    Kurzfassung: XueFu ZhuYu Decoction (XFZYD) is an effective prescription that is widely used to improve blood circulation by removing blood stasis. This study aimed to investigate the effects and the underlying molecular mechanisms of XFZYD on lung pyroptosis in cardiopulmonary bypass- (CPB-) induced acute lung injury (ALI) rats. A rat model of ALI was induced by CPB treatment after XFZYD, Ac-YVAD-CMK, and Bay-11-7082 administration. The respiratory index (RI) and oxygenation index (OI) were determined at each time point. The levels of interleukin (IL)-1β, IL-6, IL-18, and TNF-α in serum and lung were measured by enzyme-linked immunosorbent assays (ELISA). Moreover, the protein levels, neutrophil counts, and total cell of bronchoalveolar lavage fluid (BALF) were detected. Additionally, Myeloperoxidase (MPO) expression was detected by immunohistochemical assay. Lung injury was evaluated with the wet/dry (W/D) ratio and pathologic changes, respectively. Besides, the expression of NLRP3 inflammasome and IkB-α/NF-κB pathway proteins was estimated by immunofluorescence, quantitative real-time PCR (qRT-PCR), and Western blotting assays, respectively. XFZYD pretreatment significantly ameliorated pulmonary ventilation function and reduced the CPB-induced lung histopathological injury, inflammatory cell infiltration in BALF and lung, and the apoptosis of lung cells. Interestingly, XFZYD decreased the CPB-induced NLRP3, ASC, Caspase-1 p20, Pro-GSDMD, GSDMD p30, IL-18, IL-1β p-P65, and p-IKBα mRNA or protein levels in lung tissues in ALI model rats. In summary, these findings suggest that XFZYD effectively mitigates NLRP3 inflammasome-dependent pyroptosis in CPB-induced ALI model rats, possibly by inhibiting the IkB-α/NF-κB pathway in the lung.

    Kurzfassung: Objective. To examine the expression of transforming growth factor-β (TGF-β) in the periapical granulation tissue and serum of patients with chronic apical periodontitis and to conduct immunohistochemical analysis so as to explore the relationship between TGF-β and the degree of periapical lesions. Methods. Periapical granulation tissues of 20 cases of chronic apical periodontitis were collected as the experimental group. Healthy gingival tissues without eruption of third molars of 5 cases were collected as the control group. Immunohistochemistry, enzyme-linked immunosorbent assay, and real-time PCR (RT-PCR) were utilized to determine the expression of TGF-β mRNA and protein, and the difference in the expression of TGF-β was compared between groups. In the experimental group, oral CBCT was taken to measure the periapical bone resorption area. Spearman’s correlation method was applied to analyze the correlation between TGF-β protein and gene expression levels and periapical bone resorption area. Results. Immunohistochemistry and enzyme-linked immunosorbent assay demonstrated that the expression of TGF-β protein in chronic apical periodontitis tissue and serum was higher than that in the controls P 〈 0.05 . RT-PCR revealed that the expression of TGF-β mRNA was higher in chronic apical periodontitis tissue than that of the controls P 〈 0.05 . Spearman’s correlation analysis showed that in the experimental group, the mRNA expression of TGF-β was positively correlated with the periapical bone resorption area P 〈 0.01 , and the protein expression level was not correlated with the periapical bone resorption area P 〈 0.05 . Conclusion. The increased expression of TGF-β in the periapical granulation tissue and serum of patients with chronic apical periodontitis has a certain correlation with the progression of periapical periodontitis. The correlation between TGF-β at the mRNA level and the degree of early stage disease as well as the high expression of TGF-β in inflammatory cells in immunohistochemistry have confirmed that TGF-β promotes bone resorption in early periapical periodontitis, and its mechanism of action deserves further investigation.

    Kurzfassung: Background. Pathological neovascularization, which involves a disruption in the balance between angiogenic and antiangiogenic factors under pathological conditions, is the basis of many intraocular diseases. Pigment epithelium-derived factor (PEDF) is a potent natural, endogenous inhibitor of neovascularization because of its antiangiogenic and neuroprotective benefits. However, its application is restricted by its instability and short half-life. The present study is aimed at investigating the cytotoxicity and antiangiogenic effects of PEDF-loaded PEGylated nanoparticles (NP-PEG-PEDF) on high glucose-stimulated human umbilical vein endothelial cells (HUVECs). Methods. In this study, NP-PEG-PEDF were fabricated using the multiple emulsion method for the first time. HUVECs were cultured in a high concentration of glucose (30 mmol/L D-glucose), simulating diabetic conditions. The antiangiogenic effects of vascular endothelial growth factor (VEGF), pure PEDF, and NP-PEG-PEDF on proliferation, migration, and tube formation were evaluated. VEGF secretion in high glucose-stimulated HUVECs was further tested in vitro. Results. NP-PEG-PEDF exhibited low cytotoxicity in HUVECs. Our results indicated that in vitro, NP-PEG-PEDF attenuated diabetes-induced HUVEC proliferation, migration, and tube formation and suppressed VEGF secretion. The apoptosis of diabetes-induced HUVECs occurred in a dose-dependent manner, which showed a statistically significant difference compared with the PEDF treatment group. Conclusion. Our study is the first to demonstrate that NP-PEG-PEDF exert antiangiogenic effects on high glucose-stimulated HUVECs and have the potential to alleviate microvascular dysfunction. These data suggest that the NP-PEG-PEDF delivery system may offer an innovative therapeutic strategy for preventing neovascularization of the fundus.

    Kurzfassung: Irisin plays a crucial role in the glucose metabolism of mammals and fish. However, the mechanism by which irisin regulates hepatic glucose metabolism is not entirely known. To clarify irisin’s function and underlying mechanisms in common carp glucose metabolism, the FNDC5 expression patterns were detected after insulin/glucagon injection, long-term high-glucose, and high-fat diet feeding. In vivo, the activity of PFK and the synthesis of liver glycogen were significantly increased, while the enzyme activity of PEPCK was reduced after intraperitoneal injection with irisin. In contrast, irisin siRNA attenuated the brain and liver FNDC5a and FNDC5b expression, and the serum glucose was increased. Glut2, gs, hk, and pfk were considerably downregulated in the brain and liver, while pygl, pepck, and g6pase were remarkably upregulated after siRNA treatment. In vitro, irisin activates the phosphorylation level of AMPK and PI3K/Akt in primary hepatocytes. Compound C and wortmannin inhibit AMPK and PI3K/Akt, reversing irisin-induced expression of g6pase, fbpase, pepck, gk, gs, pfk, hk, and gsk3b. At the same time, compound C attenuated irisin-induced expression of pi3k and akt. Our research reveals the mechanisms of irisinA and irisinB as glucose metabolism regulators in teleost for the first time.

    Kurzfassung: Various applications of the Internet of Things assisted by deep learning such as autonomous driving and smart furniture have gradually penetrated people’s social life. These applications not only provide people with great convenience but also promote the progress and development of society. However, how to ensure that the important personal privacy information in the big data of the Internet of Things will not be leaked when it is stored and shared on the cloud is a challenging issue. The main challenges include (1) the changes in access rights caused by the flow of manufacturers or company personnel while sharing and (2) the lack of limitation on time and frequency. We propose a data privacy protection scheme based on time and decryption frequency limitation that can be applied in the Internet of Things. Legitimate users can obtain the original data, while users without a homomorphic encryption key can perform operation training on the homomorphic ciphertext. On the one hand, this scheme does not affect the training of the neural network model, on the other hand, it improves the confidentiality of data. Besides that, this scheme introduces a secure two-party agreement to improve security while generating keys. While revoking, each attribute is specified for the validity period in advance. Once the validity period expires, the attribute will be revoked. By using storage lists and setting tokens to limit the number of user accesses, it effectively solves the problem of data leakage that may be caused by multiple accesses in a long time. The theoretical analysis demonstrates that the proposed scheme can not only ensure safety but also improve efficiency.
