Abstract: The treatment goal for spinal cord injury (SCI) is to repair neurites and suppress cellular apoptosis. This study is to investigate the effects of nuclear heme oxidase-1 (HO-1) on the acute spinal cord injury and the related mechanisms. The rat model of the SCI was established. On day 7, before model establishment, the adenovirus vector carrying nuclear HO-1 (Ad-GFP-HO-1C Δ 23) was injected into the animals into the tenth thoracic spine (T10) segment by the intrathecal injection. Starting from after the model establishment to day 28, the recovery of motor function was assessed by the Basso-Beattie-Bresnahan (BBB) scoring method. Immunofluorescence was performed to detect the expression patterns of nuclear and cytoplasmic proteins. HE and Nissl staining methods were used to evaluate the structural damage and the number of surviving neurons near the injured area. The TUNEL method was conducted to evaluate the apoptotic degree. Protein expression levels were detected with the Western blot analysis. The BBB assay scores in the nuclear HO-1 group were significantly higher than the blank and adenovirus control groups. Moreover, compared to the blank and adenovirus control groups, the neuronal apoptosis in the nuclear HO-1 group was significantly alleviated. Furthermore, the expression levels of the endoplasmic reticulum stress-related proteins, i.e., CHOP, GRP78, and caspase-12, were significantly decreased in the nuclear HO-1 group. Nuclear HO-1 significantly improves the SCI, promotes the functional recovery, inhibits the endoplasmic reticulum stress, and alleviates the apoptotic process after SCI.

Abstract: Alpine meadows are one major type of pastureland on the Tibetan Plateau. However, few studies have evaluated the response of soil respiration ( R s ) to grazing along an elevation gradient in an alpine meadow on the Tibetan Plateau. Here three fenced enclosures were established in an alpine meadow at three elevations (i.e., 4313 m, 4513 m, and 4693 m) in July 2008. We measured R s inside and outside the three fenced enclosures in July–September, 2010-2011. Topsoil (0–20 cm) samples were gathered in July, August, and September, 2011. There were no significant differences for R s , dissolved organic C (DOC), and belowground root biomass (BGB) between the grazed and ungrazed soils. Soil respiration was positively correlated with soil organic C (SOC), microbial biomass (MBC), DOC, and BGB. In addition, both R s and BGB increased with total N (TN), the ratio of SOC to TN, ammonium N ( NH 4 + -N), and the ratio of NH 4 + -N to nitrate N. Our findings suggested that the negligible response of R s to grazing could be directly attributed to that of respiration substrate and that soil N may indirectly affect R s by its effect on BGB.

Abstract: Objective. Multidrug-resistant tuberculosis (MDR-TB) causes persistent infection and challenges tuberculosis control worldwide. T cell-mediated immunity plays a critical role in controlling Mycobacterium tuberculosis (Mtb) infection, and therefore, enhancing Mtb-specific T cell immune responses represents a promising therapeutic strategy against TB. Cytokine-induced killer (CIK) immunotherapy is based on autologous infusion of in vitro expanded bulk T cells, which include both pathogen-specific and nonspecific T cells from patient peripheral blood mononuclear cells (PBMC) into TB patients. Preclinical mouse studies have shown that the adoptive T cell therapy inhibited Mtb infection. However, the efficacy of CIK immunotherapy in the treatment of MDR-TB infection has not been evaluated in clinical trials. Methods. We performed a retrospective study of MDR-TB patients who received CIK immunotherapy in combination with anti-TB chemotherapy and those who had standard chemotherapy. Results. Our results showed that CIK immunotherapy in combination with anti-TB chemotherapy treatment increased the conversion rate of sputum smear and Mtb culture, alleviated symptoms, improved lesion absorption, and increased recovery. The kinetics of serology and immunology index monitoring data showed good safety profiles for the CIK treatment. Conclusion. Our study has provided strong evidence that CIK immunotherapy in combination with anti-TB chemotherapy is beneficial for MDR-TB patients. A multicenter clinical trial is warranted to evaluate CIK as a new immune therapy for MDR-TB.

Abstract: In order to determine the influence of heat on the stability of the saline soil subgrade slope in cold regions. Firstly, four typical representative saline soils were selected as subgrade fillers, and geotextiles were added at the bottom of the subgrade as a study subject. A two-dimensional numerical model for the temperature field of subgrade soil was established based on COMSOL Multiphysics. Secondly, combined with the main strength parameters of subgrade soil at different temperatures, an isothermal stratification-strength parameter function calculation model was proposed. Then, the SLOPE/W module in GeoStudio is used to stratify the established subgrade model according to isotherms and calculate the stability safety factor of saline soil subgrades in cold regions under different months, slopes, and subgrade heights, respectively. Finally, the influence of submergence at the foot of the slope on the stability of the subgrade slope is discussed. The results show: After the cold season coming, with the decreasing of temperature, the stability of HC-1 and HC-2 saline soil subgrade slope increased to different extents when the ground temperature dropped to -10°C. When the local temperature dropped below -10°C, the stability has slight decline; CS-1 and CS-2 saline soil subgrade in the ground temperature dropped to 10°C (HC-1 and HC-2 denote two groups of saline soil with high chloride-salt content (≥15%) and low sulfate salt content (≤0.5%), whereas CS-1 and CS-2 denote two groups of saline soil with low chloride-salt (≤5%)), the slope stability was significantly improved, and the stability of CS-1 increases slightly with further decrease in temperature.

Abstract: We have built a rat’s model to investigate whether the hypothermia induced by adenosine 5′-monophosphate (5′-AMP) (AIH) could attenuate acute lung injury induced by LPS in rats. We detected the inflammatory cytokine levels in the plasma and bronchoalveolar lavage fluid samples, and we analyzed the pathological changes in the lungs. We have found that AIH can effectively inhibit acute inflammatory reactions and protect the lung from acute injury induced by LPS in rats.

Abstract: Secondary salinization is a common problem in saline soil projects. In order to grasp the mechanism of water and salt migration of high-chlorine saline soil during the cooling process, the saline soils along the Qarhan-Golmud Highway in the Qinghai-Tibet Plateau were selected as test samples. Firstly, the basic physical parameter test and the soluble salt chemical experiment were carried out and obtained liquid and plastic limits, dry density, etc. Secondly, freezing temperature experiments and water-salt migration experiments under one-way cooling conditions were conducted according to the actual environmental conditions, and after the temperature gradient line of the soil sample was stable, water content and labile salt chemistry experiments were conducted to obtain the distribution of water and salt contents of soil samples. Finally, the effect of crystallization-water phase transition on water and salt migration and the effect of chloride salt on the temperature of crystallization-water phase transition were considered, and a mathematical model applicable to the water and salt migration of highly chlorinated saline soils under the effect of unidirectional cooling was established and solved with COMSOL Multiphysics software, and the correctness of the model was verified by comparing the simulation results with the experimental results. The study found that (1) during the one-way cooling process, both water and salt showed a tendency to migrate to the cold end. The MC (saline soil with medium chlorine content) with an initial water content of 16.9% and Cl− content of 3.373% was measured to reach a 17.6% water content and 3.76% Cl− content at the cold (top) end after the experiment. The HC (saline soil with high chlorine content) with an initial water content of 6.6% and Cl− content of 17.928% was measured to reach a 6.83% water content and 18.8% Cl− content after the experiment and (2) after the one-way cooling experiment of the MC, the water content at a distance of 1–2 cm from the cold end has abrupt changes, which may be caused by a small amount of crystallization—water phase transition at this location. At the same time, according to the temperature change graph during the cooling process, the phase change temperature was set to −9°C in the numerical simulation process to match the experimental results.

Abstract: In order to grasp the characteristics and mechanism of saline soil deformation under the coupling action of cooling and dynamic load, three types of subgrade fillings from the Qarhan-Golmud Highway in Qinghai-Tibet Plateau were selected as experimental soil samples for the experimental study. Firstly, the freezing temperature experiment was carried out on SS (sandy silt) and HS (high-sulfate silty clay). The test results showed that the freezing temperature of SS is -0.32°C, while that of HC (high-chloride silty clay) and HS will not freeze at -20°C, due to the presence of salt. Secondly, the three soil samples were subjected to deformation characteristic test under coupling action of cooling and dynamic load, respectively, and the time history curve of temperature gradient change, the time history curve of the change rate of deformation, and deformation rate were summarized. Finally, the model of deformation rate vs. time and the model of change rate of deformation vs. deformation rate under the coupling action of cooling and dynamic load are proposed. The test results found that (1) the cooling rate of the temperature gradient curve of the three soil samples showed a rapid cooling rate in the early stage, and it tends to stabilize in the later stage. The distance of the 0°C line from the top gradually decreases, which is affected by the freezing temperature and the salt content. (2) Affected by the freezing temperature and salt type, SS exhibits frost heave, and HS and HC appear to settlement. The final deformation is 1.0%, -0.73%, and -1.10%, respectively. (3) The model of deformation rate vs. time and the model of change rate of deformation vs. deformation rate under the coupling action of cooling and dynamic load were proposed and verified, which are helpful for the evaluation of engineering stability on saline soil subgrade fillings.
