions and molecular mechanisms of XF in the treatment of CRS. TNF, INS, CCL2, CXCL8, IL-10, VEGFA, and IL-6 have all been identified as potential targets for anti-inflammatory and immune-boosting effects. This network pharmacology prediction could be useful in manifesting the molecular mechanisms of the Chinese herbal compound XF for CRS.

    Abstract: In order to study the damage characteristics of polymer plates under the impact of the underwater explosion, the underwater contact and near-field explosion tests of polymer plates were conducted using different explosive quantities. In this paper, eight polymer plates with the size of 500 mm × 500 mm × 60 mm were made, and eight groups of explosion tests were carried out by using the rock emulsion explosive and nonconductive detonators. The damage modes and spatial distribution characteristics of the polymer plate generated by the underwater contact and near-field explosion impact with different explosive quantities are compared and analyzed. In addition, the characteristics of the shock wave propagation in the plates are investigated. It can be observed that the main damage mode of polymer plate is overall damage under the contact underwater explosion. For the near-field explosion, the main damage mode changes to overall failure, and the damage of contact explosion to polymer plate is greater than that of underwater near-field explosion. The polymer plate can reduce and delay the shock wave effectively, but the effect decreases with the increase of explosive quantity in the underwater contact explosion.

    Abstract: Background. Lung adenocarcinoma is one of the most commonly diagnosed malignancies worldwide. Macrophage plays crucial roles in the tumor microenvironment, but its autocrine network and communications with tumor cell are still unclear. Methods. We acquired single-cell RNA sequencing (scRNA-seq) ( n = 30 ) and bulk RNA sequencing ( n = 1480 ) samples of lung adenocarcinoma patients from previous literatures and publicly available databases. Various cell subtypes were identified, including macrophages. Differentially expressed ligand-receptor gene pairs were obtained to explore cell-to-cell communications between macrophages and tumor cells. Furthermore, a machine-learning predictive model based on ligand-receptor interactions was built and validated. Results. A total of 159,219 single cells (18,248 tumor cells and 29,520 macrophages) were selected in this study. We identified significantly correlated autocrine ligand-receptor gene pairs in tumor cells and macrophages, respectively. Furthermore, we explored the cell-to-cell communications between macrophages and tumor cells and detected significantly correlated ligand-receptor signaling pairs. We determined that some of the hub gene pairs were associated with patient prognosis and constructed a machine-learning model based on the intercellular interaction network. Conclusion. We revealed significant cell-to-cell communications (both autocrine and paracrine network) within macrophages and tumor cells in lung adenocarcinoma. Hub genes with prognostic significance in the network were also identified.

    Abstract: Product weight is one of the most important properties for an injection-molded part. The determination of process parameters for obtaining an accurate weight is therefore essential. This study proposed a new optimization strategy for the injection-molding process in which the parameter optimization problem is converted to a weight classification problem. Injection-molded parts are produced under varying parameters and labeled as positive or negative compared with the standard weight, and the weight error of each sample is calculated. A support vector classifier (SVC) method is applied to construct a classification hyperplane in which the weight error is supposed to be zero. A particle swarm optimization (PSO) algorithm contributes to the tuning of the hyperparameters of the SVC model in order to minimize the error between the SVC prediction results and the experimental results. The proposed method is verified to be highly accurate, and its average weight error is 0.0212%. This method only requires a small amount of experiment samples and thus can reduce cost and time. This method has the potential to be widely promoted in the optimization of injection-molding process parameters.

    Abstract: Injection molding is one of the most significant material processing methods for mass production of plastic products. It is widely used in various industry sectors, and its products are ubiquitous in our daily life. The settings and optimization of the injection molding process dictate the geometric precision and mechanical properties of the final products. Therefore, sensing, optimization, and control of the injection molding process have a crucial influence on product quality and have become an active research field with abundant literature. This paper defines the concept of intelligent injection molding as the integral application of these three procedures—sensing, optimization, and control. This paper reviews recent studies on methods for the detection of relevant physical variables, optimization of process parameters, and control strategies of machine variables in the molding process. Finally, conclusions are drawn to discuss future research directions and technologies, as well as algorithms worthy of being explored and developed.

    Abstract: Background. The tumor immune microenvironment (TIME) played an important role in immunotherapy prognosis and treatment response. Immune cells constitute a large part of the tumor microenvironment and regulate tumor progression. Our research is dedicated to studying the infiltrating immune cell in lung adenocarcinoma (LUAD) and seeking potential targets. Methods. The scRNA-seq data were collected from our FDZSH and two public datasets. The code for cell-type mapping algorithms was downloaded from the CIBERSORTx portal. The bioinformatics data of LUAD patients could be approached from The Cancer Genome Atlas (TCGA) portal. Weighted gene coexpression network analysis (WGCNA) and least absolute shrinkage and selection operator (LASSO) analyses were performed to construct a risk model. TIMER2 and TIDE helped with the immune infiltration estimation, while PROGENy helped the cancer-related pathways’ enrichment analysis. GSE31210 dataset and IMVigor ICB therapy cohort validated our findings as the external validation datasets. Results. We clustered the scRNA-seq dataset (integrating our FDZSH datasets and other public datasets) into 23 subpopulations. After curated cell annotation, we implemented Cibersort and WGCNA analysis to anchor the brown module and natural killer cell cluster1 due to the most relationship with tumor trait. The overlap of the brown module gene, natural killer cell signature, and DEGs of tumor and adjacent normal samples was screened by LASSO Cox regression. The obtained 5-gene risk model showed an excellent prognostic performance in the validation dataset. Furthermore, there was a correlation between risk score and tumor-infiltrating immune cells and tumor genomics abnormity. Patients with higher risk scores had a significantly lower immunotherapy response rate. Conclusion. Our observations implied that immune cells played a pivotal role in TIME and established a 5-gene signature (including IDH2, ADRB2, SFTPC, CCDC69, and CCND2) on the basement of nature killer markers targeted by WGCNA analysis. The significance of clinical outcome and immunotherapy response prediction was validated robustly.

    Abstract: Hydraulic fracturing with multiple clusters has been a significant way to improve fracture complexity and achieve high utilization of shale formation. This technology has been widely applied in the main shale area of North America. In Changning shale block of China, it, as a promising treatment technology, is being used in horizontal well now. Due to the anisotropy of mechanical property and the stress shadowing effect between multiclusters, fractures would extend nonuniformly and even some clusters are invalid, leading to a poor treatment performance. In this work, based on the geology and engineering characteristics of Changning shale block, different cluster number, cluster spacing, perforation distribution, and flow rate were discussed by the numerical simulation method to clarify multifracture propagation. It is implied that with the reduction of cluster number and the growth of cluster spacing and flow rate, the length and average width of interior fractures are inclined to increase due to the mitigation of stress shadowing effect, contributing to the lower standard deviation (SD) of fracture length, but too small cluster number or too large cluster spacing is not recommended. Besides, the perforation distribution with more perforations in interior fractures can get larger length and average width of interior fractures compared with another two perforation distributions because of more fractional flow rates obtained, which results in more even fracture propagations. In Changning shale block, multicluster hydraulic fracturing with 4-6 clusters in a stage has been employed in 300-400 m well spacing, and diversion technology, limited-entry perforation (36-48 perforations per stage), high flow rate (16 m3/min), and small-sized ceramic proppant (100 mesh) are used to get better shale gas production. To promote the even propagation of fractures further, nonuniform perforation distribution should be introduced in the target shale area.

    Abstract: Taken overlying strata of fully mechanized top coal caving mining (FMTCCM) in 15 m extra thick coal seam as the research object, the comprehensive research methods such as field investigation, theoretical calculation, and numerical analysis are used to systematically analyze. During the mining of extra thick coal seam, the overlying strata form the structure of lower cantilever beam and upper hinged rock beam. The downward transmission caused by the interaction of this combined structure is the fundamental reason for the strong periodic ground pressure behavior of working face and roadway blow. The movement process of overlying strata movement is divided into four stages, and dynamic distribution characteristics of lateral abutment pressure in different stages are obtained. It is considered that the gob side roadway can be in a relatively stable overburden structure and stress environment during the stable stage of abutment pressure. The distribution range of the internal and external stress fields is determined, which provides a theoretical basis for the reasonable roadway layout. At last, the fracture position and abutment pressure evolution process of overlying strata along the goaf side of the extra thick coal seam are further verified by drilling stress measurement.
