    Abstract: Background . Tumor necrosis factor- α -induced protein 8-like 2 (TIPE2 or TNFAIP8L2) is a negative regulator of natural and adaptive immunity. The role of TIPE2 in type 2 diabetes mellitus (T2DM) remains unknown, although TIPE2 plays key roles in preserving inflammatory homeostasis. Methods . TIPE2 expression was measured by Western blotting and real-time polymerase chain reaction (RT-PCR) in peripheral blood mononuclear cells (PBMCs) isolated from T2DM patients and healthy controls, and tumor necrosis factor- α (TNF- α ), high-sensitivity C-reactive protein (hsCRP), interleukin 6 (IL-6), and other related biometabolic parameters were detected using a nephelometer or by ELISA. Differentiated THP-1 cells were exposed to siTIPE2 and TIPE2 adenovirus. Results . TIPE2 was significantly increased in PBMCs from T2DM patients compared with those from healthy controls and was negatively correlated with serum TNF- α , IL-6, and hsCRP concentrations but positively correlated with HbA1c and LDL-C in T2DM patients. High glucose treatment (50 mmol/L) can upregulate the expression of TIPE2 and cytokine secretion in differentiated THP-1 cells. siTIPE2 infection exacerbated the increased TNF- α and IL-6 concentrations in differentiated THP-1 cells under high glucose conditions (50 mmol/L), while infection with TIPE2 adenovirus reversed the increased TNF- α concentration. Conclusions . The present study indicates that TIPE2 may participate in T2DM by regulating TNF- α production.

    Abstract: Electroacupuncture (EA) can effectively modulate pain perception and pain-related negative affect; however, we do not know whether the effect of EA on sensation and affect is parallel, or dissociated, interactional. In this study, we observed the effects of the anterior cingulate cortex (ACC) lesion and the primary somatosensory cortex (S1) activation on pain perception, pain-related affection, and neural oscillation in S1. ACC lesions did not affect pain perception but relieved pain-paired aversion. S1 activation increased pain perception and anxious behavior. EA can mitigate pain perception regardless of whether there is an ACC lesion. Chronic pain may increase the delta and theta band oscillatory activity in the S1 brain region and decrease the oscillatory activity in the alpha, beta, and gamma bands. EA intervention may inhibit the oscillatory activity of the alpha and beta bands. These results suggest that EA may mitigate chronic pain by relieving pain perception and reducing pain-related affection through different mechanisms. This evidence builds upon findings from previous studies of chronic pain and EA treatment.

    Abstract: This study described a TaqMan based real-time fluorescent quantitative PCR (qPCR) method to detect porcine cytomegalovirus (PCMV) infection, targeting the conserved region of the DNA polymerase (DPOL) gene. The standard curve showed a linear regression relationship with a coefficient of 0.999 and a slope of y  = −3.249 x  + 38.958 corresponding to the amplification efficiency at 99.8%. The limit of the qPCR method was 51.9 copies/ μ l. The established qPCR method showed excellent specificity, with no cross-reaction observed with common porcine pathogens. The coefficient of variation for intra-assay and interassay variability ranged up to 1.51% and 2.24%, respectively. PCMV positive signals can be found in semen using this qPCR method, which suggested that we should pay more attention to PCMV contamination in semen in order to eliminate PCMV infection in artificial insemination and xenotransplantation.

    Abstract: Objectives. To study the quantification of the components in rat plasma after oral administration of Shenyanyihao oral solution. Methods. Shenyanyihao oral solution has been traditionally used for the treatments of chronic nephritis in clinics. Stachydrine, Danshensu, chlorogenic acid, protocatechuic acid, plantamajoside, aesculetin, isoquercitrin, ferulic acid, baicalin, and baicalein are regarded as the main compounds in Shenyanyihao oral solution. A sensitive, efficient, and precise UPLC-MS/MS method was established and validated for the quantification of the components in rat plasma after oral administration of Shenyanyihao oral solution. Results. The main pharmacokinetic parameters of the components were acquired based on the analysis of the plasma sample by a noncompartmental method using the WinNonlin7.0 pharmacokinetic program. Danshensu, protocatechuic acid, isoquercitrin, and ferulic acid from Shenyanyihao oral solution were quickly absorbed, and their peak concentration occurred at less than 0.5 h. The pharmacokinetic parameter of the average t 1 / 2 from Danshensu was 3.91 h in rats, and it was the most rapid distribution and elimination among the components. In addition, the C max of stachydrine and baicalin were revealed as the higher plasma concentrations in rats. Conclusions. This pharmacokinetic study seems to be useful for a further clinical study of Shenyanyihao oral solution in the treatments of chronic nephritis.

    Abstract: Objectives . To investigate the mechanism of the protective effect of Zishenwan on pyelonephritis rats. Methods . In the rat model of pyelonephritis, protective effects of Zishenwan, the content of secretory immunoglobulin A (SIg A), and interleukins were detected by ELISA. The expressions of TLR4-NF κ B pathway were detected by Western blot in renal and urinary tract mucosa. The protective effect and influence on TLR4-NF κ B pathway of Zishenwan were studied. Results . Zishenwan protected rats from pyelonephritis which related to the increase of SIgA, the regulation of interleukins, and the inhibition of TLR4-NF κ B pathway. Serum containing Zishenwan can significantly decrease LPS-induced expression of TLR4, MyD88, and NF κ B in vitro . And the inhibition of TLR4 signal by Zishenwan related to the degradation of TRAF3 and TRAF6. Conclusions . Zishenwan protected rats from urinary tract infection by clearance of bacteria and decrease of tissue damage. 20S proteasomes mediated the degradation of TRAF3 which is important to the decrease of tissue damage from Zishenwan.

    Abstract: This study was to explore the risk factors and prognosis of early neurological deterioration (END) after intravenous thrombolysis in patients with cerebral ischemic stroke (CIS) with the guidance of magnetic resonance imaging (MRI) under the compressed sensing-MRI (CSMRI) algorithm. 187 patients with CIS in the hospital were selected and grouped into a deterioration group and a control group according to whether they had END. The CSMRI algorithm was constructed and compared with digital television (DTV) algorithm and Bayesian compressed sensing (BCS) algorithm. It was found that the reconstruction time of CSMRI algorithm in platform I (1134.9 s) and platform II (2615.8 s) was visibly lower than that of DTV algorithm (2634.6 s, 3963.4 s) and BCS algorithm (5631.5 s, 7412.3 s), showing statistically obvious differences ( P 〈 0.05 ). In addition, the reconstruction efficiency of the CSMRI algorithm was the best. After 4 hours of intravenous thrombolysis, the stroke scale score (12.3 scores) of the deterioration group was much higher than that of the control group (8.4 scores) ( P 〈 0.05 ). The occlusion of responsible great vessel in the deterioration group (30 cases, 83.33%) was obviously higher in contrast to that in the control group (74 cases, 49%) ( P 〈 0.05 ). Stroke scale score and occlusion of responsible great vessel were risk factors for EBD after intravenous thrombolysis.
