    Abstract: Protein kinase A (PKA)/cAMP response element-binding (CREB) protein signaling pathway, contributing to impaired neurogenesis parallel to depressive-like behaviors, has been identified as the crucial factor involved in the antidepressant response of acupuncture. However, the molecular mechanisms associated with antidepressant response of acupuncture, neurogenesis, and depressive-like behaviors ameliorating remain unexplored. The objective was to identify the mechanisms underlying the antidepressant response of acupuncture through PKA signaling pathway in depression rats by employing the PKA signaling pathway inhibitor H89 in in vivo experiments. Our results indicated that the expression of hippocampal PKA- α and p-CREB was significantly downregulated by chronic unpredicted mild stress (CUMS) procedures. Importantly, acupuncture reversed the downregulation of PKA- α and p-CREB. The expression of PKA- α was upregulated by fluoxetine, but not p-CREB. No significant difference was found between Acu and FLX groups on the expression of PKA- α and p-CREB. Interestingly, H89 inhibited the effects of acupuncture or fluoxetine on upregulating the expression of p-CREB, but not PKA- α . There was no significant difference in expression of CREB among the groups. Conclusively, our findings further support the hypothesis that acupuncture could ameliorate depressive-like behaviors by regulating PKA/CREB signaling pathway, which might be mainly mediated by regulating the phosphorylation level of CREB.

    Abstract: Aiming at the problem of optimal path planning for deviation correction of full-width horizontal axis roadheader in the confined roadway of the coal mine, a deviation correction path planning method of full-width horizontal axis roadheader based on improved particle swarm optimization (I-PSO) algorithm is proposed. First, according to the driving characteristics of roadheader and deviation correction strategy, the mathematical model of full-width horizontal axis roadheader is established, and the constraints based on pose analysis are determined. Then, taking the shortest total distance from the initial position to the terminal position of roadheader as the objective function of path planning, an optimization model of deviation correction path planning is established. Finally, the I-PSO algorithm is proposed to quickly and accurately solve the deviation correction path model, in order to plan a safe and shorter distance path of full-width horizontal axis roadheader. In the I-PSO algorithm, the fitness difference of particles is used to dynamically adjust the inertia weight parameters to optimize the exploration of the whole or part of activity space, and the learning factor is adjusted to speed up particle convergence and optimization of particles, and a random factor is introduced to update the position of particles to avoid local traps as much as possible. The performance test of the I-PSO algorithm shows that the I-PSO algorithm is superior to other algorithms. The EJM340/4-2 full-width horizontal axis roadheader is used as the simulation object to verify the effectiveness of the I-PSO algorithm in the deviation correction path planning of roadheader. The simulation results show that the I-PSO algorithm proposed can quickly and accurately plan a safe and shorter distance path than the WPSO algorithm and CPSO algorithm and has better convergence and stability. It has laid a good foundation for the intelligent navigation of full-width horizontal axis roadheader in coal mine.

    Abstract: Rapid and accurate diagnosis of pulmonary tuberculosis (PTB) is an unresolved problem worldwide, especially for sputum smear− (S−) cases. In this study, five antigen genes including Rv3871, Rv3874, Rv3875, Rv3876, and Rv3879 were cloned from Mycobacterium tuberculosis ( Mtb ) RD1 and overexpressed to generate antigen fragments. These antigens and their combinations were investigated for PTB serodiagnosis. 298 serum samples were collected from active PTB patients, including 117 sputum smear+ (S+) and sputum culture+ (C+) cases, 101 S−/C+ cases, and 80 S−/C− cases. The serum IgG levels of the five antigens were measured by ELISA. Based on IgG levels, the sensitivity/specificity of Rv3871, Rv3874, Rv3875, Rv3876, and Rv3879 for PTB detection was 81.21%/74.74%, 63.09%/94.78%, 32.21%/87.37%, 62.42%/85.26%, and 83.56%/83.16%, respectively. Furthermore, the optimal result for PTB diagnosis was achieved by combining antigens Rv3871, Rv3876, and Rv3879. In addition, the IgG levels of Rv3871, Rv3876, and Rv3879 were found to be higher in S−/C+ PTB patients than in other PTB populations. More importantly, combination of the three antigens demonstrated superior diagnostic performance for both S−/C+ and S−/C− PTB. In conclusion, the combination of Rv3871, Rv3876, and Rv3879 induced higher IgG response in sputum S−/C+ PTB patients and represents a promising biomarker combination for diagnosing of PTB.

    Abstract: A damper of the hydraulically interconnected suspension (HIS) system, as a quarter HIS, is prototyped and its damping characteristic is tested to characterize the damping property. The force-velocity characteristic of the prototype is analyzed based on a set of testing results and accordingly a piecewise hysteresis model for the damper is proposed. The proposed equivalent parametric model consists of two parts: hysteresis model in low speed region and saturation model in high speed region which are used to describe the hysteresis phenomenon in low speed and nonhysteresis phenomenon in high speed, respectively. The parameters of the model are identified based on genetic algorithm by setting the constraints of parameters according to their physical significances and the corresponding testing results. The advantages of the model are highlighted by comparing to the nonhysteresis model and the permanent hysteresis model. The numerical simulation results are compared with the testing results to validate the accuracy and effectiveness of the proposed model. Finally, to further verify the proposed model’s wide applicability under different excitation conditions, its results are compared to the testing results in three-dimensional space. The research in this paper is significant for the dynamic analysis of the HIS vehicle.

    Abstract: Nanocrystalline La 2 Sn 2 O 7 and La 2 Sn 1.8 Co 0.2 O 7 with a phase-pure pyrochlore structure were synthesized by a hydrothermal method, and their catalytic activity was investigated for soot combustion. The as-synthesized catalysts presented relatively larger surface area, and pore volume, which was benefit to the gas molecule diffusion in the reaction. A uniform spherical structure with particle size of 200–500 nm was found in SEM. The samples via hydrothermal route are more active for catalytic soot combustion, ascribing to the spherical morphology, high surface area and improved oxygen mobility. After Co, the reducibility was improved and surface oxygen vacancy was produced, resulting in the enhanced activity and selectivity to CO 2 formation.

    Abstract: Planetary gearbox is widely used in various low-speed machines due to its large transmission ratio. However, the gears in a planetary gearbox are prone to the mechanical faults due to the complex dynamic heavy load. Vibration frequencies caused by an early tooth root crack of sun gear are usually difficult to accurately extract, so its fault diagnosis is one of the main challenges of planetary gearbox reliability. In this paper, a simplified tooth root crack model of sun gear is proposed, and then a rigid-flexible coupled dynamics model of the whole planetary gear system is constructed. By the numerical simulation, the fault frequencies caused by a tooth root crack of sun gear are obtained. A Variational Mode Decomposition (VMD) algorithm for the vibration frequency extraction is proposed. The measured vibration signals are decomposed into the sparse Intrinsic Mode Functions (IMFs) by the VMD, and then the IMFs are further analyzed by the spectral method to accurately extract the crack-induced frequency components. The experimental results show that the proposed dynamics model and VMD method are feasible; an error between the characteristic frequencies from the tested signal analysis and the theoretical calculation is less than 1%.

    Abstract: Background. Renal carcinoma is one of the most common malignant tumors in the urinary system. Autophagy can be both activated and inhibited in renal carcinoma, and it plays a double-edged role in the development of renal carcinoma. In the early stage of cancer, autophagy can suppress tumors. In the late stage, autophagy contributes to the survival of tumor cells in an unfavorable environment, and some autophagy-related proteins P62, LC3B, and beclin-1 have become indicators of the prognosis of patients with renal carcinoma. Aim. To demonstrate that ursolic acid activates autophagy in renal carcinoma 786-O cells by inhibiting the hedgehog signaling pathway. Methods. The effect of ursolic acid on the viability of 786-O cells was determined by the MTT method; the effect of ursolic acid on the proliferation and migration of 786-O cells was examined by crystalline violet staining and scratch assay, respectively. For the study of autophagy, we firstly screened the time points. Western blot assay was used to detect the expression level of autophagic protein P62 at different time points of ursolic acid on 786-O. Then, the Cell MeterTM Autophagy Assay Kit was used to detect the effect of different doses of ursolic acid on the autophagic fluorescence intensity of 786-O cells; the Western blot method was used to detect the effect of different doses of ursolic acid on the expression levels of LC3II and P62 proteins in 786-O cells. Further, AdPlus-mCherry-GFP-LC3B adenovirus transfection was used to detect the effect of ursolic acid on the autophagic flow of 786-O cells; ursolic acid was combined with the autophagy inhibitor chloroquine (CQ) to detect the expression level of autophagy protein LC3II by Western blot. In terms of mechanism, the effect of ursolic acid on hedgehog signaling pathway-related proteins in 786-O cells was detected by Western blot. Results. Ursolic acid inhibited the activity, proliferation, and migration of 786-O cells, enhanced the fluorescence intensity of autophagosomes in 786-O cells, increased the expression level of autophagy marker protein LC3II, and inhibited the expression level of P62 in a time and dose-dependent manner; ursolic acid activated the autophagic flow in 786-O cells, which showed that ursolic acid caused the accumulation of autophagic fluorescent spots and enhanced the fluorescence intensity of autophagosomes. Ursolic acid activated the autophagic flow in 786-O cells, as evidenced by the accumulation of autophagic fluorescent spots and enhanced fluorescence intensity of autophagosomes, and the combined use of the autophagy inhibitor CQ increased the expression level of LC3II compared to ursolic acid alone; ursolic acid decreased the expression levels of PTCH1, GLI1, SMO, SHH, and c-Myc and increased the expression level of Sufu in the hedgehog signaling pathway. Conclusion. Ursolic acid activates autophagy in renal carcinoma 786-O cells, probably by inhibiting hedgehog signaling pathway activity.

    Abstract: Background . As the malignant tumor with the highest incidence in teenagers, osteosarcoma has become a major problem in oncology research. In addition to surgical management, the pharmacotherapeutic strategy for osteosarcoma treatment is an attractive way to explore. It has been demonstrated that biochanin A has an antitumor capacity on multiple kinds of solid tumor, including osteosarcoma. But the precise mechanism of biochanin A against osteosarcoma is still needed to be discovered. Objective . To identify the potential therapeutic targets of biochanin A in treating osteosarcoma. Methods . In present study, an integrated approach including network pharmacology and molecular docking technique was conducted, which mainly comprises target prediction, network construction, gene ontology, and pathway enrichment. CCK8 test was employed to evaluate the cell viability of MG63 cells. Western-blot was used to verify the target proteins of biochanin A. Results . Ninety-six and 114 proteins were obtained as the targets of biochanin A and osteosarcoma, respectively. TP53, IGF1, JUN, BGLAP, ATM, MAPK1, ATF3, H2AFX, BAX, CDKN2A, and EGF were identified as the potential targets of biochanin A against osteosarcoma. Based on the western-blot detection, the expression of BGLAP, BAX, and ATF3 in MG63 cell line changed under the treatment of biochanin A. Conclusion . Biochanin A can effectively suppress the proliferation of osteosarcoma and regulate the expression of BGLAP, BAX, and ATF3, which may act as the potential therapeutic targets of osteosarcoma.

    Abstract: To reduce the risk of roof falling and rib spalling during equipment retracement in thin coal seam faces with soft roofs, the 25070 working face of Xuehu Coal Mine was taken as the research object, and theoretical analysis, numerical simulation, and field practice methods were used. Under different space relationships between retracement channels and main roof fracture, the load of hydraulic supports was quantitatively analyzed. The relationship between the working resistance of the hydraulic support and the sinking rotation angle of the immediate roof was analyzed, and a reasonable time for the arrangement of the retracement channel was determined. The sublevel excavation technology and the combined support technology of roof anchor cables and coal rib anchors were proposed. The field application shows that the falling height and rate of movable prop of the hydraulic supports, working resistance of the hydraulic supports, and the roof subsidence all meet the requirements of safety production during the terminal mining period, and the surrounding rock control effect of retracement channel was determined to be good. The safe and efficient mining of the coal mine is ensured, and the research results can provide guidance for similar working faces.

    Abstract: Aiming at the problem of low diagnosis efficiency and accuracy, due to noise and cross aliasing among various faults when diagnosing composite faults of rolling bearing under actual working conditions, a composite fault diagnosis method of rolling bearing based on optimized wavelet packet autoregressive (AR) spectral energy entropy and adaptive no velocity term particle swarm optimization-self organizing map-back propagation neural network (ANVTPSO-SOM-BPNN) is proposed. The energy entropy feature is extracted from the bearing vibration signal through wavelet packet AR spectrum, and SOM and BPNN are combined to form a series network. For PSO, the velocity term is discarded and the inertia weight and learning factor are adaptively adjusted. Finally, the Dempster-Shafer (D-S) evidence fusion diagnosis is carried out. To get closer to the application condition, the data are collected near and far away from the fault point for the composite fault diagnosis, which verifies the effectiveness of the proposed method.
