    Abstract: Our previous studies showed that α 7 nicotinic acetylcholine receptor (nAchR) agonist nicotine has stimulatory effects on murine bone marrow-derived semimature DCs, but the effect of nicotine on peripheral blood mononuclear cell- (PBMC-) derived human semimature dendritic cells (hu-imDCs) is still to be clarified. In the present study, hu-imDCs (cultured 4 days) were conferred with   ex vivo lower dose nicotine stimulation and the effect of nicotine on surface molecules expression, the ability of cross-presentation, DCs-mediated PBMC priming, and activated signaling pathways were determined. We could demonstrate that the treatment with nicotine resulted in increased surface molecules expression, enhanced hu-imDCs-mediated PBMC proliferation, upregulated release of IL-12 in the supernatant of cocultured DCs-PBMC, and augmented phosphorylation of Akt and ribosomal protein S6. Nicotine associated with traces of LPS efficiently enhanced endosomal translocation of internalized ovalbumin (OVA) and increased TAP-OVA colocalization. Importantly, the upregulation of nicotine-increased surface molecules upregulation was significantly abrogated by the inhibition of Akt kinase. These findings demonstrate that ex vivo nicotine stimulation augments hu-imDCs surface molecules expression via Akt-S6 pathway, combined with increased Ag-presentation result in augmented efficacy of DCs-mediated PBMC proliferation and Th1 polarization.

    Abstract: The construction industry is facing a data tsunami, while emerging information technologies (IT) show great potential for the effective processing of these data or information. However, a comprehensive review for technological change, the resulting process, and organizational changes in the Big Data context, especially from the angle of whole lifecycle of construction project, is lacking. To fill the void, related works published in the databases of Web of Science, Science Direct, and American Society of Civil Engineers library are systematically reviewed. The general trend in emerging IT application in terms of construction project management (CPM) phases, technology and application, and research topics are revealed. Following this analysis, the particularized proposals in relation to each of the main topics within CPM is discussed. Furthermore, according to the advances and limitations of the current literature, corresponding future agendas such as the implementation of comprehensive data-driven CPM scenario are proposed to bridge the gaps between theoretical research and practical demands.

    Abstract: Human platelets express Toll-like receptors (TLR) 4. However, the mechanism by which TLR4 directly affects platelet aggregation and blood coagulation remains to be explored. Therefore, in this study, we evaluated the platelet TLR4 expression in patients who underwent CABG surgery; we explored the correlation between platelet TLR4 expression and the early outcomes in hospital of patients. Additionally, C57BL/6 and C57BL/6- Tlr LPS −/− mice were used to explore the roles of platelet TLR4 in coagulation by platelet aggregometry and rotation thromboelastometry. In conclusion, our results highlight the important roles of TLR4 in blood coagulation and platelet function. Of clinical relevance, we also explored novel roles for platelet TLR4 that are associated with early outcomes in cardiac surgery.

    Abstract: The shear strength of the soil refers to the ultimate strength of the soil against shear failure, which is one of the important indicators used to measure slope stability. This paper presents a simulation of direct shear tests on root-soil composites with different root embedding angles under different stress conditions. By comparing and analyzing the simulation results of ABAQUS software and the laboratory test results, the enhancement effect of plant roots on soil shear strength was explored. Conclusions can be drawn as follows: the excellent agreement between numerical models and laboratory shear tests suggested that the developed model can quickly and conveniently predict the shear strength of the root-soil composites. The shear strength was related to the rooting arrangement. For a single root system, when the inclination angle of the root was about 64° to the shear direction, the shear resistance of soil was much improved, while the root reinforcement had less effect when the inclination angle was greater than 90°. In the case of multiple roots, the hybrid rooting method can more effectively improve the shear resistance of the root-soil composite. Therefore, in the practical application of using the root to strengthen the soil, the angle of a single root and arrangement of multiple roots should be comprehensively considered.

    Abstract: The strong interest in graphene has motivated large effort in the scalable production of high-quality material. The potential of chemical vapor deposition on Cu foil to produce such graphene is impeded by lacking understanding of the relation between catalyst properties and graphene performance. We here present a systematic analysis of the catalyst morphology and its effect on electrical properties of graphene. We find that nanometer sized particles increase the density of bilayer regions but have no significant effect on carrier transport. Long wavelength roughness (waviness), on the other hand, generates defective graphitic regions that deteriorate carrier mobility. These findings shed light on the graphene formation process on Cu substrates and open a route to improve graphene quality for electronics applications.

    Abstract: The present work investigates the validity of electrospun borneol-polyvinylpyrrolidone (PVP) nanocomposites in enhancing drug dissolution rates and improving drug physical stability. Based on hydrogen bonding interactions and via an electrospinning process, borneol and PVP can form stable nanofiber-based composites. FESEM observations demonstrate that composite nanofibers with uniform structure could be generated with a high content of borneol up to 33.3% (w/w). Borneol is well distributed in the PVP matrix molecularly to form the amorphous composites, as verified by DSC and XRD results. The composites can both enhance the dissolution profiles of borneol and increase its physical stability against sublimation for long-time storage by immobilization of borneol molecules with PVP. The incorporation of borneol in the PVP matrix weakens the tensile properties of nanofibers, and the mechanism is discussed. Electrospun nanocomposites can be alternative candidates for developing novel nano-drug delivery systems with high performance.

    Abstract: We would like to highlight the application of natural products to hepatocellular carcinoma (HCC). We will focus on the natural products known as flavonoids, which target this disease at different stages of hepatocarcinogenesis. In spite of the use of chemotherapy and radiotherapy in treating HCC, patients with HCC still face poor prognosis because of the nature of multidrug resistance and toxicity derived from chemotherapy and radiotherapy. Flavonoids can be found in many vegetables, fruits, and herbal medicines that exert their different anticancer effects via different intracellular signaling pathways and serve as antioxidants. In this review, we will discuss seven common flavonoids that exert different biological effects against HCC via different pathways.

    Abstract: Background . Exophthalmos occurs in patients with primary hyperthyroidism. There were few studies about the changes of brain functional networks of patients with exophthalmos of primary hyperthyroidism (EOPH). However, differences in spontaneous brain activity in patients with EOPH remain unclear. Objective . This study explored alterations in the brain functional networks of patients with EOPH using a voxel-wise degree centrality (DC) method. Methods . A total of 20 patients with EOPH (8 men and 12 women) were enrolled. In addition, 20 patients with primary hyperthyroidism without exophthalmos, matched in age, sex, and education status, were enrolled as a control group. The Hospital Anxiety and Depression Scale was used to assess the anxiety and depression status of participants. All participants were examined using resting-state functional MRI. Changes in spontaneous brain activity were investigated using the DC method. To distinguish between the DC values of the patients with EOPH and those of the control group, we analyzed the receiver operating characteristic (ROC) curve. The interrelationships between the DC values and clinical variables in the patients with EOPH were evaluated using Pearson’s correlation coefficient. Results . Patients with EOPH exhibited notably lower DC values in the cerebellum posterior lobe than the control group. In addition, there were negative correlations between the anxiety scores (AS) and the depression scores (DS) and DC values of the cerebellum posterior lobe. The ROC curve analysis of the cerebellum posterior lobe demonstrated that the area under the curve method had a high diagnostic accuracy. Conclusions . Our study was the first, to our knowledge, to explore changes in the brains of patients with EOPH using the DC method. The DC value was significantly different in the cerebellum posterior lobe in patients with EOPH, indicating that the cerebellum posterior lobe is associated with EOPH.

    Abstract: Pancreatic cancer has higher incidence and mortality rates worldwide. PW06 [(E)-3-(9-ethyl-9H-carbazol-3-yl)-1-(2,5-dimethoxyphenyl) prop-2-en-1-one] is a carbazole derivative containing chalcone moiety which was designed for inhibiting tumorigenesis in human pancreatic cancer. This study is aimed at investigating PW06-induced anticancer effects in human pancreatic cancer MIA PaCa-2 cells in vitro. The results showed PW06 potent antiproliferative/cytotoxic activities and induced cell morphological changes in a human pancreatic cancer cell line (MIA PaCa-2), and these effects are concentration-dependent (IC50 is 0.43 μM). Annexin V and DAPI staining assays indicated that PW06 induced apoptotic cell death and DNA condensation. Western blotting indicated that PW06 increased the proapoptotic proteins such as Bak and Bad but decreased the antiapoptotic protein such as Bcl-2 and Bcl-xL. Moreover, PW06 increased the active form of caspase-8, caspase-9, and caspase-3, PARP, releasing cytochrome c, AIF, and Endo G from mitochondria in MIA PaCa-2 cells. Confocal laser microscopy assay also confirmed that PW06 increased Bak and decreased Bcl-xL. Also, the cells were pretreated with inhibitors of caspase-3, caspase-8, and caspase-9 and then were treated with PW06, resulting in increased viable cell number compared to PW06 treated only. Furthermore, PW06 showed a potent binding ability with hydrophobic interactions in the core site of the Fas-Fas death domains (FADD). In conclusion, PW06 can potent binding ability to the Fas-FADD which led to antiproliferative, cytotoxic activities, and apoptosis induction accompanied by the caspase-dependent and mitochondria-dependent pathways in human pancreatic cancer MIA PaCa-2 cells.

    Abstract: Quercetin, a naturally occurring flavonoid, is mainly extracted from tea, onions, and apples. It has the underlying neuroprotective effect on experimental ischemic stroke. A systematic review and meta-analysis were used to assess quercetin’s efficacy and possible mechanisms in treating focal cerebral ischemia. Compared with the control group, twelve studies reported a remarkable function of quercetin in improving the neurological function score (NFS) ( P 〈 0.05 ), and twelve studies reported a significant effect on reducing infarct volume ( P 〈 0.05 ). Moreover, two and three studies showed that quercetin could alleviate blood-brain barrier (BBB) permeability and brain water content, respectively. The mechanisms of quercetin against focal cerebral ischemia are diverse, involving antioxidation, antiapoptotic, anti-inflammation, and calcium overload reduction. On the whole, the present study suggested that quercetin can exert a protective effect on experimental ischemic stroke. Although the effect size may be overestimated because of the quality of studies and possible publication bias, these results indicated that quercetin might be a promising neuroprotective agent for human ischemic stroke.
